Characteristics of Fermentation and Wine Quality (시판 와인효모의 발효 특성과 포도주 품질)

  • This study investigated the effect of 12 different commercial yeast strains on the characteristics of fermentation and wine quality. All yeast strains had more rapid fermentation at higher temperatures. Wines fermented with the AR2, EC-1118, Premier Cuvee, and RC212 strains had faster sedimentation rates than wines fermented with the other strains. Wines fermented with EC-1118, Montrachet, and Primeur had low titratable acidity whereas wines fermented with D47 and VR5 had high titratable acidity. There was a correlation (r = 0.826) between tannin content and wine redness. Wines fermented with Fermivin, Montrachet, Primeur, VR5, Noble, and Merit strains produced lower levels of sulfur dioxide during fermentation. Wines fermented with D47, K1V-1116, AR2, and VR5 had high concentrations of glycerol, a compound known to add to "mouth feel". Wines fermented with the Fermivin, Montrachet, and Noble strains had lower concentrations of volatile acids than wines fermented with the other strains.

Economic Evaluation with Uncertainty Analysis of Glycerol Steam Reforming for the H2 Production Capacity of 300 m3 h-1 (수소 생산 규모 300 m3 h-1급 글리세롤 수증기 개질반응에 대한 경제적 불확실성 분석)

  • In this paper, an economic evaluation with the uncertainty analysis using a Monte-Carlo simulation method was performed for the glycerol steam reforming to produce $H_2$ at a capacity of $300m^3h^{-1}$. Fluctuations in a unit $H_2$ production cost were identified based on the variation of key economic factors at ${\pm}10-{\pm}40%$ and the probability of 30.9% was obtained for a previously reported unit $H_2$ production cost of 5.10 $ $kgH{_2}^{-1}$. In addition, fluctuations in the B/C ratio were obtained by varying the fixed capital investment (${\pm}20%$), cost of manufacturing (${\pm}20%$), revenue (${\pm}20%$), and discount rate (2-10%) and the probability ranging from 17 to 55% was observed to meet a minimum B/C ratio of 1 for the economic feasibility of the glycerol steam reforming to produce $H_2$.

Effect of Various Additives on the Production of Recombinant HBsAg during Methanol Induction in Pichia pastoris (Pichia pastoris에서 메탄올 유도시 첨가물이 재조합 HBsAg 생산에 미치는 영향)

  • Methanol induction conditions with various additives for the enhanced production of recombinant hepatitis B surface antigen(HBsAg) were investigated in Pichia pastoris, which can utilize methanol as a carbon source and produce recombinant proteins under the control of strong, tightly-regulated alcohol oxidase(AOX) promoter. The presence of non-methanol carbon sources such as glycerol and glucose fully repressed the expression of AOX promoter. Various additives were tested to improve the production of recombinant protein and it was found that sorbitol could be a good carbon source during methanol induction period. An optimized concentration of amino acid mixture enhanced the production of HBsAg significantly. Pluronic F-68, a non-ionic surfactant, also improved the production of HBsAg without inhibiting cell growth. Addition of oleic acid at 0.01%(v/v) during the induction period showed positive effect on the production of HBsAg. Finally, 1.2%(v/v) of trace salts enhanced the production of HBsAg 1.9 times compared to that of control culture.

Polyvinylchloride Plasticized with Acetylated Monoglycerides Derived from Plant Oil (아세틸화 모노글리세라이드계 가소제 합성 및 PVC 가소성능에 관한 연구)

  • To replace phthalate plasticizer for PVC, acetylated monoglyceride (AMG) plasticizers were prepared from plant oil and their plasticization effects were also investigated. Transesterification of coconut oil by glycerol followed by acetylation with acetic anhydride gave AMG-CoCo (Coco : Coconut Oil). In addition, AMG-GMO (GMO : Glycerol monooleate) and AMG-GMO-Epoxy were synthesized by acetylation and epoxidation with glycerol monooleate. It was found that the thermal stability of AMG plasticizers increased in the following order: AMG-GMO-Epoxy > AMG-GMO > AMG-CoCo and all three plasticizers were thermally more stable than those of common petroleum-based plasticizer DOP (Dioctyl phthalate). The tensile strain values of the PVC containing AMG compounds were ca. 770~810%, while tensile strength values were ca. 19~22 MPa, which were higher than those of PVC containing DOP. DMA (Dynamic Mechanical Analysis) results showed that the miscibility of AMG-GMO-Epoxy in PVC was excellent and the $T_g$ of PVC containing AMG-GMO-Epoxy at 50 phr decreased down to $24^{\circ}C$. Finally, the leaching experiment result showed that the weight loss values of PVC containing AMG-GMO and AMG-GMO-Epoxy at 50 phr were as low as 2 and 1%, respectively, indicating that they have high water migration resistance. The above findings suggested that AMG-GMO-Epoxy could be one of plant oil-based PVC plasticizers to replace DOP.

Effects of Temperature on Diacylglycerol Production by Enzymatic Soli-Phase Glycerolysis of Hydrogenated Beef Tallow (온도조절이 고상계에 경화우지로부터 디글리세리드의 효소적생산에 미치는 영향)

  • Diglyceride was prepared by reaction of hydrogenated beef tallow and glycerol (GL) in the presence of a Pseudomonas lipase. Both substrates were mixed at the ratio of GL/Triglyceride of 0.5 which is the stoichiometric molar ratio for the complete conversion of triglyceride (TG) to diglyceride (DG). DG can be obtained by solid phase-glycerolysis of hydrogenated beef tallow without use of organic solvents or emulsifiers by careful control of reaction temperature. Optimized reaction temperature condition was as follows: An initial incubation at$60^{\circ}C$ for 2h followed by the first temperature shift down to $55^{\circ}C$ for 4h, and then the second shift down to $50^{\circ}C$ for up to 3 days. There was a large decrease in the content of TG during the first $60^{\circ}C$ incubation for 2h. Even a prolonged incubation at $60^{\circ}C$ could not make a change of the composition of the reaction mixture at liquid state. By controlling the temperature lower than $60^{\circ}C$, reaction mixtures were solidified. The reaction temperature at $50^{\circ}C$ below the melting temperature of hydrogenaed beef tallow gave an 71% optimum yield of DG after 72h enzymatic glycerolysis and about 73% of total DG was 1,3-DG.

Effects of Chitosan on the Induction of Renal Dipeptidase (RDPase) from the Proximal Tubules (신장의 근위세뇨관에서 Renal Dipeptidase(RDPase)의 유도에 관한 키토산의 효과)

  • The purpose of this study was to evaluate the effects of chitosan, which is deacetylated derivative of chitin, on the renal function. Renal dipeptidase (RDPase, membrane dipeptidase, dehydropeptidase 1, EC 3.4.13.19) is glycosyl phosphatidyl-inositol (GPI)-anchored ectoenzyme of renal proximal tubular microvilli and was related with renal disease including acute renal failure, pyelitis and nephritis. The released RDPase and Udpase activities were assayed by modified fluorometric method. In vitro experimental groups were consisted of group 1, the concentration ranges of 0, 0.01, 0.05 and $0.1\%$ chitosan only, group 2, the concentration ranges of 1, 2 and 4 mM glycerol only, and group 3, the concentration ranges of 0, 0.01, 0.05 and $0.1\%$ chitosan in the presence of glycerol (4 mM). In vivo experimental groups were consisted of group 1 in which rats were treated with glycerol for the purpose of glycerol-induced renal damage, and group 2 in which rats were treated with chitosan plus glycerol. The RDPase release of 0.01, 0.05, and $0.1\%$ chitosan groups were increased in the concentration dependent manner. The RDPase release of 1, 2, and 4mM glycerol groups were decreased in the concentration dependent manner. Chitosan in the presence of glycerol restored the released RDPase activity in the proximal tubules. In vivo, chitosan inhibited the decrease of RDPase release by glycerol in the kidney and blocked the decrease of Udpase activity by glycerol in urine. These results indicated that chitosan was possible as a functional food to control renal function and its diseases.

Chromophoric Structures of Alkali Lignin (알카리리그닌의 착색구조(着色構造)에 관(關)한 연구(硏究))

  • To investigate the formation of the chromophoric structures taking place during the alkaline pulping vanillyl alcohol [${\alpha}-^{13}C$] guaiacylglycerol-${\beta}$-aryl ether [${\alpha}-^{13}C$ or ${\gamma}-^{13}C$] and phenylcoumarn [${\alpha}-^{13}C$] units as model lignins were treated with 1N sodium hydroxide at 165$^{\circ}C$ for 1.5-3 hours. From the chemical structures of the isolated products and $^{13}C$-NMR Spectra of the reaction mixtures, the main conclusion is as follows; 1) Condensation products of II-1-5 were identified from the reaction mixture of vanillyl alcohol treated with alkali and theses compounds afforded the quinonmethide structure(Fig. 3-7) by air oxidation. 2) Treatment of guaiacylglycerol-${\beta}$-aryl ether unit gave ${\varphi}$-aryl-${\beta}$-aroxy quinone structures (IV-15, IV-16), diguaiacyl-1, 4-penta-diene ${\beta}$, ${\beta}$'-diaroxyl distyrene methane unit, ${\beta}$-aroxy distyrene methane. These distyrene methanes of the compounds are transformed by air oxidation into the corresponding o-quinonemethide units (V-8, V-9). 3) On the treatment of phenylcoumaran, the stilbene derivative was formed in quantitative yield and dimerized(VI-11) in preference to oxidation to the corresponding extended quinone structures. The chromophoric structures taken place during the alkaline treatment of the model lignins are thought to be some important types in alkaline pulping on the basis of the reaction mechanism in this experiment.

Stabilization of Nanoemulsion Using PEG-free Surfactant (PEG-free 계면활성제를 사용한 Nanoemulsion의 안정화)

  • Polyethylene glycol (PEG) is widely used in cosmetics as a surfactant, detergent and emulsifier. During the manufacturing process, 1,4-dioxane, which is toxic to humans, can be produced as a by-product by dimerization of ethylene oxide. As consumers' interest in cosmetic ingredients has increased, the need for safe emulsion research without PEG ingredients in the personal care market has increased. With increasing consumer interest in cosmetic ingredients, the need for safer emulsion research without the PEG ingredient in the personal care market has increased. In this study, we aimed to develop and stabilize nanoemulsion formulation without PEG. Response Surface Methodology (RSM) was used to develop optimized nanoemulsion formulations. Surfactant content (2~4%), oil content (4~8%) and polyol content (12~24%) were set as independent variables as a result of preliminary experiments for determining independent variables and ranges. The particle size, zeta potential, turbidity, and polydispersity index of the formulation were measured as response variables. As a result of measurement of the prepared nanoemulsion by FIB (Focused ion beam), spherical particles were found to have a size distribution of 100 to 200 nm. The stability of each formulation was evaluated for 30 days at each temperature ($4^{\circ}C$, $25^{\circ}C$, and $45^{\circ}C$). The optimal formulation considering the optimum particle size, turbidity, polydispersity index and zeta potential was found to be surfactant (2%), oil (8%) and polyol (24%).

The Effect of Paljeong-San Pharmacopuncture Treatment on Glycerol-Induced Acute Renal Failure in Rats (팔정산(八正散) 약침(藥鍼)이 글리세롤로 유발된 흰쥐의 급성 신부전에 미치는 영향)

  • Objective: The primary objective this study was to evaluate the effect of Paljeong-san (PJS) pharmacopuncture treatment on against the glycerol-induced acute renal failure in rats. Methods: Glycerol injection decreased GFR (glomerular filtration rate) and increased urine volume, serum creatinine, BUN level, urine albumine secretion and fractional excretion of Na+, K+. PJS was selected in the basis of invigorating kidney which can eliminate pathogens. Rats were treated with PJS pharmacopuncture on Shin-shu (BL23) and Chon-chu (ST25) point for 3 days, followed by 50% concentration of glycerol injection ($5m{\ell}/kg$ body weight). Results: After the 3 days treatment period, Paljeong-san (PJS) pharmacopuncture treatment improved renal function. In addition, Glycerol injection increased lipid peroxidation, and decreased Na-K-ATPase in renal cortex and which were prevented by PJS treatment. Conclusion: This study suggests that Paljeong-san (PJS) pharmacopuncture treatment show favorable effect on glycerol-induced acute renal failure in rats.

Mechanical Properties and Morphology of Pectin/Starch Blend Films (펙틴/전분 블렌드 필름의 기계적 물성 및 형태학)

  • Mechanical properties and morphology of pectin/starch blend films depending upon the composition and plasticizer ratio were studied. Blends were prepared continuously in a twin-screw extruder. Films were prepared using a single-screw extruder with slit die. Most of the blends showed a good thermoplastic behavior, while the blends with very high ratio of pectin/starch had a poor thermoplasticity. Tensile properties of blend films were changed significantly by the amount of glycerol and relatively little by the ratio of pectin/starch. Differential scanning calorimetry(DSC) curves of wet blends showed a transition similar to glass transition temperature(Tg) at about $125^{\circ}C$. It was found by Scanning electron microscopy(SEM) study that there exists a good interfacial adhesion between pectin and starch.
