Emission Rate of Greenhouse Gases from Bedding Materials of Cowshed Floor: Lab-scale simulation study (우사깔짚에서 발생되는 온실가스 배출량 산정: 모의 실험결과)

  • To know the emission amount of greenhouse gases from bedding materials of cowshed floor, the emission rates of methane ($CH_4$) and nitrous oxide ($N_2O$) gases from a simulated cowshed floor (SCF) with sawdust that manure loading rate into the bedding material could be accurately controlled were assessed in this study. The manure loading rates of Korean beef and Holstein dairy cattle into the SCF of $0.258m^2$ surface area with 10 to 15 cm height sawdust were $1.586kg/m^2/d$ and $3.588kg/m^2/d$, respectively, and those were calculated on the basis of "Standard model for sustainable livestock" and "Data for excretion amount of manure from livestock". All experiments were done in triplicates in three different seasons (May to July, Sep. to Nov., and Feb. to Apr.) using 12 SCFs. The effects of bedding material thickness on $CH_4$ and $N_2O$ emission from SCFs for both Korean beef cattle and Holstein dairy cattle were not statistically significant (p<0.05). Emission amount of $CH_4$ and $N_2O$ per square meter of SCF for Holstein dairy cattle was 7.5 and 1.2 times higher than that of Korean beef cattle, respectively. The yearly $CH_4$ amount per head was 17.7 times higher in Holstein dairy cattle, obtaining 130.4 g/head/year from SCF for Holstein dairy cattle and 7.4 g/head/year from SCF for Korean beef cattle, and $N_2O$ was also 3.8 times higher in Holstein dairy cattle (3,267 g/head/year in Korean beef cattle and 14,719 g/head/year in Holstein dairy cattle). However, the $N_2O$-N per loaded nitrogen into SCF was higher in Korean beef cattle, having 0.2148 and 0.1632 kg $N_2O$-N/kg N in Korean beef cattle and Holstein dairy cattle, respectively, and those values were 3.07 and 2.33 times higher than that of Intergovernmental Panel on Climate Change (IPCC) 2006 guideline (GL) (0.07 kg $N_2O$-N/kg N).

Predictive indicators of coronary artery complications in Kawasaki disease (가와사키 병 환아에서 관상동맥 합병증의 예측인자)

  • Purpose : Kawasaki disease—the most common cause of acquired heart disease in children—incidence is increasing yearly. Therefore, we evaluated the predictive indicators of coronary complications of Kawasaki disease based on clinical and laboratory data. Methods : Between January 2005 and March 2008, of the 201 children with Kawasaki disease treated at the Gil Hospital of Gachon University of Medicine and Science, 51 had coronary artery lesions (Group II) and 150 had no lesions (Group I). The reasons for coronary artery lesions were deduced from the clinical and laboratory data. Results : Analysis of the 2 groups revealed that fever duration and days of fever after and before initial intravenous gammaglobulin (IVIG) treatment were significantly longer in Group 2 than in Group I. IVIG infusions were statistically higher in Group II than in Group I. As per the laboratory data, C-reactive protein (CRP) value was significantly higher in Group II. Collectively, >10 days of fever duration, >48 h of fever duration after, and >10 days of fever before IVIG treatment increased the risk of coronary artery lesions 6-, 5-, and 3.5-fold, respectively. Furthermore, additional IVIG courses and higher CRP level increased the risk of coronary artery lesions 4-fold and 2-3-fold, respectively. Conclusion : The following 3 factors were responsible for increased risk of coronary artery lesions in children with Kawasaki disease: fever duration and days of fever after and before IVIG treatment. To identifythe predictive indicators of coronary complications, it is necessary to further elucidate the relationship between well-known forecasting factors.

Production of a Fermented Korean Pear Puree using a New Strain Leuconostoc mesenteroides KACC 91495P Isolated from Kimchi (김치에서 신규 Leuconostoc mesenteroides KACC 91495P 균주의 분리 및 이를 이용한 배 발효물의 제조)

  • A lactic acid bacterial strain showing fast growth and high acid production in Korean pear puree was isolated from Kimchi. This strain was analyzed by API 50 CHL kit and 16S rRNA sequencing analysis and identified as Leuconostoc (Ln.) mesenteroides KACC 91495P. Korean pear puree was fermented using Ln. mesenteroides KACC 91495P strain at $30^{\circ}C$ for 18 h. The changes of pH, titratable acidity and viable cell number during fermentation were investigated. The pH and titratable acidity were reached to pH 3.86 and 1.09% after 18 h fermentation, respectively. The viable cell population of Ln. mesenteroides KACC 91495P was rapidly increased to $2.0{\times}10^9\;CFU/g$ during the 9 h of cultivation. The contents of lactic acid, acetic acid and malic acid were determined to be 0.213, 0.259, and 0.217% after 18 h fermentation, respectively. The content of polyphenolic compounds, known as antioxidants, in pear puree were enhanced by Ln. mesenteroides KACC 91495P cultivation. The level of total polyphenolic compounds was increased to around 140% of initial concentration. When the fermented pear puree was kept at $4^{\circ}C$, pH, titratable acidity and number of viable cells population were nearly maintained for 13 days.

Production of Tannin from Hairy Root Cultures of Rheum undulatum L. (대황 모상근의 배양에 의한 tannin 생산)

  • Hairy root cultures of Rheum undulatum L. induced by a co-culture with Agrobacterium rhizogenes ATCC15834 were established and the production of tannin in hairy roots was investigated. The growth of hairy roots was maximized in WPM liquid medium supplemented with 2 mg/L IAA, and 3% sucrose (pH 5.7). The highest yields of tannin were obtained from WPM medium containing 0.5 mg/L ABA, and 5% sucrose (pH 5.5). The growth and tannin production in hairy root cultures were influenced by the addition of elicitors to the medium. The addition of 50 mg/L chitosan enhanced the production of tannin with about 1.7 fold.

Effects of Temperature and Photoperiod on the Growth of Tatary Buckwheat(Fagopyrum tataricum) (온도 및 일장처리가 쓴 메밀(Fagopyrum tataricum)의 생육에 미치는 영향)

  • This study was carried out to investigate the effects of temperature and photoperiod on the growth of Fagopyrum tataricum. It showed a tendency to promote the germination rate and plant height of tartary buckwheat under high temperature. Plant height and number of leaves were promoted in the order of 25 &gt; 20 &gt; 15 &gt; 3$0^{\circ}C$ under the different temperature after 30 days of the planting. In the 10, 12, 14hr photoperiods at 2$0^{\circ}C$, plant height, number of leaves and dry weight were increased as much as photoperiod became long except by 16hr. Rutin contents was not regular both common and tartary buckwheat under the photoperiods. The rutin content in leaves was higher than stem. Compared to yield of tartary and common buckwheat, plant height, number of leaves, number of branch and stem diameter of tartary buckwheat increased more than common buckwheat. Especially, number of leaves was increased about 2.5 times more than common buckwheat. And there was a difference in about 2.1 times in grain weight and about 5.4 times in number of grains. Rutin content was 1469.8mg/l00g in grain of tartary buckwheat. It was about 60 times higher than 22mg/100g in grain of common buckwheat.

Laparoscopy Application to Determine Estrous Cycle in Korean Black Goats (복강경을 이용한 한국흑염소의 성주기 판정)

  • This study was performed to determine the estrous cycles by macroscopic observation of the ovarian changes using the laparoscopy and to make use of these results for embryo transfer in Korean black goat (Copra hircus aegagrus). Laparoscopic examinations of the ovaries were performed from 2 days after $CIDR^(R)$ removal to 22 days after ovulation. The serial morphological changes of follicles and corpus luteum (CL) were observed. CL was classified corpus hemorrhagicum(CH), corpus luteum (CL) and corpus albicans (CA) by its maturation and regression. On the day before ovulation (Day 0), Graafian follicles (GF) were found on one or both ovaries. On the day (Day 1) and $2^{nd}$day (Day 2) of ovulation, and ovulation depression (OD) and an early stage corpus hemorrhagicum $(CH_1)$ were observed at the site of GF, respectively. On Days 3 to 4, more developed and enlarged corpus hemorrhagicum $(CH_2\;and\;CH_3)$ arised from the ovulation of the GF with well vascularization. On Days 5 to 6, it was identified that mature corpus luteum $(CL_3)$ was grown on the ovary, and fully developed CL with adjacent follicles were occupied most part of the ovary on Days 17 and 18. Then the size of CL was diminished, and completely luteal regression $(CL_1\;or\;CA)$ with new large follicle was identified on Days 20 and 22. From these results, the 4 stages of the estrous cycle in Korean black goats were 1) estrus (Day 0) for 1 day, 2) metestrus $(Day\;1{\sim}4)$ for 4 days (stage of CH development), 3) diestrus $(Day\;5{\sim}16/17)$ for 12 or 13 days (luteal stage), and 4) proestrus $(Day\;17/18{\sim}20/22)$ for 4 or 5 days (stage of luteal regression and follicular growing). Laparoscopy for observation of ovarian changes was invasive than laparotomy. Additionally, it had advantages of reduced adhesion and quick operation time. It was considered that laparoscopic examination of ovarian changes will be useful for embryo transfer in the Korean black goats.

Shoot Cutting Effects on the Productivity and Nutrient Removal of Some Wetland Plants (습지식물의 지상부 제거가 생산력과 영양염류 제거량에 미치는 효과)

  • This work focused on the effects of the timing and the frequency of shoot cutting to maximize the productivity and the nutrient removal of three emergent macrophytes, Phragmites communis, Zizania latifolia and Typha angustifolia in natural wetlands. Shoot cutting significantly enhanced biomass production and resulted in more nitrogen and phosphorus removal from water in these three experimental species, compared to those of control. However, the frequency and the timing of shoot cutting, and the enhancement ratio were different among three species. For Phragmites stands, the highest productivity was 1.9 times of control in June treatment of the first year experiment, while 1.3 times in May treatment of the second year experiment. Zizania and Typha stands were both 1.2 times of control in August treatment and June and August treatment. Calculating the total annual removal rate of nitrogen and phosphorus based on the highest productivities among treatments, in Phragmites stands, 2.0 times of nitrogen and 1.8 times of phosphorus were removed in the first year, and both 1.4 times in the second year experiment. Likewise, for nitrogen and phosphorus; 2.4 and L.8 times in Zizania stands, and 1.8 and 1.9 times in Typha stands were removed. Overall, these results suggested that cutting treatment of shoots be effective. Thus, shoot cutting of two times during a growing season were recommended to maximize the effects: that is, in May or June, and October for Phragmites stands, and in August and October fur Zizania and Typha stands.

Effect of Macromolecules Supplemented to NCSU-23 and PZM Culture Media on in vitro Development of Porcine Embryos (NCSU-23과 PZM 배양액내 첨가된 Macromolcule이 돼지 체외수정란의 발육에 미치는 영향)

  • Three experiments were performed to develop an optimal culture system of in-vitro derived porcine embryos. In experiment 1, embryos were cultured in two different basic culture media (NCSU-23 and PZH) supplemented with BSA (4mg/ml), FBS (10%), PVA (3mg/ml). Although no difference on the percentage of embryos developed to blastocysts was found (P>0.05), more 2-cell embryos (59.5% vs. 44.7 to 52.0%) were obtained when embryos were cultured in NCSU-23 added with BSA. In experiment 2, identical treatment was conducted using PZM medium. More (P<0.05) 2-cell embryos (54.4% vs. 40.7 to 44.5%) and blastocysts (12.5% vs. 5.8 to 9.0) were produced when embryos were cultured in PZM added with BSA (4mg/ml). In experiment 3, embryos were cultured in media as follows; 1) NCSU-23 without BSA for 192 hours; 2) NCSU-23 with BSA for 192 hours; 3) NCSU-23 with BSA for 96 h and then subsequently injected with FBS into culture drops. Although no difference on the percentage of embryos developed to blastocysts was found (P>0.05), more 2-cell embryos (63.8 to 69.2% vs. 52.0%) were obtained when embryos were cultured both in NCSU-23 added with BSA throughout culture duration and in FBS supplemented medium. In conclusion, no difference was found when porcine preimplantation embryos were cultured in NCSU-23 added with various macromolecules. However, when PZM medium was used, PZM medium supplemented with BSA showed more potential to support embryo development. Finally, FBS supplemented into culture drop 96h post-insemination did not show any benefits on embryo development.

Sensitivity dependence on the effective surface area for cholesterol biosensor fabricated by silanization process (실란화 공정으로 제작된 콜레스테롤 센서의 전극 표면적에 따른 감도 특성)

  • 센서의 소형화 되는 추세에 전극 면적은 sensitivity의 중요한 요인이다. 본 연구에서는 콜레스테롤을 측정하기 위해 각각 planar 전극과 porous 전극에 효소 고정화 방법으로 covalent binding인 silanization 공정을 이용하여 전극 면적에 따른 전기화학적 감도를 비교하였다. Handles-Sevcik equation을 이용하여 전극 면적을 구한 결과 planar 전극의 경우 0.1608 $cm^2$, porous 전극의 경우 0.5054 $cm^2$로 porous 전극 면적이 planar전극에 비해 약 3.1배 증가하였다. 또, planar 센서의 sensitivity는 0.08567 ${\mu}A/mM{\cdot}cm^2$이고 porous 센서의 sensitivity는 planar sensor에 비해 약 3.1배 증가한 0.2656 ${\mu}A/mM{\cdot}cm^2$였다. 따라서 porous sensor의 sensitivity의 증가는 전극 면적의 증가에 따른 것이라고 할 수 있다.

Quench characteristics of the SFCL using YBCO film (YBCO 박막형 초전도 한류기의 퀜치 특성)

  • YBCO film을 이용하여 2 stripe-meander 형태의 초전도 한류기를 제작하고 시험하였다. 초전도 film의 hot spot에 의해 발생하는 열을 분산시키기 위해 gold shunt layer를 입혔다. 사고각 $0^{\circ}$에서 사고발생 후 최초 quench 전류값은 $9.56A_{peak}$이었고 약 0.63 msec동안 fast quench 가 진행되고 이후에는 gold층의 열발생에 의한 저항증가가 약 3주기동안 진행된 후 안정되었다. 사고각 $45^{\circ}$$90^{\circ}$ 에서는 전류가 상승하는 도중 혹은 최고지점에서 사고가 발생했기 때문에 fast quench time이 각각 0.56 msec와 0.26msec를 보였다. 초전도 한류기를 적용하였을 때 사고전류는 사고직후 약 5배, 3주기후에는 약 8.5배까지 효과적으로 제한함을 알 수 있었다.
