스트레스-유도 열충격단백질 27(Heat Shock Protein 27)의 활성과 물리치료의 상관성 (The Activation of Stress-induced Heat Shock Protein 27 and the Relationship of Physical Therapy)

  • Purpose: Heat shock proteins (HSPs) are a group of proteins that are activated when cells are exposed to a variety of environmental stresses, such as infection, inflammation, exposure to toxins, starvation, hypoxia, brain injury, or water deprivation. The activation of HSPs by environmental stress plays a key role in signal transduction, including cytoprotection, molecular chaperone, anti-apoptotic effect, and anti-aging effects. However, the precise mechanism for the action of small HSPs, such as HSP27 and mitogen-activated protein kinases (MAPKs: extracellular-regulated protein kinase 1/2 (ERK1/2), p38MAPK, stress-activated protein kinase/c-Jun N-terminal kinase (SAPK/JNK), is not completely understood, particularly in application of cell stimulators including platelet-derived growth factor (PDGF), angiotensin II (AngII), tumor necrosis factor $\alpha$ (TNF$\alpha$), and $H_2O_2$. This study examined the relationship between stimulators-induced enzymatic activity of HSP27 and MAPKs from rat smooth and skeletal muscles. Methods: 2-dimensional electrophoresis (2DE) and matrix assisted laser desorption ionizationtime-of-flight/time-of-flight (MALDI-TOF/TOF) analysis were used to identify HSP27 from the intact vascular smooth and skeletal muscles. Three isoforms of HSP27 were detected on silver-stained gels of the whole protein extracts from the rat aortic smooth and skeletal muscle strips. Results: The expression of PDGF, AngII, TNF$\alpha$, and $H_2O_2$-induced activation of HSP27, p38MAPK, ERK1/2, and SAPK/JNK was higher in the smooth muscle cells than the control. SB203580 (30${\mu}$M), a p38MAPK inhibitor, increased the level of HSP27 phosphorylation induced by stimulators in smooth muscle cells. Furthermore, the age-related and starvation-induced activation of HSP27 was higher in skeletal muscle cells (L6 myoblast cell lines) and muscle strips than the control. Conclusion: These results suggest, in part, that the activity of HSP27 and MAPKs affect stressors, such as PDGF, AngII, TNF$\alpha$, $H_2O_2$, and starvation in rat smooth and skeletal muscles. However, more systemic research will be needed into physical therapy, including thermotherapy, electrotherapy, radiotherapy and others.

금은화가 LPS로 유발된 급성 폐 손상에 미치는 영향 (Effects of Lonicerae Flos Extracts on LPS-induced Acute Lung Injury)

  • Objective : The object of this study was to observe the effects of Lonicerae Flos (LF) aqueous extracts on lipopolysaccharide (LPS)-induced rat acute lung injury. Method : Five different dosages of LF extracts were orally administered once a day for 28 days before LPS treatments, and then all rats were sacrificed after 5 hour-treatment of LPS. Eight groups of 16 rats each were used in the present study. The following parameters caused by LPS treatment were observed ; body weights, lung weights, pulmonary transcapillary albumin transit, arterial gas parameters (pH, $PaO_2$ and $PaCO_2$) bronchoalveolar lavage fluid (BALF) protein lactate dehydrogenase (LDH), and proinflammatory cytokines tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\beta}$) contents, total cell numbers, neutrophil and alveolar macrophage ratios, lung malondialdehyde (MDA), myeloperoxidase (MPO), proinflammatory cytokines TNF-${\alpha}$ and IL-$1{\beta}$ contents. In addition, the histopathologic changes were observed in the lung in terms of luminal surface of alveolus, thickness of alveolar septum, number of polymorphonuclear neutrophils. Result : As results of LPS-injection, dramatical increases in lung weights, pulmonary transcapillary albumin transit increases, increases in $PaCO_2$, decreases in pH of arterial blood and $PaO_2$, increases of BALF protein, LDH, TNF-${\alpha}$ and IL-$1{\beta}$ contents, total cells, neutrophil and alveolar macrophage ratios, TNF-${\alpha}$ and IL-$1{\beta}$ contents increases were detected with decreases in LSA and increases of alveolar septum and PMNs numbers, respectively as compared with intact control. These are means that acute lung injuries (resembling acute respiratory distress syndrome) are induced by treatment of LPS mediated by inflammatory responses, oxidative stress and related lipid peroxidation in the present study. However, these LPS-induced acute lung injuries were inhibited by 28 days continuous pretreatment of 250 and 500mg/kg of LF extracts. Because of lower three dosages of LF treated groups, 31.25 and 62.5 and 125mg/kg did not showed any favorable effects as compared with LPS control, the effective dosages of LF in LPS-induced acute lung injuries in the present study, is considered as about 125mg/kg. The effects of 250mg/kg of LF extracts showed almost similar effects with ${\alpha}$-lipoic acid 60mg/kg in preventing LPS-induced acute lung injuries. Conclusion : It seems that LF play a role in protecting the acute respiratory distress syndrome caused by LPS.

Effect of Hijikia fusiforme extracts on degenerative osteoarthritis in vitro and in vivo models

  • BACKGROUND/OBJECTIVES: The inhibitory effect of Hijikia fusiforme (HF) extracts on degenerative osteoarthritis was examined in primary cultured rat cartilage cells and a monosodium iodoacetate (MIA)-induced osteoarthritis rat model. MATERIALS/METHODS: In vitro, cell survival and the expression of matrix metalloproteinases (MMPs), collagen type I, collagen type II, aggrecan, and tissue inhibitor of metalloproteinases (TIMPs) was measured after $H_2O_2$ ($800{\mu}M$, 2 hr) treatment in primary chondrocytes. In vivo animal study, osteoarthritis was induced by intra-articular injection of MIA into knee joints of rats, and then RH500, HFE250 and HFE500 were administered orally once a day for 28 days. To determine the anti-inflammatory effects of HFE, nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$) expression were measured. In addition, real-time PCR was performed to measure the genetic expression of MMPs, collagen type I, collagen type II, aggrecan, and TIMPs. RESULTS: In the in vitro assay, cell survival after $H_2O_2$ treatment was increased by HFE extract (20% EtOH). In addition, anabolic factors (genetic expression of collagen type I, II, and aggrecan) were increased by HFE extract (20% EtOH). However, the genetic expression of MMP-3 and 7, known as catabolic factors were significantly inhibited by treatment with HFE extract (20% EtOH). In the in vivo assay, anabolic factors (genetic expression of collagen type I, II, aggrecan, and TIMPs) were increased by oral administration of HFE extract. However, the genetic expression of MMP-3 and 7, known as catabolic factors, and production of NO and $PGE_2$ were significantly inhibited by treatment with oral administration of HFE extract. CONCLUSION: HFE extract inhibited articular cartilage degeneration through preventing extracellular matrix degradation and chondrocyte injury.

호도약침(胡桃藥鍼)이 인간(人間)의 신경교종 세포(細胞)에 유발된 저산소증(低酸素症)에 대한 방어효과(防禦效果) (Effect of Juglans sinensis Dode extract on chemical hypoxia-induced cell injury in human glioma cells)

  • 이 논문(論文)은 활성 산소(ROS)의 작용(作用)을 규명하고 호도약침액(胡桃藥鍼液)이 인간의 신경교종 세포인 A172에서 화학적(化學的) 저산소증(低酸素症)으로 유발된 세포 사멸에 대해 효능이 있는지를 연구(硏究)한 것이다. 화학적(化學的) 저산소증(低酸素症)은 세포내 미토콘드리아의 전자 수송을 방해하는 antimycin A를 가진 배양세포에 의해 유발(誘發)하였다. 화학적(化學的) 저산소증(低酸素症)에 노출된 세포(細胞)는 시간과 그 양에 따라서 세포 사멸의 결과(結果)가 다르게 나타난다. 화학적 저산소증에 의해서 ROS의 생산이 증가하는데 이것은 $H_2O_2$ 소거(消去) Catalase(과산화수소를 물과 산소로 분해하는 효소)에 의해 방지(防止)된다. Catalase는 화학적 저산소증에 의해 유발(誘發)된 세포 사멸을 방지하는데 비해 DMTU는 효과적이지 않다. 지질(脂質)에 녹는 산화방지제 DPPD와 물에 녹는 산화방지제 Trolox는 세포사멸을 방지하는데 효과(效果)가 없다. 호도약침액(胡桃藥鍼液)은 그 양(量)에 의존적으로 저산소증에 의해 유발된 세포 사멸을 방지하는 효과가 있다. 즉 화학적 저산소증으로 유도된 ROS의 발생을 막고, $H_2O_2$로 유도된 세포사멸을 방지하는데 이것은 화학적 저산소증과 $H_2O_2$의해 유도된 세포사멸에 대해 호도약침액(胡桃藥鍼液)이 방지효과(防止效果)가 있다는 것을 의미한다. 이러한 결과(結果)들은 $H_2O_2$가 지질 과산화와는 무관한 메카니즘으로 저산소증(低酸素症)으로 유발(誘發)된 세포사멸을 중재하고, 따라서 호도약침액(胡桃藥鍼液)은 지질막의 과산화를 방지하기 보다는 ROS를 직접적으로 소거(消去)함으로써 방지 효과가 있다는 것을 의미한다. 더구나 화학적(化學的) 저산소증(低酸素症)은 caspase와 무관한 메카니즘으로 apoptosis를 유발(誘發)한다.

HaCaT 피부각질세포에서 들깻잎 추출물의 산화적 스트레스에 대한 항산화 효과 (Protective Effects of Perilla frutescens Britt var. japonica Extracts from Oxidative Stress in Human HaCaT Keratinocytes)

  • 들깻잎 메탄올추출물(PLME)이 가지는 산화적 스트레스 개선효과를 확인하기 위하여 $H_2O_2$으로 유도된 산화적 스트레스에 대한 HaCaT 피부 각질세포의 보호효과를 조사하였다. 또한 PLME의 항산화 능력을 확인하기 위하여 DPPH, hydroxyl free radical 소거능 및 총 항산화물질(페놀류, 플라보노이드류 및 아스코르브산) 함량을 조사하였다. $H_2O_2$(500 ${\mu}M$)에 의한 산화적 스트레스가 유발된 HaCaT세포에 PLME를 처리한 결과, 농도 의존적으로 세포의 생존율이 증가하였고, 세포 지질과산화물질 MDA의 생성효과는 PLME 처리에 의해 유의적으로 감소하는 것을 관찰하였다. 또한 $H_2O_2$로 인하여 세포내 항산화효소인 SOD, GSH-px와 CAT 등의 활성이 감소된 HaCaT세포에 PLME를 처리했을 때, 이들 효소의 활성이 농도 의존적으로 증가되었다. PLME의 DPPH와 hydroxyl radical 소거능을 측정한 결과, 농도 의존적으로 radical 소거능이 증가함을 알 수 있었다. 50 ${\mu}g/mL$ 이상 농도의 PLME의 DPPH 소거능은 60%의 저해율을 나타낸 천연항산화제인 아스코르브산(50 ${\mu}g/mL$)과 유사한 효과를 보였고, ${\cdot}OH$ radical 소거능은 아스코르브산(50 ${\mu}g/mL$)보다 높은 결과를 나타냈다. 또한 PLME가 함유하고 있는 항산화물질인 폴리페놀류, 플라보노이드류, 아스코르브산의 함량을 측정한 결과 총 페놀류화합물은 $52.2{\pm}1.1$ mg GAE/g, 총 플라보노이드화합물은 $33.7{\pm}4.7$ mg RUE/g, 아스코르브산의 함량은 $17.0{\pm}0.5$ mg AA/g으로 나타났다. HaCaT 세포에서 $H_2O_2$에 의해 발생하는 산화적 스트레스에 대한 보호 효과를 측정한 결과 PLME는 세포 사멸을 방지하고, 세포 지질과산화물질(MDA)의 생성을 억제하여 세포내 항산화효소의 활성을 증가시키는 효과를 가지는 것으로 보인다. 이상의 결과로 들깻잎 메탄올추출물은 인체 피부각질 세포에 대한 보호 작용과 in vitro에서의 항산화 능력이 있는 것으로 확인되었다.

Alaria esculenta Extract Protects against Oxidative Damage by Inducing Heme Oxygenase-1 Expression via Akt and Nrf2

  • Alaria esculenta is a brown seaweed found in the Arctic. This study investigated the protective effect of A. esculenta extract (AEE) against oxidant-mediated injury and its mode of action in RAW264.7 macrophages. The methyl thiazolyl tetrazolium (MTT) assay showed that $H_2O_2$ treatment reduced cell viability, whereas AEE protected cells from $H_2O_2$-mediated cytotoxicity in a dose-dependent manner. Because heme oxygenase-1 (HO-1) is known to protect cells against oxidative damage, we investigated the effect of AEE on HO-1 gene expression and HO enzyme activity. The protective effect of AEE against $H_2O_2$-induced injury was correlated with increased HO enzyme activity. AEE also induced HO-1 mRNA and protein expression, as determined RT-PCR and Western blotting, respectively. To characterize the mechanisms by which AEE induces HO-1 gene expression, we examined the effect of AEE on the nuclear translocation of NF-E2-related factor-2 (Nrf2) and Akt phosphorylation. AEE treatment activated upstream signaling for HO-1 gene expression, including the nuclear translocation of Nrf2 and Akt phosphorylation. Collectively, these results suggest that AEE has anti-oxidant activity that is mediated, at least in part, via the activation of Nrf2 and Akt and the subsequent induction of HO-1 gene expression.

청국장 메탄올 추출물이 T98G 세포와 허혈성 뇌졸중 백서에 미치는 영향 (The Effects of Methanol Extract from Cheonggukjang in T98G Cells and Early Stage of Focal Ischemia Rodent Models)

  • This study was conducted to evaluate the neuroprotective effects of Cheonggukjang extract in in-vitro and in-vivo models. T98G-human glioblastoma cells were pretreated with various concentrations (1~10 mg/mL) of Cheonggukjang extract for 24 h and then exposed to $H_2O_2$ (1 mM) for 3 h. The neuroprotective effects of Cheonggukjang extract were measured using a CCK-8 kit assay, total antioxidant capacity (TAC) assay, reactive oxygen species (ROS) assay, and lactate dehydrogenase (LDH) release assay. The early stage focal ischemia rodent model was used as the in-vivo neurotoxicity model. Various concentrations (10~200 mg) of Cheonggukjang extract were administered to the animal models for 1 week. Peripheral blood was analyzed for glutathione peroxidase (GPx) expression by ELISA, and infarct volume reduction was analyzed by TTC staining. Cheonggukjang extract significantly (p<0.05) increased cell viability in T98G cells against $H_2O_2$ as well as against the induced neurotoxicity. Indeed, treatment with the Cheonggukjang extract induced a decrease in ROS and LDH expression and increased TAC significantly (p<0.05). However, Cheonggukjang extract did not induce a decrease in infarct volume or an increase in GPx expression in the in-vivo model. Despite the limitation in neuroprotection, Cheonggukjang extract may be useful for treating ROS injury.

Tat-indoleamine 2,3-dioxygenase 1 elicits neuroprotective effects on ischemic injury

  • It is well known that oxidative stress participates in neuronal cell death caused production of reactive oxygen species (ROS). The increased ROS is a major contributor to the development of ischemic injury. Indoleamine 2,3-dioxygenase 1 (IDO-1) is involved in the kynurenine pathway in tryptophan metabolism and plays a role as an anti-oxidant. However, whether IDO-1 would inhibit hippocampal cell death is poorly known. Therefore, we explored the effects of cell permeable Tat-IDO-1 protein against oxidative stress-induced HT-22 cells and in a cerebral ischemia/reperfusion injury model. Transduced Tat-IDO-1 reduced cell death, ROS production, and DNA fragmentation and inhibited mitogen-activated protein kinases (MAPKs) activation in H2O2 exposed HT-22 cells. In the cerebral ischemia/reperfusion injury model, Tat-IDO-1 transduced into the brain and passing by means of the blood-brain barrier (BBB) significantly prevented hippocampal neuronal cell death. These results suggest that Tat-IDO-1 may present an alternative strategy to improve from the ischemic injury.

과산화수소로 유도된 SH-SY5Y 신경세포 사멸에 대한 오미자·칠해목 추출혼합물의 보호효과 (Neuroprotective Effects of Schisandra chinensis and Ribes fasciculatum Extract on Hydrogen Peroxide-Mediated Oxidative Stress in Neuroblastic SH-SY5Y Cell Line)

  • In neuronal cell deaths, oxidative stress is normally implicated with a most of these deaths occurring in neurodegenerative disorders such as the Alzheimer's and Parkinson's diseases. In this study, the neuroprotective effects of Schisandra chinensis (SC) and Ribes fasciculatum (RF) extracts on hydrogen peroxide ($H_2O_2$)-induced oxidative stress in neuroblastic cell line were investigated. For an hour, hydrogen peroxide of $100{\mu}M$ concentration, was induced on neuroblastic cells, causing apoptic cell death. For the neuroprotection, a sample of neuroblastic cells had been pre-treated with SC and RF extracts for 24 hours before application of the hydrogen peroxide. No neurotoxic effects were observed in the cells that had been treated by SC and RF. This prove that the treatment of SC and RF extract prevented apoptotic cell death of neuroblastic cell line exposed to oxidative injury. In addition, applying both SC and RF extracts at a 7:3 ratio increased the neuronal cell survival rate, compared to individual treatments of SC and RF extract. This study suggests that SC and RF extracts may be potential therapeutic agents for the prevention of neuronal cell death.

Up-regulation of Heme Oxygenase-1 by Korean Red Ginseng Water Extract as a Cytoprotective Effect in Human Endothelial Cells

  • Korean red ginseng (KRG) is used worldwide as a popular traditional herbal medicine. KRG has shown beneficial effects on cardiovascular diseases, such as atherosclerosis, diabetes, and hypertension. Up-regulation of a cytoprotective protein, heme oxygenase (HO)-1, is considered to augment the cellular defense against various agents that may induce cytotoxic injury. In the present study, we demonstrate that KRG water extract induces HO-1 expression in human umbilical vein endothelial cells (HUVECs) and possible involvement of the anti-oxidant transcription factor nuclear factor-eythroid 2-related factor 2 (Nrf2). KRG-induced HO-1 expression was examined by western blots, reverse transcriptase polymerase chain reaction and immunofluorescence staining. Specific silencing of Nrf2 genes with Nrf2-siRNA in HUVECs abolished HO-1 expression. In addition, the HO inhibitor zinc protoporphyrin blunted the preventive effect of KRG on $H_2O_2$-induced cell death, as demonstrated by terminal transferase dUTP nick end labeling assay. Taken together, these results suggest that KRG may exert a vasculoprotective effect through Nrf2-mediated HO-1 induction in human endothelial cell by inhibition of cell death.
