This study was carried out to evaluate cytotoxic effects of Ajuga multiflora Bunge extracts on murine leukemia tumor $(P388D_1)$ cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay. The comparison of $IC_{50}$ values of Ajuga multiflora Bunge extracts in L1210 and $P388D_1$ cell lines showed that their susceptibility to these extracts decreased in the following order: Adriamycin>methanol extract>chloroform extract>ethyl acetate extract>hexane extract>water extract by the MTT assay. In order to develop an antimicrobial agent, dried Ajuga multiflora Bunge was extracted with several solvents, and then antimicrobial activity was investigated. The minimal inhibitory concentration (MIC) of the extract against microorganisms were also examined. Antimicrobial activities of amocla and ketoconazole as references were compared to those of extracts of $H_2O$, n-hexane, chloroform, ethyl acetate and methanol. The antimicrobial activity of all extracts from the sample had growth inhibition activity against gram-negative bacteria, gram-positive bacteria and fungi $(MIC>200\;{\mu}g/ml)$. These results suggest that the methanol soluble extract of Ajuga multiflora Bunge may be a valuable choice for the studies on the treatment of murine leukemia tumor cell lines.

Water environment and freshwater algae were studied in the upstream of the Tamjin River Dam. Among the environmental factors, DO concentration in the Tamjin River ranged from 9.0 mg $O_2/l$ to 9.2 mg $O_2/l$, pH from 7.0 to 7.1 and conductivity from $98{\mu}S/cm$ to $100{\mu}S/cm$. Average concentration of $NH_4$ and $NO_3$ ranged from $40{\mu}g\;N/l$ to $56{\mu}g\;N/l$ and from $489{\mu}g\;N/l$ to $611{\mu}g\;N/l$, respectively. $NO_3$ was more plentiful above 9~15 fold than that of $NH_4$. Average concentrations of soluble reactive phosphorus and soluble reactive silicon were $2{\mu}g\;P/l$ and 1.6 mg Si/l, respectively. Particulaly, Si nutrient increased by heavy rain events during summer season. The ratios of N/P and Si/P ranged from 248 to 261 and from 640 to 740, respectively. It is assumed that P would be limiting nutrient on the freshwater algal growth. Average content of planktonic chlorophyll-a ranged from $5{\mu}g/l$ to $13{\mu}g/l$. Mean contents of chlorophyll-a, phaeo-pigment and ash-free dry matter of periphyton were $50.3mg/m^2$, $11.9mg/m^2$, $11.5g/m^2$ in the main stream and $30.1mg/m^2$, $5.6mg/m^2$, $7.8g/m^2$ in the tributary. By comparison of the epilithon biomass, the main stream was higher with 1.5~2.1 fold than the tributary. The impotant algae were composed of diatom Achnanthes linearis, A. minutissima, Fragilaria crotonensis, Gomphonema gracile, Tabellaria flocculosa and blue-green algae Microcystis aeruginosa. In the relative abundance of the phytoplankton and epilithon, the serial dominance were diatom > green algae > blue-green algae, and diatoms were very abundant in comparison with other algal phylum.
Protective effects of natural components including genistein (4',5,7-trihydroxyisoflavone) from Glycine max MERRILL on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. Genistein $(10{\sim}100\;{\mu}m)$ suppressed photohemolysis in a concentration-dependent manner, and was more effective than the lipid peroxidation chain blocker, ${\alpha}$-tocopherol (Vit. E). Glycoside of genistein, genistin, the water-soluble antioxidant, L-ascorbate, and the iron chelator, myo-inositol hexaphosphoric acid dodecasodium salt (sodium phytate) did not exhibit protective effect against photohemolysis. L-Ascorbate and sodium phytate stimulated photohemolysis at high concentration $(500\;{\mu}m)$. ${\alpha}$-Carotene 3,3'-diol (lutein), a singlet oxygen $(^1O_2)$ quencher, exhibited pronounced protective effect, an indication that $^1O_2$ is important in photohemolysis sensitized by rose-bengal. Reactive oxygen scavenging activities $(OSC_{50})$ of natural antioxidants including genistein on reactive oxygen species (ROS) generated in $Fe^{3+}-EDTA/H_2O_2$ system using the luminol-dependent chemiluminescence assay were in the order of sodium phytate > L-ascorbate > ${\alpha}$-tocopherol > genistein > genistin. $OSC_{50}$ value of genistein, genistin, ${\alpha}$-tocopherol, L-ascorbate, and sodium phytate were 41.0, 109.0, 9.0, 5.2, and $0.56{\mu}m$ respectively. The order of free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity $(FSC_{50})$ was L-ascorbate > ${\alpha}$-tocopherol > genistein > genistin. These results indicate that genistein can function as an antioxidant in biological systems, particularly skin exposed to solar UV radiation by scavenging $^1O_2$ and other ROS, and to protect cellular membranes against ROS.

Muskmelon(Cucumis melo L.)은 독특한 향기와 높은 당도로 각광받고 있는 과일로 주로 지중해, 아시아, 북유럽과 미주지역에서 광범위하게 재배되고 있으며 우리나라에서는 주로 시설 내에서 재배되고 있다. 점차 그 수요가 증가하고 있는 fresh-cut제품은 유통, 저장 기간 동안 제품의 신선함 유지가 요구된다. 본 연구에서는 가공 전 열처리가 fresh-cut muskmelon의 품질에 미치는 영향과 최적 열처리 조건을 조사하였다. Half-cut melon을 대상으로 blanching 온도 설정을 위한 예비 실험을 한 결과 5$0^{\circ}C$ 열처리한 구가 향, 연화 저해 정도, 그리고 전체적인 기호도에 대한 관능평가 가장 우수하였다. 이를 바탕으로 5$0^{\circ}C$의 water bath상에서 whole melon을 10, 20, 30, 40분 blanching하여 5$^{\circ}C$에서 24시간 저장한 후 cork borer를 이용하여 cylinder 형의 fresh-cut melon을 가공하였다. 가공한 melon은 5$^{\circ}C$에서 6일 저장하면서 L-value, chroma-value, soluble solid, pH, 경도, 호흡 변화 등을 통해 품질변화를 측정하였다. 저장 6일후 L-value의 변화는 대조구에서 가장 크게 나타났으며, chroma-value는 저장 시간이 경과함에 따라 감소하였으며 10분 열처리 한 구에서 변화정도가 가장 낮게 나타났다. 수용성 고형분 함량은 저장기간의 경과에 따른 큰 변화는 없었으나 40분 열처리한 구에서 수용성 고형분 함량이 가장 낮았으며 20, 30분 열처리한 구에서 높게 나타났다. 저장 중 fresh-cut melon의 pH는 증가하는 경향을 보였으며 10분 열처리한 구에서 가장 낮게 나타났다. 호흡변화는 저장 기간이 경과함에 따라 $O_2$는 감소하고 $CO_2$는 증가했으며 특히 20분 열처리한 구에서 $CO_2$가 크게 증가한 것으로 나타났다. 증가 정도가 가장 크게 나타났다. 열처리한 fresh-cut melon의 hardness는 저장 3일 후 대조구와 40분 열처리 구에서 가장 낮아 연화현상을 확인했으며 20분 열처리구에서 초기 hardness가 유지되는 것으로 확인했다. 따라서 fresh-cut melon을 위한 가공 전 열처리를 위해서는 5$0^{\circ}C$에서 10분 열처리가 품질 변화를 유지하는데 적당하다고 판단된다.
Monascus pilosus (KCCM 60160) in submerged culture was optimized based on culture medium and fermentation conditions. Monacolin-K (Iovastatin), one of the cholesterol lowing-agent which was produced by Monascus pilosus may maintain a healthy lipid level by inhibiting the biosynthesis of cholesterol. Plackett-Burman design and response surface method were employed to study the culture medium for the desirable monacolin-K production. As a result of experimental designs, optimized production medium components and concentrations (g/L) were determined on soluble starch 96, malt extract 44.5, beef extract 30.23, yeast extract 15, $(NH_4)_2SO_4$ 4.03, $Na_2HPO_4{\cdot}12H_2O$ 0.5, L-Histidine 3.0, $KHSO_4$ 1.0, respectively. Monacolin-K production was improved about 3 times in comparison with shake flask fermentation of the basic production medium. The effect of agitation speed (300, 350, 400 and 450 rpm) on the monacolin-K production were also observed in a batch fermenter. Maximum monacolin-K production with the basic production medium was 68 mg/L when agitation speed was 500 rpm. And it was found that all spherical pellets (average diameter of $1.0{\sim}1.5mm$) were dominant during fermentation. Based on the results, the maximum production of 185 mg/L of monacolin-K with the optimized production medium was obtained at pH (controlled) 6.5, agitation rate 400 rpm, aeration rate 1 vvm, and inoculum size 3%.
We synthesized Pd(II) complexes containing sulfoxide and cycloamine series on the basis of the fact that many workers had the foresight to suggest anticancer activites using Pd(II) complexes with sulfur, nitrogen, or nucleic acid relatives as ligands particulary. The identification and characterization of these complexes are followed; content rates of atoms and molecular weights are confirmed by Elemental Analysis and Mass spectra, bonding of metal and ligands by UV-Vis. spectra and NMR spectra, and donor sites of ligands and configuration of complexes by IR spectra. These are yellow crystals, soluble in methanol and chloroform, and insoluble in H2O and the other common organic solvents.
Antitumor polyacetylenic alcohol, panaxytriol (Cl7 H26O3), was isolated and purified from a powder of the root of Pnnnx tin.1.encl C.A. Meyer. Panaxytriol possesses unusual property of being soluble in both water and organic solvents. Panaxytriol inhibited the growth of various kinds of human cultured cell lines in dose-dependent fashion in vitro. The in vivo effects of panaxytriol were tested against C57BL/6 mice transplanted with Bl6 melanomas. Panaxytriol (8 and 40 mg/kg) administered intra-muscularly(im) produced significant tumor growth delays in mice. Although a detailed mechanism of growth inhibition by panaxytriol is unknown, preliminary results appear to implicates a surface membrane site of action. And its action seems to be more dose-dependent than time-dependent. Finally, panaxytriol pharmacokinetics was evaluated in mice given single 8 mg/kg doses intraperitoneally (ip) or im. Serum panaxytriol content was measured using both tumor growth inhibitory assay and a gas chromatographic method. The maximum serum panaxytriol content after ip and im administration was 35.0 and 1.6 $\mu$g/ml respectively. These results indicate that the compound may act as cytotoxic substance even in patients.
Limitation of photosynthesis in detached Chinese cabbage (Brassica campestris L.) leaves was induced by feeding of mannose (25 mM) for 12 h in the light, and changes in the basic thylakoid functions under this condition were investigated. The acid soluble phosphate contend and CO2 uptake rate was decreased by 66% and 67%, respectively. However, the starch content was increased by 24% compared to those of controls. From the fast induction curves of chlorophyll fluorescence, dark level fluorescence (Fo) slightly increased while intermediate plateau fluorescence level (FI) to peak level fluorescence (Fp) transient was significantly decreased with a slight decrease in the Fo-to-FI transient. This data means that reduction of secondary electron acceptor of PSII (QB) might be more severely inhibited than that of primary electron acceptor of PSII (QA) by decrease in phosphate level. The strong decline of (Fv)m//Fm ratio suggests that efficiency of excitation energy capture by PSII was decreased markedly. The quenching of Fo (qO), an indicator of state transition, was also occurred over the slow induction kinetics of chlorophyll fluorescence. From quenching analysis, fluorescence was dominantly quenched by nonphotochemical quenchings (qE+qT). These results showed that the capture and transfer efficiency of excitation energy to PSII reaction center in thylakoid was decreased with the decline of leaf phosphate level, and that the state transition was occurred during the induction of photosynthesis under these conditions.

Bio-Green(B.G.) functional water was manufactured by Kyungwon Enterprise Co. through a series of processes ; water longrightarrow ultra-purification longrightarrow adding catalysts longrightarrow energy imprinting fermenting with energized water + zeolite and others + photosynthetic bacteria in fermenter longrightarrow filtering. Control(0), 5 or 10 liters of B.G. functional water were supplied to the orchard soil under canopy of 10 year old 'Tsugaru'/M26 apple trees on March 20, May 20 and June 20, 1995, respectively. Some orchard soil characteristics, not only pH, but also Ca and Mg of exchangeable cations were increased by supply with B.G. functional water. However, P$_2$O$_{5}$, K, and B contents were not influenced by the treatment. At harvest time soluble solid content of flesh and anthocyanin of fruit skin were increased by the treatment. B.G. functional water treatment showed higher root activities, and photosynthesis of leaves than that of control. Also B.G. functional water treatment showed higher Ca content in fruit skin and flesh tissues, whereas not affected N, K, and Mg contents. During storage at 4$^{\circ}C$ cold room, the more volume of B.G. functional water supply showed lower bitter pit symptom. Respiration and ethylene evolution in fruit were decreased, while fruit firmness increased by the treatment during storage.e.
