Isolation and Characterization of 𝛽-Glucosidase-Producing Yeast, Rhodotorula sp. GYP-1 (𝛽-Glucosidase 생성 효모 Rhodotorula sp. GYP-1의 분리 및 특성)

  • Nine microbial strains were isolated from the byproduct of ginseng processing and field of ginseng cultivation. Two strains among them were confirmed. Phylogenetic analysis of these 𝛽-Glucosidase strains confirmed that strain GYP-1 belongs to the Rhodotorula and strain GYP-3-3 belong to genus Brachybacterium. Rhodotorula sp. GYP-1 was finally selected due to its high biomass production. The 𝛽-Glucosidase activity of Rhodotorula sp. GPY-1 was assessed at 30 ℃, and Higher than 70% of the enzyme activity was maintained at the temperature range of 20-40℃. Although the optimum pH for the highest enzyme activity was pH 5.0, the enzyme was stable throughout the pH range of 5.0-8.0. In addition, Rhodotorula sp. demonstrated antifungal activity against the ginseng root rot disease caused by Botrytis.

The Hair Growth Effects of Wheat Bran (밀기울의 모발 성장 효과)

  • This study was conducted to evaluate the effect of wheat bran on the promotion of hair growth. When rat vibrissa follicles were treated with n-hexane fraction of wheat bran, the hair-fiber lengths of the vibrissa follicles significantly increased. Moreover, n-hexane fraction of wheat bran was found to significantly induce the telogen-anagen transition in C57BL/6 mice. The fraction increased the proliferation of immortalized vibrissa dermal papilla cells (DPCs) in a dose dependent manner. To elucidate the molecular mechanisms in relation to proliferation of DPCs by the fraction of wheat bran, we examined the expression of cell cycle proteins and wnt/${\beta}$-catenin signaling components. Western blot analysis revealed that the proliferation of DPC by n-hexane fraction of wheat bran was accompanied by increased the level of cyclin D1, cyclin E, phospho-CDK2 and phospho-pRB. In addition, the fraction of wheat bran increased the level of phospho(ser552)-${\beta}$-catenin, phospho(ser675)-${\beta}$-catenin and phospho(ser9)-GSK$3{\beta}$. These results suggest that the hair growing potential of wheat bran mediated by proliferation of DPCs via the regulation of cell cycle proteins and Wnt/${\beta}$-catenin signaling.

Effects of Extraction Temperature and Time on Saponin Content and Quality in Raw Ginseng (Panax ginseng) Water Extract (수삼의 추출 온도 및 시간이 물 추출액의 사포닌 함량 및 품질에 미치는 영향)

  • In this study, raw ginseng water extract solutions were analyzed to set up the functional saponin content and quality optimization condition. The highest saponin content among the total raw ginseng water extracts was $74.6\;mg/100\;m{\ell}$ which was extracted at $75^{\circ}C$ for 24 hours. In addition, the saponin content decreased according to the increased extraction temperature and time. The highest total content of $Rb_2$ and Re was $19.9\;mg/100\;m{\ell}$ at $75^{\circ}C$ for 12 hours which decreased according to the increased extracted temperature and time. The highest prosapogenin ($Rg_2\;+\;Rg_3\;+\;Rh_1$) content among the total raw ginseng water extracts was $28.6\;mg/100\;m{\ell}$ which was extracted at $85^{\circ}C$ for 36 hours. The reducing sugar content, sweetness and turbidity were increased according to the increased extraction temperature and time. But pH were decreased according to the increased extracted time.

A Geochemical Study on the Behaviors of Major and Trace Elements in the Ulsan Granite and Its Contact Serpentinite (울산화강암 및 인접 사문암 중 주/미량 원소의 거동에 관한 지화학적 연구)

  • Geochemical characteristics of iron-related Ulsan granite was studied in comparison with the Cretaceous granitoids from the metallogenic provinces of copper, lead-zinc and lead-zinc/molybdenum in the Gyeongsang Basin, and the variation of cheminal compositions at the Ulsan granite/serpentinite contact was investigated. Ulsan granite is plotted in the regions of granite and granodiorite of Streckeisen's diagram. It shows differentiation trend of calc-alkali magma, and the magmatic evolution from granodiorite to granite is consistant with the general crystallization path of the Cretaceous the granitoids in Gyeongsang Basin. Differentiation index(D.I.) of Ulsan granite is 86~95, which is higher than those of Jindong granites (D.I.=45~70) and Onjongri granites (D.I.=67~84), and there are differences in the content of some major and trace elements between Ulsan granite and other Cretaceous granitoids. At the Ulsan granite/serpentinite contact $SiO_2$, $K_2O$, $Na_2O$, $Al_2O_3$, Rb, Sr, Ba which are abundant in Ulsan granite decrease toward serpentinite, while T.Fe, MgO, Ni, Cr which are abundant in serpentinite decrease toward Ulsan granite. Therefore, the geochemical characteristics of Ulsan granite is applicable to distinguish iron province from different metallogenic provinces where other Cretaceous granitoids occur in the Gyeongsang Basin, and it is possible to find serpentinite which was intruded by granite on the basis of chemical variations.

Measurement of hydrogen content in a-C:H films prepared by ECR-PECVD (ECR-PECVD 방법으로 증착된 a-C:H 박막의 수소함량 측정)

  • Hydrogenated amorphous carbon (a-C:H) films were deposited by ECR-PECVD (electron cyclotron resonance-plasma enhanced chemical vapor deposition) method with deposition conditions such as ECR plasma source power, gas composition of methane and hydrogen, deposition time and substrate bias voltage. The hydrogen content in the films has been measured by ERDA (elastic recoil detection analysis) using 2.5 MeV $He^{++}$ ion beam. From the results of AES (Auger electron spectroscopy), RBS (Rutherford backscattering spectrometry) and ERDA, the composition elements of deposited film were confirmed the carbon atom and the hydrogen atom. It was observed by FTIR (Fourier transform infrared) that the hydrogen contents in the film varied according to the deposition conditions. In deposition condition of substrate bias voltage, the hydrogen contents were decreased remarkably because the amount of dehydrogenation in films was increased as the substrate bias voltage increased. In the rest deposition conditions, the hydrogen contents in the film were measured in the range 45~55%.

Experimental Investigation on Deformation Capacity of CFT Column to H-Steel Beam Connections (콘크리트충전 각형기둥-H형강보 접합부의 변형능력에 관한 실험적 연구)

  • A test program was conducted on full-scale steel moment connections constructed using a T-stiffener. In the T-stiffener connection, the beam-to-column connection was reinforced with the horizontal and vertical element of the T-stiffener to resist moment under severe cyclic loads. A total of five specimens were tested in this study together with a concrete-filled tubular(CFT) column(${\sqsubset}-500{\times}500{\times}12$) and a steel beam($H-506{\times}201{\times}11{\times}19$). For the specimens, the T-stiffener was combined with RBS (also known as "Dog-bone") detail or Horizontal Element Hole(HEH) detail constructed to enhance deformation capacity. The test program showed excellent seismic performance for specimens constructed with an RBS or an HEH. except the specimens had brittle failure of VE. The test results also showed that the connections all developed maximum moments at the face of the column. Such moments were at least 15% and as much as 36% larger than the plastic moment capacity of the beam. based on the actual yield stress of the beam steel.

EFFECTS OF THE HERBICIDE, BUTACHLOR, ON NITROGEN FIXATION IN PHOTOTROPHIC NONSULFUR BACTERIA

  • In an effort to identify possible microbes for seeking bioagents for remediation of herbicide-contaminated soils, seven species of phototrophic nonsulfur bacteria (Rhodobacter capsulatus and sphaeroides, Rhodospirillum rubrum, Rhodopseudomonas acidophila, blastica and viridis, Rhodomicrobium vannielii) were grown in the presence of the herbicide, butachlor, and bacterial growth rates and nitrogen fixation were measured with different carbon sources. Under general conditions, all species showed 17-53% reductions in growth rate following butachlor treatment. Under nitrogen-fixing conditions, Rb. capsulatus and Rs. rubrum showed 1-4% increases in the growth rates and 2-10% increases in nitrogen-fixing abilities, while the other 5 species showed decreases of 17-47% and 17-85%, respectively. The finding that Rp. acidophila, Rp. blastica, Rp. viridis and Rm. vannielii showed stronger inhibitions of nitrogenase activity seems to indicate that species in genera Rhodobacter and Rhodospirillum are less influenced by butachlor than those in Rhodopseudomonas and Rhodomicrobium in terms of nitrogen-fixing ability. Overall, nitrogenase activity was closely correlated with both growth rate and glutamine synthetase activity (representing nitrogen metabolism). When the carbon sources were compared, pyruvate (three carbons) was best for all species in terms of growth rate and nitrogen fixation, with malate (four carbons) showing intermediate values and ribose(five carbons) showing the lowest; these trends did not change in response to butachlor treatment. We verified that each of the 7 species had a plasmid ($12.2{\sim}23.5\;Kb$). We found that all 7 species could use butachlor as a sole carbon source and 3 species were controlled by plasmid-born genes, but it is doubtful whether plasmid-born genes were responsible to nitrogen fixation.

Role of $Ca^{2+}$ and Calmodulin on the Initiation of Sperm Motility in Salmonid Fishes

  • $K^+$ efflux through a certain type of $K^+$ channels causes the change of membrane potential and leads to cAMP synthesis in the transmembrane cell signaling for the initiation of sperm motility in the salmonid fishes. The addition of $Ca^{2+}$ conferred motility to the trout sperm that were immobilized by external $K^+$ and other alkaline metals, $Rb^+$ and $Cs^{2+}$, suggesting the participation of external $Ca^{2+}$ in the initiation of sperm motility. L-type $Ca^{2+}$ channel blockers such as nifedipine, nimodipine, and FS-2 inhibited the motility, but N-type $Ca^{2+}$ channel blocker, w-conotoxin MvIIA, did not. On the other hand, the membrane hyperpolarization and cAMP synthesis were suppressed by $Ca^{2+}$ channel blockers, nifedipine, and trifluoroperazine. Furthermore, these suppressions were relieved by the addition of $K^+$ ionophore, valinomycin. Inhibitors of calmodulin, such as W-7, trifluoperazine, and calrnidazol-C1, inhibited the sperm motility, membrane hyperpolarization, and cAMP synthesis. The results suggest that $Ca^{2+}$ influx through $Ca^{2+}$ channels that are sensitive to specific $Ca^{2+}$ channel blockers and calmodulin participate in the changes of membrane potential, leading to synthesis of cAMP in the cell signaling for the initiation of trout sperm motility.

  • The determination of the neuromuscular blocking agents vecuronium bromide(VeBr) in biological fluids has been investigated. The method depends on the formation of insoluble red complex between vecuronium bromide and rose bengal in aqueous layer. The amount of vecuronium bromide was calculated from that of extracted rose bengal which was determined by spectrofluorimetry or HPLC/fluorescence detection method. It was possible to analyze VeBr in the range of $2{\sim}32{\mu}g/ml$(r=0.998 for water soln., 0.999 for urine, 0.996 for plasma). This method was applied to the analysis of VeBr in biological fluids, urine and plasma.

Parental Age-Related Risk of Retinoblastoma in Iranian Children

  • Background: Retinoblastoma is a rare malignant intraocular neoplasm. About 90% of cases feature a germline mutation in the RB1 gene and these will develop retinoblastoma during their early childhood. An association between mutations in germline cells and aging has been demonstrated. This suggests a higher incidence of childhood cancer including retinoblastoma among children of older parents. Materials and Methods: In the present study we aimed to determine the association of paternal and maternal age with an increased risk of retinoblastoma in a case-control study in Iranian population. The study was carried out on 240 persons who were born during 1984-2012 in Mahak and Mofid hospitals in Tehran, Iran. The statistical analysis included studying the mean age of parents and in order to know whether parental age of patients is different from parental age of control group, (t-test) compare averages test is used perfectly. By binary logistic regression, odds ratios (ORs) and 95% confidence intervals (CIs) were calculated. Results: The results of statistical analysis including the study of mean parental age by the use of (t-test) compare averages test showed a significant difference between parental ages of patients and controls. Logistic regression showed that coefficients were significant for maternal but not paternal age. Conclusions: Our findings indicate that advanced maternal age can increase the risk of retinoblastoma in offspring, but the paternal age has no significant effect.
