A Study on Comparison Experiments for Judgement to the Nondestructive Analysis of Gold Plated Silverware Remains (비파괴분석법에 의한 은제금도금유물의 판단을 위한 비교실험 연구)

  • If the nondestructive analysis is carried out by using an X-ray Fluorescence Analyzer(XRF) for ingredient analysis with the target of gold plated silverware that plates gold on a silver basis, a singular point showing big difference at the value of weight percent(wt.%) according to the electron orbit of the principal ingredient can be confirmed. The present research made reproduced samples and carried out comparative analysis in order to suggest a basis that can judge whether the technique of gold plated silverware was used in case of remains uneasy to understand with eyes by using this singular point. As a result, the same result as analysis results of actual remains could be seen in specimens manufactured by a mercury amalgam plating method. However, in case of gold and silver alloy specimens, it could be confirmed that the difference according to the electron orbit of the principal ingredients is slight unlike plated specimens. The reason appearing at plated specimens seems to be generated while absorbing XRF beam at plated layers by double structure due to plating instead of single material. Accordingly, if this result appearing at nondestructive analysis is used, it was confirmed that the application of the technique of gold plated silverware can be judged without observing its section.

Stability Evaluation on Measuring Water-soluble Chloride Anions from Iron Artifacts (철제유물의 수용성 염소이온 측정방법에 대한 안정성 평가)

  • The most ideal method to measure the water-soluble $Cl^-$ ion eluted from iron artifacts is conducting the analysis on desalting solution by Ion Chromatography. But most institutes related to cultural heritages use Cl meter by reason of lack of budget and experts. This study evaluated reliability and stability between Cl meter and Ion Chromatography by doing cross-validation with results from two methods to detect $Cl^-$ ion of desalting solution. From D.I water, extremely small quantities of $Cl^-$ ion was detected by the influence of remaining water-soluble $Cl^-$ ion at the electrode of Cl meter and water-soluble $Cl^-$ which remains in Sodium sesquicarbonate, components of reagent was detected as well. The first desalting solution had the most $Cl^-$ ions, $Cl^-$ ion slightly decreased from the second to the fourth desalting solution and tend to decrease again at the stage of dealkalified in D.I water. Each Cl meter has the standard deviation according to the measured numbers and the higher concentration of $Cl^-$ ion the desalting solution has, the wider the deviation is. But when the concentration of $Cl^-$ ion is low, it was stable to use Cl meter to detect the concentration of $Cl^-$ ion from iron artifacts because there is the small deviation, It is thought that conductivity meter method is not suitable for measuring $Cl^-$ ion, because the electrical conductivity of alkaline solution is too high to measure $Cl^-$ ion.

  • Molten salt electrorefining process achieves uranium deposits at cathode using an electrochemical processing of spent nuclear fuel. In order to recover pure uranium from cathode deposit containing about 30wt% salt, the adhered salt should be removed by cathode process (CP). The CP has been regarded as one of the bottle-neck of the pyroprocess as the large amount of uranium is treated in this step and the operation parameters are crucial to determine the final purity of the product. Currently, related research activities are mainly based on experiments consequently it is hard to observe processing variables such as temperature, pressure and salt gas behavior during the operation of the cathode process. Hence, in this study operation procedure of cathode process is numerically described by using appropriate mathematical model. The key parameters of this research are the amount of evaporation at the distillation part, diffusion coefficient of gas phase salt in cathode processor and phase change rate at condensation part. Each of these conditions were composed by Hertz-Langmuir equation, Chapman-Enskog theory, and interphase mass flow application in ANSYS-CFX. And physical properties of salt were taken from the data base in HSC Chemistry. In this study, calculation results on the salt gas behavior and optimal operating condition are discussed. The numerical analysis results could be used to closely understand the physical phenomenon during CP and for further scale up to commercial level.

A Basic Study on Separation of U and Nd From LiCl-KCl-UCl3-NdCl3 System (LiCl-KCl-UCl3-NdCl3 system에서 U 및 Nd 분리에 관한 기초연구)

  • In case of high contents of rare earths in the LiCl-KCl salt, it is not easy to recover U and TRU metals as a usable resource form from LiCl-KCl eutectic salts generated from the pyroprocessing of spent nuclear fuel. In this study, a conversion of $UCl_3$ into an oxide form using $K_2CO_3$ and an electrodeposition of $NdCl_3$ into a metal form in $LiCl-KCl-UCl_3-NdCl_3$ system were conducted to resolve the problem. Before conducting the conversion, experimental conditions for the conversion were determined by performing a thermodynamic equilibrium calculation. In this study, almost all of $UCl_3$ disappeared in the LiCl-KCl salt when the injection of $K_2CO_3$ reached theoretical equivalent for the conversion, and then $NdCl_3$ was effectively electrodeposited as a metal form using liquid zinc cathode. After that, the LiCl-KCl salt became transparent, and uranium oxides were precipitated to the bottom of the LiCl-KCl salt. These results will be utilized in designing a process to separate U and rare earths in LiCl-KCl salt.

Effects of Auxin, GA and Cytokinin on the Protein Synthesis (Accumulation) of Soybean (Auxin, GA 및 Cytokinin이 대두의 단백질합성 (축적)에 미치는 영향)

  • Aqueous solutions of 2,4-D, BA or $GA_3(10^{-6},\;10^{-5},\;and\;10^{-4}M,\;respectively)$ were sprayed onto soybean (Glycine max) plants in the flowering stage, and proteins of immature (33days after flowering) and mature (77days after flowering) seeds were analyzed by electrophoresis to elucidate the effects of the growth reguators on protein synthesis or protein accumulation in the seeds. Accumulations of some proteins were altered by 2,4-D or BA at certain concentrations, but no proteins were affected by $GA_3$. The ${\alpha}\;and\;{\alpha}'$ subunits of 7S and acidic subunit of 11S disappeared in mature seeds after treatments at the flowering stage with 2,4-D or BA. The presence of ${\alpha}\;and\;{\alpha}'$ subunits of 7S and acidic subunit of 11S in immature seeds indicated that the absence of the above polypeptides in mature seeds did not result from inhibitions in syntheses of the polypeptides by the growth regulators. Disappearance of the above proteins in mature seeds seemed to be concerned with the action of specific proteolytic enzyme (s) (metalloendopeptidase?), and 2,4-D and BA might promote gene expression or activation of the enzyme.

A Development of Telephone for the Hearing Impaired to Improve Listening Ability of Telephone Speech (난청인의 통화 청취도 향상을 위한 전화기 개발)

  • We developed a new hearing aid telephone which helps the hearing impaired person to improve the listening ability of telephone speech. Recently, the hearing impaired person and the elderly who has hearing loss have been continuously increased and their desire for participating society as a producer has been increased also. So they strong1y want the hearing aid devices which make compensation fortheir handicap. The hearing aid telephone is one of the basic aid devices that helps the hearing impaired to communicate well with other poeple and to acquire easily useful information through the phone. We analyze the hearing ability of the hearing impaired, design the new model of the hearing aid telephone and test the telephone in three fields-electrical, word perception, user test. Our new tolephone has lour band pass filter channels and the center frequencies of these filters are 500, 1000, 2000, 3000Hz which are considered psychoacoustic factors and telephone line characteristics. The hearing impaired can adjust the total gain characteristics of receiving sound to his hearing ability by setting four volumes in the telelphone. This procedure is called fitting which is a very important factor for the hearing impaired to take meaning of speech. The total gain of this telephone is over 20dB from 250Hz to 3200Hz range. From the results of the tests we certify that our new model is better for the hearing impaired to understand the meaning or telephone speech than the old general models. The next step of developing the hearing aid telephone is to study about compressing sidetone and noise, dividing frequency bands, selecting hearing aid pattern and compensating psychoacoustic loudness. we expect that the advanced hearing aid telephone can be developed by the research about speech perception characteristics of the hearing impaired in engineering and clinical side.

Tracking Interdisciplinary Relationships between Scientific Researches in Energy Fields using Bibliometic Analysis (과학기술논문을 통한 에너지 연구 분야의 다학제 동태 추적분석)

  • In this paper, we attempted to analysis and track dynamic properties of the interdisciplinary relationship between scientific research areas in energy field using bibliometric analysis. We created network maps with SCs (subject categories) defined from $WoS^{\circledR}$ (Web of Science, Thomson Scientific ISI, Philadelphia, USA) using co-occurrence analysis method in order to identify overall disciplines directly linked with energy fields and investigate the change of interaction between SCs as a function of time. From the results of this study, thermodynamics, fuels, chemistry/chemical engineering, and electrochemistry have differentiated into more specific disciplines while the strength of interaction with energy field gradually has increased. Meanwhile, "nuclear physics" was developed to "nuclear science & technology" toward applicable target sector and also the interaction of "environmental science" with "energy generation area" among various energy disciplines recently showed the radical increase as compared with the values of 4 years ago. Finally, through combinative reviews of today's energy policy established by South Korea government, this study will give a help for keeping up with national energy agenda meeting the diverse characteristics of academic disciplines of energy field. In addition, our results support that the use of such network analysis based on bibliometric analysis to discern shifts in academic R&D strategies and target sectors.

An Analysis of the Linked Structure for Technology-Industry in National R&D Projects (국가 R&D과제의 기술-산업 연계구조분석)

  • Technology is closely related to industrial development and various studies have been performed to understand the linked structure for knowledge flow between the technology and industry. The research, however, wasn't carried out to flow for Korea National Research and Development projects. In this study, linked structure for technology-industry was discussed by utilizing patent data performed in actual National R&D using NTIS Information of the national research and development, and then it was analyzed how knowledge flows between the technology and industry are flowing. It should be defined that the individual applications expected by researchers at the start of the research and technology-industry applications actually applied from the research performances after research was completed. As a result, it was confirmed in most projects the flow of knowledge was occurring to predicted industries before the start of the R&D. However, the technology was applied to unexpected industry in three industries such as Y09(medical, precision and optical instruments), Y10(electrical and mechanical equipment), Y11(automotive and transportation equipment). The results of this study will be able to contribute to planning for efficient investment strategy of technology-industry by investigating the technology-industry knowledge flow relations of national R&D projects.

Purification and Properties of Sunflower Seed $\alpha$-Galactosidase by Affinity Chromatography. (Galactomannan 이용에 관한 연구;Affinity Chromatography법에 의한 해바라기씨 유래 $\alpha$-Galactosidase의 정제 및 성질)

  • An ${\alpha}$-D-galactosidase (${\alpha}$-D-galactoside galactohydrolase, EC 3. 2. 1. 22) from sunflower seed was purified by affinity chromatography using N-$\varepsilon$-aminocaproyl-${\alpha}$-D-galactopyranosylamine coupled to sepharose and its properties were examined. The specific activity of the purified enzyme, tested with p-nitrophenyl-${\alpha}$-D-galactopyranoside as substrate, was 291.66 units/mg protein, representing an 115-folds purification of the original crude extract. The final preparation obtained from by Sephadex G-25 chromatography showed a single band on SDS-polyacrylamide gel electrophoresis. The molecular weight was determined to be 42,000 by SDS-polyacrylamide gel electrophoresis. The purified galactosidase showed maximum activity at pH 4.5 and 55$^{\circ}C$, and was stable in the pH and temperature ranges of 4.0 to 5.0 and 30 to 55$^{\circ}C$, respectively. The enzyme activity was inhibited by Ag$\^$2+/, Hg$\^$2+/ and Co$\^$2+/. The enzyme activity was not affected considerably by treatment with other metal compounds. The enzyme liberated galactose from melibiose, raffinose, copra galactomannan, guar gum and locust bean gum by TLC, and also the hydrolysis rate of substrate was compared by HPLC.

  • [ ${\alpha}$ ]Galactosidase was purified from a culture filtrate of Mortierella sp. by CM-sephadex C-50, and subsequent Sephadex G-100 column chromatography. The final preparation thus obtained showed a single band on SDS-polyacrylamide gel electrophoresis. The molecular weight was determined to be 56 kDa. $Gal^3Man^4$ ($6^3$-mono-O-${\alpha}$-D-galactopyranosyl-4-O-${\beta}$-D-mannotetraose), $Gal^{2,3}Man_5$ ($6^{2,3}$-di-O-${\alpha}$-D-galactopyranosyl-4-O-${\beta}$-D-mannopentaose), $Gal_2Man_3$ ($6^2$-mono-O-${\alpha}$-D-galactopyranosyl-4-O-${\beta}$-D-mannotriose), $Gal^2Man_6$ ($6^2$-mono-O-${\alpha}$-D-galactopyranosyl-4-O-${\beta}$-D-mannohexaose) and $Gal^2Man_5$ ($6^2$-mono-O-${\alpha}$-D-galactopyranosyl-4-O-${\beta}$-D-mannopentaose), prepared from 3 types of microbial ${\beta}$-mannnanase, were used as substrates. $Gal^3Man_4$ and $Gal^2Man_3$ had a stubbed ${\alpha}$-galactosyl residue on the $2^{nd}$ and $3^{rd}$ mannose from the reducing end of mannotetraose and mannotriose, thus ${\alpha}$-galactosidase showed a preference for stubbed ${\alpha}$-galactosyl residue. ${\alpha}$-Galactosidase hydrolyzed $Gal^3Man_4$ more rapidly than $Gal^2Man_3$. However, ${\alpha}$-galactosidase hardly acted on $Gal^{2,3}Man_5$, $Gal^2Man_6$ or $Gal^2Man_5$. The enzyme hydrolyzed melibiose to galactose and glucose, raffinose to galactose and sucrose, and also stachyose to galactose and raffinose.
