Utilizing Natural and Engineered Peroxiredoxins As Intracellular Peroxide Reporters

  • It is increasingly apparent that nature evolved peroxiredoxins not only as $H_2O_2$ scavengers but also as highly sensitive $H_2O_2$ sensors and signal transducers. Here we ask whether the $H_2O_2$ sensing role of Prx can be exploited to develop probes that allow to monitor intracellular $H_2O_2$ levels with unprecedented sensitivity. Indeed, simple gel shift assays visualizing the oxidation of endogenous 2-Cys peroxiredoxins have already been used to detect subtle changes in intracellular $H_2O_2$ concentration. The challenge however is to create a genetically encoded probe that offers real-time measurements of $H_2O_2$ levels in intact cells via the Prx oxidation state. We discuss potential design strategies for Prx-based probes based on either the redoxsensitive fluorophore roGFP or the conformation-sensitive fluorophore cpYFP. Furthermore, we outline the structural and chemical complexities which need to be addressed when using Prx as a sensing moiety for $H_2O_2$ probes. We suggest experimental strategies to investigate the influence of these complexities on probe behavior. In doing so, we hope to stimulate the development of Prx-based probes which may spearhead the further study of cellular $H_2O_2$ homeostasis and Prx signaling.

Inhibition Effect of Zizania latifolia on Apoptosis Induced by $H_2O_2$ in Neuro2A Cell ($H_2O_2$로 유발된 Neuro2A 신경세포고사에 대한 줄풀의 억제 효과)

  • The purpose of this study was to examine the inhibition effect of Zizania latifolia that has been used heart disease, Diabetes Mellitus and Skin disease for a long time on apoptosis induced by $H_2O_2$ in Neuro2A cell. Neuro2A cells were cultivated in RPMI(GibcoBRL) with $5\%$, FBS and treated with $H_2O_2$, and Zizania latifolia. We measured the cell viability and analyzed DNA fragmentation. Activity of PARP, Cytochrome C, caspase-9, caspase-3, p53, p21, Bax and Bcl-2 in the cell was examined by using western blot. The cell viability in Zizania latifolia treatment (60ug/ml<) decreased significantly compared with that of none treatment. (p<0.001) Zizania latifolia increased cell viability about twice as much as that being injury by $H_2O_2$. (Zizania Latifolia 20ug/ml, $H_2O_2$ 200uM, P<0.001) DNA fragmentation developed by $H_2O_2$, but was not developed in Zizania latifolia treatment. PARP, Cytochrome C, caspase-9 and caspase-3 activated all by $H_2O_2$ but were not activated in Zizania latifolia treatment.. P53, P2l and Bu activated by $H_2O_2$, and Bcl-2 got into inactivation. But the opposite results appeared in Zizania latifolia treatment. In conclusion, these results suggest that Zizania latifolia inhibit the development of DNA fragmentation and apoptosis by $H_2O_2$ and the antioxidant action of Zizania latifolia is effective. More researches about effect of Zizania latifolia are considered to need.

Acoustic parameters that differentiate /o/ from /u/ in Seoul Korean (서울말 /ㅗ/와 /ㅜ/를 구별하는 음향변수)

  • Earlier studies reported that the /o/ and /u/ phonemes of Seoul Korean were currently merging in the F1/F2 space. However, studies on perception tests have shown that rates of correctness were high, even in cases where the two vowels overlapped. This study explores whether there is another acoustic parameter that differentiates /o/ from /u/, besides the F1/F2 contrast. Seventy-five native speakers of Seoul Korean, born between 1953 and 1999, participated in a production test. The data collected were analyzed in terms of F1 and F2, H1-H2, and F0. The result shows that the /o/ and /u/ of female speakers almost overlap in the F1/F2 space for all ages, while H1-H2 values are significantly different between the two vowels regardless of age. On the other hand, the /o/ and /u/ of male speakers are largely well separated in the F1/F2 space, while the H1-H2 values between the two vowels are very close at all ages. F0 effect is relatively small for both male and female speakers, even though there is a statistically significant difference. The result of this study provides evidence that female speakers use phonation differences to distinguish /o/ from /u/, and that the F1/F2 contrast has been replaced by H1-H2 values.

  • This study was investigated to find storage material of thermal energy storage system for a vehicle with the basic material of $Ba(OH)_2{\cdot}8H_2O$ and to test a feasibility of it. Experiment was investigated usability for long time and state change and thermal property after cycle with $Ba(OH)_2{\cdot}8H_2O$ and misxture doping additive to it. The result of this research indicated the mixture adding $Sr(OH)_2{\cdot}8H_2O$ to $Ba(OH)_2{\cdot}8H_2O$ have high feasibility as storage material for thermal energy storage system. This mixture did not exhibit the state change during 1300 cycles and the rate of decrease of heat realese energy was about 2%, relatively low value.

Time variations of H2O and SiO masers in the proto-Planetary Nebula OH 231.8+4.2

  • H2O (22 GHz) and SiO masers (43, 86, 129 GHz) in the bipolar proto-planetary nebula OH 231.8+4.2 were simultaneously monitored using the 21-m antennas of the Korean VLBI Network in 2009-2015. Both species exhibit periodic flux variations that correlate with the central star's optical light curve, with a phase delay of up to 0.15 for the maser flux variations with respect to the optical light curve. The flux densities of SiO v = 2, J = 1→0 and H2O masers decrease with time, implying that they may disappear in 10-20 yr. However, there seems to have been a transient episode of intense H2O maser emission around 2010. We also found a systematic behaviour in the velocity profiles of these masers. The velocities of the H2O maser components appear to be remarkably constant, suggesting ballistic motion for the bipolar outflow in this nebula. On the other hand, those of the SiO maser clumps show a systematic radial acceleration of the individual clumps, converging to the outflow velocity of the H2O maser clumps. Measuring the full widths at zero power of the detected lines, we estimated the expansion velocities of the compact bipolar outflow traced by H2O maser and SiO thermal line, and discussed the possibility of the expanding SiO maser region in the equatorial direction. All of our analyses support that the central host star of OH231.8 is close to the tip of the AGB phase and that the mass-loss rate recently started to decrease because of incipient post-AGB evolution.

Experimental and model study on the mixing effect of injection method in UV/H2O2 process

  • The appropriate injection of H2O2 is essential to produce hydroxyl radicals (OH·) by mixing H2O2 quickly and exposing the resulting H2O2 solution to UV irradiation. This study focused on evaluating mixing device of H2O2 as a design factor of UV/H2O2 AOP pilot plant using a surface water. The experimental investigation involved both experimental and model-based analyses to evaluate the mixing effect of different devices available for the H2O2 injection of a tubular hollow pipe, elliptical type of inline mixer, and nozzle-type injection mixer. Computational fluid dynamics analysis was employed to model and simulate the mixing devices. The results showed that the elliptical type of inline mixer showed the highest uniformity of 95%, followed by the nozzle mixer with 83%, and the hollow pipe with only 18%, after passing through each mixing device. These results indicated that the elliptical type of inline mixer was the most effective in mixing H2O2 in a bulk. Regarding the pressure drops between the inlet and outlet of pipe, the elliptical-type inline mixer exhibited the highest pressure drop of 15.8 kPa, which was unfavorable for operation. On the other hand, the nozzle mixer and hollow pipe showed similar pressure drops of 0.4 kPa and 0.3 kPa, respectively. Experimental study showed that the elliptical type of inline and nozzle-type injection mixers worked well for low concentration (less than 5mg/L) of H2O2 injection within 10% of the input value, indicating that both mixers were appropriate for required H2O2 concentration and mixing intensity of UV/ H2O2 AOP process. Additionally, the elliptical-type inline mixer proved to be more stable than the nozzle-type injection mixer when dealing with highly concentrated pollutants entering the UV/H2O2 AOP process. It is recommended to use a suitable mixing device to meet the desired range of H2O2 concentration in AOP process.

A Study on the Treatment of Dyeing Wastewater Using TiO2/UV (TiO2/UV 산화기술을 이용한 염색폐수처리에 관한 연구)

  • This research uses the $TiO_2$/UV process to verify the most suitable condition and possibility to dispose dyeing wastewater that contains pigment and a large amount of pollutants. For this, this research has enforced experiments that compare photo adsorption, photolysis, and photo catalyst oxidation reaction, and also evaluated and analyzed the change of pH and $TiO_2$ dosage, irradiation rates of ultraviolet rays and the dosage change and injection method of $H_2O_2$. According to the results of the dyeing wastewater experiment of storehouse catalyst that uses the new form of $TiO_2$, the photo catalyst oxidation reaction proved to be more effective than photo adsorption and photolysis; 35%, 21% in the case of $TCOD_{cr}$ and 39%, 28% in the case of chromaticity. Taking into consideration the reaction time, amount of photo catalyst reaction and irradiation amount of ultraviolet rays, the decomposition efficiency of pH change proved to be most effective at pH 4. On the whole, the acidity area proved to be effective in dyeing water exclusion than neutral and alkalinity areas. Having evaluated the influence of $TiO_2$ dosage, not only does the decomposition efficiency continuously improve as the $TiO_2$ dosage increases but the shielding effect does not occur also when the $TiO_2$ is at a fixed state. The influence of ultraviolet irradiation amount concluded in the result that as the ultraviolet irradiation amount increases the decomposition efficiency continually increased, but in the case of chromaticity when the irradiation amount was higher than 37.8mW/cm2 the removal efficiency is slowed remarkably. The influence of $H_2O_2$ dosage evaluation reached the results that although the decomposition efficiency increases with the increase of $H_2O_2$ dosage, when above 150mg (total dosage: 1200mg) $H_2O_2$ consumes OH radical itself and reduces the decomposition efficiency. Also in the case of the $H_2O_2$ injection method rather than injecting in the whole amount of $H_2O_2$ (1200mg) needed at the beginning all at once, injecting divided quantities of $H_2O_2$ whenever the electric current density falls below 10mgfl reduces the wases of OH radical due to an excess of $H_2O_2$ and in tum heightens the decomposition efficiency.

Removal of Diclofenac, Ibuprofen and Naproxen using Oxidation Processes (산화공정에서의 Diclofenac, Ibuprofen 및 Naproxen의 제거특성 평가)

  • In order to evaluate a removal characteristic of diclofenac, ibuprofen and naproxen by oxidizing agents, $Cl_2,\;O_3$ and $O_3/H_2O_2$ are used as oxidants in this study. In case of that $Cl_2$ is used for oxidizing pharmaceuticals, ibuprofen is not removed entirely at $Cl_2$ dose range of 0.5~5.0 mg/L for 60 minutes, however, removal tendency of diclofenac and naproxen are so obviously at $Cl_2$ dose higher than 0.5 mg/L. In addition, as $Cl_2$ dose and contact time are increased, the removal rate of diclofenac and naproxen is enhanced. When $O_3$ is used as oxidizing agent, ibuprofen is not eliminated at $O_3$ dose range of 0.2~5.0 mg/L. On the contrary, 72~100% of diclofenac and 49~100% of naproxen are removed at $O_3$ dose of 0.2~5.0 mg/L. From experiments using $O_3/H_2O_2$ as an oxidant, we can find that $O_3/H_2O_2$ is much more effective than $O_3$ only for removal of diclofenac and naproxen. Moreover, the efficiency is raised according to increase of $H_2O_2$ dose, however, experiments using $O_3/H_2O_2$ show that oxidation of pharmaceuticals is less effective as $H_2O_2$ to $O_3$ ratio increased to above approximately 1.0. On reaction rate constant and half-life of diclofenac, ibuprofen and naproxen depending on $Cl_2$, $O_3$ and $O_3/H_2O_2$ dose, an oxidation of pharmaceuticals by $Cl_2$ and $O_3$ particularly has a comparatively high reaction rate constant and short half-life comparing $O_3/H_2O_2$. From above results, we can fine that diclofenac and naproxen can be easily eliminated in oxidation processes.

Propofol protects against oxidative-stress-induced COS-7 cell apoptosis by inducing autophagy

  • Background: In oxidative stress, reactive oxygen species (ROS) production contributes to cellular dysfunction and initiates the apoptotic cascade. Autophagy is considered the mechanism that decreases ROS concentration and oxidative damage. Propofol shows antioxidant properties, but the mechanisms underlying the effect of propofol preconditioning (PPC) on oxidative injury remain unclear. Therefore, we investigated whether PPC protects against cell damage from hydrogen peroxide ($H_2O_2$)-induced oxidative stress and influences cellular autophagy. Method: COS-7 cells were randomly divided into the following groups: control, cells were incubated in normoxia (5% $CO_2$, 21% $O_2$, and 74% $N_2$) for 24 h without propofol; $H_2O_2$, cells were exposed to $H_2O_2$ ($400{\mu}M$) for 2 h; $PPC+H_2O_2$, cells pretreated with propofol were exposed to $H_2O_2$; and 3-methyladenine $(3-MA)+PPC+H_2O_2$, cells pretreated with 3-MA (1 mM) for 1 h and propofol were exposed to $H_2O_2$. Cell viability was determined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide thiazolyl blue (MTT) reduction. Apoptosis was determined using Hoechst 33342 staining and fluorescence microscopy. The relationship between PPC and autophagy was detected using western blot analysis. Results: Cell viability decreased more significantly in the $H_2O_2$ group than in the control group, but it was improved by PPC ($100{\mu}M$). Pretreatment with propofol effectively decreased $H_2O_2$-induced COS-7 cell apoptosis. However, pretreatment with 3-MA inhibited the protective effect of propofol during apoptosis. Western blot analysis showed that the level of autophagy-related proteins was higher in the $PPC+H_2O_2$ group than that in the $H_2O_2$ group. Conclusion: PPC has a protective effect on $H_2O_2$-induced COS-7 cell apoptosis, which is mediated by autophagy activation.

Fast temporal detection of intracellular hydrogen peroxide by HyPer

  • HyPer is the genetically encoded biosensor of intracellular hydrogen peroxide ($H_2O_2$), the most stable of the reactive oxygen species (ROS) generated by living cells. HyPer has a high sensitivity and specificity for detecting intracellular $H_2O_2$ by confocal laser microscopy. However, it was not known whether high speed ratiometric imaging of $H_2O_2$ by HyPer is possible. We thus investigated the sensitivity of HyPer in detecting changes to the intracellular $H_2O_2$ levels in HEK293 and PC12 cells using a microfluorometer imaging system. Increase in the HyPer ratio were clearly evident on stimulations of more than $100{\mu}M$ $H_2O_2$ and fast changes in the HyPer ratio were observed on ratiometric fluorescent images after $H_2O_2$ treatment. These results suggest that HyPer is a potent biosensor of the fast temporal production of intracellular $H_2O_2$.
