Quality Properties of Pork Fed with Glucosamine Derivatives (GD) as Dietary Supplementation (글루코사민 유도체(GD) 급여 돈육의 육질 특성)

  • The feeding group, composed of weaning pigs fed 3 mL of glucosamine derivatives, was compared for the carcass and quality characteristics with the unfeeding control group for 25∼70 days since they were born. The results were as follows; There were no significant differences in carcass weight and back-fat thickness between the feeding group and the unfeeding group although the feeding group had low ranges of standard error when compared to the control. Feeding group had higher incidence frequencies of A grade (42.7%) than the control (29.2%). The feeding group and unfeeding group had no significant differences in meat color, cooking loss, WBS, pH, WHC and purge loss. Results from this study showed that feeding GD had effect on the decrease the market weight and production of consist carcass weight, however, the feeding GD had no effect on pork quality during rearing times after weaning.

  • 약 먹는 것을 좋아하는 김간난 할머니 (72세, 중구 신당동)는 하루에 먹어야 하는 약만 해도 몇 가지나 된다. 당뇨약, 고혈압약, 고지혈증약, 관절염약을 복용하고 있으며 그 외에도 글루코사민, 종합 영양제, 홍삼 액기스도 챙겨먹고 있다. 양로원이나 친구를 만날 때면 박카스 등 각종 드링크제도 자주 마시고 있다. 그러나 이 아플 때만 약을 먹고 약 먹는 것을 잊어버릴 때도 많아 꾸준하게 복용하고 있지는 않다.

  • 키틴은 N-아세틸-D-글루코사민 잔기가 다수 beta-(1, 4) 결합하고 있는 다당류이고, 그 탈아세틸화물을 키토산이라 한다. 키토산도 D-글루코사민의 beta-(1, 4) 중합체이지만 보통 약간의 아세틸기를 가지고 있는 것을 키토산이라 부르고 있다. 현재로는 제조 키틴의 대부분이 키토산으로 변환되어 응집제로서 폐수처리에 이용되고 있고, 기타 많은 용도가 개발되어 있지만 아직 연구단계에 있다. 따라서 키틴이나 키토산 분자의 잠재적 기능을 개발하여 인간생활에 유효하게 이용하도록 한다는 것은 폐기물 처리나 미이용 지원의 개발이라는 측면에서 볼 때 중요한 문제라고 볼 수 있다. 따라서 본고에서는 키틴의 생물계에서의 분포, 입체배좌, 제조법, 물성 및 기능성 그리고 그 용도에 대해 간추렸다.

  • Recombinant Corynebacterium glutamicum producing N-acetylglucosamine (GlcNAc) was constructed by metabolic engineering. To construct a basal strain producing GlcNAc, the genes nagA, nagB, and nanE encoding N-acetylglucosamine-6-phosphate deacetylase, glucosamine-6-phosphate deaminase, and N-acetylmannosamine-6-phosphate epimerase, respectively, were sequentially deleted from C. glutamicum ATCC 13032, yielding strain KG208. In addition, the genes glmS and gna1 encoding glucosamine-6-phosphate synthase and glucosamine-6-phosphate N-acetyltransferase, which originated from C. glutamicum and Saccharomyces cerevisiae, respectively, were expressed in several expression vectors. Among several combinations of glmS and gna1 expression, recombinant cells expressing glmS and gna1 under control of the ilvC promoter produced 1.77 g/l of GlcNAc and 0.63 g/l of glucosamine in flask cultures.

  • Recently, westernized diet and lifestyles have led to obesity and various adult diseases resulting in a negative influence on the quality of life. There has been an increased interest in choosing proper diet and regular exercise in order to lead a healthy life. The number of people looking for dietary supplements has increased steadily. Dietary supplements are products intended to help maintain or improve the health of consumers. However, if customers take dietary supplements excessively, they may be harmful due to side effects, misuse, abuse and overdose. I performed a toxicologic review of the dietary supplements, glucosamine and chitosan, which are widely used in the country in order to provide the proper understanding of safety of these products.

Effect of Antimicrobial Activity of the Glutaraldehyde Cross-linked Glucosamine (글루코사민-글루타르알데히드 가교결합체의 항균 효과)

  • This study was investigated the antimicrobial activity of glutaraldehyde cross-linked glucosamine. Glutaraldehyde was used as a cross-linker which specifically combines an amine-group of molecules. To optimize the mixing ratio of glutaraldehyde and glucosamine, mixing ratio was set up 1:1, 2:1, 3:1 and 0.5:1 in molarity, respectively. The optimum mixing ratio of glucosamine and glutaraldehyde was found to be 3:1 using thin layer chromatography based on the production of complex. Glucosamine-glutaraldehyde cross-linked complex (Ggcc) revealed significant antimicrobial activity toward PWG than F1, both microbial strains were isolated from porcine semen as antibiotics resistance bacteria (ARB). These results clearly demonstrate that Ggcc has potential bactericidal activity toward ARB in porcine semen.

  • The structure of glycan residues attached to glycoproteins can influence the biological activity, stability, and safety of pharmaceutical proteins delivered from transgenic pig milk. The production of therapeutic glycoprotein in transgenic livestock animals is limited, as the glycosylation of mammary gland cells and the production of glycoproteins with the desired homogeneous glycoform remain a challenge. The ${\beta}$-1,3-N-acetylglucosaminylatransferase1 (B3GNT1) gene is an important enzyme that attaches N-acetylglucosamine (GlcNAc) to galactose (Gal) residues for protein glycosylation; however, there is limited information about pig glycosyltransferases. Therefore, we cloned the pig B3GNT1 (pB3GNT1) and investigated its functional properties that could attach N-acetylglucosamine to galactose residue. Using several different primers, a partial pB3GNT1 mRNA sequence containing the full open reading frame (ORF) was isolated from liver tissue. The ORF of pB3GNT1 contained 1,248 nucleotides and encoded 415 amino acid residues. Organ-dependent expression of the pB3GNT1 gene was confirmed in various organs from adult and juvenile pigs. The pB3GNT1 mRNA expression level was high in the muscles of the heart and small intestine but was lower in the lungs. For functional characterization of pB3GNT1, we established a stable expression of the pB3GNT1 gene in the porcine kidney cell line (PK-15). As a result, it was suggested that the glycosylation pattern of pB3GNT1 expression in PK-15 cells did not affect the total sialic acid level but increased the poly N-acetyllactosamine level. The results of this study can be used to produce glycoproteins with improved properties and therapeutic potential for the generation of desired glycosylation using transgenic pigs as bioreactors.
