Changes in Enzyme Activity and Sensory Characteristics of Kochujang with Different Ratios of Added Deoduk (Codonopsis lanceolata) Root Powder (더덕 분말 첨가량을 달리한 고추장의 효소력 변화 및 관능적 특성)

  • Deoduk (Codonopsis lanceolata) root powder was added to traditional kochujang to improve the quality of traditional kochujang. The microbial characteristics, enzyme activities, and sensory characteristics were investigated during fermentation. The aerobic bacterial count in kochujang was not remarkabley different, and yeast and mold decreased during fermentation. Yeast and mold in the kochujang with 3 and 5% added deoduk root powder were 5 log scale at 8 weeks, whereas those in the control reached the same level at 6 weeks. ${\alpha}$, ${\beta}$-Amylase activity was at its highest level at 4 weeks during fermentation, and glucose and fructose contents showed the same results. The major free sugars in kochujang were glucose and fructose, and their contents increased rapidly at 2 weeks. Free sugar contents of kochujang with added deoduk was higher than that in the control. The sensory evaluation results showed that 1 and 3% deoduk kochujang had higher scores for taste and overall acceptance than those in the control. In particular, 1% deoduk kochujang had the highest scores.

  • Molds cause severe cheese deterioration, even though some white and blue molds are used for the manufacture of Camembert and Blue cheese, respectively. The species of Geotrichum, Moniliella, Aspergillus, Penicillium, Mucor, Fusarium, Phoma, and Cladosporium are the main fungi that affect contamination during cheese ripening. Once deteriorated by fungal spoilage, cheese becomes toxic and inedible. Fungal deterioration of cheese decreases the nutritional value, flavor profiles, physicochemical and organoleptic properties, and increases toxicity and infectious disease. Fungal contamination during cheese ripening is highly damaging to cheese production in Korean farmstead milk processing companies. Therefore, these companies hesitate to develop natural and ripened cheese varieties. This article discusses the recent and ongoing developments in the removal techniques of fungal contamination during cheese ripening. There are 2 categories of antifungal agents: chemical and natural. Major chemical agents are preservatives (propionic acid, sodium propionate, and calcium propionate) and ethanol. Among the natural agents, grapefruit seed extract, phytoncide, essential oils, and garlic have been investigated as natural antifungal agents. Additionally, some studies have shown that antibiotics such as natamycin and Delvocid$^{(R)}$, have antifungal activities for cheese contaminated with fungi. Microbial resources such as probiotic lactic acid bacteria, Propionibacterium, lactic acid bacteria from Kimchi, and bacteriocin are well known as antifungal agents. In addition, ozonization treatment has been reported to inhibit the growth activity of cheese-contaminating fungi.

Isolation and Identification of Protease Producing Bacteria in Kimchi (김치에서 단백질분해효소활성 균주분리 및 동정)

  • Strains producing the protease, which is essential for growth of lactic acid bacteria and fermented kimchi, were screened and identified. Among five types of selected pulmuone kimchis (Jeonlado kimchi, ripened kimchi, yeolmu kimchi, kakdugi, and baechu kimchi), nine strains of bacteria were screened and identified by whole cell protein pattern and API test. The nine strains consisted of one of Lactobacillus sp., one of Leuconostoc sp., six of Bacillus sp., and one of Brevibacillus sp. The protease activities of these strains were compared with known strains (Bac. subtilis KCCM 12248 and Bac. licheniformis KCCM 11851) producing protease. Among tested strains, K-2 (Brevibacillus sp.) showed the highest value (0.11 unit/mg) in protease activity.

Quality Characteristics of Umbilicaria esculenta Yakju Added with Grapefruit Seed Extract during Storage (자몽씨 추출물을 첨가한 석이버섯주의 저장 중 품질 특성)

  • This study was performed to investigate changes in characteristics of Umbilicaria esculenta Yakju added with 0.5% and 1% grapefruit seed extracts (GSE) during storage for 75 days at $25^{\circ}C$ and 165 days at $4^{\circ}C$. Quality characteristics of pH, titratable acidity, turbidity, total aerobic microbes, and ethanol, glucose, acetic acid, and lactic acid contents were analyzed. Control group showed sharply increased pH, titratable acidity, turbidity, and acetic acid and lactic acid has contents while the 0.5% and 1% GSE groups added to Yakju did not show significant changes in pattern (P<0.05). Total aerobic microbes increased rapidly during storage from 15 days in the control group, and the 0.5% and 1% GSE groups showed gradually increased total aerobic microbes from 30 days of storage compared to the control group (P<0.05). In addition, in all test groups, coli, yeast, and fungus were not detected. Ethanol content slightly increased during storage and glucose content slightly decreased, but there were no large changes during storage between the control group and GSE groups. Therefore, 3 months and $4^{\circ}C$ are considered safe for 6 months of storage, and GSE is effective to improve shelf-life and quality improvement of Yakju.

Changes in the quality characteristics of kimchi broths and growth of the isolated strains due to blue light emitting diode irradiation (Blue LED (Light emitting diode) 조사에 의한 배추김치와 백김치 발효액의 품질 특성 및 분리 균주의 생육 변화)

  • Kimchi is a widely consumed traditional Korean food, and its probiotic properties have received great attention. In this study, changes in the quality characteristics of fermentation broths obtained from two types of Chinese cabbage kimchi (red with red pepper and white without red pepper) were assessed after the administration of blue light emitting diode (BLED) irradiation at 4℃; characteristics assessed included acidity, chromaticity, antioxidant activity, and growth modulation of isolated microorganisms. The pH of the white kimchi (WK) broth decreased with time; the decrease was delayed significantly under BLED irradiation (p<0.05). BLED irradiation decreased the L (lightness) and b (yellowness) values and increased a (redness) in WK, whereas the a and b values of the red kimchi (RK) broth increased with BLED irradiation. Growth stimulation of lactic acid bacteria by BLED irradiation was observed in both WK and RK. The numbers of yeast and mold were also increased in RK (p<0.05), but not in WK. There was no change observed in the scavenging activities against ABTS (2,2'-azido-bis(3-ethylbenzthiazoline-6-sulfonic acid) radicals in both kimchi broths after BLED treatment. The results of this study indicated that BLED irradiation could modulate the fermentation process and the quality characteristics of kimchi during storage.

Effect of packaging conditions on the quality changes of fermented soy paste and red pepper paste (포장조건에 따른 한국전통 된장과 고추장의 품질변화)

  • 180g of fermented soy paste and 150g of red pepper paste were packaged in glass jars of 232 mL with different conditions of active packaging and then stored at $13^{\circ}C$ for about 170 and 128 days, respectively. During the storage, package atmosphere, surface color, pH, acidity and microbial flora were monitored to see the effect of packaging conditions. Test packaging conditions include package equipped with $Ca(OH)_2\;as\;CO_2$ absorber, package with pinhole and closed control one. Closed control packages of soypaste and red pepper paste showed the increased $CO_2$ partial pressure, the decreased $O_2$ partial pressure and the constant $N_2$ partial pressure to produce high pressure buildup with storage. The paste packages with $Ca(OH)_2$ maintained relatively low $CO_2$ partial pressure and thus the package pressure close to normal atmospheric pressure for initial storage period of 70 days. The packages with air pinhole channel had the partial pressures of $O_2\;and\;N_2$ decreased with storage time, while $CO_2$ partial pressure first increased to a maximum and then slowly decreased thereafter Without any pressure increase the packages with pinhole gave the lowest quality changes possibly due to the effect of package atmosphere, but it had problem of mold contamination and growth for soy paste after 120 days. There were no difference in microbial flora between the packages after about 70 day storage.

김치의 저장성에 미치는 포장내 표면처리 효과

  • 알코올 분무, 셀로판지 부착 등의 표면처리가 포장김치의 표면갈변과, pH, 산도, 균수 및 관능적 품질에 미치는 영향을 조사하였다. 실험에 사용된 포장김치는 200 g 들이 플라스틱 용기에 김치를 담은 후 무처리, 70% 알코올로 표면을 1회 분무한 것, 김치표면에 폴리에틸렌필름(두께 0.04 mm)을 부착하여 저장실 (10$\pm$1$^{\circ}C$)에서 21일간 저장하면서 3일 간격으로 조사하였다. 그 결과, 무처리와 알코올을 분무한 경우는 저장 15일째에 pH가 4.0수준에 도달하였으나 김치표면에 polyethylene film을 부착한 경우는 21일째까지도 pH가 4.0 이상을 유지하였다. 산도의 경우도 pH와 동일한 경향을 나타내어 polyethylene film으로 표면처리함으로서 김치의 숙성이 지연되었다. 무처리와 알코올을 처리한 것은 12일째부터 김치표면이 갈변하기 시작하였으며 18일째는 그 갈변정도가 심하였다. 그러나 polyethylene film을 표면에 부착한 경우는 저장 21일째까지도 갈변정토가 미약하였다. 저장기간에 따른 총균수의 변화는 무처리와 알코올을 처리한 경우가 polyethylene film을 부착한 경우에 비하여 증가폭이 컸으나 젖산균수는 반대로 polyethylene film을 부착한 경우가 무처리 또는 알코올을 분무한 경우에 비하여 높아 김치의 품질이 양호하였다. 무처리와 알코올을 분무한 김치의 표면에는 저장 15일 이후에 산막표모 및 곰팡이의 균사체가 번식하는 김치도 발견되었다. 처리별 이미 및 이취의 발생정도를 조사한 결과 무처리와 알코올을 처리한 경우는 저장 15일경에 특히 용기 표면에 위치하는 김치에서 이미와 이취가 있었으나 polyethylene film을 부착한 경우는 이미와 이취가 전혀 나타나지 않았다.채반에 넣고 밀도를 측정한 결과 1.002g/㎤을 나타냈다. 즉 물의 이론밀도인 1g/㎤에 근접한 값을 보여 정확한 밀도 계측이 가능한 것으로 판단되었다. 또한 밀도 계측시스템의 측정 반복간 정밀도를 파악하기 위해 수박 6개를 임으로 선정하여 3반복 측정 시험한 결과, 측정표준편차가 0.001~0.004g/㎤로 해상도보다는 다소 높았으나 대체로 양호한 결과를 나타냈다. 수박 35개를 이용하여 개발 계측시스템과 사람이 직접 부력법으로 밀도를 측정 비교한 결과, 계측시스템에 의해 측정된 수박 밀도가 사람이 측정했을 때 보다 낮게 측정되었다. 수박의 외관인자(무게, 길이, 직경, 체적), 밀도와 당도의 상관관계 구명시험을 위해 원예연구소 시험포장에서 재배된 삼복꿀수박 총 74개를 공시재료로 하였고, 시험은 출수일별로 10~14개씩 수확하여 외관인자, 밀도, 당도를 각각 측정하고, 이들 인자들간의 상관관계를 구명하였다. 외관인자들간에는 높은 상관관계를 보였으나, 외관인자들과 밀도, 외관인자들과 당도, 밀도와 당도와는 매우 낮은 상관관계를 나타냈다. 점성에 영향을 미치는 가장 주요한 조건이라고 생각된다.환원당인 sucrose 함량은 계속 증가하였고 fructose, glucose, sorbitol의 함량(추황의 sorbitol을 제외)은 생장이 촉진됨에 따라 증가하다가 다시 점차적으로 감소하였다. 이러한 결과는 총당과 환원당의 측정결과와 일치한 것으로 나타났다. 결론적으로 배의 성장에 따라 산 함량은 감소하였고 당 함량은 증가하였다.luco-pyranoside, quercetin 7-O- -glucopyranoside, acacetin 7-O-$\beta$-D-glucuronide and apigenin-6-C-$\beta$-D-glucopyranosyl-8-C-$\b

Effect of Prunus mume Extract on Shelf-life of Fermented Dairy Product (매실추출물이 발효유제품의 Shelf-life에 미치는 영향)

  • In order to examine the antimicrobial effect on dairy processing facilities and products, Prunus mume extinct (PME) was applied to the pilot plant system of dairy industry and yogurt, PME showed thermal and pH stability in the wide spectrum of temperature ($40{\sim}150^{\circ}C$) and pH ($4{\sim}10$) and remarkable antimicrobial activities against dairy spoilage microorganisms. As the result of aseptic treatment of dairy processing facilities with PME microbial colony including coliform bacteria was not detected canpore to those detected in the control. In the level of PME concentration which inhibit the growth of putrefactive microorganisms we could produce yogurt with good scores of sensory evaluation.

Physicochemical and Microbiological Properties of Korean Traditional Meju (한국 재래식 메주의 이화학적 및 미생물학적 특성)

  • The objective of this study was to obtain basic data on Korean traditional meju collected in 17 regions of Korea, to define and control meju quality. The moisture, crude fat, crude protein, and amino nitrogen contents of meju were 9.83-36.24%(w/w), 17.46-28.74%(w/w), 42.00-45.54%(w/w), and 223.65-1137.68 mg%, respectively. Meju was the enzyme source which made the soy sauce and doenjang. The $\alpha$-amylase, $\beta$-amylase, and protease levels were 130.32-1254.45, 30.07-167.88 and 72.53-340.04 units, respectively. Regional enzyme activities differed widely. Bacterial levels were $4.8{\times}10^7-2.6{\times}10^{10}cfu/g$, and molds and yeasts were at $4.3{\times}10^4-7.9{\times}10^6cfu/g$.

Evaluation of Functional Properties of Onion, Rosemary, and Thyme Extracts in Onion Kimchi (양파, 로즈마리, 타임의 기능성에 관한 연구)

  • This study was conducted to evaluate the functional properties of onion, rosemary and thyme extracts which are the ingredients of onion Kimchi. Onion extract showed a significant difference in antioxidative effect based on peroxide value and thiocyanate method. Antioxidative activity of rosemary extract was similar to those of BHA and BHT. but thyme extract did not show any antioxidative effect.. Onion, rosemary, and thyme extracts showed antimicrobial activities against gram positive bacteria but onion and thyme extracts did not against E. coli, Candida, and molds. Rosemary extract demonstrated a strong activity against L. plantarum which is a major lactobacillus in Kimchi fermentation, and Micrococcus luteus. Onion, rosemary, and thyme extracts showed an effect or hangover relief effect by lowering the alcohol concentration of blood in rat. Blood pressure of the male spontaneous hypertensive rat was suppressed by onion extract after 3 days of feeding, but rosemary and thyme extracts were not effective for lowering blood pressure.
