Evaluation of the Fermentation Ability of Lactic Acid Bacteria to Manufacture Highest Quality Rice Straw Silage (고품질 볏짚 사일리지 제조를 위한 다양한 유산균주 평가)

  • The main objective of this study was to determine the quality of rice straw silage made with various lactic acid bacteria (LAB) during fermentation. Five strains of LAB (Lactobacillus plantarum CMRT, L. leuconostoc mesenteroides M17, L. sakei C11, M5, SP2) were used in this study. With regard to odor, ready-made CMRT (a comparison strain) had the highest value from 10-60 days, followed by M17. The pH level of all silages made with five strains (apart from CMRT) ranged from 4.02 to 4.59. Of these, M17 rapidly lowered the pH value in the silage. Crude protein (CP) content was significantly lower (p < 0.01) in ready-made CMRT compared to the other bacteria. C11 fermentation produced the highest content of silage, with a score of 5.56. Acetic acid and butyric acid were not detected in any of the silage products. The lactic acid levels in silages produced by M17, M5, C11, and SP2 were comparatively higher than that produced by CMRT. The total digestible nutrient (TDN) content levels and relative fees value (RFV) were he highest in the silage of C11 (69.65 and 155.56, respectively), followed by M17. Based on odorl, pH, protein, organic acid, and feed value, we suggest tha the M17 strain is a suitable substitute for CMRT that can be used to generate high quality rice straw silage.

  • This study was conducted to study the environment effects on live weight, carcass weight, dressing percentage, eye muscle area, backfat thickness, and marbling score, which are the carcass traits of Hanwoo, based on the estimates and all the possible regression for the selection of variable and significance test for 231,382 heads that underwent the carcass measurements. The average and standard deviation for the live weight, carcass weight, dressing percentage, eye muscle area, backfat thickness, and marbling score were 654.79${\pm}$91.61 kg, 362.30${\pm}$67.15 kg, 59.52${\pm}$0.03%, 81.79${\pm}$12.21 $cm^2$, 11.39${\pm}$5.40 mm, 4.38${\pm}$2.29, respectively. The live weight, carcass weight, dressing percentage, eye muscle area, backfat thickness, and marbling score for cow were 532.79${\pm}$78.38 kg, 313.40${\pm}$44.90 kg, 56.50${\pm}$0.03%, 75.24${\pm}$10.69 $cm^2$, 11.82${\pm}$5.10 mm, 4.30${\pm}$2.06, respectively, while for bull were 619.74${\pm}$93.27 kg, 376.89${\pm}$48.62 kg, 58.61${\pm}$0.02%, 85.61${\pm}$10.46 $cm^2$, 5.64${\pm}$2.71 mm, 1.41${\pm}$0.83, respectively, and for steer were 681.78${\pm}$70.72 kg, 415.23${\pm}$49.43 kg, 60.19${\pm}$0.02%, 88.29${\pm}$10.27 $cm^2$, 12.71${\pm}$5.23 mm, 5.42${\pm}$1.99, respectively. In the environmental variables selection based on the variables selection method, the examination by carcass traits suggested that the most appropriate model could be determined when five variables were selected for the live weight, carcass weight, dressing percentage, eye muscle area, and four variables for backfat thickness, and marbling score. When they were considered at a time altogether based on multiple traits, it was deemed to be desirable to insert all five variables into the variables for analysis. In addition, high significance was found by carcass traits.

Influence of Potassium Concentrations in Fertilizer Solution on the Growth, Appearance of Physiological Disorder and Tissue Nutrient Contents of Eggplant (Solanum melogena L.) (칼륨 시비농도가 가지의 생육과 생리장해 발현 및 무기원소 함량에 미치는 영향)

  • Objective of this research was to investigate the effect of various potassium concentrations in fertilizer solution on growth of and nutrient uptake by 'Chugyang' eggplant ($Solanum$ $melongena$ L.). Tissue analyses were conducted to determine the threshold levels of potassium in plants when disorders develop for potassium. The lower leaves of K deficient plants became spotted, yellowing in color, and finally necrosis occurred. The necrosis spread inward and upward. K toxic plants developed necrotic spot at the margin of old leaves and the surface of old leaves were twisted. The tissue K contents in the most recently fully expanded leaves and dry weight of full above ground plant tissue at 35 days after transplanting showed linear and quadratic response to elevated K concentrations with the equation of y=1.127+0.3147x ($R^2$=0.8916) and y=14.92+2.2743x-$0.1402x_2$ ($R^2$=0.8659). When 10% reduction in dry weight set to threshold levels, optimum tissue K contents are in the range from 2.1 to 5.1%. The yield through 150 days after transplanting and tissue K contents at 150 days after transplanting also showed quadratic and linear responses to elevated potassium concentrations in fertigation solution with the equation of y=153.24+345.5x-$18.46x^2$ ($R^2$=0.8620) and y=0.9921+0.3860x ($R^2$=0.9611), respectively. When the 10% reduction in yield is set to threshold levels, the tissue K contents for maximum yield should be around 3.4% to 5.9% in most recently fully expanded leaves.

Cutoff Values of Body Mass Index and Body Fat Measures for Metabolic Syndrome in Korean Population (한국인에서 대사증후군의 선별검사로서 체성분 분석 및 체질량지수의 타당성연구)

  • This study investigated cut-off values of body mass index (BMI) and body fat measures for metabolic syndrome (MS) in elderly Koreas. Questionnaire surveys, anthropometric measurements, medical examinations, and body composition analyses were conducted for 10,077 subjects aged 40-65 years in the health examinee cohort in Korea between 2004 and 2006. Cut-off values were identified using receiver-operating characteristic (ROC) curve analysis for both men and women. Stratified analyses by weight range (<60, 60-69, 70-79, and 80-89, ${\geq}90\;kg$ for men; <50, 50-59, 60-69, 70-79, ${\geq}80\;kg$ for women) were conducted. Among male subjects, the cut-off points were $25.5\;kg/m^2$ for BMI with a sensitivity of 70.0% and a specificity of 72.0%, and 26.1% for body fat percentage with a sensitivity of 60.6% and a specificity of 76.4%. Among female subjects, the cutoff points were $24.1\;kg/m^2$ for BMI with a sensitivity of 73.3% and a specificity of 68.8%, and 31.5% for body fat percentage with a sensitivity of 76.7% and a specificity of 65.6%. Stratified analysis by weight range showed that the cutoff points of BMI and body fat measures tended to higher as weight level increased. The results of our study suggest cut-off values of BMI and body fat measure for MS were similar to the general obesity criteria in Korea.

Antibacterial, Antioxidative Activity and Component Analysis of Pinus koraiensis Leaf Extracts (잣나무 잎 추출물의 항균 및 항산화 활성과 성분분석)

  • In this study, the antibacterial, antioxidative effect and component analysis of Pinus koraiensis leaf extracts were investigated. MIC values of the ethyl acetate fraction from P. koraiensis leaf extracts on P. acnes, S. aureus, P. ovale, and E. coli were 0.06 %, 0.25 %, 0.13 % and 0.50 %, respectively. The results showed that the antibacterial activity of the ethyl acetate fraction on P. acnes, P. ovale. and S. aureus was more prominent. The aglycone fraction of P. koraiensis leaf extracts ($22.93\;{\mu}g/mL$) showed more higher free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of P. koraiensis leaf extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system was investigated using the luminol-dependent chemiluminescence assay. The 50 % ethanol extract ($0.70\;{\mu}g/mL$) showed the most prominent ROS scavenging activity. Also the ethyl acetate ($1.04\;{\mu}g/mL$) and the aglycone fraction ($1.43\;{\mu}g/mL$) showed very high antoxidant activity. The protective effects of extract/fractions of P. koraiensis leaf extracts on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The P. koraiensis leaf extracts showed cellular membrane protective effects in a concentration dependent manner ($5{\sim}50\;{\mu}g/mL$). TLC and HPLC chromatogram of the ethyl acetate fraction obtained from hydrolysis of P. koraiensis leaf extracts revealed 2 main bands (PK-4, PK-6) and peaks (peak 1, peak 2), which were identified as kaempferol-3-O-glucoside (PK-6, peak 1) and kaempferol-3-O-arabinoside (PK-4, peak 2) by LC/ESI-MS/MS, respectively. These results indicate that extract/fractions of P. koraiensis can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging ROS, and protect cellular membrane against ROS. Extract/fractions of P. koraiensis can be applicable to new cosmeceuticals for antioxidant, antiaging, and antibacterial activity.

A Survey on Dietary Habits in Gyeongnam and the Development of the Nutrition Education Curriculum with Teacher's Guide for Obese Elementary School Children (경남지역 일부 초등학교 비만아동의 식습관 분석 및 영양교육을 위한 교수학습과정안 개발)

  • The purpose of this study was to develop a nutrition education curriculum with teacher's guide which includes discretionary activities for obese children. A survey was carried out to investigate the recognition of body image and food behaviors according to the obesity index (mild, moderate, severe) in school children (4~6th grade, 158 boys and 60 girls) who were selected based on a physical examination in May, 2006 in the Gyeongnam province. Next, a nutrition education curriculum with teacher's guide was developed on the basis of the findings from the survey and from preceding researches. The results are summarized as follow. The results of this study showed the existence of some nutritional problems such as overeating, prejudice, skipping meals, snacking patterns, etc, which indicate the need for nutritional management for obese children. Most overweight children (80.3%) showed the most interest in the nutrition education program, particularly with regards to dieting for weight control (64.7%). The developed nutrition education curriculum consisted of 8 main subjects and 13 subtitles. The curriculum was prepared for 13 lessons and included songs and singing, making-up lyrics, games about nutrition, discussions of the experience of eating (satiety, thirst, hunger), debates on dietary habits, writing and others to promote the interest for learning. We aimed to develop this program in an attempt to improve the dietary habits of obese school children. This is very important because once a dietary habit is formed in adults, it is difficult to change and the best adjustable stage is during childhood. Therefore, early nutrition education during elementary school can change and build-up the awareness of health in young elementary school children.

Antiemetic Effect of Dexamethasone in Dogs Sedated with Xylazine (Xylazine hydrochloride로 진정시킨 개에 대한 Dexamethasone의 항구토 효과)

  • This prospective study aimed to assess the efficacy of dexamethasone to prevent xylazine induced emesis in dogs. The antiemetic effect of graded, single high-dose intravenous dexamethasone against xylazine hydrochloride was studied. Clinically healthy mixed breed dogs that weighed $ 4.64\pm1.25kg$ were used in this study. Food and water were given 3 hours before the experiment. Venous blood specimens were collected from all experimental animals for hema-tological and blood chemical test pre- and post-experiment. Twenty-eight experimental animals were randomly divided into 4 groups; the group treated with 0.2ml/kg of normal saline (Control group), the groups treated with 1mg/kg (D1 group), 2mg/kg (D2 group) and 4 mg/kg of dexamethasone (D4 group). Three doses of the dexamethasone or normal saline was administered intravenousely to each group and after 5 minutes, xylazine (2.2 mg/kg) was administered intramuscularly. The time until onset of the first emetic episode and rate of emesis were investigated. At the same time, the extent of sedation was scored subjectively 5, 15, 30 and 60 minutes after injection of xylazine hydrochloride using Visual Sedation Score. The time until onset of the first emetic episode was $203.25\pm11.35$ sec in Control group, $187.33\pm48.0l$ sec in D1 group and 218.33± 13.58 sec in D2 group. The rate of xylazine induced emesis were $57\%$ in Control group and $43\%$ in D1 and D2 group respectively. On the other hand, any emetic episodes were not observed in 04 group. At extent of sedation score, all experimental animals especially including the animals in D1 group were highly sedated at 15 minutes after administration of xylazine hydrochloride. Hematological and blood chemical values showed normal ranges pre- and post-experiment. We concluded that prior treatment with 4 mg/kg of dexamethasone hardly caused xylazine-induced emesis without disturbing the sedative effect of xylazine in dogs.

Acupuncture for Prehypertension and Stage 1 Hypertension in Postmenopausal Women: Protocol for a Randomized Controlled Pilot Trial (폐경 후 여성의 전단계 및 1기 고혈압에 대한 침 치료: 다기관 무작위 대조 예비연구)

  • Objectives : This study aims to evaluate the effectiveness and safety of acupuncture and explore the appropriate number of treatment for postmenopausal women diagnosed with prehypertension and stage 1 hypertension. Methods : A 4-arm randomized open label pilot trial will be performed at 2 centers. Sixty participants will be divided into 2 treatment groups and 2 control groups. Treatment groups will receive acupuncture at 8 points(bilateral GB20, LI11, ST36, SP6) for 4 weeks(treatment group A, 10 total sessions) or 8 weeks(treatment group B, 20 total sessions), while maintaining usual care. Control groups will not receive acupuncture but will be under usual care for 16 weeks(control group C) or 20 weeks(control group D). Each patient's living habits will be corrected and drugs that may affect blood pressure(BP) will be prohibited. Treatment group A and control group C will be evaluated at 4, 8, 12, and 16 weeks after randomization, while treatment group B and control group D will be evaluated at 4, 8, 12, 16, and 20 weeks after randomization. The major outcome variable is the magnitude of change in diastolic BP levels at 4 weeks after randomization; auxiliary outcome variables are (1) diastolic BP change at 8, 16, and 20 weeks, (2) systolic BP change, (3) BP control rate, (4) lipid profiles, and (5) high-sensitivity C-reactive protein. Patient safety will be assessed at every visit. Results and Conclusions : The study findings may help develop evidence for the effectiveness and safety of acupuncture for BP control.

  • Translationally controlled tumor protein (TCTP) is one of the most abundant proteins in various eukaryotic organisms. TCTPs play important roles in cell physiological processes in cancer, cell proliferation, gene regulation, and heat shock response. TCTP is also considered an important factor in the resistance to oxidative stress induced by dithiothreitol or hydrogen peroxide (H2O2). Arctic calanoid copepods have a variety of antioxidant defense systems to regulate the levels of potentially harmful reactive oxygen species generated by ultraviolet radiation in the Arctic marine ecosystem. However, information on the antioxidant activity of TCTP in the Arctic Calanus glacialis is still scarce. To understand the putative antioxidant function of the Arctic copepod C. glacialis TCTP (Cg-TCTP), its gene was cloned and sequenced. The Cg-TCTP comprised 522 bp and encoded a 174-amino acid putative protein with a calculated molecular weight of ~23 kDa. The recombinant Cg-TCTP (Cg-r TCTP) gene was overexpressed in Escherichia coli (BL21), and Cg-rTCTP-transformed cells were grown in the presence or absence of H2O2. Cg-rTCTP-transformed E. coli showed increased tolerance to high H2O2 concentrations. Therefore, TCTP may be an important antioxidant protein related to tolerance of the Arctic copepod C. glacialis to oxidative stress in the harsh environment of the Arctic Ocean.

Analysis of Lymphocyte Subsets in Peripheral Blood after Radiotherapy (악성 종양 환자에서 방사선 치료 전, 후의 림프구 아형 분석)

  • Purpose : To evaluate the changes of differential counts and lymphocyte subsets in cancer patients' leukocyte before and after radiotherapy. Materials and Methods : From Dec. 1994 to Mar 1995, the changes of leukocyte and its subsets in 16 patients who received radiotherapy in the Dept. of Radiation Oncology of Dong-A University Hospital were investigated. Radiation was delivered from 2700 cGy to 6660 cGy with median dose of 5400 cGy. The results of pre- and Post-radiotherapy were analyzed by paired T-test. The results of patients Who received < 50 Gy and $\geq$ 50 Gy were analyzed by Wilcoxon test. Results : Before and after radiotherapy, there was not any significant differences in the counts of leukocyte, granulocyte and monocyte. A remarkable decrease was noted in lymphocyte counts after radiotherapy(p=0.015). T cells, B cells and natural killer cells were also decreased in number after radiotherapy but it was not significant statistically. 1 helper cells and T suppressor cells were also decreased in number(p>0.05). The ratio of T helper/suppressor cell was decreased from 1.52 to 1, 11 and it was significant statistically(p=0.016). The portion of T suppressor cell among all T cells was increased after radiotherapy (p=0.0195). No significant difference was observed in the analysis of leukocyte and its subsets between patients who received < 50 Gy and $\geq$ 50 Gy, Conclusion : Radiotherapy caused remarkable decrease in lymphocyte count and its subsets. Among all lymphocyte subsets, T helper cell might be the most vulnerable to radiation, considering decreased ratio of T helper/suppressor cell count after radiotherapy.
