Biogas Production by Anaerobic Co-digestion of Livestock Manure Slurry with Fruits Pomace (가축분뇨와 과실착즙박의 혼합 혐기소화에 따른 바이오가스 생산)

  • This study is conducted to investigate the effects of anaerobic treatments of swine manure slurry alone and combination of livestock manure slurry and fruit pomace on biogas production. Anaerobic co-digestion was evaluated in mesophilic tank reactors for 96 day-incubation period. The organic matter loading of anaerobic digestion was 1 kg of volatile solids(VS) per $1m^3{\cdot}day$. The highest methane production was achieved from the combination of swine manure slury and mandarin pomace(70:30) treatment, whereas the lowest daily and cumulative methane yields was observed in swine manure slurry alone treatment. More than two-fold increase in bio-gas and methane production was obtained by combination of livestock manure slurry and mandarin pomace treatment, compared to the swine manure slurry alone treatment. The co-digestion of livestock manure and fruits pomace has advantages to enhance the production of methane gas, compared to digestion of swine manure slurry alone.

  • 농업에 있어 해충 방제는 중요한 일이다. 특히, 볼록총채벌레는 최근 감귤원 해충 피해의 주요 해충으로 인식되어 주기적인 예찰이 이루어지고 있으나 성충의 크기가 0.8mm 정도로 작아 육안 식별에 어려움이 있다. 본 논문에서는 예찰 트랩에 포집된 볼록총채벌레를 자동으로 판별하기 위한 전처리 알고리즘을 제안하였다. 광학 현미경의 배율을 30, 40, 50배로 조정하여 영상을 획득하였고, 제안한 알고리즘을 적용하여 판별 성능을 측정한 결과, 50배율에서 가장 높은 판별율을 보였으나 더 많은 볼록총채벌레 영상을 대상으로 알고리즘을 테스트하고 개선할 필요가 있다.

제주도 농업용 관정의 양수량 산정과 지하수 이용특성 연구

  • 제주도내에 개발ㆍ이용 중인 사설 농업용 44개 관정을 대상으로 2002년 11월부터 2003년 11월까지 1년간 지하수 이용량 모니터링 조사를 실시하였다. 재배작물 유형별 지하수 이용특성을 평가한 결과, 밭작물은 대체로 9월에서 11월 초순까지 집중적으로 지하수를 이용하고 있으며, 시설하우스의 경우에는 2월에서부터 10월말까지 비교적 지속적으로 지하수를 이용하고 있는 것으로 분석되었으나, 노지 감귤원은 가뭄이 아닌 경우에는 농약살포시 매우 소량의 지하수를 이용하고 있으며 이용시기도 매우 불규칙한 것으로 조사되었다. 평균 총 가동 일수는 시설하우스 및 밭작물에 혼용으로 사용하고 있는 지하수 관정의 128일로 최대치를 나타내고 있으며, 연간 지하수 총 이용량의 경우도 관정 가동 일수와 같은 경향을 나타내고 있다. 단위 면적(ha) 당 지하수 일 평균 이용량은 시설하우스가 74m$^3$/일으로 가장 많았고 밭작물이 50m$^3$/일로 조사되었다. 원격검침 시스템에 의해 얻어진 모니터링 테이터로부터 산정된 양수량과 계량기 검침에 의한 양수량과의 관계를 비교 분석한 결과, 상관계수가 $R^2$ =0.997을 나타내 수중모터펌프 가동시간 모니터링에 의한 양수량 산정방법이 매우 유효함이 입증되었다.

Antioxidant efficacy of Jeju crop extracts using Jeju lava seawater as a solvent (제주 용암해수를 용매로 한 제주 작물 추출물의 항산화 효능)

  • Jeju lava seawater's abundant minerals are known to exert antioxidant effects that remove the free radicals responsible for aging. Therefore, lava seawater reportedly possesses high commercial value as a functional food material. This study compared and analyzed the antioxidant activities of extracts from crops produced in Jeju (carrots, blueberries, and mandarins) using distilled water and lava seawater as solvents. Lava seawater extracts exhibited higher total polyphenol and flavonoid contents of blueberries and mandarins than distilled water extracts. Furthermore, the antioxidant enzymatic and 2, 2-diphenyl-1-picrylhydrazyl radical-scavenging activities of these crops were higher in lava seawater extracts than in distilled water extracts. In particular, using Vero cells, the ROS-scavenging efficacies of blueberries and mandarins were found to be higher in lava seawater extracts. Meanwhile, the antioxidant activities of carrots were higher in lava seawater extracts, despite no difference in total polyphenol and flavonoid contents. These results suggest that lava seawater exhibits favorable potential as a solvent in the functional food industry, and lava seawater-based Jeju crop extracts are potentially useful as functional food ingredients.

Evaluating the Impacts of Long-Term Use of Agricultural Chemicals on a Soil Ecosystem by Structural Analysis of Bacterial Community (세균군집의 구조분석을 통한 장기간 농약사용이 토양생태계에 미치는 영향 평가)

  • In this study bacterial community was analyzed to evaluate the impacts of long-term use of agricultural chemicals on a soil ecosystem as well as to obtain fundamental data on the relationship. Sequences of 16S rRNA clones from a non-agricultural site and a tangerine orchard soil which has a history of long-term use of agricultural chemicals over 30 years were analyzed. This revealed that bacterial community containing 5 divisions and 18 genera was distributed in a tangerine orchard soil, while bacterial community containing 9 divisions and 44 genera was distributed. In a tangerine orchard soil site, the most abundant bacteria in subdivision level were placed into Proteobacteria γ group which occupied 56% of total clones. The other bacterial clones from the ocrhcard soil exposed to agricultural chemicals over 30 years were Acidobacteria group (25%), Fimicutes group (5%), Planctomycetes group (2%), Proteobacteria α (1%), δ group (1%), and Cyanobacteria group (1%). Whereas, the clones were from the non-agricultural site were distributed among the division or subdivision Acidobacteria group (14%), Planctomycetes group (13%), Proteobacteria α (10%), β (9%), δ (9%), Fimicutes group (8%), Verrucomicrobia group (8%), Actinobacteria group (6%), Proteobacteria γ group (3%), Bacteroidetes group (3%), Gemmatimonadetes group (3%), and Cyanobacteria group (1%). This finding suggests the possibility that long-term application of agricultural chemicals or fertilizers on a tangerine orchard might result in drastic reduction or alteration in the composition of the bacterial community in the contaminated soil site.

Nocturnal temperature distribution on orange orchards in Cheju Island (II) (제주도 감귤 과수원의 야간 기온 분포(II))

  • The Characteristics of nocturnal temperature fields were analyzed to understand the factors of freezing-and-cold damages on orange orchards in Cheju Island. Temperatures were measured from January 7th through 27, 1995 at 25 sites in an area of 1x1.25$ extrm{km}^2$, Wasan-ri, Chochon-up, Pukcheju-kun located on the northeastern slope of Mt.Hanla. Several other weather elements such as wind and cloud were observed as supplementary data. Surface weather maps were also analyzed to clarify the influence of prevailing pressure patterns on the temperature fields. The vertical temperature profiles were obtained at the height from the ground up to 360 cm in 30 cm intervals at site 3, a frost hollow, and site 10 on the upper slope. The results show that freezing damages occured in the hollows, terrain depressions, rather than at the upper slope due to nocturnal radiation cooling as well as accumulation of cold airflow from Mt.Hanla. Windbreaks of densely planted Japanese ceders with stone-walls also roled as obstacles to the cold airflow in nights with Clear skies and light winds. The maximum intensity of temperature inversion in hollows, quasi-cold air lake, was 3.1$^{\circ}C$. Cold air from Mt.Hanla was trapped in the depressions up to a height of 90cm forming frost pocket. Man-made facilities such as shelterbelt or stone-wall which are built to prevent the penetration of cold north-westeries in winter aggravated the cold damage. The differance of daily minimum temperatures between before and behind shelterbelts was 2.$0^{\circ}C$. The man-made convection by smudgin which raised the temperatures up to 3.8$^{\circ}C$ can reduce the cold damage in the hellows.

Isolation and Characterization of an Antagonistic Endophytic Bacterium Bacillus velezensis CB3 the Control of Citrus Green Mold Pathogen Penicillium digitatum (감귤저장병 병원균 Penicillium digitatum 방제를 위한 길항 내생세균 Bacillus velezensis CB3의 분리 및 특성 규명)

  • In order to develop environment friendly fungicide for the control of citrus green mold (Penicillium digitatum) using endophytic bacteria, the 21 bacterial isolates were isolated from citrus leaves in seven different orchards in Jeju Province. Among the 21 bacterial isolates, 5 bacterial isolates presented antifungal activity against green mold pathogen P. digitatum. The CB3 isolate, which showed the most strong antagonistic effect, was selected through opposite culture against the pathogen. The rod-shaped, gram-positive bacterium CB3 was identified as Bacillus velezensis based on morphological, physiological characteristics, 16S rDNA, and gyr A gene sequence analysis. The isolate CB3 showed strong antifungal activity against two citrus postharvest pathogen P. digitatum. Citrus fruits were treated by wound inoculation with P. digitatum pathogen, and the control efficacy of CB3 culture broth was 66.7% ($1{\times}10^8$ cfu/ml). In conclusion, The stability of CB3 and its strong antifungal activity also lead us to believe that it has potential for application as an environment friendly biological control agent.

Chemical Characteristics and Water Dispersible Colloid Content of Jeju Citrus Orchard Soils (제주도 감귤원 토양의 화학적 특성과 물 분산성 콜로이드 함량)

  • Water-dispersible colloids are suspected to facilitate transport of contaminants to groundwater. This study evaluated some soil chemical properties in relation to the stability of colloids in soils of Jeju citrus orchards. Thirty surface soil samples were collected, and pH, organic matter content oxalate-extractable Al and Fe contents, and water-dispersible colloid content were measured. In soils of higher pH, water-dispersible colloid contents were higher. The stability of colloids was found to be significantly promoted at pH above 5$\sim$6. Since organic matter can act as a flocculant organic matter content significantly enhanced the colloid stability. In soils of less than 5% organic C, water-dispersible colloid content was expected to be significantly higher. In soils of higher oxalate-extractable Al and Fe contents, colloids remaining in suspension were lower. This indicated that amorphous oxides and hydroxides play important stabilizing roles in soil structure and can stabilize soil clay against dispersion. Therefore in soils of higher pH, lower organic matter, and lower amorphous clay minerals, the stability of water-dispersible colloids and the potential of colloid-mediated transport of organic chemicals to groundwater could be higher.

Comparison of Predation Rates of Three Phytoseiid Mite Species on Citrus Red Mite (Panonychus citri McGregor) on Citrus Tree (감귤원에 발생하는 귤응애에 대한 3종 이리응애의 포식량 비교)

  • Predation amount and density suppression ability of three species of phytoseiid mites (one native, Amblyseius womersleys Schica; two introduced species, Amblyseius fallacis Garman and Typhlodromus occidentalis Nesbit) on Panonychus citri McGregor were examined in laboratory condition (25$\pm$1$\^{C}$, RH 65$\pm$5%, 16L:8D). A. fallacis and T. occidentalis consumed 20.1 and 9.1 eggs of P. citri, respectively, whereas A. womersleyi consumed 1.1 eggs. When each larva, protonymph and deutonymph of P. citri were supplied as prey for 24 hours. A. womersleyi consumed 23.8, 16.5 and 9.0, and A. fallacis consumed 26.1, 18.2 and 7.4, respectively. However, T. occidentalis consumed only 7.8, 4.2 and 4.2. respectively Density suppression ability of A. womersleyi and A. fallacis against P. citri was very high at the release ratios of 20 : 1 (prey : predator). However, T. occidentalis did not regulate P. citri population effectively Developmental periods from egg to adult of A. womersleyi and A. fallacis was 6.1 and 5.9 days at 25$\^{C}$, respectively.

Characteristics of Bacillus sphaericus PSB-13 as Phosphate Solublizing Bacterium Isolated from Citrus Orchard Soil (감귤원 토양에서 분리한 인산염 가용화 미생물 Bacillus sphaericus PSB-13의 특성)

  • This study was carried out to measure insoluble phosphorus fractions content fixed in different soil type and isolate a superior phosphate solublizing bacteria(PSB) producing free phosphate in citrus orchard soil. Distribution of insoluble phosphate fraction ordered Al-P>Ca-P>Fe-P in the investigated citrus orchards. Insoluble phosphate fraction such as Al-P, Ca-P, Fe-P were higher in volcanic ash than in non-volcanic ash soil. A PSB with high holo zone in PDA-P medium isolated from citrus orchard soil. This strain identificated by MIDI system as Bacillus sphaericus. The optimum growth of pH and temperature were at 4~5, $30^{\circ}C$, respectively. When Bacillus sphaericus cultured at $25^{\circ}C$, 150 rpm condition in LB broth medium included different phosphate. Bacillus sphaericus produced free phosphate in the culture broth medium from tricalcium-phosphate(207.0 ppm), aluminium phosphate(324.5 ppm) and hydroxyapatite(334.8 ppm) and Phosphatase activity of Bacillus sphaericus was higher at $35^{\circ}C$ culture condition than that of $25^{\circ}C$. Two type preparation inoculated Bacillus sphaericus made with carrier materials such as Bentonite, $CaCO_3$, Sodium alginate. Density of PSB in this preparation conserved at $10^5c.f.u.\;g^{-1}$ level during storage in different temperature condition for 7 month. It also showed that free phosphate produced at PDA-P medium.
