Mass Spectrometry-Based Strategy for Effective Disulfide Bond Identification (질량분석기를 활용한 효과적 이황화결합 분석법 개발)

  • The determination of disulfide bonds is important for comprehensive understanding of the chemical structure of protein. So far, many strategies for the disulfide bond analysis have been suggested in terms of speed and sensitivity. However, most of these strategies have not considered free thiol residues in the target protein in the process of determining the disulfide bond. We suggested the strategy which was composed of four steps for the identification of disulfide bonds; the first step was the prediction of possible disulfide bonds, the second step was the determination of free cysteine residues, the third step was the analysis of disulfide bond using a high-resolution mass spectrometry, and the final step was the determination of disulfide bonds based on the comprehensive verification. In this study, we performed the characterization of disulfide bonds for the recombinant protein (HRPE1), where 1 and 5 inter- and intra-chain disulfide bonds were identified, respectively.

Effect of Silk in Silk/PLGA Hybrid Films on Attachment and Proliferation of Human Aortic Endothelial Cells (실크/PLGA 하이브리드 필름에서 실크가 인간 대동맥 내피세포의 부착과 증식에 미치는 효과)

  • The vascular endothelial cells are the inner layers of blood vessels. It regulates the function of blood vessels and proliferation of vascular smooth muscle cells. Poly(lactide-co-glycolic acid) (PLGA) is a biodegradable synthetic polymer with a well-controlled degradation rate and an acceptable mechanical strength. It can be easily fabricated into many shapes. Silk consists of 18 amino acids. It found important for attaching cells cultured in vitro, and maintaining cell functions. In this study, we fabricated silk/PLGA biomaterial hybrid films of 0, 10, 20, 40 and 80 wt% silk. We performed MTT, SEM, ELISA, and immunocytochemistry analyses. We confirmed the adhesion and the proliferation of HAECs on silk/PLGA according to the content of silk, and 40 wt% silk/PLGA hybrid films have superior adhesion and proliferation properties. These results demonstrate that silk/PLGA hybrid films provide suitable surfaces for HAECs, and there is the effect of silk on cell growth and proliferation.

  • 한국어는 다른 언어와는 달리 초성, 중성, 종성의 자소가 모여서 한 음절을 이룬다. 음절을 이루는 자소는 그 발생의 확률적 성질에 따라 확률변수로 간주된다. 음절 안에서 자소간의 발생의 상관관계는 자소간 조건부 확률 및 엔트로피로 표시된다. 음절이 모여서 단어를 이루고 단어를 이루는 음절은 그 발생의 확률적 성질에 따라 확률변수로 간주된다. 한국어 단어안에서 음절간의 발생의 상관관계는 음절간 조건부 확률 및 엔트로피로 표시된다. 수 있다. 그런데 가능한 음절의 종류가 매우 많기 때문에 음절 발생의 상관관계를 표시하는 지표로서 음절간 조건부 확률 대신 초성, 중성, 종성 단위의 조건부 확률을 사용하는 것이 음절간의 발생의 상관관계를 표시하는데 효과적이다. 이러한 한국어의 정보이론적 연구를 위하여서는 기초자료로서 한국어 단어의 빈도분포가 필요하다. 한국어 단어의 빈도분포의 포괄적인 조사는 1956년의 "우리말 말수 사용의 잦기 조사"가 유일한 실정이다. 시간 경과에 따른 한국어의 정보이론적 특성 변화의 분석을 위하여서는 한국어 단어 빈도의 주기적인 조사가 필요하다. 한국어에서 초성, 중성, 종성단위의 정보이론적 연구결과는 한국어 음성인식 및 함성, 자연언어처리, 암호법, 언어학, 음성학, 한국어부호 표준화 연구등에 이용될 것으로 기대된다. 남북한의 언어는 분단이 지속됨에 따라 상호 이질화가 진행되고 있다. 이러한 이질화를 극복하려는 부분적인 노력으로 남북한 언어의 한국어 영문표기의 단일화 등이 있었다. 이러한 노력에 병행하여 남한과 북한의 언어에 대한 정보이론적 비교 연구도 있어야 할 것이다. 정보를 효과적으로 캐싱할 수 있도록 인접한 데이터를 클러스터링해서 브로드캐스팅하여 이동 호스트의 구성 시간(setup time)을 최소화하였다. 그리고, 맨하탄거리(Manhattan Distance)를 사용해서 위치 의존 질의에서 사용하는 데이타를 캐싱하고 질의를 처리하는 방법을 제안한다. 맨하탄 거리를 이용해서 캐싱하면 도로에 인접해서 위치한 데이타를 효과적으로 캐싱할 수 있다. 또한, 거리 계산 방법으로 맨하탄 거리를 사용하면 도심에서 실제 이동 거리와 비슷한 값을 알 수 있고, 직선 거리 계산식에 비해서 계산식도 간단하기 때문에 시스템 계산량도 줄일 수 있다. 기준으로 라이신 부산물은 어분 단백질을 40%까지 대체가 가능하였으며, 아울러 높은 라이신 부산물의 대체 수준에 있어서 사료효율과 단백질 전환효율을 고려한다면 아미노산 첨가(라이신과 아르지닌)와 중화 효과에 좋은 결과가 있을 것으로 사료된다.의한 적정 양성수용밀도는 각고 5~6cm 크기의 경우 10~15개체가 적합하였다. 수증별 성장은 15~20 m 수층에서 빨랐으며, 성장촉진과 폐사를 줄이기 위해서는 고수온이 지속되는 7~10월에는 20~30m수층으로 채롱을 내려 양성하고 그 외 시기에는 15 m층 내외가 좋은 것으로 나타났다. 상품으로 출하 가능한 크기 인 각고 10 cm이상, 전중량 140 g 내외로 성장시 키기까지는 채묘후 22개월이 소요되었고, 출하시기는 전중량 증가가 최대에 이르는 3월에서 4월 중순이 경제적일 것으로 판단된다.er 90 % of good relative dynamic modulus of elasticity due

Isolation and identification of soycurd forming lactic acid bacteria which produce GABA from kimchi (김치로부터 GABA를 생산하는 커드 형성 젖산균의 분리 및 동정)

  • Gamma amino butyric acid (GABA), known as a non-protein amino acid and major inhibitory neurotransmitter in the brain, has several functional properties such as neurotransmission, induction of hypotension, tranquilizer, and diuretic effects. The purpose of this study was to isolate and identify lactic acid bacteria, producing high GABA in fermented soy curd. Thirty-two strains of tofu-forming lactic acid bacteria were isolated from kimchi which a traditional Korean food fermented with many kind of microorganism. Among 32 strains, four strains (strain No. 10, 104, 214, 249) formed firm soycurd. In order to select lactic acid bacteria having high GABA producing potential, the isolated strains were cultured in the soymilk and fermented for 48 hr at $37^{\circ}C$. A strain No. 383, which showed highest GABA contents in fermented soycurd, was identified as L. sakei by 16S rDNA sequencing and API analysis, and named as L. sakei 383. L. sakei 383 showed optimal growth up to 24 hr at $35^{\circ}C$ in MRS broth. The optimal time and temperature for GABA production were 18 hr and $35^{\circ}C$ in soymilk. In the optimal condition time and temperature, GABA content of fermented soycurd by L. sakei 383 was 8.65 mg/100 g.

Classification of Antimicrobial Peptides among the Innate Immune Modulators (선천성 면역조절자인 항생펩타이드 분류)

  • Multidrug-resistant super bacterial, fungal, viral, and parasitic infections are major health threaten pathogens. However, to overcome the present healthcare situation, among the leading alternatives to current drugs are antimicrobial peptides (AMPs), which are abundantly produced via various species in nature. AMPs, small host defense proteins, are in charge of the innate immunity for the protection of multicellular organisms such as fish, amphibian, reptile, plants and animals from infection. The number of AMPs identified per year has increased steadily since the 1980s. Over 2,000 natural AMPs from bacteria, protozoa, fungi, plants, and animals have been listed into the antimicrobial peptide database (APD). The majority of these AMPs (>86%) possess 11–50 amino acids with a net charge from 0 to +7 and hydrophobic percentages between 31–70%. This report classified AMP into several categories including biological source, biological functions, peptide properties, covalent bonding pattern, and 3D structure. AMP functions not only antimicrobial activity but facilitates cell biological activity such as chemotatic activity. In addition, fibroblastic reticular cell (FRC) originated from mouse lymph node stroma induced the expression of AMP in inflammatory condition. AMP induced from FRC contained whey acidic protein (WAP) domain. It suggests that the classification of AMP will be done by protein domain.

Homology Modeling and Characterization of Oligoalginate Lyase from the Alginolytic Marine Bacterium Sphingomonas sp. Strain MJ-3 (알긴산을 분해하는 해양미생물인 Sphingomonas sp. MJ-3 균주의 올리고알긴산 분해효소의 상동성 모델링 및 특성연구)

  • Alginates are found in marine brown seaweeds and in extracellular biofilms secreted by some bacteria. Previously, we reported an oligoalginate lyase from Sphingomonas sp. MJ-3 (MJ3-Oal) that had an exolytic activity and protein sequence homology with endolytic polymannuronate (polyM) lyase in the N-terminal region. In this study, the MJ3-Oal was tested for both exolytic and endolytic activity by homology modeling using the crystal structure of Alg17c from Saccharophagus degradans 2-40T. The tyrosine residue at the $426^{th}$ position, which possibly formed a hydrogen bond with the substrate, was mutated to phenylalanine. The FPLC profiles showed that MJ3-Oal degraded alginate quickly to monomers as a final product through the oligmers, whereas the Tyr426Phe mutant showed only exolytic alginate lyase activity. $^1H$-NMR spectra also showed that MJ3-Oal degraded the endoglycosidic bond of polyM and polyMG (polymannuronate-guluronate) blocks. These results indicate that oligoalginate lyase from Sphingomonas sp. MJ-3 probably catalyzes the degradation of both exo- and endo-glycosidic bonds of alginate.

Cloning and Characterization of BTG-1 Gene from Pacific Oyster (Crassostrea gigas) (참굴(Crassostrea gigas)의 BTG1 유전자의 특성)

  • BTG 1 (B-cell translocation gene 1) gene was first identified as a translocation gene in a case of B-cell chronic lympocytic leukemia. BTG1 is a member of the BTG/TOB family with sharing a conserved N-terminal region, which shows anti-proliferation properties and is able to stimulate cell differentiation. In this study, we identified and characterized the pacific oyster Crassostrea gigas BTG1 (cg-BTG1) gene from the gill cDNA library by an Expressed Sequence Tag (EST) analysis and its nucleotide sequence was determined. The cg-BTG1 gene encodes a predicted protein of 182 amino acids with 57% 56% identities to its zebrafish and human counterparts, and is an intron-less gene, which was confirmed by PCR analysis of genomic DNA. Maximal homologies were shown in conserved Box A and B. The deduced amino acid sequence shares high identity with other BTG1 genes of human, rat, mouse and zebrafish. The phylogenic analysis and sequence comparison of cg-BTG1 with other BTG1 were found to be closely related to the BTG1 gene structure. In addition, the predicted promoter region and the different transcription-factor binding site like an activator protein-1 (AP-1) response element involved in negative regulation and serum response element (SRE) were able to be identified by the genomic DNA walking experiment. The quantitative real-time PCR analysis showed that the mRNA of cg-BTG1 gene was expressed in gill, heart, digestive gland, intestine, stomach and mantle. The cg-BTG1 gene was expressed mainly in heart and mantle.

Food Component Characteristics of Cold Air Dried Anchovies (냉풍건조 멸치의 식품성분 특성)

  • This study was conducted to evaluate the quality of the cold air dried anchovies (CA), and compared with those of sun-dried anchovies (SA) and hot air dried anchovies (HA). Peroxide value increased, while ratio of percentage of (20:5+22:6) to that of 16:0 decreased during boiling and drying. The extents of change were CA>SA>HA in the order. In the case of CA, lightness was higher, but redness, yellowness and degree of browning was lower than in SA and HA. From the results of lipid properties, color test and sensory evaluation on color and rancidity odor, lipid deterioration was the lowest in CA of all boiled dried anchovy. Favorite properties such as a hot-water soluble nitrogen content and sensory evaluation on color and appearance of CA was superior to that of SA and HA. The nutritional properties (total amino acid contents, mineral contents and fatty acid compositions) of CA was similar to that of SA, but was superior to that of HA. Consequently, CA was recognized as a boiled dried anchovy with the most quality.

MOLECULAR CLONING AND SEQUENCE ANALYSIS OF THE GENE FOR THE HEMIN-BINDING PROTEIN FROM Prevotella intermedia (Prevotella intermedia에서의 Hemin 결합 단백질 유전자의 분리 및 염기서열 분석)

  • Prevotella intermedia is one of the most frequently implicated pathogens in human periodontal disease and has a requirement for hemin for growth. This study has identified a hemin-binding P. intermedia protein by expression of a P. intermedia genomic library in Escherichia coli, a bacterium which does not require or transport exogenous hemin. The genomic library of P. intermedia was constructed into plasmid pUC18, transformed into Escherichia coli strain $DH5{\alpha}$, and screened for recombinant clones using heminbinding activity by plating onto hemin-containing agar. Approximately 5,000 recombinant E. coli colonies were screened onto LB-amp-hemin agar, single clone(pHem1) was exhibited a clearly pigmented phonotype. The 2.5kb insert DNA of pHem1 was determined by restriction enzyme mapping. Southern blot analysis of BamHI, BglII, EcoRI, HindIII and PstI-digested P. intermedia DNA indicated that single copy of the gene was present in the genome. Northern blot analysis revealed that the size of transcript was approximately 1.8 kb. The cloned gene contained a single ORF, consisting of approximately 850-residue amino acids. A BLAST search of the Institute for Genomic Research genes with similar nucleotide sequence revealed no significant similarity It needs further investigation to clarify the mechanisms of heme uptake in P. intermedia.

Molecular Isolation and Characterization of the 2CysPrx Gene from Salicornia herbacea (퉁퉁마디로부터 2CysPrx 유전자 분리 및 특성 분석)

  • This study is focused on the investigation of the genes which are induced by various stresses of the halophyte Salicornia herbacea. One of the factors influencing in the germination of Salicornia herbacea is salt stress. The highest germination rate was found in the condition without NaCl, and the upper limit of the NaCl concentration for the germination of Salicornia herbacea was 7%. The optimal temperature of $20^{\circ}C$showed a germination rate of 98%. Among genes induced by stress the 2CysPrx gene was cloned and analyzed for this study. The 2CysPrx gene has two cysteine conserved residues and is composed of 275 amino acids with molecular weight of 30.1kDa. The 2CysPrx gene appeared to be one copy in the genome and consists of 6 introns and 7 exons. Quantitative real-time PCR revealed that the highest transcription rate induced by NaCl and $H_2O_2$ appeared to be at the concentration of 3.5% NaCl and 40mM $H_2O_2$, respectively. The amount of transcript induced by high temperature($40^{\circ}C$) and $75{\mu}M$ of ABA was respectively highest. The gene at low temperature ($4^{\circ}C$) appeared not to be expressed. We are conducting to clone other peroxyredoxin genes induced by various environmental stresses.
