Effect of trans Fatty acid containing Fats on Cholesterol Metabolism and Hepatic Membrane Fluidity in Rats (trans 지방산 함유 지방이 콜레스테롤대사와 간 세포막 유동성에 미치는 영향)

  • Although negative evidences of trans fatty acids(tFAs) are gradually increasing, the hypercholesterolemic effect of tFAs are controversial and its biological significances are still not known. The objective of this study was to examine the effect of dietary tFAs on cholesterol metabolism and membrane fluidity in rats. Animals were fed diets containing 0.5% cholesterol and 20% test fats(margarine, butter, corn oil) for 8 weeks. Each test fats(margarine, butter, corn oil) contained 25%, 8.5% and no tFAs, respectively, Plasma total triglyceride(TG) were not different among diet treatments. Feeding trans fat diets (margarine and butter) lowered plasma cholesterol. Specially, butter diet elevated LDL-cholesterol and decreased HDL-cholesterol levels, resulting in the highest atherogenic index among diet treatments. Hepatic cholesterol concentration and HMG CoA reductase activity were also decreased, whereas fecal excretion of cholesterol was increased in trans fat-fed animals. trans fat containing diets also decreased hepatic membrane fluidity. From these results, it can be concluded that hypercholesterolemic effect of tFAs can be modulated by the fatty acid composition of diets, but its spatial cofiguration may help behave like saturated fatty acid in membrane fluidity.

Physicochemical Composition of Broccoli Sprouts (브로콜리 싹의 이화학적 성분)

  • This study was conducted to investigate the major chemical components of dried broccoli (Brassica oleracea var. italica Plenck) sprouts. The proximate compositions of broccoli sprouts as dry matter basis were 2.04% of moisture content, 22.04% of crude protein, 12.80% of crude fat, 6.25% of crude ash, and 56.87% of carbohydrate, respectively. The major free sugars were identified as ribose, glucose and fructose. Analysing total amino acids, 15 kinds of components isolated from broccoli sprouts. The essential amino acid contained in broccoli sprouts accounted for 45.62% of total amino acid, while the non-essential amino acid accounted for 54.38%. Fatty acids were 3.19% of saturated fatty acids, 14.42% of monounsaturated fatty acids and 82.39% of polyunsaturated fatty acids. Cis-11,14-eicosatrienoic acid, linoleic acid and oleic acid were the major fatty acids among 14 fatty acids detected in dried broccoli sprouts. Lactic acid was the major organic acids. Antioxidative and Antimicrobial Activities of Pleurotus eryngii Extracts Prepared from Different Aerial Part (부위별 새송이버섯 추출물의 항산화 및 향균효과)

  • Antioxidative and antimicrobial activities were measured for the Pleurotus eryngii (P. eryngii) solvent extracts in order to discover new functional activities. In P. eryngii, the powder moisture was 9.0%, and the carbohydrate, crude protein, crude ash and crude fat contents were 63.06, 20.70, 5.20 and 2.0%, respectively. Among the detected minerals, potassium (K) had the highest levels and manganese (Mn) the lowest. The amount of polyphenol in EtEx (Ethanol Extract) was 387 mg% for the whole body, 158 mg% for the stipe, and 593 mg% for the pileus. Higher levels of polyphenol in the entire body were found in the BuEx (Butanol Extract) (594 mg%) and WaEx (Water extract) (404 mg%) of the P. eryngii powder. BuEx had the highest level in the pileus, and EtEx and BuEx were higher than the other extracts in the stipe. The electron donating ability (EDA) of EtEx of the P. eryngii powder was the highest, at 91.12%, for the whole body, while it was the lowest, at 62.90%, in the stipe. In addition, the EDA of WaEx was 90.39% for the whole body. These EDA values were similar to those for tocopherol (93.93%) and BHT (96.72%), supporting the potential of these extracts to act as antioxidants. A number of the extracts were certified to have antimicrobial activities for small number of microorganisms, especially for gram-negative microorganisms. In other words, BuEx and EAEx in the pileus and WaEx in the stipe were found to inhibit the growth of Pseudomonas aeruginosa (gram negative). Additionally, EtEx and WaEx in all parts were shown to act as antimicrobial agents for Escherichia coli of gram negative.

  • 하천주변 저지대에 주택이나 공장 등의 시설물 집중과 산지의 개발 및 인구 집중으로 인한 도시화 지역의 증가로 홍수 피해는 더욱 커지고 있다. 특히, 상습침수지구의 주민들은 항상 재해로부터 위험을 느끼고 있으므로 새로운 재해관리체계를 구축하여 이에 대한 확고한 대책이 필요하다. 최근 인터넷 사용의 증가로 많은 사용자들이 웹을 통해 다양한 데이터를 공유하고 있으며, 이러한 추세는 더욱 더 증가할 것이다. 인터넷은 다양한 정보를 네트워크를 통해 사용자에게 실시간으로 제공하고 있으며, 통신기술의 발전, 네트워크 통합화 속에서 그 내용과 방식이 더욱 다양해지고 있다. GIS분야에 있어서도 인터넷 네트워크를 이용하여 분산되어 있는 많은 조직이나 사용자들에게 그 기능과 서비스를 제공하는 추세로 바뀌고 있다. Internet GIS는 원격 지리정보 데이타에 대한 접근, 전송, 분석 및 GIS를 표현하는 수단으로 인터넷을 이용하는 특별한 GIS 도구이다. Internet GIS는 전통적인 GIS 소프트웨어가 가지고 있는 대부분의 기능은 물론 인터넷 및 그와 관련된 WWW 및 FTP 프로토콜의 장점을 가지는 부가적인 기능들을 포함할 수 있다. 이들 부가적인 기능은 원격 데이타 및 응용 프로그램의 교환, 지역 컴퓨터에 GIS 응용 프로그램 없이 GIS 분석 기능을 수행, 인터넷상에서 상호작용하는 지도 및 데이타를 표현하는 기능들을 포함한다. Internet GIS는 객체지향적이고 상호운영적이며, 분산적이라는 주요한 특징을 갖는다. 인터넷에서 각각의 GIS 데이타 및 기능성은 하나의 객체로서 서로 다른 서버에 위치하며 필요시 조합 또는 통합되어 운영된다. Internet GIS를 이용한 상습침수지구 지역정보 제공사이트를 구축하기 위해서는 인터넷으로 서비스 할 수 있는 인터넷용 상습침수지구 GIS기본도를 구축이 필요하다. 인터넷 서비스를 위한 상습침수지구 기본도는 또 다른 형태의 주제도라고 볼 수 있으며, 이를 구축하기 위해서는 자료변환 및 가공이 필요하다. 즉, 각 상습침수지구에 필요한 지형도는 국립지리원에서 제작된 1:5,000 수치지형도가 있으나 이는 자료가 방대하고 상습침수지구에 필요하지 않은 자료들을 많이 포함하고 있으므로 상습침수지구의 데이터를 인터넷을 통해 서비스하기 위해서는 많은 불필요한 레이어의 삭제, 서비스 속도를 고려한 데이터의 일반화작업, 지도의 축소.확대 등 자료제공 방식에 따른 작업 그리고 가시성을 고려한 심볼 및 색채 디자인 등의 작업이 수반되어야 하며, 이들을 고려한 인터넷용 GIS기본도를 신규 제작한다. 상습침수지구와 관련된 각종 GIS데이타와 각 기관이 보유하고 있는 공공정보 가운데 공간정보와 연계되어야 하는 자료를 인터넷 GIS를 이용하여 효율적으로 관리하기 위해서는 단계별 구축전략이 필요하다. 따라서 본 논문에서는 인터넷 GIS를 이용하여 상습침수구역관련 정보를 검색, 처리 및 분석할 수 있는 상습침수 구역 종합정보화 시스템을 구축토록 하였다.

Liquid Culture of Basidiomycetes on Natural Media (천연물을 이용한 담자균의 균사체 배양)

  • This study was designed to develop functional drink from jujube extract through a simple submerged culture of three basidiomycetes species. The optimum Brix and pH of the jujube extract for culturing the Ganoderma lucidum appeared to be 5 Brix and pH 4. Ten days of culture period produced maximum mycelium. The optimum Brix and pH of the jujube extract for culturing the Coriolus versicolor appeared to be 5 Brix and pH 5. Ten days of culture period produced maximum mycelium. The optimum Brix and pH of the jujube extract for culturing the Phellinus igniarius appeared to be 3 Brix and pH 5. For the maximum mycelial production eighteen days of culture period was required for Phellinus igniarius. The antitumor activity of the polysaccharides extracted from the fermented drinks was demonstrated through the tumor cell line experiments. The $IC_{50}$ values of the jujube drinks fermented with Ganoderma lucidum and Phellinus igniarius against stomach cancer cell line appeared to be one fourth that of the jujube drink which was not fermented with basidiomycetes.

Effect of Chemical and Anti-fogging Agent Treated MA Packaging Film on Freshness Extension of 'Fuji' Apples (산, 염기 처리된 제올라이트와 결로 방지제를 함유한 기능성 MA포장재가 '후지' 사과의 선도유지에 미치는 영향)

  • To investigate the effects of functional MA films (FMA) masterbatched by zeolite powder treated with 1 N HCl, 0.5 N NaCl solution and anti-fogging agent (NA) compared with control on the freshness extension of 'Fuji' apples during storage at $15^{\circ}C$. Preference, weight loss, total ascorbic acid, sugar content, acidity, change of gas composition in package were measured. After 150 days of storage, the weight loss of control (L) apples was 1%, that of apple in FMA film (NA) was 1.5% after 150 days. Total ascorbic acid content of apples in control (L) after 150 days was 2.09 mg%, those of apple in FMA film (NA) was 2.72 mg%. The titratable acidity of apple in FMA film was higher than that in control, while soluble solids content of apples in FMA film was lower than that in control after 150 days. Ethylene gas adsorbability in control was 192.2 ppm and those in FMA film was 141.4 ppm after 150 days. Overall, apples in FMA film was better than that of control. It was verified that apples packed with LLDPE film (control) lasted about 130days, while those with FMA films lasted about 170 days. Also, FMA films treated with anti-fogging agent few different compare to nontreated film, but commerdity on the display in market was considered higher than that of non-treated anti-fogging agent.

Increase of Bioactive Flavonoid Aglycone Extractable from Korean Citrus Peel by Carbohydrate-Hydrol-ysing Enzymes (당 분해효소를 이용한 감귤 Flavonoid 무배당체 함량의 증가)

  • Flavonoid compounds show several biological activities and generally exist in the forms of glycones linking sugar moiety to main structure. Flavonoid glycones such as naringin and hesperidin in korean citrus peel are slower absorbed and consequently less active than their aglycone, naringenin and hesperetin, respectively. Therefare to increase the content of flavonoid aglycone in korean citrus peel, we used commercial carbohydrate-hydrolysing enzymes, AMG 300 L, Pectinex 100 L, and Viscozyme for transforming flavonoid glycones to aglycones. Optimal conditions of enzyme reaction were pH 5.0-7.0, $5\%$ enzyme, and 24-48 hrs. The content of naringenin and hesperetin as flavonoid aglycones in untreated citrus peel is $100\~200\;ng/g$ of dried citrus peel. In case of enzyme-treated citrus peel the content of naringenin and hesperetin increased to $1,539\∼6,674\;ng/g\;and\;1,974\∼8,906\;ng/g$ of dried citrus peel, respectively. Finally the content of flavonoid aglycones could be extracted to 10-80 times. Now enzyme-treated citrus peel may be applied to use for functional food because of its higher flavonoid aglycones as more active compounds.

Quality Improvement of Korean Pickled Cucumber using Cuttle Bone Powder Treated with Acetic Acid (유기산처리 갑오징어갑을 이용한 오이지의 품질개선)

  • Preparation method and characteristics of high quality Korean pickled cucumber made with acetic acid-treated cuttle bone powder (KPCCP) were examined and compared to those of Korean pickled cucumber prepared by traditional methods (control). Optimal concentration of acetic acid-treated cuttle bone powder for KPCCP preparation was revealed as 10%. Salt concentration of KPCCP fermented for 3 days was 4.5%, slightly higher than that of the control, whereas that of KPCCP fermented for over 6 day was about 4.8%, similar to that of the control. The pH, greenness, and hardness of korean pickled cucumber decreased during fermentation, with the control showing a greater extent of decrease than KPCCP Total acidity and alcohol insoluble calcium contents increased during fermentation, with KPCCP showing higher extent of increase. These results indicate that quality of KPCCP is superior to that of Korean pickled cucumber prepared by traditional methods.

Preparation and Characteristics of Soybean Curd Using Cuttle Bone Powder Treated with Acetic Acid (유기산처리 갑오징어갑 분말을 이용한 두부의 제조 및 특성)

  • Soybean curd was prepared using cuttlebone powder treated with acetic acid (ATC), and its characteristics were determined. Soybean protein was coagulated by adding calcium chloride, magnesium chloride, and ATC at 0.7, 1.2, and 0.9%, respectively, or higher. Yield, white index, texture, and sensory evaluations revealed the optimal concentration of ATC for preparation of soybean curd was 2.0% of total soybean weight. The proximate compositions of soybean curd prepared by adding 2.O% ATC were 75.9%, 12.6%, 1.9%, 5.3%, 6.60, and $1.0×10^4$ CFU/g in for moisture, crude protein, crude ash, crude lipid, pH, and viable cell count, respectively. Although white index and mineral content of both soybean curds were similar, texture of ATC-prepared soybean curd was superior to that of soybean curd prepared with 1.5% calcium chloride.

곶감의 고품질화 기술개발

  • 감 과실(Diospyros kaki folium)은 아열대로부터 온대에 이르는 넓은 지역에서 재배되고 우리나라에서는 고래로부터 중요한 과실의 하나이다. 감은 예로부터 화상, 동상, 해독 및 지혈 등의 치료에 널리 이용되어져 왔으며 특히 곶감꼭지는 시체(枾滯)라 하여 달여서 먹으면 딸꾹질을 멎게 하는데 특효가 있다고 알려져 있다. $\ulcorner$본초비효$\lrcorner$ 에서는 생감을 깍아 말린 곶감은 숙혈을 없애고, 폐열, 혈토, 구역질, 장풍과 치질을 다스리는데 쓰인다고 하였다. 또한 감은 비타민 C의 함량이 많아 감을 먹으면 감기 예방에 좋다고 알려져 왔으며 주로 생식용으로 이용되어져 왔으나 최근 감을 이용한 다양한 가공제품 및 새로운 기능성 식품의 개발을 위한 연구가 활발히 이루어지고 있다. 감의 국내 총 생산량의 약60% 이상이 영남 지방에서 집중되어 재배 생산되고 잇으며 감과실의 비중은 다른 어느 지방보다 크다고 하겠다. 감은 단감과 떫은감이 있는데 단감은 주로 생과로 이용되고 있으며 떫은감은 떫은 맛을 가지기 때문에 생과로서 이용하는데 어려움이 있다. 상주는 백미, 누애고치, 곶감 등 희게 보이는 특산물이 유명한 삼백(三白)의 고장으로 미곡 생산량 전국 9위, 떫은감 2위, 곶감은 저눅 1위, 배는 2위, 사과 5위, 포도 4위를 점유하는 농업의 비중이 매우 높은 지역으로 농산물 시장 개방 확대와 과잉생산에 따른 가격하락의 피해를 예상된다. 종래 곶감의 위생적 문제점을 보완하고 공장 규모로 생산된, 품질이 규격화되고 유통기한이 설정된 고품질 전통 곶감의 제조 및 이에 따른 기술개발이 시급히 요구된다.idin C-1-3'-3" -3/'S _0-trigallate 즉, gallate가 붙은 호합물에서100rM의 농도에서 66%와 63%의 강한 저해효과를 나타났다. Tyrosinase 저해활성을 측정한 결과는pocyanidin C-1-3'-3" -3.'S _0-trigallate는 100rM에서 70%의 강한 저해효과를 나타냈으며,epigallocatechin-(4$\beta$$\longrightarrow$8)-epigallo-catechin-(4$\beta$$\longrightarrow$8)-catechin는 51%의 저해효과를 나타내었다. 산업적응용을 위해 분획한 폴리페놀군은 미백효과 검증실험인 tyrosinase 저해율 측정평가에서 폴리페놀 함량이 가장 높은 Fraction 111의 경우 Sooppm에서 74.2%의 높은 저해율을 나타내었다. 항산화력 실험에서는500pw1이상에서 강한 활성능을 보인 SOD 유사활성능을 제외한 나머지 DPPH와 xanthine oxidase 저해효과에서는 Fraction II와III 모두가50ppm이상에서 80% 이상의 높은 유리라디칼 소거능력을 나타내었다. 그리고 각 Fraction별 항균력 측정 결과 Fraction 르와 111이 우수하게 나타났고 항균활성은 그람음성균보다 그람양성균에서 효과적이었으며, 농도별 항균력시험 결과 농도가 증가할수록 비례하여 저해율도 증가함을 알 수 있었다. 첨가농도를 달리하여 미생물의 생육도를 측정한 결과, fraction II磎꼭\ulcorner경우 그람양성균에 대해 500 ppm 이상에서 뚜렷한 증식억제효과를 나타내었다.
