A case report of Cheyletiella infestation on a Whippet dog in Korea (Whippet종 개에서의 Cheyletiella yasguri(진드기목: 발톱진드기과) 감염증례 보고)

  • A clinical case of CheMletietlo infestation on a dog born and raised in Korea is reported. A three-year old female Whippet was hospitalized due to a multiple fracture and displacement of the left scapula caused by a recent car accident The mite infestation was not noticed at the time of hospitalization. The dog underwent multiple operations involving internal fixation of the fractured scapula with wire and a plate, followed by extensive chemotherapy with antibiotics and prednisolone. After two months of hospitalization. a pruritic dermatitis near the left scapula developed. Multiple white dandruff-like ilakes were seen on the hair coat. especially over the dorsal spine and neck, and the dog expressed increased pruritus by frequently licking and scratching the affected areas. Local dense accumulations of skin debris that became crusty were also observed. Microscopic examination or a skin scraping revealed a heavy infestation of cheyletiella vnsfuri. as idrntified by the prrsrnce of holes of the palpi and the heart-shaped sensory organ on menu I. Immnosuppression elicited by the extensive administration of prednisolone was suspected for the initiation of the generalized mite infestation.

Black Leaf Mold of Tomato Caused by Pseudocercospora fuligena in Korea (Pseudocercospora fuligena에 의한 토마토 검은잎곰팡이병)

  • In September 2011, black leaf mold caused by Pseudocercospora fuligena occurred on tomato plants growing in protected cultivation conditions in Boryong and Buyeo, Chungnam Province, Korea. Symptoms of the disease initially appeared as foliar chlorotic spots on upper leaf surfaces, turned light brown and then black on lower leaf surfaces as the fungus sporulates profusely. The causal fungus was isolated from the diseased plants and identified as P. fuligena based on morphological characteristics. Pathogenicity of the fungus was proved by artificial inoculation in the green house. This is the first report of the occurrence of black leaf mold of tomato caused by P. fuligena in Korea.

Detection of Poplar Mosaic Virus in Korea (한국에서의 포플러 모자이크 바이러스 검출(検出))

  • Several plants of Populus deltoides 'R-89' exhibiting symptoms of poplar mosaic described earlier by several researchers (1,2,3) were observed in the poplar nursery of the Institute of Forest Genetics at Suweon, Korea during September, 1980. The symptoms produced a mosaic or discolored yellow spottings in mature leaves with veinal and petiolar necrosis and leaf curling (Fig. 1 and 2). Leaves with mosaic symptoms were collected and leaf-dip preparations from chlorotic area were examined for the presence of virus particles with an electron microscope. Electron micrographs of the negatively stained leaf-dip preparations revealed numerous elongated virus particles ranging 200-1300nm in length (Fig. 3). Of 156 panicles measured randomly, 68 particles (43.6%) were in the range of 660-670nm in length (Fig. 4). The values obtained fell well within the range of those described for poplar mosaic virus (PMV) by other researchers (1. 2, 3). No viruslike particles were observed from poplar leaves without mosaic symptoms. Further studies on the properties of the PMV isolate and control of the disease are underway.

Pinus densiflora S. et Z. Population in Hong-do Island in Korea (홍도(紅島)의 적송집단(赤松集團))

  • Pine forests existing in Hong-do island were examined as regarding such characteristics, tree form, needle length, branch diameter, bark colors, ratio of summer wood to springwood, location and number of resin canal and other measurements. Hong-do island, located the furthest west part of Korea, suports only Pinus densiflora but very few Pinus thunbergii, in fact the present authors could not find any black pine individuals at all through investigation. Checking 14 individual trees of red pine, more than age of 20, of the genetic contamination by black pine, in a certain degree, exist. Here, a very dense foliage type and a very sparse foliage type tree of red pine growing nearby was examined and the unique fact was that each of those type has only one (average number 1.3) accessory resin canal, while other trees have three as minimum or more. The present authors thought this island had not been covered by any Pinus thunbergii but by Pinus densiflora. It was thought that Pinus thunbergii stands grown along the sea-coast of Korean Peninsula are pollen donors to this island.

Improved Detection and Purification of Grapevine Leafroll-associated 3 Closterovirus Using Tissue Culture (포도 조직배양에 의한 Grapevine Leafroll-associated 3 Closterovirus의 증식과 검출효율 증대)

  • Grapevine leafroll-associated 3 closterovirus (GLRaV-3) is phloem limited virus, and one of the most severe viral diseases found in Korea. However, nonhomogenous distribution and low concentration and seasonal variations of GLRaV-3 in grapevines remain as main problems which prevent the introduction and molecular biology or serology experiments. Virus-infected plantlet in vitro was obtained from node tissue cuttings, which was GLRaV-3 infected 'kyoho' vines. The amount of purified virus was highest in vitro plantlet. Moreover, viruses seem to be relatively homogeneously distributed in all organs including leaf, stem and callus derived from in vitro plantlets. RT-PCR detection using in vitro plantlet tissue as template was most effective. When comparing ELISA to RT-PCR, RT-PCR detection was 1,000 times as effective as ELISA. These results can be explained by improved quality such as tenderness or less tannins in plantlet in vitro. In conclusion, until infected herbaceous host will be available, tissue culture can be usefully adopted as a technique for a good source of GLRaV-3 closterovirus for further studies.

  • In this study, whether Giemsa staining solution can accurately determine bacterial contamination of liquid spawn for Flammulina velutipes in a short period of time was investigated. Giemsa solution staining cells of blood, bone marrow, lymph node, malaria parasites, rickettsia et al. was prepared by dissolving basic methylene azul and methylene blue, and acidic eosine in methyl alcohol-glycerine. Supernatant samples of Flammulina velutipes liquid spawn cultured under explosive aeration were placed on a slide, mixed with Gimesa solution and examined with optical microscope after staining. In 40 to 60 seconds bacterial cells were distinguishable from soybean meal residual and hyphal cell fragments. Thus we conclude that microscopy using Gimesa staining solution is a quick, simple and accurate method for the mushroom growers to effectively use to detect bacterial contamination of the liquid spawn.

Primary Cultures of Drosophila melanogaster Gut Cells for Studies of Intestinal Stem Cell Regulation (장줄기세포 조절 연구를 위한 초파리 장세포의 일차배양)

  • $Drosophila$ $melanogaster$ has been used as a useful model to study development and disease. In this study, we established the primary culture method of $Drosophila$ in the intestine to understand how intestinal stem cells (ISCs) mediate tissue repair during infection and disease. To obtain intestinal cells, we separated intestines from adult flies and isolated single cells by enzymatic treatment. The survival of cultured cells was measured using MTS-analysis. The maximum growth rate of the cells was observed on the 9th day after seeding. In addition, the presence of ISCs and enteroendocrine cells was confirmed by delta and prospero staining. Accordingly, we supposed that $Drosophila$ $melanogaster$ gut cells established in this study are probably useful in studies about intestinal stem cell regulation and various diseases occurring in the intestine.

Production of Putative Somatic Hybrid of Petunia hybrida and. Nicotiana sanderae by Protoplast Fusion (Petunia hybrida와 Nicotiana sanderae의 원형질체융합에 의한 잠정적 체세포잡종 식물체 생산)

  • The experiment were carried out to obtain a somatic hybrid through protoplast fusion between P.hybrid and N. sanderae. The isoenzyme pattern he chromosome number and the phonotype were observed for genetic study on the regenerants obtained from the fusion product cultures. Putative somatic hybrids possessed all the bands that appeared in both mother plane. A specific band was found on the top of the banding pattern which was assumed to be a marker band of somatic hybrid between two genera. Aspartate amninotransferase isoenzyme bands which were found in both mother plane were also revealed in the putative somatic hybrids or deleted in the upper part of H. sanderae band pattern. The chromosome number of P.hybrida was 2n=14, while N, sanderae was 2n=18,but the number of the putative somatic hybrids ranged from n=32 to 36. The phonotype of putative somatic hybrids was intermediate of the mother plants.

  • Both bark and wood sections obtained, by cutting with a razor blade, from living tissue of Cronartium ribicola cankers of Korean white pine (Pinus koraiensis) were transferred to a mixture of 2 parts of chloroform and 1 part of methanol, and the sections were stained using a modified lactophenol cotton blue. The formula for this staining is as follows: Lactic acid 20gm, phenol crystal 20gm, cotton blue 0.05gm, and 60% EtOH 100ml. The rust hyphae and haustoria were stained blue, and the wide hyphae with straight or curved haustoria could be distingwished from the pine tissue.

Thinning Stage on the Development of Fruit Structure in 'Fuji' Apples (적뢰 및 적과에 따른 사과 '후지' 과실의 조직 발달)

  • This study was carried out to verify the effect of fruit thinning on fruit development, and the results were as follows. The number of cells between fruit epidermis and periphery vascular bundle during 35 to 184 days after full bloom (DAFB) were constant with 14~17 cells, regardless of thinning at pink bud stage, fruitlet stage or non-thinning. However, the distance between epidermis and vascular bundle was longest in the fruit thinned at pink stage followed by fruits thinned at fruitlet stage and control. Therefore, it was seemed that the fruit size increment during 35 to 184 DAFB was due to the increment of cell size. Thinning time affected fruit size and the earlier the thinning times were the bigger the fruits were. However, there were no direct relationships between thinning time and the starch or tannin particle development in the cells of the fruit.
