1,3-치환-5-chloropyrazole-4-carboxylic acid oxime ester의 합성과 살균력 (Synthesis and Antifungal Activity of 1,3-substituted-5-chloropyrazole-4-carboxylic acid Oxime Esters)

  • 생리활성을 갖는 신규물질 탐색의 일환으로 1,3 치환 -5-chloropyrazole-4-carbonyl chloride를 합성한 뒤 8종의 aldoxime 또는 ketoxime과 친핵 반응시켜 15종의 pyrazole-oxime ester를 얻었으며, 이들의 구조를 $^1H,\;^{13}C-NMR$ 및 IR 등으로 확인하였다. 이 유도체들의 R. solani, P. oryzae, B. cinerea, P. graminearum, P. capsici 및 G. cingulata에 대한 균사생육억제효과를 검정한 결과, oxime 부위$(R^3)$가 친전자성기로 치환된 방향족인 경우가(III, XIII, XIV) 포화알킬기인 경우보다 우수한 효과를 보여주었다.

한국산 개비자(Cephalotaxus koreans)에서의 Harringtonine과 Homoharringtonine의 확인 및 함량 분석 (Identification of Harringtonine and Homoharringtonine and Their Contents in Korean Native Plumyew(Cephalotaxus koreana))

  • 한국산 개비자로부터 alkaloid계통의 성분인 har­r ringtonine과 homoharringtonine의 혼합물을 col umn chromatography (CHCb: MeOH == 19: 1, Rf== 0 0.28)로 분리, 정제하였다. 이 두 물질의 혼합물의 화학 구조는 IH-NMR에 의해 결정하였으며 이미 보고된 spectra와 이 두 물질의 혼합물의 spectra를 비교하여 harringtonine과 homoharringtonine 임을 확인하였다. 한국산 개비자의 잎, 줄기, 뿌리에서의 harringtonine과 homoharringtonine의 함량은 고속 액체 크래마토그래피에 의해 결정하였다. 두 물질의 함량은 서식지, 식물의 부위에 따라 상이하였다. Harringtonine의 함량은 줄기보다 잎과 뿌리에서 높 았고, homoharringtonine의 함량은 harringtonine 함량보다 낮았다. 팔꽁산, 덕유산, 백양산, 지리산 빛 남해에서의 잎의 homoharringtonine 함량은 줄기와 뿌리에서 높게 나왔으며, 조계산과 진도에서는 잎의 함량이 뿌리와 줄기에서 보다 낮았다.

오메프라졸과 Hydroxypropyl-$\beta$-cyclodextrin의 포접화합물의 형성과 특성 (Complexation and Properties of Omeprazole with Hydroxypropyl-$\beta$-cyclodextrin)

  • Inclusion complex of omeprazole(OMZ) with hydroxypropyl-$\beta$-cyclodextrin(HP-$\beta$-CD) was prepared by freeze-drying method. The complexation was confirmed by means of UV, CD, IR, DSC, XRD and $^{1}$H-NMR spectra. The benzimidazole moiety of OMZ might be found to be included into the cavity of HP-$\beta$-CD and the inclusion complex appeared to be $A_{L}$ type. The stoichiometric ratio of OMZ : HP-$\beta$-CD was found to be 1:1, and the stability constants of the inclusion complex by solubility and UV method were about 34 and 45 M$^{-1}$, respectively. The dissolution of OMZ was significantly enhanced from powder and. yablet of OMZ-HP-$\beta$-CD complex when compared to those of OMZ alone, and oil-to-water partition coefficient of OMZ-HP-$\beta$-CD complex was significantly higher than that of OMZ alone. Therefore, it was expected that the bioavailability of OMZ could be improved markedly by inclusion complexation of OMZ with HP-$\beta$-CD.

한국산 유색미의 Anthocyanin 색소 동정 (Identification of Anthocyanins from Korean Pigmented Rice)

  • 한국산 유색미(수원 415호)로부터 안토시아닌 색소를 분석하였다. 1%HCl-methanol로 추출된 색소액을 자외선-가시광선 분광분석기로 측정한 결과 280 nm와 530 nm에서 최대흡광도를 보여 안토시아닌임을 확인 하였다. 분리된 색소를 산 분해하여 얻은 aglycon을 여러종류의 용매계로 전개시켜 $R_1$값을 비교하고, 흡광성질 및 $^1H-NMR$ 자료를 분석한 결과 aglycon은 cyanidin 및 malvidin으로 동정되었고, 결합당을 종이 크로마토그라피로 분석한 결과 monoglucose로 확인되었다. 따라서 한국산 유색미(수원 415호)의 안토시아닌 색소는 cyanidin-3-glucoside 및 malvidin-3-glucoside로 추정된다.

  • 전분은 친환경적인 천연자원이다. 건축 재료로서 활용 가능한 전분-아크릴 코팅졸을 유화중합으로 합성하였다. 제조된 합성물은 IR, $^1H$-NMR을 이용하여 구조를 분석하였고, X-ray Diffraction, 발포 시험, 백색도 시험, 광택도 시험과 인장 강도를 측정을 통하여 물리적 특성을 조사하였다. 전분-아크릴 폴리머 메트릭스에서 전분의 무정형 상태는 XRD결과로 알 수 있었다. 에멀젼을 60% $CaCO_3$ 수용액과 1%, 3%, 5% 발포제와 배합하였다. 그 결과 전분과 발포제의 양이 증가할수록 발포율이 증가하였다. 전분과 발포제의 양이 증가하면 인장 강도가 증가하였다. 그러나 전분과 발포제의 증가로 인해 백색도와 광택도는 감소하였다.

Evaluation of Hydration Effect on Human Skin by $^1H$ MRS at 14.1T

  • Purpose: We achieved high resolution MR imaging and spectra of human skin in vitro with using a 14.1 T MRI/MRS system, and evaluated the hydration effect of various cosmetic products by measuring the skin's. moisture concentration. Materials and Methods: We used the Bruker 14.1 T MRI/MRS system with a vertical standard bore that was equipped with a DMX spectrometer gradient system (200 G/cm at a maximum 40 A), RF resonators (2, 5 and 10 mm) and Para Vision software. Spin echo and fast spin echo pulse sequences were employed for obtaining the high resolution MR images. The 3D-localized point resolved spectroscopy (PRESS) method was used to acquire the MR spectra. Results: The high resolution MR images and spectra of human skin in vitro were successfully obtained on a 14.1 T system. The water concentration of human skin after applying a moisturizer was higher than that before applying a moisturizer. Conclusions: The present study demonstrated that the high-resolution MR images and spectra of human skin from a high field NMR instrument could be applicable to evaluating the hydration state of the stratum corneum.

Metabolic Profiling of Urine Samples from Colorectal Cancer Patients Before and After Surgical Treatments

  • Metabolites of urine samples from 6 colorectal cancer patients were analyzed by two-dimensional NMR spectroscopy, where the samples were collected before and after the surgical treatments per patient. NMR data were analyzed with the help of the metabolome database and the statistics software. Urine samples before and after the treatments showed significantly different metabolic profiles from each other. We were able to compare 10 different metabolites. Most of the assigned metabolites of every patient showed a tendency of increase after the surgery except for a few cases. The amount of changes in individual metabolites varied significantly from patient to patient, but the combination of such changes could be used to distinguish the condition before the surgery from after, which could be done by PCA analysis. The analysis via $^{1}H-^{13}C$ HSQC spectra proved to be applicable in assessing and classifying global metabolic profiles of the urines from colorectal cancer patients.

  • Synthesis of N-(1H-benzimidazol-2-yl)-6-substituted-1,3-benzothiazol-2-amines and 6-substituted-N-(4,5-dihydro-1H-imidazol-2-yl)-1,3-benzothiazol-2-amines by the reaction of substituted 2-aminobenzothiazoles with carbon disulphide and methyl iodide followed by the reaction with o-phenylene diamine/ethylene diamine are reported. All the synthesized compounds were characterized by elemental analysis, IR spectra and $^1H$ NMR spectral studies. The potent antibacterial and entomological (antifeedant, acaricidal, contact toxicity and stomach toxicity) activities of the synthesized compounds were investigated.

Effect of Cholesterol on the Phase Change of Lipid Membranes by Antimicrobial Peptides

  • Membrane disruption by an antimicrobial peptide (AMP) was investigated by measuring the $^2H$ solid-state nuclear magnetic resonance spectra of 1-palmitoyl-$d_{31}$-2-oleoyl-sn-glycero-3-phosphatidylcholine (POPC_$d_{31}$) in mixtures of POPC_$d_{31}$/cholesterol and either magainin 2 or aurein 3.3 deposited on thin cover-glass plates. The line shapes of the experimental $^2H$ solid-state nuclear magnetic resonance (SSNMR) spectra were best simulated by assuming the coexistence of a mosaic spread of bilayers containing pore structures and a fasttumbling isotropic phase or a hexagonal phase. Within a few days of incubation in a hydration chamber, an isotropic phase and a pore structure were induced by magainin 2, while in case of aurein 3.3 only an isotopic phase was induced in the presence of a bilayer phase. After an incubation period of over 100 days, alignment of the bilayers increased and the amount of the pore structure decreased in case of magainin 2. In contrast with magainin 2, aurein 3.3 induced a hexagonal phase at the peptide-to-lipid ratio of 1/20 and, interestingly, cholesterol was not found in the hexagonal phase induced by aurein 3.3. The experimental results indicate that magainin 2 is more effective in disrupting lipid bilayers containing cholesterol than aurein 3.3.

  • A new methacrylate copolymer that includes chalcone as a side group, poly(4-methacryloyloxyphenyl-4'-methoxystyryl ketone-co-styrene) was synthesized by free radical copolymerization. FTIR and $^1H$ NMR spectroscopic techniques were used to characterize the homopolymers and copolymers. The copolymerizations were carried out to high conversions. Copolymer compositions were established by $^1H$ NMR spectra analysis. The monomer reactivity ratios for copolymer system were determined by the linearized Kelen $T{\ddot{u}}d{\ddot{o}}s$, and Extended Kelen $T{\ddot{u}}d{\ddot{o}}s$ methods and a non-linear least squares method. The molecular weights and polydispersity index of copolymers were measured by using the gel permeation chromatography (GPC). The effect of copolymer compositions on their thermal behavior were studied by differential scanning calorimetry and thermogravimetric analysis methods. The optical properties of the resulting copolymer were also investigated.
