Functional Characterization of the Extracts from Nipa Palm, Molokhia, and Finger Root for Cosmetic Ingredients (니파팜, 몰로키아, 핑거루트 추출물의 화장품 소재로서의 기능적 특성 분석)

  • This study was conducted to evaluate the applicability of the extracts from nipa palm, molokhia, and finger root in functional cosmetics as a natural active ingredient. The extracts were obtained through the processes of heating under reflux with ethanol, filtration, concentration, and freeze-drying. UV absorption and blocking effects of the extracts were examined by using the UV-vis spectrophotometer equipped with an integrating sphere. Antioxidant activity and its stability between the extracts were compared using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay. Also, total polyphenol content in the extracts was determined quantitatively using the Folin-Ciocalteu reagent, with gallic acid as the standard. Antibacterial activity of the extracts was investigated by the disc diffusion test against Staphylococcus aureus (gram-positive) and Escherichia coli (gram-negative). Finally, collagenase inhibitor assay was performed to examine the anti-wrinkle effect of the extracts. From the results of this study, the extract of nipa palm showed the potential for use in cosmetics as an antioxidant and anti-wrinkle agent, and the extract of finger root as a sunscreen and antibacterial agent.

Determination of Siderophore from Bacillus Mojavensis Using Liquid Chromatography quadrupole Time-of-flight Tandem Mass Spectrometry (액체크로마토그래피-사중극 비행시간형 탠덤질량분석기를 이용한 Bacillus mojavensis 균주 속 사이드로포어 규명)

  • Recently, it has been reported that Bacillus mojavensis possesses antifungal properties and plant growth-promoting activities, which are similar to the characteristics of siderophore. In this study, the siderophore produced by B. mojavensis was assessed using a solid phase extraction (SPE) cartridge and liquid chromatography quadrupole time-of-flight tandem mass spectrometry (Q-TOF MS/MS). After B. mojavensis was incubated in phenol medium for 16 hr and lyophilized, the sample was dissolved in water and loaded to an SPE cartridge to remove interferences. The cartridge was washed with 5% methanol in water and eluted with 2% formic acid in methanol sequentially. The eluted solution was evaporated under a stream of nitrogen gas and reconstituted in methanol. The reconstituted sample was filtered, and $1{\mu}l$ of the sample was assessed using Q-TOF MS/MS. The mass spectrometer was operated using the positive electrospray ionization mode. Based on the mass spectrum and tandem mass spectrum, the siderophore produced by B. mojavensis was bacillibactin, one of the catechol types of siderophore with a molecular weight of 882.2556. This siderophore analysis could provide a justification for the study of B. mojavensis as a functional food and for pharmaceutical applications.

Antioxidant and Cytotoxic Activities of Hot Water and Ethanol Extracts From Caesalpinia sappan (소목의 열수 및 에탄올 추출물의 항산화 및 항암활성)

  • Caesalpinia sappan L. is an oriental medicinal plant distributed in the Asia Pacific region including India, Malaysia, and China. The dried heartwood of Caesalpinia sappan has been traditionally used as an analgesic and anti-inflammatory drug. In this study, the effects of extract methods of C. sappan on contents of total polyphenols and flavonoids, antioxidant activity, and cytotoxic activity were evaluated. As a result, hot water extract from C. sappan (CSWE) significantly exhibited contents of total polyphenols and flavonoids (22.6 mg GAE/g and 14.5 mg QE/g) higher than 70% ethanol extract (CSEE) (17.6 mg GAE/g and 13.2 mg QE/g). However, CSEE showed greater antioxidant activity than CSWE in both DPPH and ABTS. Also, the cytotoxicity of C. sappan against three kinds of cancer cell lines was higher in CSEE than in CSWE. These results show that ethanol extract is a better extract method than hot water method to maintain antioxidant and anti-cancer activities.

Studies on the Phenolic Compound Content and Antioxidant Efficacy of Rubus idaeus (Raspberry) Cultivar 'Willamette' Fruit Extract Using n-Hexane and Ethyl Acetate (n-Hexane 및 Ethyl Acetate를 이용한 Willamette 품종 라즈베리추출물의 페놀 화합물 함량 및 항산화 효능 연구)

  • In this study, Rubus idaeus (Raspberry) cultivar 'Willamette' fruit extract(RIFE) was prepared from the freeze-dried raspberry powder, n-hexane and ethyl acetate, and then the phenolic compound content, ferric reducing ability, and radical scavenging ability were measured. The raspberry cultivar 'willamette', 'polka', and 'polana' compound fruit extract did not show cytotoxicity up to the concentration of 10%. As a result of conducting an experiment at the concentration, it was confirmed that the total phenolic compound content was 375.3 ppm, and the total flavonoid content was 43.46 ppm, and the ferric reducing ability by ferric reducing antioxidant power (FRAP) reagent was equivalent to FeSO4 0.532 mM. It was confirmed that 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability was 94.5 ± 0.7%, and the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging ability was 99.4 ± 2.82%, and the nitric oxide (NO) radical scavenging activity was 88.5 ± 0.4%. When compared with the L-ascorbic acid 'standard' solution, DPPH radical scavenging ability was between 25 - 50 ppm / ABTS radical scavenging ability was close to 100 ppm / NO radical scavenging ability was more than 1,000 ppm. These results suggest that the raspberry cultivar 'willamette' fruit extract could be applied as an effective cosmetic material with antioxidant activity.

  • South Korea, as the 10th country to join the Artemis program led by NASA, is actively supporting various researches related to the lunar exploration. In particular, the utilization of water as a resource in the Moon has been focused since it was discovered that ice exists at the lunar pole as a form of frozen soil. Information on the thermal conductivity of lunar regolith can be used to estimate the existence for ice water extraction by thermal mining. In this study, the vacuum pressure effect on thermal conductivity of KLS-1 was investigated with a DTVC (Dusty Thermal Vacuum Chamber). The reliability of KLS-1 was reconfirmed through comparison with thermal conductivity of known standard lunar regolith simulants such as JSC-1A. An empirical equation to assess thermal conductivity considering dry unit weight and vacuum pressure was proposed. The results from this study can be implemented to simulate lunar cryogenic environment using the DTVC.

Bark Extractives of Several Populus Trees (몇가지 사시나무속 수종 수피의 추출성분)

  • The bark of P. alba × glandulosa, P. euramericana and P. nigra × maximounczii F1, several Populus trees, were collected, extracted with acetone-H2O(7:3, v/v), fractionated with hexane, chloroform and ethylacetate, and freeze dried to give some dark brown powder. Each fraction of the powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluents and then identified by thin layer chromatography using TBA and 6% acetic acid as developing solvents. The structures of the isolated compounds were characterized by 1H, 13C and 2D-NMR tools including mass spectrometry. Most of the compounds were flavonoids and salicin derivatives as follows: (+)-catechin, taxifolin, aromadendrin, eriodictyol, naringenin, sakuranetin, sakuranetin-5-O-𝛽-D-glucopyranoside, neosaturanin, salireposide, p-coumaric acid, and aesculin from P. alba × glandulosa, (+)-catechin, salireposide, populoside and salicortin from P. euramericana and (+)-catechin, quercetin, padmatin, salireposide, populoside and salicortin from P. nigra × maximounczii F1.

Characterization of Weissella sp. Strains Isolated from Fermented Squid and the Antibacterial Activities of Fermented Rice against Harmful Bacteria (발효된 오징어젓갈에서 분리된 Weisella sp. 균주의 특성 분석 및 쌀발효물의 유해세균에 대한 항균활성 검증)

  • The purposes of this study were to isolate and characterize lactic acid bacteria with antibacterial activity against Methicillin-resistant Staphylococcus aureus (MRSA) from fermented food and to confirm the antibacterial activities of fermented rice products using the isolated lactic acid bacterium. Three bacteria, namely, Weissella sp. ISF-1, ISF-2, and ISF-3, were selected from fermented squid based on the 16S rRNA gene sequence. All three strains grew well in an MRS medium containing 5% (w/v) NaCl and showed antibacterial activity against Bacillus cereus, Staphylococcus aureus, and MRSA. Their growth was excellent at 0% ~ 5% (w/v) NaCl and relatively good up to 7% (w/v) NaCl. The initial pH of 8 was optimal for their growth, and good growth was also observed at pH 6, 7, and 9. The lyophilisates of the fermented rice using Weissella sp. ISF-1 showed antibacterial activities against B. cereus, S. aureus, and MRSA. We inferred that isolated lactic acid bacteria could be useful in the development of probiotics and biopreservatives for foods and in the treatment of MRSA and may increase the value of rice products.

A Study on Increased Properties of Cellulose-Based Biodegradable Polymer Composites (셀룰로오스 기반 생분해성 고분자 복합재의 물성 증가에 관한 연구)

  • Growing environmental concerns regarding pollution caused by conventional plastics have increased interest in biodegradable polymers as alternative materials. The purpose of this study is to develop a 100% biodegradable nanocomposite material by introducing organic nucleating agents into the biodegradable and thermoplastic resin, poly(lactic acid), to improve its properties. Accordingly, cellulose nanofibers, an eco-friendly material, were adopted as a substitute for inorganic nucleating agents. To achieve a uniform dispersion of cellulose nanofibers (CNFs) within PLA, the aqueous solution of nanofibers was lyophilized to maintain their fibrous shape. Then, they were subjected to primary mixing using a twin-screw extruder. Test specimens with double mixing were then produced by injection molding. Differential scanning calorimetry was employed to confirm the reinforced physical properties, and it was found that the addition of 1 wt% CNFs acted as a reinforcing material and nucleating agent, reducing the cold crystallization temperature by approximately 14℃ and increasing the degree of crystallization. This study provides an environmentally friendly alternative for developing plastic materials with enhanced properties, which can contribute to a sustainable future without consuming inorganic nucleating agents. It serves as a basis for developing 100% biodegradable green nanocomposites.

Comparison of Antioxidant Compounds and Activities of Different Parts of 'Shine Muscat' Grape with Extraction Condition (포도 '샤인머스켓'의 부위별 추출조건에 따른 생리활성 물질 함량 및 항산화 활성)

  • 껍질째 먹는 청포도 품종인 '샤인머스켓'은 망고포도라고 불릴 정도로 단맛이 강하고 풍부한 머스켓향을 가지는 것이 특징이다. 또한 포도알이 크고 무핵처리에 의해 씨가 제거되어 소비자 만족도가 높아 최근 각광받고 있는 포도 품종이다. 샤인머스켓에 대한 국내연구는 주로 재배조건에 따른 과실특성 위주로 보고되고 있어, 본 연구는 소비가 늘고 있는 샤인머스켓의 기능성에 대한 기초자료를 제시하고자 부위별 추출조건에 따른 총폴리페놀, 총플라보노이드 함량 및 항산화 활성을 조사하였다. 샤인머스켓은 2019년 11월에 경북 상주지역에서 생산된 것으로, 과피와 과육의 2가지로 나누어 동결건조하였으며, 추출용매(증류수, 에탄올, 메탄올)와 추출용매농도(30%, 50%, 70%, 100%) 조건에 따라 시료 무게대비 10배의 용매를 가한 후 초음파방법으로 2시간 추출하였으며, 이를 2회 반복하였다. 부위별 총폴리페놀과 총플라보노이드 함량은 과육보다 과피에서 유의적으로 높게 나타났으며, 항산화 활성 또한 과피에서 월등히 높은 값을 나타냈다. 추출조건에 따른 생리활성 물질의 함량과 항산화 활성은 과육과 과피에서 다르게 나타났는데, 과육의 경우 메탄올 추출 시 활성이 높게 나타난 반면, 과피는 에탄올 추출 조건에서 활성이 더 높게 나타났다. 과육의 경우, 70% 메탄올 추출 조건에서 가장 높은 활성을 보였으며, 총폴리페놀 0.99 mg CE/g dw, 총플라보노이드 0.31 mg CE/g dw, DPPH 라디칼 소거능 2.94 mM TE/g dw, ABTS 라디칼 소거능 6.82mM TE/g dw, ORAC 11.40 mM TE/g dw, FRAP 4.92 mM Fe(II)/g dw이었다. 이와 달리 과피는 30% 에탄올 추출 조건에서의 활성이 유의적으로 높게 나타났으며, 총폴리페놀 10.23 mg CE/g dw, 총 플라보노이드 5.22 mg CE/g dw, DPPH 라디칼 소거능 44.72 mM TE/g dw, ABTS 라디칼 소거능 123.25 mM TE/g dw, ORAC 208.38 mM TE/g dw, FRAP 96.24 mM Fe(II)/g dw이었다. 이상의 결과를 볼 때, 껍질째 샤인머스켓을 섭취하는 것은 기능성 측면에서 유익하며, 추후 부위를 나누지 않은 샤인머스켓을 대상으로 추출조건 설정 및 기능성 평가에 대한 연구를 진행할 예정이다.

A Genotoxicity Study of Transgenic Tomatoes using CRISPR/Cas9 (CRISPR/Cas9에 의한 유전자교정 토마토의 유전독성평가)

  • Tomato is a widely distributed, cultivated, and commercialized vegetable crop. Recently, an increasing trend has been observed in the consumption of transgenic crops with enhanced functional components. However, consumer concerns regarding genotoxicity have been increasing. This study examined the genotoxicity of transgenic tomato (LTT) using the CRISPR/Cas9 system through a bacterial reverse mutation assay, chromosomal aberration assay, and mammalian micronucleus test. In the bacterial reverse mutation assay, LTT did not induce mutagenicity in Salmonella typhimurium strains TA98, TA100, TA1535, and TA1537, or Escherichia coli WP2uvrA, irrespective of the presence or absence of S9. LTT did not cause clastogenic or aneugenic chromosomal abnormalities during metaphase in CHL cells. Moreover, LTT did not increase the frequency of micronucleated polychromatic erythrocytes in the polychromatic erythrocytes. These findings can be used as a foundation to assess the genotoxicity of transgenic crops using the CRISPR/Cas9 system in the future.
