Thermostability of Polygalacturonase from Chinese Cabbage (배추 Polygalacturonase의 열안정성)

  • Polygalacturonase(PG) was extracted from Chinese cabbage and partially purified by ammonium sulfate fractionation and ion-exchange chromatography. Three fractions, D-PG, C-1 PG, and C-2 PG, were separated by CM-Sephadex C-25 column chromatography and FPLC Mono Q HR 5/5 or Mono S HR 5/5 column and their physicochemical characteristics were investigated. All three fractions had optimum pH and temperature of 4.5 and $65^{\circ}C$, and were stable in the range of pH $4.5{\sim}8.0$. These fractions were inhibited by 0.8mM $CaCl_2$ or 0.6M NaCl. In the thermal inactivation of PC isozymes at $65{\sim}80^{\circ}C$, z-values were $8.4{\sim}9.3^{\circ}C$ and D-values at $80^{\circ}C$ were In the range of $120{\sim}126s$. Thermodynamic constants were calculated from thermal inactivation data of the isozymes and were applicable to estimation of thermal process time of retort pouched Kimchi.

Comparative Analysis of the Characteristics of Natural Zeolites from Mongolia, Korea, and the United State (몽골, 한국, 미국 천연 제올라이트의 특성 비교 분석)

  • The 16 natural zeolites collected from Mongolia (6 types), the United States (1 type), and Korea (9 types) were characterized by XRD, XRF, TGA, DTA, and CEC analysis. All 16 samples are composite minerals. Two or more mineral phases co-exist and consist primarily of minerals such as clinoptilolite, heulandite, mordenite, and chabazite. In certain samples, minerals like illite and quartz were present as impurities. The XRF analysis showed that the 16 natural zeolites contain SiO2, Al2O3, K2O, CaO, Na2O, MgO, and Fe2O3 oxides. The cation exchange capacity of the U-1 sample was 223.3 meg/100 g, which is higher than the rest of the samples. M-6 sample in Mongolian natural zeolite and K-1 sample in Korean natural zeolite showed the highest cation exchange capacity at 166.6 meg/100 g. As a result of thermal differential and thermos gravimetric analysis, all 16 samples showed excellent thermal stability up to 600℃.

  • The methods for determining C-14 and tritium contents in the spent nuclear fuel sample were developed. The carbon-14($^{14}CO_2$) released during the dissolution of the spent fuel sample and $CaCO_3$ ($CO_2$ carrier) with 8 M $HNO_3$ at $90^{\circ}C$ was collected in trap containing 1.5 M NaOH. The volatile radioactive iodine evolved when the spent fuel was dissolved, was trapped on to Ag-silicagel (Ag-impregnated silicagel) adsorbent in column which is connected to two NaOH traps. The solutions which contain tritium as HTO after fuel dissolution were decontaminated by deionization with a mixture of cation and anion exchange resins and inorganic ionexchangers. The amount of C-14 in the trap solutions and the HTO concentration in the resulting deionization water were then determined by liquid scintillation counting.

Adsorption Characteristics of Heavy Metals on Clay Minerals (점토광물에 의한 중금속 흡착 특성)

  • This research was designed to investigate the removal of heavy metals, such as $Al^{3+}$, $Cu^{2+}$, $Mn^{2+}$, $Pb^{2+}$ and $Zn^{2+}$, by adsorption on clay minerals. Bentonite(Raw-Bentonite), $Ca^{2+}$ and $Na^+$ ion exchanged bentonite(Ca- and Na-Bentonite) and montmorillonite, such as KSF and K10 from Sigma Aldrich, were used as adsorbents. The component of five inorganic adsorbents was analyzed by XRF, and the concentration of metal ions was measured by ICP. The cation exchange capacity(CEC) and the particle charge of adsorbents were measured. The initial concentration range of metal ions was $10{\sim}100$ mg/L. From the experimental results, it was shown that the adsorption equilibrium was attained after $1{\sim}2$ hours. The maximum percentage removal of $Al^{3+}$, $Cu^{2+}$, $Pb^{2+}$ and $Zn^{2+}$ on Na-Bentonite were more than 98% and that of $Mn^{2+}$ was 66%. $Al^{3+}$ was leached out from KSF with the higher concentration of hydrogen ion. Percentage removals of $Pb^{2+}$ and $Zn^{2+}$ on KSF were 88% and 59%, respectively. In general, the percentage removal of metal ions was decreased with the higher initial concentration of metal ions. The adsorption capacity of metal ions on Na-Bentonite was $1.3{\sim}19$ mg/g. Freundlich equation was used to fit the acquired experimental data. As the results, the adsorption capacity of metal ions was in the order of Na-Bentonite$\gg$Raw-Bentonite$\cong$K10>Ca-Bentonite>KSF. Freundlich constant, K of Na-Bentonite was the largest for metal ions. The order K of Na-Bentonite was Al>Cu>Pb>Zn>Mn, and the adsorption intensity(1/n) was determined to be $0.2{\sim}0.39$.

Vegetation and Soil Properties of a Forest Wetland in Jangdo, Sinan-Gun (신안군 장도 산지습지 식생과 토양특성)

  • This study was carried out to investigate soil properties, ordination, and vegetation of a forest wetland in Jangdo, Sinan-Gun. Peculiar species such as Epilobium pyrricholophum and Lycopus ramosissim us were found in the forest wetland of Jangdo, and Hosta yingeri and Carpinus turczaninovii for coreana that are an endemic species of Korea were also found. The vascular plants of 40 families 62 genera 57 species 9 varieties 1 form, total 67 taxa were accounted for. The communities were classified as Salix koreensis-Isachne globosa community, Isachne globosa community, and Miscathus sinensis var purpurascens community. Soil organic matter, total nitrogen, available phosphorous concentrations, and cation exchange capacity each ranged from 20.6 to 72.4%, 0.74 to 2.13%, 33.3 to 114.6 ppm, and 25.5 to 94.3 me/100g, respectively. Soil pH ranged from 5.10 to 5.42. Soil texture was clay loam. Results of the correlation between Jangdo forest community and environmental factor are as follows; Soil pH was the most effective factor for plant community distribution. The Salix koreensis-Isachne globosa community was found where it had the highest soil organic matter, nitrogen, and exchangeable Na, Ca, Mg concentration, ana CEC among the three communities. Miscathus sinensis var. purpurascens community was found where it had the lowest soil organic matter, nitrogen, and exchangeable Na, Ca, Mg concentration, and CEC among the three plant communities.

Li+-exchanged Zeolites X and Y (FAU) from Undried Formamide Solution

  • Two single-crystals of fully dehydrated, partially $Li^+$-exchanged zeolites X (Si/Al = 1.09, crystal 1) and Y (Si/Al = 1.56, crystal 2), were prepared by flow method using 0.1 M $LiNO_3$ at 393 K for 48 h, respectively, followed by vacuum dehydration at 673 K and $1{\times}10^{-6}$ Torr. Their structures were determined by single-crystal X-ray diffraction techniques in the cubic space group $Fd\bar{3}$ and $Fd\bar{3}m$ at 100(1) K for crystals 1 and 2, respectively. They were refined to the final error indices $R_1/wR_2$ = 0.065/0.211 and 0.043/0.169 for crystals 1 and 2, respectively. In crystal 1, about 53 $Li^+$ ions per unit cell are found at three distinct positions; 9 at site I', 19 at another site I', and the remaining 25 at site II. The residual 25 $Na^+$ ions occupy three equipoints; 2 are at site I, 7 at site II, and 16 at site III'. In crystal 2, about 31 $Li^+$ ions per unit cell occupy sites I' and II with occupancies at 22 and 9, respectively; 3, 4, 23, and 3 $Na^+$ ions are found at sites I, I', II, and III', respectively. The extent of $Li^+$ ion exchange into zeolite X (crystal 1) is higher than that of zeolite Y (crystal 2), ca. 73% and 56% in crystals 1 and 2, respectively.

Purification and Characterization of an Extracellular Protease from Bacillus pumilus CN8

  • The protease produced by a Bacillus pumilus CN8 strain was purified by DEAE-Cellulose-52 ion exchange. It has a molecular weight of approximately 96,920 Dalton. In the present study, this protease showed strong activity over a broad range of pH (6.5-9.5) and temperature from $40^{\circ}C$ to $60^{\circ}C$, and the protease performed the maximal activity at pH 7.3 at $42^{\circ}C$. The effect of metal ions on protease activity showed that $K^+$ could slightly increase the protease activity, and other ions such as $Zn^{2+}$, $Fe^{2+}$, $Na^+$, $Ca^{2+}$, $Mg^{2+}$ had no significant activation or inhibition to the protease (P> 0.05), and the more important is that $Cu^{2+}$, $Mn^{2+}$, $Sn^{2+}$, $Cd^{2+}$ had a strong inhibitory effect on the protease activity.

Soil physio-chemical properties of Mt. NamSan on Kyungju in Korea (경주 남산의 토양 이화학적 특성)

  • This study was carried out in order to produce useful material for the forest multiple use and forest protection management by soil physio-chemical analysis of studied area in Mt. Namsan. The result of soil physio-chemical analysis and statical analysis represented as following. In side of physical properties of forest soil in Mt. Namsan, Soil depth was average 31.4cm and available soil depth was average 20.0cm. Soil type was Brown forest soil that representative soil type in Korea. Soil texture was sandy loom(SL) except valley area. In side of chemical properties, the range of soil acidity was 4.29 ~ 5.19 (average 4.76), organic matter content was 3.17% that compared the lowest value to organic matter content of Korea forest soil. Available phosphorus was 3.64ppm that was lower than others forest soil. Exchangeable cation content was similar to the Korea brown forest soil. Cation exchange capacity(CEC) was $8.22cmol_c/kg$ in Mt. Narnsan.

Biochemical Characters of Polygalacturonase Produced by Botryosphaeria dothidea (사과 겹무늬썩음병균(Botryosphaeria dothidea)이 생산하는 Polygalacturonase의 생화학적 특성)

  • The polygalacturonase (PG) production in rotten apples by Botryosphaeria dothidea was purified by using gel filtration and ion exchange column chromatography, and the biochemical characters of PG were investigated. The purified PG appeared as a single band on sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) with approximate molecular weight of 49 kilodalton (kDa). The molecular weight was equal to the native molecular weight estimated by gel filtration. The Km and Vmax values of PG were 0.51 mg/ml and 90.9 $\mu$M/min/ml, respectively. Optimum pH was 4.0~5.0, and the PG activity was stable from pH 5.0~10.0. Optimum temperature of the enzyme activity was 4$0^{\circ}C$. The PG activity was relatively stable at 2$0^{\circ}C$, but it was reduced 45% at 4$0^{\circ}C$ and completely inactivated at 8$0^{\circ}C$. The PG activity was considerably inhibited by Cu2+, Zn2+, SDS and EDTA, whereas it was not effected by Ca2+, K+, Mg2+ or Na+ ions.

Influence of Molten KNO3 Flow Conditions on Mechanical Properties during Fabrication of Chemically-Toughened Glass

  • In this study, we examined the influence of molten $KNO_3$ flow on mechanical properties and their deviation when a chemical toughening process was applied to soda lime silicate glass ($Na_2O-CaO-SiO_2$). $KNO_3$ melt flow was controlled using three methods: (1) glass tray rotation, (2) impeller stirring, and (3) natural convection. DOL and hardness were found to be enhanced by tray rotation because this rotation was able to maintain the concentration around the glass surface, in contrast to other methods. However, there did not appear to be a statistically significant difference in the 3-point bending strength for the three flow conditions due to the ground edge condition.
