Effects of Structural Difference of Ionic Liquids on the Catalysis of Horseradish Peroxidase

  • The dependence of the catalytic properties of horseradish peroxidase on the structural changes of ionic liquids was investigated with two water-miscible ionic liquids, N-butyl-3methypyridinium tetraftuoroborate ([$BMP_y$][$BF_4$]) and 1-butyl-3-methylimidazolium methylsulfate ([BMIM][$MeSO_4$]), each of which shares an anion ($BF_4^-$) or a cation ($BMIM^+$) with 1-butyl-3-methylimidazolium tetraftuoroborate ([BMIM][$BF_4$]), respectively. The oxidation of guaiacol (2-methoxyphenol) with $H_2O_2$was used as a model reaction. In order to minimize the effect of solution viscosity on the kinetic constants of the enzymatic catalysis, the enzymatic reactions for the kinetic study were performed in water-ionic liquid mixtures containing 25% (v/v) ionic liquid at maximum. Similarly to the previously reported results for [BMIM][$BF_4$], as the concentration of [$BMP_y$][$BF_4$] increased, the $K_m$value increased with a decrease in the $k_{cat}$value: the $K_m$value increased markedly from 2.8 mM in 100% water to 12.6 mM in 25% (v/v) ionic liquid, indicating that ionic liquid significantly weakens the binding affinity of guaiacol to the enzyme. On the contrary, [BMIM][$MeSO_4$] decreased the Km value to 1.4 mM in 25% (v/v) ionic liquid. [BMIM][$MeSO_4$] also decreased $k_{cat}$more than 3-folds [from 13.8 $s^{-1}$in 100% water to 4.1 $s^{-1}$in 25% (v/v) ionic liquid]. These results indicate that the ionic liquids interact with the enzyme at the molecular level as well as at a macroscopic thermodynamic scale. Specifically, the anionic component of the ionic liquids influenced the catalysis of horseradish peroxidase in different ways.

Economic Value of Industrial Water Use (공업용수의 수종별 경제적 가치 추정)

  • The information on the economic value of industrial water use is widely demanded in policy analysis area as well as academic research area. Therefore, this study attempts to obtain more accurate economic value of industrial water use using the economic theory. To this end, we conducted a survey of 1,017 manufacturing firms and estimated the economic value of industrial water use based on the concept of value of marginal product (VMP). Moreover, this study tried to estimate the VMP of three industrial water types, river water, precipitated water, and purified water. The results show that the VMPs of river water, precipitated water and purified water are estimated to be 769.24 won/$m^3$, 896.76 won/$m^3$ and 1,861.95 won/$m^3$, respectively. The value for the precipitated water found in this study is not significantly different from that (885.1 won/$m^3$) used in the pre-feasibility test implemented by Korea Development Institute (KDI). These imply that the latter is appropriate for evaluating the economic benefit of supplying industrial water, though it is based on 2003 Industry Survey.

  • In this paper, we analyze the telecom services and their socio-economic value for sharing spectrum, which is suitable to access. For this, we suggest a concept and a classification model of sharing spectrum and make a survey for analyzing the socio-economic value of spectrum, 910 MHz which is candidated for sharing between 1.4~4.5 GHz in Korea. Also, we propose mobile telecom service, WiFi, ASA, public service, M2M, and smart home and factory service as service for suitable to sharing spectrum.

Xanthoangelol and 4-Hydroxyderricin Are the Major Active Principles of the Inhibitory Activities against Monoamine Oxidases on Angelica keiskei K

  • Monoamine oxidase inhibitors (MAOI) have been widely used as antidepressants. Recently, there has been renewed interest in MAO inhibitors. The activity-guided fractionation of extracts from Angelica keiskei Koidzumi (A. keiskei K.) led to the isolation of two prenylated chalcones, xanthoangelol and 4-hydroxyderricin and a flavonoid, cynaroside. These three isolated compounds are the major active ingredients of A. keiskei K. to inhibit the MAOs and DBH activities. Xanthoangelol is a nonselective MAO inhibitor, and a potent dopamine ${\beta}$-hydroxylase (DBH) inhibitor. $IC_{50}$ values of xanthoangelol to MAO-A and MAO-B were calculated to be 43.4 ${\mu}M$, and 43.9 ${\mu}M$. These values were very similar to iproniazid, which is a nonselective MAO inhibitor used as a drug against depression. The $IC_{50}$ values of iproniazid were 37 ${\mu}M$, and 42.5 ${\mu}M$ in our parallel examination. Moreover, $IC_{50}$ value of xanthoangelol to DBH was calculated 0.52 ${\mu}M$. 4-Hydroxyderricin is a potent selective MAO-B inhibitor and also mildly inhibits DBH activity. The $IC_{50}$ value of 4-hydroxyderricin to MAO-B was calculated to be 3.43 ${\mu}M$ and this value was higher than that of deprenyl (0.046 ${\mu}M$) used as a positive control for selective MAO-B inhibitor in our test. Cynaroside is a most potent DBH inhibitor. The $IC_{50}$ value of cynaroside to DBH was calculated at 0.0410 ${\mu}M$. Results of this study suggest that the two prenylated chalcones, xanthoangelol and 4-hydroxyderricin isolated from A. keiskei K., are expected for potent candidates for development of combined antidepressant drug. A. keiskei K. will be an excellent new bio-functional food material that has the combined antidepressant effect.

Effect of Antioxidation and Inhibition of Melanogenesis from Ligularia stenocephala extract (곤달비추출물의 항산화와 멜라닌 생성 저해 효과)

  • In this study, we evaluated anti-oxidation and whitening effects of Ligularia stenocephala extract for use as the cosmeceuticals. L. stenocephala was extracted by three different solvents which was n-Hexane, ethyl acetate, $H_{2}O$. The free radical (1,l-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity of extract of L. stenocephala was in the order: ethyl acetate fraction of leaf ($IC_{50}$ value of 10.512ug/mL) > ethyl acetate fraction of stem ($IC_{50}$ value of 31.877ug/mL) > $H_{2}O$ fraction of leaf ($IC_{50}$ value of 129.194ug/mL). Reactive oxygen species (ROS) scavenging activity of extract of L. stenocephala was in the order: ethyl acetate fraction of leaf ($IC_{50}$ value of 0.230mg/mL) > ethyl acetate fraction of stem ($IC_{50}$ value of 0.528mg/mL) > $H_{2}O$ fraction of leaf ($IC_{50}$ value of 0.799mg/mL). Tyrosinase inhibition activity of L. stenocephala extracts was reduced 29.477% on ethyl acetate fraction of leaf, 13.583% on ethyl acetate fraction of stems. Therefore, L. stenocephala extracts may be useful as a new antioxidant and whitening agent to inhibit melanogenesis.

  • The effect of a water-miscible ionic liquid, 1-butyl-3-methylimidazolium tetrafluoroborate $([BMIM][BF_4])$, on the horseradish peroxidase (HRP)-catalyzed oxidation of 2-methoxyphenol (guaiacol) with hydrogen peroxide $(H_2O_2)$ was investigated. HRP maintains its high activity in the aqueous mixtures containing various concentrations of the ionic liquid and even in 90% (v/v) ionic liquid. In order to minimize the effect of solution viscosity on the kinetic constants of HRP catalysis, the enzymatic reactions in the subsequent kinetic study were performed in water-ionic liquid mixtures containing 25% (v/v) ionic liquid at maximum. As the concentration of $[BMIM][BF_4]$ increased for the oxidation of guaiacol by HRP, the $K_m$ value increased with a slight decrease in the $K_{cat}$ value: The $K_m$ value increased from 2.8 mM in 100% (v/v) water to 22.5mM in 25% (v/v) ionic liquid, indicating that ionic liquid significantly weakens the binding affinity of guaiacol to HRP.

Protein Adsorption on Ion Exchange Resin: Estimation of Equilibrium Isotherm Parameters from Batch Kinetic Data

  • The simple Langmuir isotherm is frequently employed to describe the equilibrium behavior of protein adsorption on a wide variety of adsorbents. The two adjustable parameters of the Langmuir isotherm - the saturation capacity, or $q_m$, and the dissociation constant, $K_d$ - are usually estimated by fitting the isotherm equation to the equilibrium data acquired from batch equilibration experiments. In this study, we have evaluated the possibility of estimating $q_m$ and $K_d$ for the adsorption of bovine serum albumin to a cation exchanger using batch kinetic data. A rate model predicated on the kinetic form of the Langmuir isotherm, with three adjustable parameters ($q_m,\;K_d$, and a rate constant), was fitted to a single kinetic profile. The value of $q_m$ determined as the result of this approach was quantitatively consistent with the $q_m$ value derived from the traditional batch equilibrium data. However, the $K_d$ value could not be retrieved from the kinetic profile, as the model fit proved insensitive to this parameter. Sensitivity analysis provided significant insight into the identifiability of the three model parameters.

Ionic Basis of Resting Membrane Potential in the Coronary Sinus Cells of the Rabbit (토끼 Coronary Sinus에서의 안정막 전압에 관한 연구)

  • Membrane potential of cells in the isolated rabbit coronary sinus was measured by conventional glass microelectrode and investigated the effect of $[K^+]_0$ variation in control, 20 mM and Ach-containing Tyrode solution. The results obtained were as follows: 1) The resting membrane potential exposed to normal Tyrode solution containing 3 mM $K^+\;was\;about\;-60{\sim}\;-65mV$. At extracellular $K^+$ concentrations from 1 to 30 mM the resting Potential was reasonably well described by Goldman -Hodgkin -Katz equation on the assumption that $[K^+]_1$ was 150 mM and that the ratio of membrane permeability coefficient for $Na^+\;and\;K^+,\;P_{Na}/P_K\;({\alpha})$ was 0.07. 2) In 20 mM Na-Tyrode solution (replacing by equimolar Tris) the resting membrane potential was hyperpolarized by 15 to 20 mV and showed slightly deviated to depolarized direction compared to the predicted value by Goldman-Hodgkin -Katz equation. 3) In the presence of $10^{-6}M$ Ach, the resting potentials at $[K^+]_0$ levels from 1 to 30 mM were well fitted with the predicted value on the assumption that $P_{Na}/P_K$ was 0.0144. It could be concluded that the low resting membrane potential of coronary sinus cells reflects a relatively high ratio $P_{Na}/P_K$ of about 0.07.

Our result is new.

Sterilization of Yakju(Rice Wine) on a Serial Multiple Electrode Pulsed Electric Field Treatment System (직렬배열 다중전극 고전압 펄스 전기장 처리장치를 이용한 약주의 살균)

  • Yakju(rice wine) was sterilized with high-voltage square-wave pulses of $1\;{\mu}s$ duration at various electric field strengths and frequencies on a serial multiple electrode pulsed electric field(PEF) treatment system consisted of 7 electrodes connected in series. The initial microbial counts of Yakju were $1.88{\times}10^3{\sim}2.13{\times}10^4$ CFU/mL for total aerobes, $1.55{\times}10^3{\sim}2.85{\times}10^4$ CFU/mL for lactic acid bacteria and $1.72{\times}10^3{\sim}2.39{\times}10^4$ CFU/mL for yeasts. The sterilization of microorganisms in Yakju was a first order reaction and the sterilization effect increased as the field strength and the frequency increased. The $D_{Hz}-value$ and the $D_{PEF}-value$ decreased with the electric field strength. Yeast showed lower $D_{PEF}-value$ than bacteria. Lactic acid bacteria showed lower $D_{PEF}-value$ than general aerobic bacteria under the electric field strength below 30 kV/cm, but higher ones under that above 40 kV/cm. The $Z_{PEF}-value$ of general aerobic bacteria, lactic acid bacteria and yeast in Yakju were 39.4, 49.3 and 47.6 kV/cm, respectively. The PEF sterilization resulted in less changes in color and sensory properties than heat sterilization, and the PEF treated Yakju showed superior quality to the heat treated one. The commercial sterilization of Yakju was accomplished with 2-cycle treatment on the tested serial PEF treatment system.
