Crude Extract and Solvent-Partitioned Fractions of the Halophyte Atriplex gmelinii Inhibit Adipogenesis in 3T3-L1 Preadipocytes (3T3-L1 지방전구세포에서 염생식물 Atriplex gmelinii의 조추출물과 용매 분획물의 지방세포분화 억제)

  • Objectives: Atriplex gmelinii C. A. Meyer is a halophyte belonging to the Chenopodiaceae family, and its young leaves and stems are used as fodder for livestock. The aim of the present study was to investigate the effects of A. gmelinii extract and its solvent fractions on lipid accumulation during adipogenesis of 3T3-L1 preadipocytes. Methods: The samples of A. gmelinii were separately extracted using methylene chloride and methanol. Subsequently, they were combined to formulate the initial extract, which was then partitioned based on polarity to prepare solvent fractions. Oil Red O staining was employed to measure lipid accumulation during the differentiation of 3T3-L1 preadipocytes. To verify cytotoxicity in 3T3-L1 cells, MTT assays were conducted. The expression levels of transcription factors in 3T3-L1 preadipocytes were measured through Western blotting analysis. Results: At 50 ㎍/mL, treatment of A. gmelinii extract and its solvent fractions during the differentiation of 3T3-L1 preadipocytes significantly diminished lipid accumulation with no noteworthy cytotoxicity on cell viability. Additionally, when investigating the biochemical pathways that underlie the prevention of lipid accumulation using solvent fractions, it was found that the n-BuOH and n-hexane fractions significantly decreased the expression of key transcription factors involved in the generation of fat, such as peroxisome proliferator-activated receptor γ (PPARγ), CCAAT/enhancer binding protein α (C/EBPα), and sterol regulatory element-binding protein-1c (SREBP1c). Conclusions: These findings indicate that A. gmelinii can effectively reduce the accumulation of fat in 3T3-L1 adipocytes, making it a potentially valuable material for mitigating and preventing obesity.

  • Ascorbic acid(AsA) is unevenly distributed throughout all body cells and fluids. Multiactivities of AsA in many biological systems and in various scientific fields were reported. In this study we aimed to clarify the inhibitory action of high concentration of AsA on the cell growth in 3T6 fibroblasts. The cells wee exposed to AsA at various concentration. It showed that 3T6 fibroblasts wee dead by the medium which contained AsA at the concentration higher than 0.5 mM. AsA caused hydrogen peroxide ($H_2O$$_2$) generation in a concentration dependent manner. These results suggested that the $H_2O$$_2$ was formed in the medium by AsA and acted as a cytotoxic gent. Moreover, it is supposed that hydroxyl radical (.OH) induced from $H_2O$$_2$also acetd as actively cytotoxic agent. This lethal effect of AsA causing the cell death was inhibited by the addition of catalase in the medium. Therefore, addition of AsA at the normal concentrations stimulate cell growth, but excess concentrations of AsA induce cell death.

Experimental Investigation on the Thermal Performance Enhancement of Cooling System for Vehicles using Water/Coolant-Based Al2O3 Nanofluids (물/부동액-기반Al2O3나노유체를 이용한 차량용 냉각시스템 성능 향상에 관한 실험적 연구)

  • In this study, the thermal performance of vehicle's cooling system is experimentally investigated using the water/coolant-based $Al_2O_3$ nanofluids as working fluids. For the purpose, the water/coolant-based $Al_2O_3$ nanofluids are prepared by twostep method with gum arabic. In order to obtain the well-suspended nanofluids, the agglomerated $Al_2O_3$ nanoparticles are precipitated using centrifugal force and the experiments are performed with supernatant of them. The thermal conductivity is measured by transient hot wire method and the thermal conductivity of nanofluids is enhanced up to 4.8% as compared to that of base fluids. Moreover, the cooling performance of water/coolant-based $Al_2O_3$ nanofluids is evaluated using vehicle's engine simulator under the constant RPM condition. The results show that the cooling performance of automobile engine increases up to 5.9% using prepared nanofluids. To investigate the effect of nanofluids on exhaust gas, the $NO_x$ emission is measured during the operation with respect to time and 10.3% of $NO_x$ emission is decreased. The experimental results imply that the water/coolant-based $Al_2O_3$ nanofluids might be used as a next-generation vehicles' coolant

  • The ethanol extract of peony root (Paeonia Lactiflora Pall, Paeoniaceae) and its major active components including gallic acid and methyl gallate were evaluated for their protective effects against free radical generation and lipid peroxidation. And protective effects against hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line were performed. The ethanol extract of peony root (PRE), gallic acid and methyl gallate were shown to possess the significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and were revealed the inhibitory effect of lipid peroxidation as expressed by malondialdehyde (MDA) formation. They were also found to strongly inhibit hydrogen peroxide-induced DNA damage from NIH/3T3 fibroblasts, assessed by single cell gel electrophoresis. Furthermore, oral administration of 50% PRE (50% ethanol extract), gallic acid and methyl gallate potently inhibited micronucleated reticulocyte (MNRET) formation of mouse peripheral blood induced by KBrO3 treatment in vivo. Therefore, PRE containing gallic acid and methyl gallate may be a useful natural antioxidant by scavenging free radicals, inhibition of lipid peroxidation and protecting oxidative DNA damage.

In addition, the protective effects against hydrogen peroxide-induced oxidative DNA damage in a mammalian cell line were examined. The ethanol extracts of the peony root (PREs) and its active constituents, gallic acid and methyl gallate, exhibited a significant free radical scavenging effect against 1,1-diphenyl-2-picryl hydrazine (DPPH) radical generation and had an inhibitory effect on lipid peroxidation, as measured by the level of malondialdehyde (MDA) formation. The PREs did not have any pro-oxidant effect. They strongly inhibited the hydrogen peroxide-induced DNA damage from NIH/3T3 fibroblasts, as assessed by single cell gel electrophoresis. Furthermore, the oral administration of 50% PRE (50% ethanol extract of peony root), gallic acid and methyl gallate potently inhibited the formation of micronucleated reticulocytes (MNRET) in the mouse peripheral blood induced by a $KBrO_3$ treatment in vivo. Therefore, PREs containing gallic acid and methyl gallate may be a useful antigenotoxic antioxidant by scavenging free radicals, inhibiting lipid peroxidation and protecting against oxidative DNA damage without exhibiting any pro-oxidant effect.

Mathematical Consideration on PV Cell Modeling (PV cell modeling의 수학적 고찰)

  • PV cell modeling is necessary both for software and hardware simulators in analyzing and testing the performance of PV generation systems. Unique I-V curve of a PV cell identifies its own characteristics by electrical equivalent model that is composed of diode constants ($I_o$, $v_t$), photo-generated current ($I_{ph}$), series resistance ($R_s$), and shunt resistance ($R_{sh}$). Photo-generated current can be easily estimated since it is proportional to irradiation level. However, other electrical parameters should be solved from the manufacturer's data sheet that is consisted with three remarkable operating points such as open circuit voltage ($V_{oc}$), short circuit current ($I_{sc}$), and maximum power voltage/current ($V_{MPP}/I_{MPP}$). This paper explains and analyzes mathematical process of a novel PV cell modeling algorithm that was proposed by the authors with the name of "K-algorithm".

Development of Bialaphos Resistant Transgenic Tabacco Plants by Pollination and Utilization of Fertilization Cycle (수분ㆍ수정 시기를 이용한 Bialaphos 저항성 형질전환 담배의 개발)

  • The herbicide bialaphos is a potent inhibitor of glutamine synthetase in higher plants. A bialaphos resistance (bar) gene encoding for an acetyltransferase was isolated from genomic DNA of Pseudomonas syringae pv tabaci. The bar gene was ligated to the binary vector pBI121. Pistils of tobacco plane were heated with the bar gene containing plasmid DNA at various times after pollination. When the treatment was applied at 30 and 40 h after pollination, a number of transgenic plants were obtained. Premary transformation (T$_{0}$ generation) and their progenies (T$_1$T$_2$) were resistant to both bialaphos and kanamycin at a dosage lathal to untransformed control plants. Stable integration of bar gene into chromosomal DNA was proven by Southern blot analysis of genomic DNA isolated from T$_1$progenies. These results show that the bialaphos resistant plane could be obtained by treatment to pistils with the exgenous bar gene through the fertilization cycle of tobacco.o.

The protective effects of Moxi-tar on injury induced by H2O2 in C6-glioma (H2O2로 유발된 뇌신경세포 상해에 대한 구진의 보호효과)

  • Objective : This study was produced to examine the effects of moxibustion that had been played important role to traditional oriental medical treatment on disease. Recently, it was reported that moxi-tar which is generated in the process of moxibustion as burning combustibles decreased NO and iNOS generation in C6-glioma and RAW 264.7 cells in our lab. Methods : C6-glioma cells were cultured in RPMI 1640 with FBS 10% in CO2 incubator. To study the protective effects of moxi-tar, we observed cell viability, DPPH activity, SOD activity, catalase activity and cell morphology after injury with $H_2O_2$. Results and Conclusions : Moxi-tar increased cell viability about twice as much as that of being injury by $H_2O_2$(moxi-tar $40{\mu}g/m{\ell}$, $H_2O_2$ $500{\mu}M$). And the results of free radical scavenger activity($80{\mu}g/m{\ell}$ : $78.91{\pm}4.4%$), SOD activity and catalase activity($80{\mu}g/m{\ell}$ : 21.6unit/mg protein) were increased by moxi-tar as dose-dependent manner. So we concluded that the effects of moxibustion which is played important role in Oriental medicine, might be free radical scavenger effects induced by moxi-tar. Conclusion : These results indicate that tBHP induces apoptosis through a lipid peroxidation-dependent mechanism and JS exerts the protective effect against the apoptosis by preventing peroxidation of membrane lipids.

Aspect ratio enhancement of ZnO nanowires using silicon microcavity

  • A great deal of attention has been focused on ZnO nanowires for various electronics and optoelectronics applications. in the pursuit of next generation nanodevices, it would be highly preferred if well-ordered ZnO nanowires of lower dimension could be fabricated on silicon. Before the growth of nanowires, silicon substrates were selectively etched using silicon nitride as masking layer. Vertical aligned ZnO nanowires were grown by metal organic chemical vapor deposition on patterned silicon substrate. The shape of nanostructures was greatly influenced by the micropatterned surface of the substrate. The aspect ratio, packing fraction and the number density of nanowires on top surface are around 10, 0.8 and $10^7\;per\;mm^2$, respectively, whereas the values are 20, 0.3 and $5\times10^7\;per\;mm^2$, respectively, towards the bottom of the cavity. XRD patterns suggest that the nanostructures have good crystallinity. High-resolution transmission electron microscopy confirmed the single crystalline growth of the ZnO nanowires along [0001] direction.
