This study was performed to determine the antimutagenic and cytotoxic effect of Aster scaber root ethanol extract on Salmonella typhymurium TA98, TA100 and cancer cell lines using Ames test and cytotoxicity assay, respectively. Cancer cell lines include chronic myelogenous leukemia(K562), human gastric carcinoma(KATOIII), human hepatocellular carcinoma(Hep3B) and human breast adenocarcinoma(MCF-7). Futher fractionations with hexane, chloroform, ethyl acetate, butanol and water from ethanol extract of Aster scaber root were performed to obtain effective fraction. Ethanol extract and ethyl acetate fraction showed 79% and 82% inhibitory effect on the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) against TA100, while 48% and 60% inhibition was observed on the mutagenesis induced by 4-nitroquinoline-l-oxide(4NQO) against TA98. In the meanwhile, ethyl acetate fraction showed 78% and 85% inhibitory effect on the mutagenesis induced by benzo(${\alpha}$)pyrene[B(${\alpha}$)P] against TA98 and TA100, respectively, while 83% inhibition was observed on the mutagenesis induced by 3-amino-l,4-dimethyl-5H-pyrido(4,3-b) indole(Trp-P-1) against TA98. Ethyl acetate fraction (0.125 mg/ml) showed the strongest cytotoxic effect against K562, KATOIII, Hep3B and MCF-7 at the same concentration compared to those of other fractions. Ethanol extract and water fraction showed the least inhibitory effect.

In this study, we investigated the effect of C. zawadskii extract on nitric oxide (NO) production, prostaglandin E2 (PGE2) production, protein and mRNA expression of inducible nitric oxide synthase (iNOS) in LPS-induced RAW 264.7 macrophage cells. C. zawadskii extract (5~50 μg/mL) significantly inhibited LPS-induced NO production in a concentration-dependent manner ranging from 23.3% to 100%. Consistent with the inhibitory effect on NO production, C. zawadskii extract inhibited the protein expression and mRNA expression of iNOS. Although flower extracts of C. zawadskii was not effective on the expression of PGE2 and COX-2, flower extracts of C. zawadskii, however, showed a strong anti-inflammatory activity through inhibition of NO production and iNOS expression. The present results suggest that C. zawadskii extract has an inhibitory effect on NO production, and thus can be used as an anti-inflammatory agent.
This study surveyed the change of housewives' purchase behaviors by food safety incidents; the outbreak of 2008 Melamine incident in Korea as for example. 565 housewives in Gunsan were interviewed in March 2009. 52.3% of respondents were regarded as unsatisfactory for food safety management in Korea. Despite the result of scientific assessment for melamine, 74.6% of respondents were yet regarded as health-threatening substance. By the point of before, during and after Melamine-related food safety incident, we were surveyed the level of purchase for melamine-related food items as five scales, the results were $2.47{\pm}0.97$, $1.80{\pm}0.92$ and $1.62{\pm}0.92$, respectively (p < 0.001). After the incident happened, the purchase level was even more reduced. This study also found that there were significance difference (p < 0.05) among the respondents' knowledge for melamine toxicity and food safety management in Korea concerning housewives' purchase behaviors, i.e. the more accurately for melamine toxicity and higher satisfactory of consciousness of food safety, there were less change of purchase behaviors. In conclusion, the consciousness of food safety and accurate knowledge of hazards were significantly affected for the change of housewives' purchase behaviors by food safety incidents. Therefore, it can be suggested that the need for more scientific risk communication strategies with consumer.
The optimization of lactic acid fermentation was conducted to produce an old antler fortified with functional ingredients. For the over-production of gamma aminobutyric acid (GABA), the extract of old antlers (OA) was fermented by Lactobacillus plantarum EJ2015 with 0.5% YE, 1.5% glucose, and 3.5% MSG at $30^{\circ}C$ for 7 days. The lactic acid fermented OA showed high viable cell counts of $2.0{\times}10^8CFU/mL$, pH 6.56 and 0.77% acidity after 7 days. Addition of Auricularia auricula-judae (AAJ) enhanced the cell growth of L. plantarum EJ2014, resulting in higher viable cell counts of $2.0{\times}10^9CFU/mL$ and acid production after fermentation for 1 day. In particular, acidity was greatly decreased after fermentation for 3 days and 1.4% GABA was produced by converting efficiently mono sodium glutamate as a substrate. Fermented OA/AAJ mixture indicated the reduced cytotoxicity compared with that of unfermented OA. The fermented OA/AAJ mixture indicated anti-inflammatory effect with less production of NO in microphage cells. The production of NO dropped to $17.75{\mu}M$ at 4 mg/mL, and to $5.58{\mu}M$ at 6 mg/mL old antler after fermentation. Thus, lactic acid fermented OA with AAJ could fortify GABA, probiotics and dietary fiber.
This study was conducted to evaluate the genotoxicity of balanced nutritional formular for patients containing various ingredients after gamma irradiation at 4 kGy. Since viable bacteria were not observed within the detection limit of 1 log CFU/g, a dose of 4 kGy was appropriate for the pasteurization of the formular. In a bacterial reverse mutation assay, both hot water and methanol extracts of the formular exhibited dose-independent responses, which was similar to those obtained from that of the negative control (distilled water or dimethyl sulfoxide). In a chromosomal aberration test using lung fibroblast cells of Chinese hamster, the numbers of normal chromosomes were comparable to those observed in the negative control, regardless of the treatment dose and metabolic activation system. Furthermore, no significant increases in the frequency of micronucleated polychromatic erythrocytes were observed relative to the control, when mice were fed with the formular at doses up to 2,000 mg/kg body weight. Therefore, the balanced nutritional formular for patients did not exhibit genotoxicity when pasteurization by gamma irradiation at 4 kGy.

Paralytic shellfish poisoning (PSP) occurs when saxitoxin (STX), which is produced by harmful algae (dinoflagellates) and then accumulated in bivalve shellfish by filter-feeding, is consumed by humans. With recent advances in analysis technology, it has been reported that dinoflagellates also produce a variety of analogues such as the gonyautoxin (GTX) group and the N-sulfo-carbamoyl toxin (C toxin) group, in addition to STX. Accordingly, CODEX and the EFSA are stepping forward to manage STX and analogues as STX groups requiring safety management. In Korea, the occurrence of dinoflagellates producing STX analogues has already been reported, and contamination of analogues (GTX group, C toxin group) in live mussels has also been reported. In this study, in order to provide the basis for systematic monitoring and safety management of STX and analogues, their physicochemical characteristics, occurrence of dinoflagellates, toxicity and toxic equivalency factor, analytical method and occurrence were widely reviewed. This review is expected to contribute to strengthening the safety management of STX and its analogues.
In this study, a quantitative risk impact assessment is performed using an ALOHA program to identify the risks when applying ammonia as fuel for combined cycle power plants as one of the solutions of climate change. The worst and the alternative accident scenarios are established for the Sejong combined cycle power plant and the effective ranges are calculated in terms of flammability, thermal radiation, overpressure and toxicity. The analysis results show that the toxic risk is the most critical and the effective distance is highly proportional to the mixing ratio of natural gas and ammonia by showing the Pearson's correlation coefficient over 98% as 0.991, 0.987 and 0.989 for the Level Of Concern(LOC)-1, LOC-2 and LOC-3, respectively. In addition, the coefficients of linearity for LOC-1, LOC-2 and LOC-3 are calculated to 133, 70 and 29, respectively so it can be confirmed that the effective distance increases as the criterion decreases.

BACKGROUND: RNA interference (RNAi) eliminates or decreases gene expression by disrupting the target mRNA or by interfering with translation. Recently, RNAi technique was applied to generate new crop traits which provide protection against pests. To establish the environmental risk assessment protocol of RNAi LMO in lab scale, we developed dsRNA expression system using E. coli and tested acute oral toxicity assay to honey. METHOD AND RESULTS: The dsRNA expression vector, L4440, was chosen and cloned 240 bp of Snf7 and GFP gene fragment. To develop the maximum dsRNA induction condition in E. coli, we tested induction time, temperature and IPTG concentration in media. To estimate the risk assessment of dsRNA to honey bee, it has been selected and cultured with dsRNA supplement for 48 hours according to OECD guideline. As a result, the optimum condition of dsRNA induction was $37^{\circ}C$, 4 hours and 0.4 mM IPTG concentration and the difference between Snf7 and GFP dsRNA molecules from E. coli was not significant in survival and behavior to honey bee. Furthermore, blast search results indicated that effective match of predicted dsRNA fragments were not existed in honey bee genome. CONCLUSION: In this study, we developed and tested the acute oral toxicity of dsRNA using E. coli expression system to honey bee.

Volatile organic compounds(VOC) present in or generated by many sources, can be toxic, mutagenic or even carcinogenic, so that control of such emissions is significant. The 6 chlorinated organic chemicals as VOC were examined in this study. Prediction of the behavior of VOC on activated carbon beds is an important part of control system design. The objective of this study was to predict gas phase sorption isotherms from physical properties and liquid phase isotherms obtained with the same adsorbent and adsorbate. One of the techniques that was investigated was quantitative structure-activity relationships(QSAR) for the predicition procedures. It was possible to estimate sorption isotherms in the gas phase($a_g$) using either connectivity index, $^2{\chi}$, and the Henry's law coefficient ($H_a$) or the solubility and the equilibrium concentration in the gas phase. As a result of study, the predictive equation based on Freundlich model for $a_g$ was ${\log}\;a_g=0.238\;^2{\chi}+0.573\;{\log}\;H_a+4.330(r^2=0.94)$. Finally, this would provide a potentially useful tool to describe and predict sorption capacity without time-consuming tests.
