참깨 리그난 성분의 화학구조와 생리활성 (Chemical Structure and Physiological Activity of Lignan Component in Sesame)

  • Sesame seed are one of the oldest oil-bearing crops known to man. Not only are they nutritious, they are physicologically beneficial. Because of these properties, the promotion of sesame as a crop is set to increase from now on. The lignan component of sesame, which has an antioxidative function, varies considerably. In addition, the seeds remain viable for longer than other crops. At low temperature, it has been reported that the seed remain viable for up to thirty years. This is due to the lignan content of the seeds. In the last twenty years, the mysteries of the natural chemistry of sesame seeds have been unravelled one by one. Up to now, the lignan component has been found to have an $\alpha$-tocopherol synergist effect, an anti-aging effect, a cancer suppressing effect, a hypertension reduction effect, an effect on the promotion of liver function, an effect on the control of the ration of unsaturated fat, and an effect on the synthesis of unsaturated fatty acids. The lignan component of sesame does not simply have an anti-oxidant function, but has diverse recently discovered physiological uses which make the study of lignan highly rewarding.

고속액체크로마토그래피를 이용한 가시오갈피의 리그난 함량 (Lignan contents in Acanthopanax senticosus by HPLC)

  • A reverse-phase system of HPLC using a linear gradient of acetonitrile and deionized water was developed for the quantification lignans, eleutherosides B and E, in Acanthopanax senticosus. The HPLC system consisted of linear gradient of acetonitrile and deionized water, and UV/VIS detection was set at 210 nm. Both eleutherosides B and E contents in different parts of A. senticosus were determined. As a result, the contents of eleutherosides B and E were measured in the leaves (trace amounts and 0.029 mg/g, respectively), stems (0.107 and 1.015 mg/g, respectively), roots (0.026 and 0.390 mg/g, respectively), and fruits (0.022 and 0.043 mg/g, respectively). Moreover, eleutherosides B and E in the water extract were found 0.011 and 0.171 mg/g, respectively.

종자류 식품에 함유된 5종 리그난의 동시 분석법 개발 (Development of Simultaneous Analytical Method for Five Lignans in Edible Seeds)

  • There has been increased interest in lignans due to their potential effect in reducing the risk of developing several diseases. To evaluate lignan contents, sensitive and accurate methods should be developed for their quantification in food. The present study aimed to validate a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the quantification of 5 lignans: lariciresinol (Lar), matairesinol (Mat), pinoresinol (Pin), secoisolariciresinol (Seco), and syringaresinol (Syr). The validation included selectivity, linearity, recovery, accuracy, and precision. The method was proved to be specific, with a linear response (R2≥0.99). The limits of detection were 0.040~0.765 ㎍/100 g and the limits of quantification were 0.114~1.532 ㎍/100 g. Recoveries were 90.588~109.053% for black sesame powder. Relative standard deviations of repeatability and reproducibility were below 5%. Total lignan contents of roasted coffee bean, oat, and blacksoy bean were 105.702 ㎍/100 g, 78.965 ㎍/100 g, and 165.521 ㎍/100 g, respectively. These results showed that LC-MS/MS analysis would be effective in producing acceptable sensitivity, accuracy, and precision in five lignan analyses.

오미자(Schisandra chinensis)의 리그난 함량과 생육특성 간의 상관관계 연구 (Study on the Correlation between the Growth Characteristics and Lignans Contents of Schisandra chinensis)

  • Schisandra chinensis is a fruit which is called Omija in South Korea. The Korean word Omija means "five flavors" (sweet, spicy, sour, bitter, and salty). The aim of his study was to investigate the correlation between growth characteristics and lignans (gomisin A, gomisin N, schisandrin) contents of Schisandra chinensis. The method for determining lignans was validated by measuring the linearity, limit of detection (LOD), limit of quantification (LOQ), precision and accuracy using UPLC-UV. Growth characteristics of S. chinensis such as number of fruits per fruit bunch, length of fruit bunch, width of fruit bunch, fresh weight of fruit bunch, length of fruit, width of fruit, fresh weight of fruit, fresh weight of 30 fruits, and sugar contents of fruit were measured. From the results of correlation analysis, it was found that the contents of lignans showed a significantly negative correlation with fresh weight and sugar contents of fruit. These results will be used to study for quality control of S. chinensis fruit.

볶음온도에 따른 참기름의 항산화성분 변화 (Studies on the Antioxidative Compounds of Sesame Oils with Roasting Temperature)

  • 볶음온도$(110^{\circ}C{\sim}230^{\circ}C)$에 따른 항산화성분의 변화를 조사한 결과, $170^{\circ}C$ 이상에서 백색도가 현저하게 감소하였다. 리그난변화에 있어서는 검출된 7종의 lignan들은 $170^{\circ}C$까지의 온도변화에서 일정하게 유지되다가 sesamolin은 $170^{\circ}C$ 이후 급격히 감소되었고, sesamin은 약간 감소경향을 나타내었다. sesamol, unknown1, unknown3은 $170^{\circ}C$ 이후 급격히 증가하였다. 토코페롤 함량변화에 있어서는 ${\gamma}$체만이 검출되었으며, 미가열참깨의 경우 70.59mg%, 온도상승에 따라 $170^{\circ}C$까지는 점차 증가하다가 $190^{\circ}C$이후 급격히 감소하여 $220^{\circ}C$에서는 33.87mg%이었고, $230^{\circ}C$에서는 26.73mg% 이었다. 산화안정성에 있어서는 미가열참깨의 경우 유도기간이 4.12hr로 가장 낮았고, 볶음온도상승에 따라 증가하다가 $170^{\circ}C$ 이후 현저히 증가하였다. 이상의 결과로부터 산화안정성과 백색도의 상관계수는 -0.993, $170^{\circ}C$ 이상에서 산화안정성과 sesamol의 상관계수는 0.934이었다. 따라서, 볶음온도에 따른 이러한 산화안정성의 현저한 증가는 sesamol, unknown1, unknown3 등 리그난의 증가에서 비롯됨을 알수 있었다.

  • Two lignanes, Comp.-I, mp 108-1$0^{\circ}C$ and Comp.-II 50-52$^{\circ}$were isolated from Korea ginseng extract by repeated column chromatographic purification. Comp.-I was identified as gomisin-N and Comp.-II as gomisin-A by spectrometric analysis, both of which have already been described as the anti-hepatotoxic lignan components of Schizandra chinesis Bail.

참식나무(Neolitsea sericea Koidz) 수피의 염착물질 분석 (Analysis of Dyeing Components from Neolitsea sericea Koidz Bark)

  • 참식나무(Neolitsea sericea Koidz) 추출물의 염색특성을 구명하기 위하여 수피의 열수추출물에 대해 검토했다. 참식나무 수피의 열수추출물로부터 디에틸에테르와 에틸아세테이트로 분획하여 얻은 fraction을 TLC 및 column chromatography 처리하여 2개의 물질을 단리하였다. 기기분석을 통하여 단리물질의 구조를 해석한 결과, 알칼로이드 계열의 4H-dibenzoquinoline-2,10-diol, 5,6,6a,7-tetrahydro-1,9-dimethoxy-6-methyl (화합물I)과 리그난 계열의 lyoniresinol (화합물II)로 동정되었다. 이들 물질이 염색에 관여하는지 확인하기 위하여 HPLC를 사용하여 검토한 결과, 화합물(I)과 화합물(II)의 retention time이 참식나무 추출물 염색액 및 탈염액의 에틸아세테이트 fraction 중 주성분 피크의 retention time과 동일한 결과를 보였다. 따라서 화합물(I)과 화합물(II)가 참식나무 수피의 주요 염착물질임을 확인할 수 있었다.

수확시기에 따른 오미자의 항산화 활성과 리그난 성분 함량 변화 (Changes of Antioxidant Activity and Lignan Contents in Schisandra chinensis by Harvesting Times)

  • We investigated antioxidant activity and lignan contents by harvesting times to expand use of Schisandra chinensis (Turcz.) Baillon. Total polyphenol and flavonoid contents of seed was higher than those of flesh but there is not much difference in harvesting times. As $RC_{50}$ value, that was, the concentration of sample required for 50% reduction of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical, was 13.7~24.2 ${\mu}g/m{\ell}$ in seed thus it showed a high antioxidant activity. Among lignan components, schizandrin content was the highest and followed by gomisin N and gomisin A in all of flesh and seed. Also these components in seed were 4~9 times more contained than those of flesh. All of them were decreased by harvesting times in flesh. But the contents of schizandrin and gomisin N were high in August 3rd and September 15th in seed, respectively. As the results, the seed of S. chinensis had high antioxidant activity and lignan contents so it could be potentially developed as a resource.

  • 참치유 보강 에멀젼에 첨가된 참기름은 클로로필의 존재 하에 $25^{\circ}C$, 1,700 lux의 빛에서 에멀젼 유지의 광산화를 유의하게 감소시켰다. 또한 참기름의 첨가는 에멀젼의 광산화 중 산성 조건에서 클로로필로부터 전환된 페오피틴의 분해속도를 낮추어 일중항산소 생성이 많았을 가능성에도 불구하고 일중항산소와 라디칼 소거능을 가진 인지방질, 토코페롤, 폴리페놀 화합물을 제공함으로써 참치유 보강 에멀젼 유지의 산화안정성을 개선하였으며 인지방질, 토코페롤에 비해 폴리페놀 화합물이 더 큰 영향을 주었다. 리그난 화합물은 에멀젼의 광산화 중 변화를 보이지 않아 물리적 기전에 의한 일중항산소 소거 작용을 암시하였다.

한국산 오미자로부터 디벤조사이클로옥타디엔 계열 리그난 화합물 정성 및 정량 분석 (Qualitative and Quantitative Analysis of Dibenzocyclooctadiene Lignans for the Fruits of Korean "Omija" (Schisandra chinensis))

  • Background : Dibenzocyclooctadiene lignans are secondary metabolites present abundantly in the fruits belonging to the genus Schisandra. According to previous studies, Schisandra lignans exhibit anti-inflammatory, anti-cancer and anti-diabetic properties, as well as an inhibitory effect on platelet aggregation. Therefore, establishing the Korean "Omija" (Schisandra chinensis) as a lignan-rich source, in addition to identifying and quantifying the lignans, is extremely valuable. Methods and Results : Dibenzocyclooctadiene lignans were analyzed with liquid chromatography using diode array detection/mass spectrometry, from methanol extracts subsequently identified by a constructed chemical library of 50 lignans. A total of 27 components of lignan including gomisin S were identified, of which schisandrin, gomisin A, gomisin N, deoxyschisandrin, ${\gamma}$-schisandrin, and schisandrin C were identified as the major components in the Korean Omija, Schisandra chinensis. These compounds were divided into two groups, S-biphenyl and R-biphenyl based on the configurations of the stereoisomers structures with contents of 661.7 and 1350.1mg per 100 g dry weight, respectively. The total lignan content averaged 2011.4mg per 100 g dry weight, of which schisandrin and gomisin N comprised the majority (771.8 and 420.5mg per 100 g dry weight respectively). Conclusions : Lignans which are present in high quantities in the ripe fruit of Schisandra chinensis are important functional compounds that play a major role in the prevention and treatment of human diseases.
