Protective Effects of Hwansodan(Huanshao-dan) Water Extract in Serum Deprivation-induced Apoptosis of PC12 Cells (환소단이 영양혈청 결핍성 PC12 신경세포의 apoptosis에 미치는 영향)

  • Objectives : This study was designed to investigate the neuroprotective effect of Hwansodan(Huanshao-dan) on the apoptosis induced by withdrawal of neurotrophic support. Methods : PCl2 pheochromocytoma cells have been used extensively as a model for studying the cellular and molecular effects of neuronal cells. The viability of cells was measured by MTT assay. We used DNA fragmentation and caspase 3-like protease activation assay. Results : The water extract of Hwansodan(Huanshao-dan) significantly showed protective effects on serum and glucose deprivation-induced apoptotic death. Hwansodan(Huanshao-dan) also prevents DNA fragmentation and caspase 3-like protease activation, representing typical neuronal apoptotic phenomena in PCl2 pheochromocytoma cells and induces tyrosine phosphorylation of proteins around 44 kDa, which was identified as ERK1 with electrophoretic gel mobility shift by Western blot. In addition, MAPK kinase(MEK) inhibitor PD98059 and Ras inactivator, ${\alpha}-hydroxyfarnesylphosphonic$ acid attenuated the neuroprotective effects of Hwansodan(Huanshao-dan) in serum-deprived PCl2 cells. Conclusions : These results indicate that Ras/MEK/ERK signaling pathway plays a key role in neuroprotective effects of Hwansodan(Huanshao-dan) in serum-deprived PCl2 cells. Taken together, we suggest the possibility that Hwansodan(Huanshao-dan) might provide a neurotrophic-like activity in PCl2 cells.

Green Tea (-)EGCG Induces the Apoptotic Death of Lung Cancer Cells via Activation of c-Jun N-terminal Kinase 1 and Activating Protein-1 (녹차의 (-)EGCG에 의한 사람 폐암 세포주 A549의 c-Jun N-terminal Kinase 1과 Activating Protein-1활성화를 통한 세포고사)

  • Green tea has been recognized as a favorite beverage for centuries in Easter and Westers cultures. Recently, anti-tumor effects of green tea constituents have received increasing attention. However, the mechanism of catechin-mediated cytotoxicity against tumor cells remains to be elusive. To elucidate the mechanical insights of anti-tumor effects, (-)epigallocatechin-gallate(EGCG) of catechin was applied to human lung cancer A549 cells. (-)EGCG induced the death of A549 cells, which was revealed as apoptosis in DNA fragmentation assay. (-)EGCG induced the activation of caspase family cysteine proteases including capase-3, -8 and -9 proteases in A549 cells. Furthermore, (-)EGCG increased the phosphotransferase activity of c-Jun N-terminal kinase 1JNK 1), which further induced tole transcriptional activation of activating protein-1(AP-1) in A549 cells. We suggest that (-)EGCG-induced apotosis of A549 cells is mediated by signaling pathway involving caspase family cysteine protease, JNK1 and transcription factor, AP-1.

  • The new knowledge in the textbooks for Physiology and Hygiene in Korea 1905-1910 was focused on the problems of hygiene. Lighting, ventilation, dryness were proposed as main conditions fer hygienic housing, for which management and planning methods were explained. This knowledge was taught as 'scientific' or 'modern' knowledge based on the physiological theory and air property, thereafter the conditions were placed as important conditions of modern housing.

  • 최근 건설경기의 난국 속에서 많은 건설기업들이 경영상의 어려움에 처해 있다. 본 연구에서는 코스피 코스닥 상장 건설기업들을 대상으로 ANP와 DEA 결합 모델을 이용하여 기업의 경영 효율성 분석을 실시한다. DEA 모형에 쓰이는 투입 및 산출 요소의 결정은 ANP 모형의 중요도 분석을 통하여 결정함으로써 기존의 방법보다 효율성 분석 결과의 신뢰도를 제고하였는데, 효율성 분석결과 비효율적 그룹에 대해서는 효율적 그룹으로의 이동을 위한 벤치마킹 대상과 개선을 위한 효율성 수치를 제공한다. 또한 연구의 결과로 나온 기업별 효율성과 주가와의 상관관계를 분석한다. 본 연구에서는 코스피 상장 건설기업 36곳과 코스닥에 상장된 건설기업 21곳인 총57개 기업을 대상으로 기업의 효율성을 평가하였는데, 효율성 분석결과 CCR 효율성이 1인 기업은 4개 기업, BCC 효율성이 1인 기업은 7개 기업, 규모의 효율성이 1인 기업은 4개 기업 이었으며 각각의 효율적 그룹군 기업과 주가와의 상관관계는 CCR, BCC 모형 모두에서 0.7 이상의 강한 양의 상관관계를 나타냈다.

The effect of Sodium chloride concentrations of Soaking-Water and Submaterials on Tongbaechu-Kimchi Taste and Fermentation (침지수의 염도 및 부재료가 통배추김치의 맛과 숙성에 미치는 영향)

  • This study was investigated to find effects of soaking-water salt concentration and submaterial on Kimchi taste and fermentation period. several parameters such as pH, acidity, reducing sugar, amino acid and sensory evaluation during fermentation at 2$0^{\circ}C$ were experimented. The results are as follows : As fermentation period increases, pH and content of reducing sugar of Kimchi have decreased. But acidity has increased. Especially, the pH of all samples rapidly decreased to a extent of pH 4.2. During fermentation the acidity and content of reducing sugar in 12% soaking-water were higher than those in 16% and 20%. Content of reducing sugar I Kimchi was observed to play an important role for the overall taste. As a result of sensory evaluation during fermentation, Kimchi in 16%, 2.6% and in 20%, 3.7%. Sensory evaluation revealed that leek-added Kimchi was highly evaluated in crispness, sour taste, flavor and overall acceptability. These results suggested that Kimchi fermented for 4days at 2$0^{\circ}C$ with 16% salt concentration of soaking water and leek as submaterials should taste most desirable.

An Investigative Analysis of Recognition and Uses for Astragalus Membranaceus in Seoul and Kyunggido Area (서울, 경기지역의 황기에 대한 인지도 및 이용실태 조사)

  • This study was conducted to investigate the recognition and uses of Astragalus membranaceus. The survey methodology employed was a questionnaire, which interviewed 272(37.8%) males and 448(62.2%) females from the Seoul and Kyunggido area. The main results are as follows: 40-50 years old like and eat Astragalus membranaceus more frequently than 10-30 years old. The most popular reason provided for the preference of Astragalus membranaceus was good health benefits. People indicating a dislike for Astragalus membranaceus had generally fewer previous experiences of eating this plant. The main purchasing place appears to be traditional markets and supermarkets. Astragalus membranaceus was acknowledged as having the following characteristics ‘excellent nutrition’(3.82), ‘natural food’(3.76), ‘possessing anti-cancer and anti-oxidization characteristics’(3.70) and ‘good for liver function, a depressant and good coelenteron effects’(3.62). However, it was not associated with a ‘cheap price’(3.02) and was regarded as ‘difficult for buy’(3.10). Recognition and eating experience was low for Astragalus membranaceus kimchi and rice bread, bread, sauce and Astragalus membranaceus added health drinks. However opinion of its taste was regarded highly.

Studies On Whole Chinese Cabbage Kimchi -An Investigation on the Method of Making Kimchi and a Taste in the Taegu Area- (통배추김치에 관한 연구 -대구지역을 중심으로 담그는 방법과 기호에 관한 실태조사-)

  • The purpose of this study was to investigate the method of making whole Chinese cabbage Kimchi, the family's taste in Kimchi and the reason for disliking the taste of Kimchi in the Taegu area. The results were as follows; 1) Most housewives learned the method of making the Kimchi from their mothers. Method was as follows; First, scatter salt on the whole Chinese cabbage and soak that for 3∼6 hours. After, wash the whole Chinese cabbage 3 times. Then, add red pepper powder, garlic, ginger and salted fermented anchovy as basic seasoning. Be sure to, mix the red pepper powder and salted fermented anchovy until they taste hot and salty. Store the whole Chinese cabbage until they ferment. Afterwards store the whole Chinese cabbage in the refrigerator. 2) It was the whole Chinese cabbage Kimchi that most family members preferred. 3) Among the family members, the old age group preferred newly made Kimchi because of the cool and fresh taste. The adult group preferred fresh taste, also smell and peculiar taste of the seasoning were important. The adolescent group preferred fresh taste and sour taste. 4) It shows that most of the family likes Kimchi, and among the family members, about 25% of the sons and daughters dislike Kimchi. And it shows that all family members dislike salty taste. 5) The older housewives’ Kimchi was preferred over the younger housewives’ Kimchi.

Zinc-induced Apoptosis in C6 glial Cells via Generation of Hydrogen Peroxide($H_2O_2$) (신경교세포주 C6 glial에서 Zinc의 Hydrogen Peroxide($H_2O_2$) 생성을 통한 세포고사)

  • Zinc is known to generate reactive oxygen species (ROS) including superoxide anion and hydrogen peroxide ($H_2O_2$), which eventually contribute to cytotoxicity in a variety of cell types. Here in, we demonstrated that zinc decreased the viability of C6 glial cells in a time and dose-dependent manner, which was revealed as apoptosis characterized by ladder-pattern fragmentation of genomic DNA. chromatin condensation and DNA fragmentation in Hoechst dye staining. Zinc-induced apoptosis of C6 glial cells was prevented by the addition of catalase and antioxidants including reduced glutathione (GSH), N-acetyl-L-cysteine (NAC) and pyrrolidinedithiocarbamate (PDTC). Wefurther confirmed that zinc decreased intrac-ellular levels of GSH and generated $H_2O_2$in C6 glial cells. Moreover, antioxidants also decreased the generation of zinc-induced $H_2O_2$ in C6 glial cells. These data indicated that zinc-induced the apoptotic death of C6 glial cells via generation of reactive oxygen species such as $H_2O_2$.

  • The development of rapid science technology and data communication technology in 1990 was infiltrated into our real lives deeply and drew forth various changes in various sides. According to that with the new definition of the character of space the users' patterns in the space were changed. We come to have a turning point of change with the introduction of personal computer and digital revolution late in the 20th century. Accordingly, this study is to analyze the connected structure of the space on a basis of the apartment model house since 2000 in order to investigate the aspect of changing the space according to the era of informationization. This analysis of space structure makes the characteristics in the era of informationization objectified. And it will understand the distinct change of space by grasping the relations with the connection of space, and predict the direction that the dwelling space heads for the future.

  • On account of the increasing crime rates annually and the consequent rising anxiety, safety has become an important topic in the aspect of environmental formation to provide decent lives to urban dwellers. As a result, great emphasis has been placed in the importance of Crime Prevention through Environmental Design (CPTED), a design method which utilizes physical environmental design to prevent crime. The definition of CPTED encompasses the technological level of crime prevention as well as comprehensive environmental design which takes into consideration fear or terror in view of human recognition and emotion. However, the current application method of CPTED in Korea is still limited to the technical supplementation of facilities, such as CCTV and streetlight, and fragmental modification like village mural paintings. Moreover, the guideline for CPTED in Korea is only a theoretical adaptation of foreign books and does not offer sufficient studies for a practical design method that will improve the life of city dwellers. The purpose of this study is to examine design methods for safe environment construction in accordance with the changing concept of CPTED by understanding the characteristic of space and analyzing the how the methods of light are expressed. This study aims to provide basic material which will serve as a guideline in creating a safe environment in the future.
