This experiment was designed to improve the production efficiency of germ-line chimeric mice from phospholipase C (PLC)-$\beta$3 or peroxiredoxin (Prx) E -targeted ($\Delta$) ES cells by the investigating the manipulation conditions and characteristics of Jl ES cells. Four targeting clones were isolated to investigate the karyotypical and morphological stability prior to injection. All clones ($\Delta$PLC$\beta$-3 C3, $\Delta$Prx II C3, C10 and I5) showed more than 80% euploidism, however, most of $\Delta$PLC$\beta$-3 C3 clones were extensively differentiated compared to the other clones. Nine of 13 $\Delta$Prx II chimeras appeared to have at least 80% chimerism, whereas $\Delta$PLC$\beta$-3 C3 chimeras had 20% chimerism at most. Therefore, the morphological stability of ES cells under stable euploidism might mainly affect the production rate of high-coat chimeric mice. To increase the collection rate of injectable blastocysts (IBs), 5 to 10 week -aged C57BL/6J female mice were sacrificed at 3.5 days post-coitum. Ten week-aged mice were the most optimal IB donors by showing the highest collection rate (2.94/mouse) of injectable blastocysts without increase of non-injectable embryos (0.29/mouse). Foster mothers might be another factor because ICR x C57BL/6J F1 foster mother showed more increased productivity in litter size (2.8 vs. 5.6) and chimera (0 vs. 35.3%) than those of ICR foster mothers. In conclusion, the efficient production of germ-line chimeras mainly depends on the maintenance of ES cell morphology during targeting procedure, and the establishment of manipulation conditions might be a key point to maximize it.
This study was carried out to examine the effect of paint dyeing by Gloiopeltis paste in the various painting sizes and concentrations of paste. In the tonality diagram of Lab, surface colors were changed to +a(red), +b(yellow) and -b(blue) by treating of Gloiopelis paste and L value was the tendency of increasing. The penetration decreased with increasing concentration of paste, except for 3% of red and yellow. While it was decreased steadily in the case of Nue. The best penetration of dye was at 3% of concentration of Gloiopeltis paste and it's viscosity was measured 1500cps. As concentration of paste increased, ${\Delta}E_{ab}$increased after 40 hr. irradiation and water fastness and washing fastness test in red and blue. But in the case of ${\Delta}E_{ab}$yellow decreased slightly or not changed.

In this study, numerical model was developed to predict behavior of several layers in a solar pond and was solved by finite difference method. The empirical correlation that described heat and salt fluxes across interfacial boundary layer between mixed layer and diffusive layer in a solar pond were obtained from experiments and utilized in developing numerical model. As the results of this study, heat and salt fluxes across the interfacial boundary layer was found to depend on density ratio ${\beta}{\Delta}M_s/{\alpha}{\Delta}T.$ It was also found that the predicted value obtained by using the modified Weinberger's stability criteria showed a good agreement with experiment data.
A specific deleted version of ARABIDOPSIS RESPONSE REGULATOR1 (ARR1) lacking the signal receiver domain (1.152 amino acids)-coding sequence, referred to as $ARR1{\Delta}DDK$, was amplified using Arabidopsis thaliana cDNA prepared from adult leaves and transferred into the genome of Nicotiana tabacum cv. Samsun under the transcriptional control of a ${\beta}$-estradiol-inducible expression system. The ectopic expression of $ARR1{\Delta}DDK$ affected the morphology of transgenic seedlings and their segments in vitro. In the presence of an inducer, ${\beta}$-estradiol, ectopic expression of $ARR1{\Delta}DDK$ induced only the formation of soft, pseudo-bulbous tissue in the root tip region of intact seedlings, which appeared similar to callus generated on a hypocotyl segment in the presence of 2,4-D and 6-benzyladenine (BA), both at $1\;{\mu}M$. Those callus tissues on the root tip region could not generate shoots unless $1\;{\mu}M$ BA was supplied. In segment culture, ectopic expression of $ARR1{\Delta}DDK$ induced calluslike tissue around the cut-end of cotyledon and hypocotyl segments with occasional shoot formation, suggesting that the expression of $ARR1{\Delta}DDK$ could substitute for the effects of cytokinin on these segments. Additionally, treatment with only ${\beta}$-estradiol induced NtWUS, a WUS ortholog in tobacco, which was detected during the process of callus tissue formation in the root tip region and also in cotyledon or hypocotyl segments. These findings suggest that the NtWUS might be associated in the transdifferentiation process caused by the functional regulation of $ARR1{\Delta}DDK$ in transgenic tobacco seedlings.
To investigate the association between Korean red ginseng (KRG) intake in HIV-1 infected patients and occurrence of grossly deleted nef genes ($g{\Delta}nef$), we characterized nef genes in 10 long-term slow progressors (LTSP) infected with HIV-1 subtype B and 34 control patients. LTSP was defined whose the annual decrease in CD4 T cells was less than $20/{\mu}l$ over 10 years in the absence of antiretroviral therapy. They were treated with KRG for a prolonged period. Nef genes were amplified from peripheral blood mononuclear cells (PBMC) using nested PCR and products were sequenced directly. Patient CD4 T cell counts decreased from $444{\pm}207/{\mu}l$ to $294{\pm}177/{\mu}l$ over $136{\pm}23$ months of KRG intake. This corresponds to an annual decrease in the level of CD4 T cells of $13.3/{\mu}l$. A total of 479 nef genes were amplified from 137 PBMC samples. Nine out of the 10 patients, 47 (34.3%) out of the 137 samples, and 92 out of the 479 genes revealed $g{\Delta}nef$. The deletion extended outside the nef gene in 25 $g{\Delta}nef$ obtained from 6 patients. The proportion of samples with $g{\Delta}nef$ (34.3%) was significantly higher than 4.8% in control patients (P<0.001). In addition, it significantly increased as the duration of KRG intake prolongs (P<0.01). These data suggest the possibility that occurrence of $g{\Delta}nef$ might be associated with long-term intake of KRG.
Sulfur-containing amino acids play important roles in good flavor generation in Maillard reaction of non-enzymatic browning, so aqueous model systems of glucosamine and cysteine were studied to investigate the effects of reaction temperature, initial pH, reaction time, and concentration ratio of glucosamine and cysteine. Response surface methodology was applied to optimize the independent reaction parameters of cysteine and glucosamine in Maillard reaction. Box-Behnken factorial design was used with 30 runs of 16 factorial levels, 8 axial levels and 6 central levels. The degree of Maillard reaction was determined by reading absorption at 425 nm in a spectrophotometer and Hunter's L, a, and b values. ${\Delta}E$ was consequently set as the fifth response factor. In the statistical analyses, determination coefficients ($R^2$) for their absorbance, Hunter's L, a, b values, and ${\Delta}E$ were 0.94, 0.79, 0.73, 0.96, and 0.79, respectively, showing that the absorbance and Hunter's b value were good dependent variables for this model system. The optimum processing parameters were determined to yield glucosamine-cysteine Maillard reaction product with higher absorbance and higher colour change. The optimum estimated absorbance was achieved at the condition of initial pH 8.0, $111^{\circ}C$ reaction temperature, 2.47 h reaction time, and 1.30 concentration ratio. The optimum condition for colour change measured by Hunter's b value was 2.41 h reaction time, $114^{\circ}C$ reaction temperature, initial pH 8.3, and 1.26 concentration ratio. These results can provide the basic information for Maillard reaction of aqueous model system between glucosamine and cysteine.

The study aimed to compare the whitening efficacy of a strip and gel containing 2.9% hydrogen peroxide, using a tooth whitening light activator. The whitening effect was compared through in vitro and in vivo studies. In the in vitro study, we used stained hydroxyapatite (HAP) specimens as artificial teeth. HAP specimens were made using HAP powder and polyvinyl alcohol solution, and stained by modified Stookey's method. A whitening gel and whitening strip were applied to the respective specimens for 20 minutes, with a light activator. The color changes were measured using a colorimeter. In the in vivo study, one group (test 1) used the gel with a light activator and the other group (test 2) used a strip with the same activator for 15 minutes a day, for four consecutive days. An organoleptic evaluation using a Vita shade guide and instrumental evaluation using a Shade eye-NCC (Shofu Co., Japan) were performed. The color change values (${\Delta}E^*$) in the in vitro study revealed the strip with the light activator to be more effective than the gel with the same activator (p<0.001). In the in vivo study, even though there was no significant difference between the groups with respect to the ${\Delta}E^*$, using either the Shade eye-NCC or the Vita shade guide, the change in yellowness (${\Delta}b^*$) was statistically significant (p=0.024). In conclusion, test 2 group that used 2.9% hydrogen peroxide strip with a light activator, showed a tendency towards increased whitening than test 1 group that used the gel with the same activator; however further studies are needed to validate the above finding.
The purpose of this study was to assess the correlation between integrated mineral loss (volume % mineral${\times}{\mu}m$, ${\Delta}Z_{TMR}$) determined using transverse microradiography (TMR) and integrated reflectivity ($dB{\times}{\mu}m$, ${\Delta}R_{OCT}$) determined using optical coherence tomography (OCT) for detecting early dental caries with lesion depth more than $200{\mu}m$. Sixty tooth specimens were made from sound bovine teeth. They were immersed in a demineralized solution for 20, 30, and 40 days. The ${\Delta}R_{OCT}$ was obtained from the cross-sectional OCT image. The ${\Delta}Z_{TMR}$ was obtained from the TMR image. The correlation between ${\Delta}R_{OCT}$ and ${\Delta}Z_{TMR}$ was examined using Pearson correlation. The Bland-Altman plot was constructed using the ${\Delta}R_{OCT}$ and ${\Delta}Z_{TMR}$ values. A significant correlation between ${\Delta}R_{OCT}$ and ${\Delta}Z_{TMR}$ was confirmed (r=0.491, p=0.003). Moreover, most of the difference between ${\Delta}R_{OCT}$ and ${\Delta}Z_{TMR}$ was included in the error section of the Bland-Altman plot. Therefore, OCT could be used as a substitute for TMR when analyzing mineral loss in early dental caries.

The characteristics of the diesel spray have affected certain aspects of engine performance, such as the power, fuel consumption, and emissions. Therefore, this study was performed to investigate the effects of various injection parameters. In order to obtain the effect of injection parameters on diesel spray characteristics, the experiment is performed by using a high temperature and pressure chamber. The behaviors of the spray are visualized by using a high speed video camera, spray angle, penetration, and various other things. The results of the experiment are summarized as follows.
