Anatomical Ultrastructure of Spermatozoa of Paralichthys olivaceus (Paralichthyidae, Perciformes) from Korea (한국산 넙치 (Paralichthys olivaceus) 정자의 미세해부학적 구조)

  • Fine structure of spermatozoa of the bastard halibut (Paralichthys olivaceus) is described. Its structure is characterized by acrosomeless, having a spherical head region and a small mid piece containing five mitochondria disposed one row. The flagellum inserts centrally on the nucleus and has lateral ribbons. The present study supports the suggestion that spermatozoal ultrastructure is of taxonomic importance and may be a phylogenetic criterion in teleostei.

Improvement of Boar Semen Quality by Sperm Selection Using Magnetic Nano-particles (마그네틱 나노비드를 이용한 돼지 정자 품질의 향상)

  • The objective of this study was to see if fairly simple magnetic nano-particle treatment enhances boar semen qualities. Boar semen samples were prepared from the swine AI center and samples were divided by 4 different motility groups (1, >90%; 2. 80~90%; 3. 70~80%; 4. <70%) using computer assisted sperm analysis (CASA) evaluation. Boar semen was extended using BTS extender and same number of magnetic nano-particles as total number of spermatozoa in each sample was treated for 20 min and collected for 5 min at room temperature. Sperm qualities such as motility and viability were evaluated by the CASA before and after treatment. Sperm abnormality and degree of agglutination were also evaluated under the microscopic examination before and after treatment. There were significant changes (p<0.05) on sperm motility from all 4 different groups in the average of 7.11% after treatment. The enhancement of sperm motility changes was more clear in the groups of lower sperm motile groups (<70% and 70~80%; 19.12±1.08% and 5.67±0.71%, p<0.05). The sperm motility character in terms of curvilinear velocity (VCL), straight line velocity (VSL), average path velocity (VAP) and linearity (LIN, %) showed also similar pattern but motility enhancement wear more clear in below 70% motile group. Average sperm viability was increased to 4% by magnetic nano-particles (p<0.05). The percentage of sperm abnormality was also reduced significantly (p<0.05) to the range of 3.7~4.5% before after treatment. The degree of sperm agglutination was also reduced in lower motility groups by the magnetic nano-particle purification.

  • Sperm morphology of Pipistrellus savii velox was examined by transmission electron microscope. The sperm head of P. savii velox was bullet in shape. The sperm head was 3.1 ${\mu}m$ in length, whose posterior 3.0 ${\mu}m$ was occupied by a nucleus with 1.8 ${\mu}m$ in width. The segmented columns were about 14~15 in number. The total number of mitochondrial gyres was 57. Number of 1, 5, and 6 of the outer dense fibers were larger than the others. A fibrous sheath and longitudinal column of the principal piece were evidence, but the fibrous sheath was not seen at the end piece. In the present study, the length of the sperm head of P. savii velox were very shorter than those of other bats.

  • 소의 위에 기생하는 쌍구흠충(Paromphistomum cewi)에서의 정자형성과정을 광학현미경과 투과전자현미경을 이용하여 관찰하였다 발생중인 모든 세포들은 중앙에 있는 세포질로부터 각 세포들로 이어지는 세포간교에 의하여 서로 연결되어 있어 분열과 분화의 동시성을 위한 영양계를 이루고 있었다 정원세포들은 4세포기까지이며 모두 기정막에 붙어 있었다. 일차정모세포는 8세포, 이차정모세포는 16세포가 영양계를 이루었고, 정세포는 32세포가 영양계를 이루었으며 이 상태에서 변태를 진행하였다 성숙한 정자의 머리는 나사모양이고 첨체는 형성되지 않았으며 2개의 축사를 가지고 있었다 축사의 단면에서 미소관의 배열상태는 9치 유형이었다.

  • 본 연구는 개의 동결정액제조 시 동결보호 희석액 내에 첨가되는 당의 종류와 조합이 동결융해 후 정자의 생존율 및 운동성에 미치는 영향과 정자동결 시 straw size, 예비동결 시 액체질소표면으로부터 straw까지의 거리 및 융해온도가 생존율에 미치는 영향에 대하여 조사하고자 실시하였다. 포유류의 경우 부생식선의 정낭선에서 분비되는 정장내에는 정자의 체외사출 후 정자의 영양물질인 fructose 가 분비되는데, 동결정액제조 시 회석액내에 첨가되는 당 성분은 fructose 및 glucose를 일반적으로 사용하고 있다.   • 본 연구는 돼지 인공수정 시 1 회 주입하는 정자농도가 번식성적에 미치는 영향을 구명하기 위하여 활력이 70%이상인 정자수 기준으로 80㎖ 병당 30.0×10/sup 8/80㎖, 25.0, 20.0, 17.5, 15.0, 12.5, 10.0 및 7.5로 농도를 각각 조절하여 12시간 간격으로 2회 인공수정하고 분만율 및 산자수를 조사하였다. 본 실험에 사용된 액상정액은 축산기술연구소 돼지 인공수정 센타의 종모돈중 통일품종 정액을 BTS(Beltsville thawing solution) 보존액으로 혼합하여 5개 농장의 암돼지에 인공수정 하였으며, 정자농도는 광전비색계(Spectronic-20, USA)를 이용하여 측정하였다. 분만율은 72.3%-89.3%로 농도별로 통계적인 유의차는 인정되지 않았다.   • This study was carried out to investigate the morphological changes of gonadotropes in pituitary gland and spermatogenic cells in testis, obtained from 150 of 3-year-old immature and mature male rainbow trout (Oncorhynchus mykiss) during the reproductive cycles from March to February in the following year. In the maturation cycle of the pituitary gonadotropes of cultured rainbow trout, three periods can be distinguished i.e. a period of resting(March-August), a period of full spermatogenesis (September-November), and a period of breeding (December-February). The ultrastructures of the gonadotropes largely parallel the cyclical changes in the tests. The seminiferous tubules contain all spermatogenetic stages and sperm cells in a period of early maturation. At first, the size of the nucleus and cytoplasm decrease gradually at every stages from spermatogonia to spermatids. In the secondary spermatocytes, the small mitochondria are located over the outer cytoplam. In spermatids, the cytoplasmic masses move toward the posterior part of the nucleus. In spermatids, the two large mitochondria are located over the cytoplasm. In spermatids, the cytoplasmic masses move towark the posterior part of the nucleus. In spermatids, the two large mitochondria are located over the cytoplasm and begin to elongate. In spermatozoa, the surface of the nucleus devreases in volume. Examination by TEM shows that the nuclear envelope and plasma membrane are slightlywrinkled and closely adhered to the nucleus of spermatozoa. Two oval mitochondria are quite separated and the flagellum is inserted into the base of the spermatozoa head.The axoneme in this fish has the typical pattern such as nine peripheral doublets and a central doublet(9+2). there are remarkable individual differences in the size and morphology of spermatozoa head as observed by transmission and scanning electron microscopy.

  • Ultrastructural characteristics of blacktip grouper, Epinephelus fasciatus spermatozoa were investigated using transmission and scanning electron microscopy. The spermatozoa of E. fasciatus consisted of a spherical head part, a midpiece with cytoplasmic canal entrance and a flagellum with lateral fins. Internal ultrastructurally, the nucleus contains high electron dense chromatin having granular particles and has no acrosome. The centriolar complex lies outside of the nuclear fossa and it is connected by the osmophilic filaments. Also the osmophilic filaments connect between the centriolar complex and the nuclear membrane. The midpiece contains eight to nine spherical mitochondria, cytoplasmic canal and necklaces. The flagellum has a typical 9+2 axonemal structure. The lateral fins contain vesicles and a typical 9+2 axonemal structure. Consequently this study contributes to comparative grouper spermatology and provide useful systematic taxonomic characters.

Effects of Straw Size and Thawing Rate on Post-thaw Quality of Bog Semen (개의 동결 정액 제조시 Straw의 크기와 융해온도가 정자의 생존율에 미치는 영향)

  • 본 연구는 개 동결 정액 융해 시 straw 크기 및 융해 속도가 융해 정자의 질(quality)에 미치는 영향을 조사하고 최적의 융해 조건을 조사하는데 그 목적이 있다. 정상적인 번식능을 가진 비글 수컷 5마리에서 정액을 채취하여 원심 분리하여 정장을 버리고 남은 정자에 동결보호제인 glycerol이 첨가된 tris-glucose-egg yolk extender를 첨가하여 동결하고 액체질소에 보관한 후 융해하였다. 동결 융해 조건에 따른 효과를 알아보기 위해 straw는 0.25 ml과 0.5 ml크기를 사용하였고 융해 조건은 $75^{\circ}C$에 10초, $55^{\circ}C$에 12초 및 $37^{\circ}C$에서 120초로 하여 융해 후 정자의 활력도(vigor), 운동성(motility), Hypo-osmotic test(HOS test)를 이용한 생존성(viability) 및 $SperMac^{\circledR}$ 염색을 하여 정자의 membrane integrity를 비교 조사하였다. 조사 결과 0.5 ml 크기의 straw를 사용한 경우 $37^{\circ}C$ 융해가 $55^{\circ}C,\;75^{\circ}C$ 융해보다, 0.25 ml 크기의 straw를 사용한 경우에는 $37^{\circ}C,\;55^{\circ}C$ 융해가 $75^{\circ}C$ 융해보다 유의적으로 높은 활력 지수 및 생존성을 보였다(P<0.05). Straw크기에 따라 비교하였을 경우 0.5 ml 군에서 유의적으로 높은 활력도, 생존성 및 membrane integrity를 보였다(P<0.05). 결론적으로 개 정액이 동결 및 융해 시 0.5ml straw를 이용하여 동결한 후 $37^{\circ}C$에서 120초 동안 융해하는 것이 최적의 조건임이 사료된다.

  • In order to obtain the basic information for the seedling production of surf clam, Spisula sachalinensis, sperm motility and optimal method for fertilization were investigated. Sperm concentration of S. sachalinensis milt was$ 2.02{\times}10^{10}\;cell/mL$ and approximately $96.0\%$ of sperm showed forward movement after exposure to seawater. When sperm and eggs obtained by incision method were fertilized in 1 hour and 4 hours, respectively, high fertilization and hatching rate were achieved. The optimal sperm concentrations and egg density for fertilization and hatching were 10$\~$100 inds./egg and 100$\~$200 inds./mL sea water, respectively.
