Purification of Oat ${\beta}-Glucan$ by ${\alpha}-Amlyase$ Treatment and Characterization of Its Physicochemical Properties (귀리 ${\beta}-glucan$${\alpha}-amlyase$를 이용한 정제와 이화학적 특성)

  • The effects of purification using ${\alpha}-amlyase$ (Termamyl 120L) on physicochemical properties of ${\beta}-glucan$ from oat bran were studied. Four fractions were selected as fraction A ($55^{\circ}C$, 15%, pH 6), fraction B ($45^{\circ}C$, 15%, pH 6), fraction C ($50^{\circ}C$, 0%, pH 7), and fraction D ($50^{\circ}C$, 10%, pH 5) from the result of physiological test, and three consecutive subfractions were obtained by repeated ${\alpha}-amlyase$ treatments on the each fractions. The contents of ${\beta}-glucan$, protein, and ash after purification were in 81.4-88.2%, 4.1-6.3% and 2.6-6.2%, respectively. The apparent viscosities of purified ${\beta}-glucan$ aqueous solutions were similar to those of hydroxy methyl cellulose. Glucose was a major monosaccharide of ${\beta}-glucan$ extracts, and xylose and arabinose were also detected as minor constituents on TLC. The average molecular weight ranged $2.0{\times}10^6-5.1{\times}10^6$ and was decreased after purification. From the result of the differential scanning calorimetry, the melting point ranged $130-140^{\circ}C$ with purification step and thermal transition enthalpy was increased. The ratio of ${\beta}-(1{\rightarrow}3)\;to\;{\beta}-(1{\rightarrow}4) $ linkages were 1:2.22-1:2.52, and increased up to 1:5.50 after purification.

Effect of Biomass and N Production by Cultivation Methods of Leguminous and Gramineae Green Manures on Rice Growth in Central Regions of Korea (중부지역 답리작에서 두과 및 화본과 녹비작물의 재배방법에 따른 biomass, 질소 함량이 벼 수량에 미치는 영향)

  • The cultivation methods are important for determining crop yield of green manure. The effect of cultivation methods of green manure crops (GMC) on biomass and rice yield was investigated. This experiment was conducted at Sinheung series (fine loamy, mixed, nonacid, mesic family of Fluvaquentic Endoaquepts from Oct. 2007 to Oct. 2008 at the National Institute of Crop Science (NICS), RDA, Suwon, Gyeonggi province, Korea. Seven GMC (hairy vetch, barley, Chinese milk vetch, rye, crimson clover, oats, rattail fescue) were cultivated and incorporated on paddy soil by broadcasting before rice harvesting (BBRH) and partial tillage seeding (PTS). Among the three leguminous GMC, the biomass and N production were the highest at the hairy vetch of PTS. Among the four gramineae GMC, the biomass and N production tended to be higher in the rye of BBRH and barley of PTS. The C/N ratio (56.5~74.2) of rye was high compared with hairy vetch (14.1). Among the GMC, the incorporation of hairy vetch increased $NH_4$-N contents in rice paddy soil at 14 and 42 days after transplanting. These results showed that hairy vetch had no significant to rice yield compared with conventional fertilization. Therefore, hairy vetch seems to be the most efficient green manure crop as an alternatives to chemical N fertilizer in the central regions of Korea.

Differences in Seed Vigor, Early Growth, and Secondary Compounds in Hulled and Dehulled Barley, Malting Barley, and Naked Oat Collected from Various Areas (맥종별 주산지와 재배한계지 수집종자의 활력, 초기생장 및 이차화합물 차이)

  • The purposes of this study were to determine how changes in temperature affect germination rates and growth of hulled and dehulled barley, malting barley, and naked oat plants, and to measure chlorophyll content, photosynthetic efficiency, and secondary compounds (total phenol, total flavonoid, and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity) in plants grown at 13℃ or 25℃). Various types of barley seeds were collected from areas with ideal conditions for barley cultivation, hereinafter referred to as IA, and also from areas where barley cultivation is more difficult due to lower temperatures, hereinafter referred to as LTA. Seeds were tested for seed vigor. While there were significant differences in the electrical conductivity values between seeds collected from certain specific areas, no significant differences were evident between IA and LTA seeds, regardless of the type of barley seed. When plants were grown at 25℃, there were no significant differences in germination rates, plant height, root length and shoot fresh weight between plants originating from IA and LTA. However, there were differences in the measured parameters of some specific seeds. Similarly, under the low temperature condition of 13℃, no differences in the emergence rate, plant height, and shoot fresh weight were evident between plants originating from IA or LTA, regardless of the type of barley. However, there were differences between some specific seeds. One parameter that did vary significantly was the emergence date. Hulled barley and malting barley emerged 5 days after sowing, whereas naked oats emerged 7 days after sowing. There were no differences in the chlorophyll content and photosynthetic efficacy, regardless of the type of barley. There were no significant differences in total phenol, total flavonoid content, and DPPH radical scavenging activity between plants originating from IA and LTA, regardless of the type of barley. However, there were differences between some specific seeds. In particular, for malting barley the total flavonoid content differed in the order of Gangjin > Changwon > Haenam = Jeonju > Naju. The results indicate that crop growth, yield and content of secondary compounds in various types of barley may be affected by climate change.

Physiological Functions of Lactic Acid Bacteria Fermented Broth Containing Fagopyrum esculentum and Saccharina japonica (메밀 및 다시마를 포함하는 유산균 발효액의 생리적 기능)

  • In this study, we investigated the potential of Lactobacillus brevis AR1 fermented broth containing various grains (Fagopyrum esculentum, Scotch oat, Sesamum indicum, Glycine max Merr, Castanea crenata, Oryza sativa L., Hordeum vulgare L., Perilla frutescens var. japonica Hara, or Triticum aestivum L.) or Saccharina japonica as a source of collagen synthesis in cosmetic products. The treatment of Lb. brevis AR1 fermented broths containing F. esculentum or S. japonica water extracts was markedly increased the synthesis of collagen in fibroblasts. The collagen synthesis capacity of the S. japonica fermentation product was higher than that of β-glucan, which was used as a positive control. Under controlled conditions in broths containing F. esculentum or the S. japonica extracts with 4% monosodium glutamate (MSG), Lb. brevis AR1 produced γ-aminobutyric acid (GABA) at a concentration of 180 mM, with an 84.5% GABA conversion rate after 72 h. Both the F. esculentum and S. japonica fermentation broths produced by Lb. brevis AR1 reduced inflammatory responses on mouse skin and did not show cell cytotoxicity in fibroblasts. These results suggest that both the F. esculentum and S. japonica fermentation products of Lb. brevis AR1 could be used as functional materials in cosmetic products to combat wrinkles and skin inflammation.

An Ecological Study on the Vegetation of Bijin and Yongcho Islets (비진도와 용초도의 식생에 관한 생태학적 연구)

  • The vegetation of Bijin and Yongcho Islets was investigated from February 1, 2000 to March 30, 2002, and forest structure was constructed. Vascular plants of this area consisted of 12 forms, 59 varieties, 476 species, 352 genera, 112 families and 37 orders. Among them, 37 species (6.8% out of the total 547 taxa) of evergreen broad leaved trees, 36 species (6.6% out of the total 547 taxa and 19.7% out of the total naturalized plants in Korea) of naturalized plants, 6 species of endemic plants and 41 species of cultivated plants were observed. Rare and endangered plants include 1 taxa: Crypsinus hastatus ('98-4). The vegetation in this study area was classified into 9 communities (Pinus thunbergii, Castanopsis cuspidata var thunbergii Camellia japonica, Neolitsea sericea, Machilus thunbergii, Quercus variabilis, Carpinus coreana, Platycarya strobilacea, Miscanthus sinensis var. purpurascens) and 1 afforestation (Alnus firma).

Effect of Temperature, Solvent Concentration, and pH on the β-Glucan Extraction (β-Glucan 추출에 미치는 온도, 용매 농도 및 pH의 영향)

  • This study investigated the effects of temperature, solvent concentration, and pH on the ${\beta}$-glucan extraction. Oat bran ${\beta}$-glucan was extracted with different extraction conditions, using various combinations of experiment factors, such as temperature (40, 45, 50, 55, and $60^{\circ}C$), ethanol concentration (0, 5, 10, 15, and 20%), and pH (5, 6, 7, 8, and 9). Under the various extraction conditions, ${\beta}$-glucan extraction rate and overall mass transfer coefficient of oat bran ${\beta}$-glucan, and viscosity of oat bran extracts were investigated. As increasing the extraction time, the extraction rate of ${\beta}$-glucan increased. The overall mass transfer coefficient of ${\beta}$-glucan ranged from $3.36{\times}10^{-6}$ to $8.55{\times}10^{-6}cm/min$, indicating the lowest at the extraction condition of $45^{\circ}C$, 15% and pH 8, and the highest at $50^{\circ}C$, 0% and pH 7. It was significantly greater with increasing extraction temperature and decreasing ethanol concentrations of extraction solvent, except for solvent pH. There were positive correlations among the overall mass transfer coefficient, the extraction rate of ${\beta}$-glucan, and the viscosity of extract.

Protective Effect of Various Grain Methanolic Extracts against UVB-induced Photo-aging in Human Skin Fibroblasts (인체 피부 섬유아세포에서 자외선 조사에 대한 다양한 곡류 메탄올 추출물의 보호 효과)

  • In the present study, we investigated the protective effect of various grain methanolic extracts against UVB-induced photo-aging in human skin fibroblasts. Various grain methanolic extracts were evaluated for their antioxidant compounds and activities. 2,2-Ddiphenyl-1-picryhydrazyl radical (DPPH) and ABTS 2,2-azino-bris-(3-ethylbenzoth iazoline-6-sulphonic acid) radical cation scavenging activities have been used to measure the relative antioxidant activities of extracts from grains. The content of total polyphenolics in the extracts were evaluated using spectrophotometric methods. Human skin fibroblast (Hs68) cells were pretreated with various grain methanolic extracts ($25{\mu}g/mL$). Skin toxicity was simulated by exposing the cells to UVB ($30mJ/cm^2$) irradiation. In response to the UVB-irradiation, an increased amount of matrix metallo-proteinases (MMPs) release was observed, whereas pretreatment of various grain methanolic extracts significantly inhibited the production of MMP-1 in Hs68 cells. We also found that pretreatment of the extracts significantly decreased UVB-induced reactive oxygen species and significantly increased total collagen content in Hs68 cells. These results provide that grains could be regarded as a potential ingredient in natural cosmetics, used for UVB protection.

Counter-measure for Reducing Residual Effects of Quinclorac to Followed-by Crops of Paddy Rice (답후작(沓後作)의 Quinclorac 잔류피해(殘留被害)에 대한 경감대책(輕減對策) 연구(硏究))

  • For reducing the residual effects of Quinclorac to followed-by crops of paddy rice, three tests by different soil improvers, different cultural performances with tillage and application of composts, and cultivation of different depletion crops were evaluated, respectively. Among seven soil improvers, activated carbon, composts and perlite showed significant promisable feasibility to reduce phytotoxicity of tomato seedlings as affected by Quinclorac residuals, And comparing to untreated check, the application of tillage and composts prior to transplanting of tomato seedling could helpful to reduce Quinclorac residuals in the soil, but tomato and lettuce, most susceptible crops to Quinclorac, were not remarkably protected by those cultural performances. Also among cultivation of depletion crops just after rice harvest, Italian ryegrass, tall fescue, rape and oats engaged to deplete and reduce Quinclorac residuals for tomato seedling growth, however alfalfa and hairy vetch, relatively susceptible plants to Quinclorac were not promisable.

Seasonal Dynamics of Enzymetic Activities and Functional Diversity in Soils under Different Organic Managements (시용 유기물을 달리한 토양에서 미생물 군락의 효소활성과 기능적 다양성의 계절적 변화)

  • Soil microbial activity and diversity are affected by organic sources applied to improve soil quality and fluctuate seasonally. We investigated the effects of municipal compost (MC), poultry litter (PL), and cover crops of spring oats and red clover (RC) on soil enzyme activities, and soil bacterial community-level physiological profiling (CLPP) in a Mexico silt loam in North Central Missouri, USA. Temporal patterns of these parameters were observed by periodic five soil sampling from spring to fall over a two year period. MC increased soil dehydrogenase (DH) activity consistently beginning about three months after MC application; fluorescein diacetate (FDA) hydrolytic activity significantly began to increase by the September of the first year but fluctuated during the following period. DH activity responded more directly to the amount or properties of organic residues in soils while FDA hydrolysis and CLPP were generally influenced by composition of organic sources, and enzyme activities and CLPP showed seasonal variation, which depended on organic sources and soil moisture. MC and cover crops may be useful organic sources for enhancing general soil microbial activity and altering soil microbial diversity, respectively. Because microbial activities and diversity are dynamic and subject to seasonal changes, the effects of organic amendments on these parameters should be investigated frequently during a growing season.

Flora in Ahnshim Wetland, Daegu Metropolitan City (대구광역시 안심습지의 식물상)

  • The purpose of this study was to suggest the raw data on establishing the domestic wetland conservation plan. The results of this study were as follows. The vascular plants were summarized as 201 taxa in Ahnshim wetland; 57 families, 154 genera, 179 species and 22 varieties. The rare and endangered plant designated by Korea Forest Service was Hydorcharis dubia. In the wetland plants, the emerged plants were showed Typha angustata, Beckmannia syzigachne, Zizania latifolia, Phragmites communis, Persicaria thunbergii, Oenanthe javanica and Veronica undulata, and the submerged plants were Potamogeton crispus, Vallisneria asiatica, Hydrilla verticillata, Ceratophyllum demersum and Myriophyllum verticillatum. And the floating leaved plants were confirmed Potamogeton distinctus, Hydrocharis dubia, Nelumbo nucifera and Nymphoides peltata, and the floating plants were Salvinia natans, Spirodela polyrhiza and Lemna paucicostata. The naturalized plants were 33 taxa; Avena fatua, Dactylis glomerata, Rumex crispus, Rumex conglomeratus, Amaranthus retroflexus, Lepidium apetalum, Tlaspi arvense, Descurainia pinnata, Potentilla supina, Vicia villosa, Robinia pseruo-acacia, Amorpha fruticosa, Trifolium repens, Medicago sativa, Ailanthus altissima, Euphorbia supina, Oenothera lamarckiana, Quamoclit angulata, Ipomoea purpurea, Solanum nigrum, Veronica perisca, Helianthus tuberosus, Ambrosia artemisifolia var. elatior, Xanthium strumarium, Erigeron annuus, Erigeron canadensis, Conyza sumatrensis, Senecio vulgaris, Bidens frondosa, Tagetes minuta, Taraxacum officinale, Taraxacum laevigatum and Sonchus oleraceus.
