The Antioxidant, Alcohol Metabolizing Enzyme, and Hepatoprotective Activities of Dendropanax morbifera Vinegar with Traditional Fermentation Methods (전통적인 발효 방법으로 제조한 황칠 식초의 항산화, 알코올 대사 효소 및 간보호 활성)

  • This study was performed to investigate the organic acids, alcohol metabolism enzyme, and antioxidative, nitrite-scavenging, and hepatoprotective effects of Dendropanax morbifera vinegar prepared by a traditional fermentation method. Among the organic acids detected, acetic acid was the highest found, at 91.72 mg/ml, followed by lactic acid (7.31 mg/ml), malic acid (1.36 mg/ml), and succinic acid (1.20 mg/ml). The total polyphenol content of the D. morbifera vinegar was 13.73 ㎍ tannic acid equivalent (TAE)/ml. The 1,1-Diphenyl-2-picrylhydrazy (DPPH) radical scavenging activity of D. morbifera vinegar was 76.04% at a 60% concentration. The superoxide dismutase (SOD) activity of D. morbifera vinegar was increased in a dose-dependent manner, which was 95.14% at a 60% concentration, while the α-glucosidase inhibitory activity of D. morbifera vinegar was 98.94% at a 10% concentration. The effects of D. morbifera vinegar on alcohol metabolism were determined by measuring the generation of reduced nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH). The ADH and ALDH activities of D. morbifera vinegar were increased in a dose-dependent manner, 43.62% and 60.39% at a 60% concentration, respectively. The D. morbifera vinegar showed significant protective effects against tacrine-induced cytotoxicity in HepG2 cells at the 0.6% concentration. These results suggest that D. morbifera vinegar has great potential as a resource for high quality functional health beverages.

The Anti-inflammatory Effects of Heat-treated Broccoli Extract (열처리된 브로콜리 추출물의 항염증 효과)

  • This study was carried out to investigate the heat-treated broccoli extraction which have a beneficial effect on the human body and which can be used safely for a long period of time without adverse side effects and also have excellent effects of protecting liver and improving liver function. The heat-treated broccoli extract does not show cytotoxicity, and thus can be used safely. In an experiment performed on an animal model with liver injury induced by a drug (APAP), it could be seen that the heat-treated cabbage extract exhibited the effects of protecting liver and improving liver function by effectively reducing AST and ALT which are liver injury markers, indicating that the heat-treated brocoli extract is effective as a pharmaceutical extraction for preventing or treating liver disease. In particular, the heat-treated broccoli extract was effective in treating inflammation of the liver by reducing the expression of the inflammatory mediators iNOS and COX-2 and the proinflammatory cytokine $IL-1{\beta}$, which are involved in acute inflammatory reactions accompanying liver injury.

Chemical components and hepato-protective effect of Lentinula edodes fermented by lactic acid bacteria (표고 유산균 발효물의 성분 및 간기능 보호 효과)

  • This study was conducted to improve the useful components and biological activity of Lentinula edodes fermented by lactic acid bacteria (LAB). Three LAB strains (Lactobacillus brevis KCCM 11904, L. plantarum KCCM 354469, and L. fermentum KCCM 12116) were inoculated and used for L. edodes hot water extract (10%, 20%, 30%) fermentation. LAB fermentation of L. edodes hot water extracts decreased pH and thus were more acidic than non-fermented L. edodes hot water extract. β-glucan and ergothioneine contents were increased by L. edodes in a concentration-dependent manner. The ergothioneine and β-glucan contents were highest in fermented with 30% L. edodes hot water extract fermented by L. plantarum and L. brevis (40.48 mg/100 g and 13.94%, respectively). The hepato-protective effect of fermented L. edodes hot water extracts by the three LAB were tested using Sprague-Dawley rat primary hepatocytes. In primary hepatocytes obtained following liver injury induced by acetaminophen, fermented L. edodes hot water extracts by the three LAB showed protective effects, as evident by reduction of the aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase liver markers. The collective results indicate that the fermented L. edodes hot water extracts obtained using LAB are potentially valuable in preventing or treating liver disease.

Protective effect of STAR of STAR series on CCl4 induced acute hepatotoxicity by regulation of reactive oxygen species (활성산소종의 조절을 통한 음료 '별의별간'의 급성간독성 보호효과)

  • STAR of STAR (SS 01-04) is a series of drinks that consist of various extracts obtained from Coriolus versicolor, Artemisia capillaris Thunb., Hovenia dulcis, Acanthopanax sessiliflorus, Lycium chinense, Citrus reticulata, Saururus chinensis, Pueraria lobata, Pyrus pyrifolia, and Oenanthe javanica. A purpose of this study was to investigate the hepatoprotective effect of SS 01-04. Antioxidant activity of the drinks was evaluated by conducting a hydroxyl radical-scavenging assay. Cytotoxicity and hepatoprotective potential were determined using HepG2 cells in vitro, while protective effects against acute hepatotoxicity was evaluated in vivo. The antioxidant activity of the SS 01-04 at concentration of 100 and 250 mg/mL was similar to that of $50{\mu}M$ vitamin C. tert-Butyl hydroperoxide (tBHP)-induced production of reactive oxygen species (ROS) was blocked by SS 01, 03 and 04 in a dose-dependent manner. Treatment with SS 04 significantly lowered the serum levels of alanine aminotransferase and aspartate aminotransferase in an animal model of carbon tetrachloride $(CCl_4)-induced$ hepatotoxicity (p<0.05). In addition, SS04 increased glutathione level while decreased malondialdehyde level in the liver considerably (p<0.05). It also inhibited the $CCl_4-induced$ increase in the levels of triglyceride and cholesterol in serum and the liver. These findings indicated that SS 01-04 possessed antioxidant activity and protect against ROS. In particular, SS 04 is potentially highly beneficial in treating liver damage as it scavenges reactive free radicals and boosts the endogenous antioxidant system.

  • Hepatoprotective activities of 26 traditional Korean herbal prescriptions described in Donghibogam were evaluated in vivo and in vitro screening system. Twenty one prescriptions appeared to be active. Six prescriptions among them showed highly significant hepatoprotective activities; Shihogyegy-Tang(柴胡桂枝湯), Soshiho-Tang(小柴胡湯), Shiho plus Younggolmoryo-Tang(柴胡加龍骨牡蠣湯), Yinjinho-Tang(茵蔯蒿湯), Yinsam-Tang(人蔘湯) and Shashim-Tang(瀉心湯). Shihogyegy-Tang was most active. They reduced the release of transaminases from dissociated hepatocytes by thioacetamide, and serum transaminases and alkaline phosphatase activities of rat with carbontetrachloride-induced hepatitis. They inhibited significantly lipid peroxidation and cellular fatty change of liver by carbontetrachloride.

The preventive effect of Daeshiho-tang on liver damage induced by acetaminophen in the rats (대시호탕이 acetaminophen으로 유도된 간독성 흰쥐에 미치는 영향)

  • Acetaminophen은 세계적으로 널리 사용되는 해열 진통제이지만, 또한 과용 및 남용, 알코올 중독과 같은 여러 원인에 의해 간독성을 유발할 수 있는 약물이다. 이러한 acetaminophen의 간독성은 CYP에 의해 생성되는 대사산물인 NAPQI와 활성산소에 의해 유발되는 것으로 알려져 있다. 본 연구에서 5주된 수컷 백서에 acetaminophen (500 mg/kg)을 투여하기 전에 대시호탕 (500 mg/kg)를 일주일간 투여하였다. 이후 GOT, GPT, GST 그리고, 조직사진으로 대시호탕의 간보호작용을 측정하였다. 또한 대시호탕의 간보호작용 기전을 항산화작용과 CYP 2E1 발현조절을 통한 NAPQI 생성억제의 두 가지 면에서 측정하였다. GOT, GPT 그리고 조직사진에서 나타난 결과들은 대시호탕이 고용량의 acetaminophen에 대한 간보호작용이 있음을 증명할 수 있었다. 또한 LPO와 catalase, 그리고 GSH 실험에서 나타난 결과들을 통해 대시호탕이 항산화작용이 있음을 알 수 있었다. 그리고 GSH, GST, RT-PCR, western blot 실험에서 대시호탕이 CYP 2E1의 발현을 조절하여 NAPQI 생성을 억제한다는 것도 알 수 있었다. 이상의 결과들을 바탕으로 대시호탕은 항산화작용에 의한 활성산소 제거력과 CYP 2E1의 발현조절을 통한 NAPQI 생성억제로 고용량 acetaminophen에 의해 유도된 간손상에 대해 유의성 있는 보호작용을 한다는 것을 알 수 있었다.

Protective Effects of Mesangi (Capsosiphon fulvecens) on Hepatotoxicity in Carbon Tetrachloride (CCI4)-Intoxicated Rats (매생이가 사염화탄소로 유발된 흰쥐의 간손상에 미치는 영향)

  • This study investigated the hepatoprotective effects of mesangi (Capsosiphon fulvecens) in carbon tetrachloride $(CCI_4)$-induced liver injury. Thirty healthy male Sprague-Dawley rats were divided into three groups : the normal (Normal), $CCI_4$-treated $(CCI_4)$, and mesangi-treated (Exp-CF) groups. Administration of $CCI_4$ increased the levels of GOT, GPT, and LDH in serum, while the levels were significantly decreased by the addition of mesangi. Higher levels of HDL-cholesterol were found in the Normal and Exp-CF groups, which has lower levels of total and LDL-cholesterol. Administration of $CCI_4$ also increased IGFBP-1 expression in serum, but it was decreased in the Exp-CF group. This suggests that $CCI_4$ is involved in the change in IGFBP-1 expression via its hepatotoxicity. The results indicate that mesangi has a hepatoprotective effect in rats given $CCI_4$.

Current Management Status of 'Day and Night Care Facilities' for Long-Term Care Insurance Benefit (노인장기요양보험 급여 주야간보호사업소의 운영현황)

  • This study was conducted to understand current management status of 'day and night care' facilities and to explore the related factors with rates of operation of them. The nationwide mailing survey was conducted with structured questionnaires from July, 14, 2010 to July, 28, 2010. The subjects were 277 facilities (response rate 24.5%). Regarding the types of operating, 79.1% of faculties was run by cooperation, and 17.8% and 3.6% for individuals and national/public institutes respectively. The average of operation rate was 70.15%. National/public institutes showed higher in the rate of operation(79.08%) than 72.49% of cooperations and 57.78% of individuals. The number of manpower was not nearly different by the types of operating bodies, but the number of nurse was significantly different among them. The national/public institutions had 1.07 nurses while individual institutions have only 0.08. We found that facilities run by national/public institutes and founded before 2008 years showed higher rate of operation. In case of providing regular monitoring and evaluation services, and music program for patients showed higher rate of operation. In addition, the number of managers, social workers, and nurses increase the rate of operation. We suggest that quality management and monitoring program for the facilities which run by individuals or established after 2008 years would be developed. We also call for development of programs for facilitating utilization of 'day and night care' facilities such as expanding the family support.
