Role of Temperature and Gibberellic Acid in Stimulating Embryo Elongation and Radicle Emergence of Hepatica asiatica Nakai Seeds

  • This research was performed to determine the temperature requirements for embryo growth and radicle emergence of Hepatica asiatica Nakai, a perennial herb native to Korea. Seed viability, embryo growth, and radicle emergence were monitored in seeds exposed to various temperatures (10, 15, $20^{\circ}C$ and $30{\rightarrow}15^{\circ}C$). Laboratory experiments at various temperatures revealed that (1) embryo elongation occurred effectively between 10 and $15^{\circ}C$; (2) radicle emergence occurred only at $15^{\circ}C$; (3) a warm stratification (2-8 weeks at $30^{\circ}C$) was not required for embryo elongation and radicle emergence, and led to inhibition of radicle emergence; (4) application of gibberellic acid ($GA_3$) promoted embryo growth, but not radicle emergence. These results suggested that H. asiatica seeds have two separate mechanisms to overcome dormancy, either by $GA_3$ (morphological dormancy) or temperature (physiological dormancy), and warm stratification is not involved in breaking radicle dormancy. These findings indicate that H. asiatica seeds have morphophysiological dormancy.

Changes of Physico-chemical Properties during the Leaf Development and Senescence of Tobacco Plant (담배의 엽 발달 및 노화과정 중 이화학성 변화)

  • This experiment was conducted to obtain the basic information on the morphological and physiological changes in tobacco leaf during the growth period by measuring the changes of chlorophyll, sugar, lipid and mineral contents in tobacco plant. Leaf length and width have been fully developed at 25 days after leaf emergence. Dry weight was rapidly increased between 10 and 15 days after leaf emergence and reached the highest at 30 days. Crude lipid content, palmitic acid, and the major saturated fatty acid were increased with progressing senescence, while unsaturated fatty acid including linolenic acid was decreased as the senescence was advanced. The total nitrogen content showed the highest value at IS days after leaf emergence. On the other hand, the total sugar content showed the highest value at 45 days after leaf emergence and glucose, fructose and sucrose were decreased with leaf development and increased at the end of senescence. The content of chlorophyll showed the highest value at 15 days after leaf emergence and began to decrease at 30 days after leaf emergence. The contents of p, Cu, Zn, and Fe in tobacco leaves were decreased by the end of senescence after leaf emergence but those of Ca, Mg, and Mn in leaves were increased.   • The emergences of Echinochloa glabrescens, Monochoria vaginalis, and Cyperus difformis seeds were little affected by fertilizer application. All shading treaments resulted in decrease in emergence of E. glabrescens, while shading up to 67% slightly stimulated emergence of M. vaginalis and C. difformis. The peak emergence of E. glabrescens was at two week after sowing in the full sunlight, 31, and 52% shading, but no peak emergence at the 67% shading. Emergence peaks of C. difformis were observed during 6 to 8 weeks after sowing for all shading levels employed.

  • The experiment were carried out to establish the optimal condition of bulblet size, low temperature treatment and time of incubation in acclimatization on stem emergence of Lilium oriental hybrids. The large size bulblet was better than middle and small size bulblets in percentage of stem emergence and weight of bulblets. The highest percentage of stem emergence was 95% in large size bulblet 'Acapulco' but small size bulblet of 'Casablanca' was no stem emergence. Low temperature treatment(5℃)for breaking dormancy was needed at least more than 9weeks. Weight of bulblets and percentage of stem emergence was not good in 5℃ treatment for 6 weeks. Germination and Seedling Emergence of Ammannia coccinea as Influenced by Environmental Factors

  • Petri dish and pot experiments were conducted to investigate germination and seedling emergence of Ammannia coccinea as influenced by environmental factors. The best germination of A. coccinea was obtained at $35/30^{\circ}C$ of temperature and 0 bar of osmotic potential, while no germination at temperatures of ${\leq}$ $15^{\circ}C$ and ${\geq}$ $40^{\circ}C$, osmotic potentials of ${\leq}$ -2.0 bar, or dark condition. The best seedling emergence was observed at $35/30^{\circ}C$, at which the first emergence of A. coccinea was observed at 7 days after sowing (DAS) with its maximum emergence reached at 10 DAS. No seedling emergence was observed at $15/10^{\circ}C$ with significant reduction at $40/35^{\circ}C$. Seedling emergence decreased with increasing soil depth, resulting in no seedling emergence at ${\geq}$ 3 cm. The Gompertz model well described the cumulative germination and seedling emergence of A. coccinea with time. Germination influenced by osmotic potential and seedling emergence influenced by soil burial depth were well described by the logistic model. Overall results indicate that A. coccinea is photoblastic and requires temperatures greater than $15^{\circ}C$, osmotic potential greater than -2.0 bar, and soil burial depth shallower than 3 cm for its germination and seedling emergence, which were faster than M. vaginalis but slower than E. crus-galli.

  • In oder to find out the effective seed germination testing method to the field emergence, an experiment was conducted ; 1. TTC testing results were higher percentage than any other germination testing methods . 2. On the corn seed , field emergence was highly correlated with germinator test, TTC test and AA test hut cold test was lower percentage . 3. Field emergence , on the soybean seeds was highly correlated with AA test and cold test but germinator test and TTC test was higher percentage .

  • Stratified American ginseng(Panax quinquefoilium L.) seeds were planted in a shaded greenhouse at four depths and in four different soil types to observe effects on emergence rate and root size. Seeding depth affected seedling emergence rates and the number of days required to complete emergence. The shape of the roots was affected by the texture of soil, especially percentage of sand.

Effect of Soil Temperatures on Emergence and Early Growth of Eleocharis kuroguwai, Potamogeton distinctus, and Sagittaria pygmaea (토양온도(土壤溫度)가 올방개, 가래 및 올미의 출아(出芽)와 초기생장(初期生長)에 미치는 영향)

  • Emergence and early growth of Eleocharis kuroguwai, Potamogeton distinctus, and Sagittaria pygmaea were examined at 10, 14, 18, 22, 26, and $30^{\circ}C$ of soil temperatures using soil temperature gradient bath. Eleocharis kuroguwai emerged at $14^{\circ}C$ or higher temperature and % emergence was increased as soil temperature increased up to $26^{\circ}C$ but decreased at $30^{\circ}C$. Percentage of emergence in Potamogeton distinctus was very low at $10^{\circ}C$ but most of them were emerged at $14^{\circ}C$ or above. Emergence rate was faster as soil temperature increased. Percentage of emergence in Sagittaria pygmaea was increased until 10 days after planting as soil temperature increased but final percentage of emergence was 100% at all temperature. Number of days required for 80% emergence was shortened as soil temperature increased and Eleocharis kuroguwai and Potamogenton distinctus required longer duration to emerge compared to Sagittaria pygmaea. Eleocharis kuroguwai and Potamogeton distinctus required higher accumulated soil temperature for emergence compared to Sagittaria pygmaea. Optimum soil temperature for early growth after emergence of Eleocharis kuroguwai, Potamogeton distinctus, and Sagittaria pygmaea were 26, 18-22, and 18-$26^{\circ}C$, respectively.

  • Adult eclosion and emergence behavior of the black pine base scale, Matsucoccus thunbergianae Miller and Park, was studied in a laboratory. Adult eclosion in males did not appear to be limited to a specific time of the day. Newly molted male adults, at room temperatures, usually remained in the cocoons as pre-emergence adults for 0.5-20. days before emerging; most males emerged within one hour after the onset of photophase, and emergence was faster at higher light intensity. Most females molted into adults within three hours after the onset of photophase, and they were active immediately after molting. Quiescence in males appears to help the scales concentrate their sexual activity within a particular period of the day as well as to protect newly molted adults until sexual and locomotive maturity.

  • The present study elucidates of the effect of less water stress in different temperature condition on the emergence of forage grasses. Water condition was controlled to 30% and 60% by water content by wet soil. The mean temperature is conducted by $10^{\circ}C$ (out side) and $20^{\circ}C$ (glass house). The results are as follows: 1. Mean emergence time and emergence day after sowing of grasses were greatly influenced by water content of soil and temperature. It was suggested that temperature was very important for the light competition with weed in the early growth of grass. 2. Accumulatied emergence of grasses was nat afected by temperature, but it was sensitively affected by water content of soil.
