The Taxonomic Consideration of Leaf Epidermal Microsturcture in Glechoma L. (Nepetinae, Lamiaceae) (긴병꽃풀속(Glechoma L., 꿀풀과)의 잎표피 미세구조에 대한 분류학적 검토)

  • A comparative micromorphological study was examined on the leaves of the genus Glechoma and related genera (Nepetinae, Lamiaceae) by scanning electron microscopy (SEM) in order to evaluate their significance in the taxonomy. The leaves of taxa Marmoritis, Nepeta sect. Glechomanthe, G. hederacea var. longituba (Korea) are revealed amphistomatic type, while the remnants of taxa had hypostomatic type. The size range of the guard cells is $12.50-28.75{\times}9.17-21.25{\mu}m$: the smallest one was found in M. pharicus ($12.50-15.83{\times}9.17-11.25{\mu}m$), while the largest one was measured to G. hederacea var. longituba (Korea: $28.75-28.88{\times}21.25-21.38{\mu}m$). The stomatal type of genera Agastache, Dracocephalum was mostly diacytic, however for the rest rarely together with anisocytic and anomocytic, except G. hederaca var. longituba (Korea), Meehania urticifolia by having combined with diacytic and anomocytic. The shapes of epidermal cells are differ from in abaxial and adaxial side, and dived with two types (e.g., platelet, stripe pattern). Five types (three glandular, two non-glandular hairs) of trichomes are distributed in leaves. Among trichomes, long and stalk capitates glandular trichome, subsessile glands are different from studied taxa so that leaf micromorphological characters are significance features in the taxonomy.

A Molecular Genetic Analysis of the Introduced Wild Boar Species (Sus scrofa coreanus) on Mount Halla, Jeju Island, Korea (제주도 한라산에 서식하는 도입종 야생멧돼지에 대한 분자유전학적 분석)

  • An wild boar species which has been known as an extinct species on Jeju Island, was recently observed in the surrounding areas of Mount Halla. Based on the molecular techniques, this study examines whether they are crossbred with domesticated pig breeds. Intraspecific genetic relationships with other wild boar populations and molecular sexing were examined as well. Total of four molecular markers on mitochondrial DNA(control region and ND2) and nuclear DNA(MC1R and KIT) were applied to test crossbreeding between with domesticated pig breeds, such as Landrace, Large White, Berkshire, Hampshire, and Duroc. All individuals of wild boar population had identical mtDNA control region(CR) sequences. In addition, the sequences were the same as those of some native pig breeds which are distributed in Northeast China, but different from those previously reported from the Korean Peninsula up to date. These results suggest that this population may have originated from a genetic lineage had been not previously studied and genetically related to Chinese native pig breeds. Molecular sexing results show that there are twice as many females as male. Thus the population is under expansion and its size will dynamically increase if not controlled.

Phylogenetic classification of Korean vascular flora according to the recent APG classification system (APG 분류체계에 따른 한국 관속식물상의 계통학적 분류)

  • A recently published Korean Flora, "The genera of vascular plants of Korea (GFK)", includes the descriptions and keys for 217 families, 1,044 genera, and 3,209 species of Korean vascular plants. We reclassified these taxa according to the recent APG classification system, which resulted in 64 orders, 204 families, 1,044 genera and 3,209 species. Twenty-two families from the GFK were abandoned because of changes to the familial delimitations in the APG system. In contrast, the number of families in the Liliaceous group was increased. The Liliaceae in the GFK included 31 genera and 109 species. These taxa are now assigned to 10 families in four different orders including Liliales, Asparagales, Alismatales, and Dioscoreales because of the drastic changes to the monocot classification system in the past 20 years. In addition, the family name of the Aucubaceae was changed to Garryaceae. As a result, the number of families in the GFK has been reduced to 204. The results were summarized in four tables and two figures at the levels of unofficial higher taxonomic hierarchies, orders, families and genera. This new information can provide a guidelines for selecting the phylogenetic analysis unit for the Korean tree of life (KTOL) project. Futhermore, the updated classification system also provides an important summarization for the systematic community for placing the Korean flora in a modern phylogenetic context.

Phylogenetic study of the fern genus Hypodematium (Hypodematiaceae), focusing on Korean native taxa (한국산 금털고사리속의 계통분류학적 연구)

  • In Korea, Hypodematium glanduloso-pilosum was formerly known as the only Korean native species in the genus. Recently, however, we reported one unrecorded taxon, H. squamuloso-pilosum Ching, which was found on rocks at a limestone mountain in Yeongwol, along with one new taxon, H. angustifolium in Okcheon. Traditionally, Hypodematium taxa are often distinguished from each other by vegetative characters such as pinnatifid lamina, glandular hairs, and narrower or linear lanceolate scales on stipe bases. H. glanduloso-pilosum, distributed widely throughout the country except on Jejudo Island in Korea, exhibiting variations in leaf segregation, indusia positions, hair distributions and size. The high variation in the morphological characters in the widely distributed taxon has caused problems delimitating three native species from each other. To evaluate the phylogenetic relationships among H. glanduloso-pilosum and taxa related to Hypodematium (all Korean native taxa), we carried out morphological and molecular analyses (cpDNA rbcL and psbA-trnH) of populations of the genus Hypodematium in Korea. Although H. glanduloso-pilosum exhibits high variations in some morphological characters, the species is characterized by stipes and indusia with densely multicellular hairs and rod-shaped glandular hairs or hairs and lanceolate or oblong lanceolate scales in rhizomes and stipe bases distinguished from those of other Korean native taxa (H. squamuloso-pilosum and H. angustifolium). In the analyses of cpDNA data, three Korean native taxa are placed in the same clade, i.e., in the glanduloso-pilosum group. Moreover, our analyses propose that H. squamuloso-pilosum (China and Korea), H. angustifolium (Korea), and H. fordii (China and Japan) share the same glanduloso-pilosum clade with H. glandulosopilosum (China, Korea, and Japan).

In Vitro Tissue Culture Frequency and Transformation of Various Cultivars of Soybean (Glycine max (L.) Merr.) (다양한 콩 자원들의 기내 조직배양 효율 및 형질전환)

  • Efficient in vitro regeneration system is essential for the successful crop breeding of soybean (Glycine max (L.) Merr.) using the new biotechnology. The genotype of donor plants strongly influences the establishment of tissue culture system. Therefore, the screening of genotypes with excellent tissue culture ability is very important for soybean genetic improvement. In this study, we report the tissue culture efficiency of 21 soybean cultivars belong to Korean soybean core-collection and two foreign cultivars (Jack and Maverick). The Kwangan, Anpyeong and Seonam are share close genetic relationship in 21 cultivars and these three cultivars were observed the high frequency of germination and regeneration. Furthermore, the high tissue culture abilities were also observed in the Williams 82 used in reference genome sequencing and the two foreign cultivars. The transformation of pBAtc:tRNA with bar gene was performed by Agrobacterium tumefaciens in the cultivars with high tissue culture ability. Transformation of the bar gene was identified by PCR analysis in Kwangan, Pungwon, Seonam, and Maverick. Our results provide useful information for the breeding of various soybean cultivars by plant biotechnology such as, genome editing.

  • The germination characteristics of the resting cysts of Pheopolykrikos hartmannii collected from the southern coastal sediments of Korea were studied at different temperature conditions, and the morphology and phylogeny of the germlings were examined. The resting cysts of Ph. hartmannii were round and characterized by a red accumulation body and many arrow-like spines and could germinate at temperature of 10 to 30℃. High germination rates (>90%) were observed at 15 and 20℃, indicating that the resting cysts could act as seed populations for the bloom initiation of Ph. hartmannii in Korean coastal waters in early summer or early fall. The morphology of the germlings was generally consistent with the previous description, and an apical groove characterized by a fully enclosed loop was observed. Phylogenetic analysis based on large SubUnit (LSU) rRNA gene sequences revealed that the germlings shared an identical sequence with the Korean and American isolates of Ph. hartmannii and was a sister clade of Polykrikos species.

Application of Molecular Biological Technique for Development of Stability Indicator in Uncontrolled Landfill (불량매립지 안정화 지표 개발을 위한 분자생물학적 기술의 적용)

  • This study was conducted for developing the stability parameter in uncontrolled landfill by using a biomolecular investigation on the microbial community growing through leachate plume. Landfill J(which is in Cheonan) and landfill T(which is in Wonju) were chosen for this study among a total of 244 closed uncontrolled landfills. It addressed the genetic diversity of the microbial community in the leachate by 165 rDNA gene cloning using PCR and compared quantitative analysis of denitrifiers and methanotrophs with the conventional water quality parameters. From the BLAST search, genes of 47.6% in landfill J, and 32.5% in landfill T, respectively, showed more than 97% of the similarity where Proteobacteria phylum was most significantly observed. It showed that the numbers of denitrification genes, i.e. nirS gene and cnorB gene in the J site are 7 and 4 times higher than those in T site, which is well reflecting from a difference of site closure showing 7 and 13 years after being closed, respectively. In addition, the quantitative analysis on methane formation gene showed that J1 spot immediately bordering with the sources has the greatest number of methane formation bacteria, and it was decreased rapidly according to distribute toward the outer boundary of landfill. The comparative investigation between the number of genes, i.e. nirS gene, cnorB gene and MCR gene, md the conventional monitoring parameters, i.e. TOC, $NH_3-N,\;NO_3-N,\;NO_2-N,\;Cl^-$, alkalinity, addressed that more than 99% of the correlation was observed except for the $NO_3-N$. It was concluded that biomolecular investigation was well consistent with the conventional monitoring parameters to interpret their influences and stability made by leachate plume formed in downgradient around the uncontrolled sites.

Classification of Cordyceps spp. by Morphological Characteristics and Protein Banding Pattern (동충하초(冬蟲夏草)(Cordyceps) 속균의 형태적인 특징과 단백질 Pattern에 의한 계통 분류)

  • Ten species of Cordyceps species were collected throughout Kangwon province including Chuncheon Dongsanmyun KNU forest experiment from June to September, 1993. Collected Cordyceps species were identified as Cordyceps militaris, C. roseostromata, C. kyushuensis, C. scarabaeicola, Phytocordyceps ninchukiospora, C. nutans, Paecilomyces tenuipes, C. sphecocephala, Hymenostilbe odonatae, Torrubiella sp.. C. militaris, type species of Cordyceps species, was mainly formed on pupae of Lepidoptera and found after the rainy season around July. Fruiting body of C. roseostromata was morphologically similar to those of C. militaris, but relatively small in size and they were also found on lawn or pupa of Lepidoptera. Fruiting body of C. scarabaeicola was found on adult Scarabaeidae specifically and collect fruiting bodies of C. kyushuensis were on larva of moth. C. nutans and C. sphecocephala had host specificity on Hemiptera and Hymenoptera, respectively. Each species formed elliptical fertile part attach to the slim and carneous stalk and they were collected the most in specimen number through whole season of the summer. Ascospore of Phytocordyceps ninchukiospora on seed was characterized by two viable, multiseptate, fusiform units linked end-to-end by a long, filiform connective. Paecilomyces tenuipes, imperfect stage of the genus Cordyceps is multi-infective fungi that attack all stages of all groups of insects. Hymenostilbe odonatae attacks only adult Odonata and Torrubiella sp. formed on spider was difficult to collect because it was found the back side of leaf. As results of cultural test PDA medium showed the best mycelial growth. In the experiment of effect of the acidity inside of the media, C. militaris was good on pH 5, C. nutans and Phytocordyceps ninchukiospora were good on pH 6 and Paecilomyces tenuipes was on pH 7 and C. scarabaeicola was on pH 9. All isolates tested showed the best mycelial growth at $20^{\circ}C$. Morphologically similar isolates were used to analyze protein banding pattern among and within species. As a result, C. militaris, C. roseostromata and C. kyushuensis were clustered into close species and C. scarabaeicola and Phytocordyceps ninchukiospora were relatively distant from those species.

Culture characteristics of Pleurotus commercial strains at different temperature (계절별 온도에 따른 느타리 품종의 재배적 특성)

  • We determined the genetic relatedness among 81 commercial varieties of oyster mushrooms by either comparison of fruiting body characteristics or using PCR amplification with URP primers. Also, we assessed their temperature adaptation by using box cultivation. Based on our data, some varieties such as Wonhyung and variety 1, Suhan and variety 20, Wang-hukpyung and variety 4, Chunchu #2 and variety 6, Shinong #11 and variety 1, Samgu #1 and variety 2, showed the close genetic relatedness between each other. It is known that mushroom culture room is affected by outer temperature because mushroom need the adequate aeration for their growth. Therefore, in aspect of saving energy, the cultivation of season favorable mushroom species has been recommended. Eight different commercial varieties including Sa-chul, Yeoreum and Bunhong(Pink) grew well at high temperature ($16^{\circ}C$). Also, about 51 different oyster mushroom strains were good at relatively high temperature. However, other 64 different oyster mushroom varieties were grew well at low temperature ($11^{\circ}C$). Fourty-nine varieties were adapted well in most temperatures. The slow growth rate were observed in some varieties including Kun(P. eryngii ), Awi(P. nebrodensis ), and Cheunbok(P. abalonus ), which are not suitable for box cultivation. We believe that the culture characterization of each mushroom varieties should help many farmers in many aspects such as saving energy, quantity improvement and new variety development etc.

Occurrence of Papaya ringspot virus Infecting Cucurbit Crops in Korea (박과작물에 발생하는 파파야원형반점바이러스의 발생 보고)

  • A flexuous rod-shaped virus was isolated from Cucurbita pepo leaves showing as green mosaic and puckering symptoms at Anseong, Korea. Based on the biological analysis, electron microscopy, and reverse transcription-polymerase chain reaction (RT-PCR), the virus isolate was identified as Papaya ringspot virus type watermelon (PRSV-W). From biological analysis, the host range of PRSV-W was limited to the families Cucurbitaceae and Chenopodiaceae. Most susceptible cucurbit species, such as Cucumis lanatus, Cucumis sativus, Cucurbita pepo, and Citrullus lanatus, showed symptoms of green mosaic, malformation, puckering, and narrow laminae by infection with PRSV-W. The local lesion were showed on the inoculated leaves of both Chenopodium amaranticolor and C. quinoa. Field survey of PRSV, Watermelon mosaic virus (WMV) and Zucchini yellow mosaic virus (ZYMV), three major viruses infecting cucurbit, was done during 2001 to 2003 on 173 commercial cucurbit cultivating fields distributed over the three regions of Gyeonggi, Gyeongbuk and Jeonnam Provinces where cucurbits are grown in different environmental conditions and cropping patterns. Typical viral symptoms were observed from 107 cultivating fields, and all three kinds of potyviruses were detected from 206 samples out of the 235 samples using RT-PCR. Watermelon mosaic virus (WMV) and Zucchini yellow mosaic virus (ZYMV) are the most widely distributed viruses in outdoor and retarding-culture fields, at an infection rating of 48 and 33 percents, respectively. PRSV was detected from 12 percent of 235 samples. The nucleotide and amino acid sequences of coat proteins (CP) of eight PRSV isolates, collected from several areas including Anseong, were determined and sequenced heterogeneity among the isolates was performed. The CP gene of PRSV showed 88.6~97.3 percent homology in nucleotide sequences and 95.1~99.3 percent homology in amino acid sequences with other PRSV isolates worldwide. The phylogenetic analysis indicated that the Korean PRSV isolates belong to the southern-east Asian cluster.
