Kaempferol Activates G2-Checkpoint of the Cell Cycle Resulting in G2-Arrest and Mitochondria-Dependent Apoptosis in Human Acute Leukemia Jurkat T Cells

  • The effect of kaempferol (3,5,7,4-tetrahydroxyflavone), a flavonoid compound that was identified in barnyard millet (Echinochloa crus-galli var. frumentacea) grains, on G2-checkpoint and apoptotic pathways was investigated in human acute leukemia Jurkat T cell clones stably transfected with an empty vector (J/Neo) or a Bcl-xL expression vector (J/Bcl-xL). Exposure of J/Neo cells to kaempeferol caused cytotoxicity and activation of the ATM/ATR-Chk1/Chk2 pathway, activating the phosphorylation of p53 (Ser-15), inhibitory phosphorylation of Cdc25C (Ser-216), and inactivation of cyclin-dependent kinase 1 (Cdk1), with resultant G2-arrest of the cell cycle. Under these conditions, apoptotic events, including upregulation of Bak and PUMA levels, Bak activation, mitochondrial membrane potential (Δψm) loss, activation of caspase-9, -8, and -3, anti-poly (ADP-ribose) polymerase (PARP) cleavage, and accumulation of apoptotic sub-G1 cells, were induced without accompanying necrosis. However, these apoptotic events, except for upregulation of Bak and PUMA levels, were completely abrogated in J/Bcl-xL cells overexpressing Bcl-xL, suggesting that the G2-arrest and the Bcl-xL-sensitive mitochondrial apoptotic events were induced, in parallel, as downstream events of the DNA-damage-mediated G2-checkpoint activation. Together these results demonstrate that kaempferol-mediated antitumor activity toward Jurkat T cells was attributable to G2-checkpoint activation, which caused not only G2-arrest of the cell cycle but also activating phosphorylation of p53 (Ser-15) and subsequent induction of mitochondria-dependent apoptotic events, including Bak and PUMA upregulation, Bak activation, Δψm loss, and caspase cascade activation.

교맥의 RBL-2H3 비만세포 탈과립과 cytokine 생산 억제 효과 (Inhibitory effect of Fagopyrum esculentum on degranulation and production of cytokine in RBL-2H3 cells)

  • Objectives : Fagopyrum esculentum(FE) has been used for removal of inflammation of internal organs and treatment of sore and ulcer by heat toxin in Korean herbal medicines. In this study, To investigated the protective effect of FE on allergic response, we determined whether FE inhibits allergic response. Methods : The effect of FE was analyzed by ELISA, RT-PCR and Western blot in RBL-2H3 cells. We investigated cell viability, ${\beta}$-hexosaminidase, as a marker of degranulation, cytokne, and intracellular ROS and MAPK and NF-${\kappa}B$ signaling. Results : We found that FE suppressed ${\beta}$-hexosaminidase release, the production of IL-4 and TNF-${\alpha}$ and intracellular ROS level in RBL-2H3 by the anti-DNP IgE plus DNP-HSA stimulation. FE also significantly inhibited cytokine mRNA expressions, such as IL-$1{\beta}$, IL-2, IL-3, IL-4, IL-5, IL-6, IL-13, TNF-${\alpha}$ and GM-CSF in RBL-2H3. In addition, PF suppressed the phospholyation of ERK1/2, JNK1/2, p38 and $I{\kappa}B{\alpha}$ and NF-${\kappa}B$ signal transduction pathway. Conclusions : Our results indicate that FE protects against allergic response and exerts an anti-inflammatory effect through the inhibition of degranulation and production of cytokines and ROS via the suppression MAPK and NF-${\kappa}B$ of signal transduction. 족태양방광경(足太陽膀胱經)에서 투사(投射)되는 신경원(神經元)의 표지부위(標識部位)에 대(對)한 연구(硏究) (Central Localization of the Neurons Projecting to the Urinary Bladder Meridian)

  • Transsynaptic tracer이며 신경친화성 virus인 pseudorabies virus(PRV)를 방광(膀胱), 방광유(膀胱兪), 위중(委中) 및 중추(中極)에 주입(注入)한 후 4일간의 생존기간이 경과한 후 희생시켜 면역조직화학침액법(免疫組織化學染色法)에 의하여 뇌척수에 표지된 공통된 영역들을 비교하여 관찰한 결과는 다음과 같다. 1. 방광벽(膀胱壁), 방광유(膀胱兪), 위중(委中) 및 중추(中極)에서 척수에 투사된 영역은 흉수(胸髓), 요수(腰髓) 및 천수(薦髓)에 모두 표지되었으며 공통적으로 표지된 부위는 척수(脊髓)의 층판 IV, V, VII, IX, X영역에 표지되었으나 주로 강하게 표지된 공통된 영역은 층판 VII의 중간외측핵, 가슴기둥 및 층판 X영역이었다. 2. 방광벽(膀胱壁), 방광유(膀胱兪), 위중(委中) 및 중추(中極)에서 뇌(腦)에 투사된 공통된 영역은 연수(延髓)에서는 A1 noradrenalin cells/C1 adrenalin cells/caudoventrolateral reticular nucleus에서 양성반응을 나타내었다. 솔기핵의 경우 아핵인 불명솔기핵, 창백솔기핵 및 큰솔기핵에서 양성반응을 보였다. 다리뇌에서는 청색반점, Barrington's nucleus, A5세포군 및 삼차신경운동핵에서 양성반응을 보였고, 중뇌에서는 눈돌림신경핵, 눈돌림신경섬유 및 다리핵에서 양성반응을 보였다. 간뇌에서는 시상하부(視床下部)의 뇌실(腦室)곁핵과 시상의 뇌실곁핵에서 양성반응을 보였고 대뇌(大腦)에서는 septal nucleus, 피질(皮質)의 뒷다리영역, 마루엽, 이마엽에서 양성반응을 보였다. 이상의 결과를 종합하면 방광(膀胱)에서 투사되는 뇌척수의 영역과 방광유(膀胱兪)나 위중(委中)에서 투사되는 공통된 표지영역들은 방광(膀胱)과 족태양방광경(足太陽膀胱經) 그리고 그 경락(經絡)의 경혈(經穴)들이 어떤 상관성(相關性)을 가지고 연결(連結)되어 있다는 사실을 실험적으로 알 수 있었다. 특히 방광(膀胱)과 방광유(膀胱兪), 위중(委中)에서 투사된 공통된 표지영역, 즉 배뇨중추인 Barrington's nucleus에 표지되는 것은 내장(內臟)-경락(經絡)이 central autonomic pathway에 의하여 서로 연결되었음을 입증하는 중요한 결과(結果)라고 사려(思慮)된다.

아까시 나무 고온추출물의 항염증 효과 (Anti-inflammatory Effect of the Robinia pseudoacacia L. High Temperature Extract)

  • 본 연구는 아까시나무의 물 추출물, 에탄올 추출물 및 고온 추출물을 이용하여 마우스 대식세포주인 Raw 264.7 세포에 대해 염증억제 효과가 있는지 알아보고자 수행하였다. RP1(아까시 나무 물 추출물), RP2(아까시 나무 에탄올 추출물) 및 RP3(아까시 나무 고온 추출물)은 세포생존율 분석에서 $200{\mu}g/m{\ell}$의 농도까지 Raw 264.7 세포에 세포독성을 나타나지 않았다. NO 생성 억제효과를 분석하였을 때 LPS 처리군과 비교하여 RP3는 약 87% 정도의 억제효과를 나타내 RP1과 RP2에 비해 NO 억제활성이 가장 높았다. 뿐만 아니라 RP3는 RP1과 RP2와 비교하여 염증매개인자의 억제율이 각각 $PGE_2$ (86.3%), $TNF-{\alpha}$ (64.1%), IL-6 (65.1%) 및 $IL-1{\beta}$ (63.3%)로 가장 높았다. 이는 RP3의 처리가 LPS에 의해 증가된 염증매개인자의 분비를 억제함을 통해 항염증 효과가 있을 것으로 생각되며, 염증관련 신호전달경로에 직접적으로 작용할 가능성이 있는 것으로 판단된다. 하지만 Raw 264.7 세포주는 염증조절복합체를 구성하는 ASC 단백질이 발현되지 않아 다른 신호전달 경로를 통해 염증매개인자를 분비하기 때문에, 설치류의 대식세포를 직접 일차배양(primary culture)하여 이에 관련된 신호전달경로를 확인하는 추가 실험이 필요하다고 사료된다.

지하수 중 탄소원으로 fumarate 주입과 유전자분석을 통한 질산성질소 자연저감도 평가 (Evaluation of Natural Attenuation by Addition of Fumarate as Carbon Source and Gene Analysis in Groundwater Sample)

  • In the results of monitoring nitrate concentration in more than 8,000 groundwater wells around agro-livestock, the average and maximum nitrate concentration was 9.4 mg/L and 101.2 mg/L, respectively. Since about 31% of the monitoring wells was exceed the quality standard for drinking water, nitrate control such as remediation or source regulation is required to conserve safe-groundwater in South Korea. Typical nitrate-treatment technologies include ion exchange, reverse osmosis, and biological denitrification. Among the treatment methods, biological denitrification by indigenous microorganism has environmental and economic advantages for the complete elimination of nitrate because of lower operating costs compared to other methods. Major mechanism of the process is microbial reduction of nitrate to nitrite and nitrogen gas. Three functional genes (nosZ, nirK, nirS) that encode for the enzyme involved in the pathway. In this work, we tried to develop simple process to determine possibility of natural denitrification reaction by monitoring the functional gene. For the work, the functional genes in nitrate-contaminated groundwater were monitored by using PCR with specific target primers. In the result, functional genes (nosZ and nirK) encoding denitrification enzymes were detected in the groundwater samples. This method can help to determine the possibility of natural-nitrate degradation in target groundwater wells without multiplex experimental process. In addition, for field-remediation application we selected nitrate-contaminated site where 200~600 mg/L of nitrate is continuously detected. To determine the possibility of nitrate-degradation by stimulated-natural attenuation, groundwater was sampled in two different wells of the site and nitrate concentration of the samples was 300 mg/L and 616 mg/L, respectively. Fumarate for different C/N ratio was added into microcosm bottles containing the groundwater to examine denitrification rate depending on carbon concentration. In the result, once 1.5 times more than amount of fumarate stoichiometry required was added, the 616 mg/L of nitrate and 300 mg/L of nitrate were completely degraded in 8 days and 30 days. The nitrite, byproduct of denitrification process, was also completely degraded during the experimental period.

옥수수수염 추출물이 SW480 Colon Cancer Cell에서 NF-κB와 염증성 사이토카인 발현에 미치는 영향 (The Effect of Saccharin on the Gene Expression of NF-κB and Inflammatory Cytokines in LPS-Stimulated SW480 Colon Cancer Cells)

  • There have been no published studies concerning the anti-inflammatory effects of corn silk on colon cancer cells. Thus, this study was conducted to investigate the effect of corn silk extract containing high levels of maysin on inflammation and its mechanism of action in colon cancer cells. SW 480 human colon cancer cells were treated with $1{\mu}g/mL$ of lipopolysaccharide (LPS) to induce inflammation, and next they were treated with different concentrations of corn silk extract (0, 5, 10 and $15{\mu}g/mL$). The concentrations of nitric oxide (NO) were determined. The mRNA expressions of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), tumor necrosis factor ${\alpha}$ ($TNF-{\alpha}$), interleukin-1beta ($IL-1{\beta}$) and interleukin-6 (IL-6), were determined. Western blot analysis was performed to determine the protein expressions of nuclear factor-kappa B ($NF-{\kappa}B$) and mitogen-activated protein kinases, and the latter consists of extracellular signal-related kinase (ERK), c-jun NH2-terminal kinase (JNK) and p38 MAP kinase (p38). The concentration of NO and the mRNA expression of iNOS were significantly and dose-dependently decreased in the corn silk-treated groups (P<0.05). The mRNA expression of $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6 were significantly increased in the LPS-treated group (P<0.05), but these expressions were significantly and dose-dependently decreased in the corn silk treated groups (P<0.05). The protein expressions of $NF-{\kappa}B$ (in a dose-dependent fashion), ERK (at 10 and $15{\mu}g/mL$), JNK (at $15{\mu}g/mL$) and p38 (at 10 and $15{\mu}g/mL$) were significantly decreased with corn silk treatments (P<0.05). In conclusion, corn silk extract containing high levels of maysin seems to inhibit the LPS-induced inflammatory responses in SW480 colon cancer cells via the $NF-{\kappa}B$ pathway.

Assessment of Environmental Radioactivity Surveillance Results around Korean Nuclear Power Utilization Facilities in 2017

  • Background: Government conducts environmental radioactivity surveillance for verification purpose around nuclear facilities based on the Nuclear Safety Law and issues a surveillance report every year. This study aims to evaluate the short and the long-term fluctuation of radionuclides detected above MDC and their origins using concentration ratios between these radionuclides. Materials and Methods: Sample media for verification surveillance are air, rainwater, groundwater, soil, and milk for terrestrial samples, and seawater, marine sediment, fish, and seaweed for marine samples. Gamma-emitting radionuclides including $^{137}Cs$, $^{90}Sr$, Pu, $^3H$, and $^{14}C$ are evaluated in these samples. Results and Discussion: According to the result of the environmental radioactivity verification surveillance in the vicinity of nuclear power facilities in 2017, the anthropogenic radionuclides were not detected in most of the environmental samples except for the detection of a trace level of $^{137}Cs$, $^{90}Sr$, Pu, and $^{131}I$ in some samples. Radioactivity concentration ratios between the anthropogenic radionuclides ($^{137}Cs/^{90}Sr$, $^{137}Cs/^{239+240}Pu$, $^{90}Sr/^{239+240}Pu$) were similar to those reported in the environmental samples, which were affected by the global fallout of the past nuclear weapon test, and Pu atomic ratios ($^{240}Pu/^{239}Pu$) in the terrestrial sample and marine sample showed significant differences due to the different input pathway and the Pu source. Radioactive iodine ($^{131}I$) was detected at the range of < $5.6-190mBq{\cdot}kg-fresh^{-1}$ in the gulfweed and sea trumpet collected from the area of Kori and Wolsong intake and discharge. A high level of $^3H$ was observed in the air (Sangbong: $0.688{\pm}0.841Bq{\cdot}m^{-3}$) and the precipitation (Meteorology Post: $199{\pm}126Bq{\cdot}L^{-1}$) samples of the Wolsong nuclear power plant (NPP). $^3H$ concentration in the precipitation and pine needle samples showed typical variation pattern with the distance and the wind direction from the stack due to the gaseous release of $^3H$ in Wolsong NPP. Conclusion: Except for the detection of a trace level of $^{137}Cs$, $^{90}Sr$, Pu, and $^{131}I$ in some samples, anthropogenic radionuclides were below MDC in most of the environmental samples. Overall, no unusual radionuclides and abnormal concentration were detected in the 2017's surveillance result for verification. This research will be available in the assessment of environment around nuclear facilities in the event of radioactive material release.

생쥐 소장 카할세포에서 발생하는 향도잡이 기능의 올란자핀 효능에 관한 연구 (Effects of Olanzapine on the Pacemaker Potentials of Interstitial Cells of Cajal in Murine Small Intestine)

  • 카할세포는 위장관에서 향도잡이 역할을 한다. 본연구에서는 생쥐 소장 카할세포에서 발생되는 향도잡이 기능에서 올란자핀의 역할을 연구하였다. 패치클램프 방법을 사용하여 향도잡이 전압을 측정하였다. 올란자핀에 의해서 카할세포 향도잡이 전압이 탈분극 되었으며, 이 탈분극은 무스카린성 3번 수용체 억제제에 의해서 억제 되었다. 세포내 $GDP{\beta}S$을 넣어주니 올란자핀에 의해 향도잡이 전압 탈분극이 억제되었다. 또한, 세포밖 $Na^+$ 농도 감소와 비선택성 양이온 통로 억제제에 의해서 올란자핀에 의한 향도잡이 전압 탈분극이 억제 되었다. 세포내 PLC기전의 억제제인 U-73122에 의해서 올란자핀에 의한 향도잡이 전압 탈분극이 억제 되었다. 이러한 결과로 올란자핀은 무스카린성 3번 수용체를 통해서 세포내 G 단백질과 PLC기전 및 세포밖 $Na^+$이 관여함을 알 수 있었다. 따라서 올란자핀은 카할세포를 통해서 장운동성을 조절 할 수 있을 것으로 생각된다.

Gintonin-mediated release of astrocytic vascular endothelial growth factor protects cortical astrocytes from hypoxia-induced cell damages

  • Background: Gintonin is a ginseng-derived exogenous ligand of the G protein-coupled lysophosphatidic acid (LPA) receptor. We previously reported that gintonin stimulates gliotransmitter release in primary cortical astrocytes. Astrocytes play key roles in the functions of neurovascular systems. Although vascular endothelial growth factor (VEGF) is known to influence the normal growth and maintenance of cranial blood vessels and the nervous system, there is little information about the effect of gintonin on VEGF regulation in primary astrocytes, under normal and hypoxic conditions. Methods: Using primary cortical astrocytes of mice, the effects of gintonin on the release, expression, and distribution of VEGF were examined. We further investigated whether the gintonin-mediated VEGF release protects astrocytes from hypoxia. Results: Gintonin administration stimulated the release and expression of VEGF from astrocytes in a concentration- and time-dependent manner. The gintonin-mediated increase in the release of VEGF was inhibited by the LPA1/3 receptor antagonist, Ki16425; phospholipase C inhibitor, U73122; inositol 1,4,5- triphosphate receptor antagonist, 2-APB; and intracellular $Ca^{2+}$ chelator, BAPTA. Hypoxia further stimulated astrocytic VEGF release. Gintonin treatment stimulated additional VEGF release and restored cell viability that had decreased due to hypoxia, via the VEGF receptor pathway. Altogether, the regulation of VEGF release and expression and astrocytic protection mediated by gintonin under hypoxia are achieved via the LPA receptor-VEGF signaling pathways. Conclusion: The present study shows that the gintonin-mediated regulation of VEGF in cortical astrocytes might be neuroprotective against hypoxic insults and could explain the molecular basis of the beneficial effects of ginseng on the central nervous system.

산마늘로부터 단리한 kaempferol과 quercetin의 콜레스테롤 저하 활성 (Cholesterol inhibitory activities of kaempferol and quercetin isolated from Allium victorialis var. platyphyllum)

  • 식용 임산자원인 산채류를 기능성 식품으로 개발하고자 산마늘을 비롯한 총 13종의 에탄올 조추출물에 대한 콜레스테롤 저하활성을 검정하였다. 즉, 콜레스테롤 생합성 과정 초기에 관여하는 3-hydroxy-3-methylglutaryl coenzyme A reductase(HMG-CoA reductase)와 후기에 관여하는 squalene synthase의 효소 활성을 조사한 결과 산마늘 잎 에탄올 추출물이 두 효소의 활성을 공히 70% 이상 저해하여 활성이 가장 우수한 것으로 나타났다. 그리고 이러한 콜레스테롤 저하활성과 관련이 있는 물질을 탐색하고자 산마늘로부터 물질 단리를 시도하여 디클로로메탄 가용부로부터 kaempferol과 quercetin을 단리하였다. 또한 유전자 레벨에서의 콜레스테롤 저하 활성을 조사하기 위해 단리물질과 분획물을 C100세포(햄스터 유래 HMG-CoA reductase 고발현 세포주)에 각각 5 ㎍/㎖과 10 ㎍/㎖로 24시간 처리하여 HMG-CoA reductase와 squalene synthase의 mRNA 발현 정도를 조사하였다. 그 결과 10 ㎍/㎖로 kaempferol과 quercetin을 처리한 경우 두 효소의 mRNA가 전혀 발현하지 않는 것으로 나타나 유전자 레벨에서의 콜레스테롤 생합성 저해 효과를 확인할 수 있었다. 이상의 결과 산마늘 잎 에탄올 추출물은 콜레스테롤 생합성에 관여하는 HMG-CoA reductase와 squalene synthase의 활성을 저해하며 이러한 활성 저해 효과는 kaempferol과 quercetin에 기인하는 것으로 사료되었다. 특히 kaempferol과 quercetin은 여러 식물의 성분으로서 이미 알려진 화합물이지만 콜레스테롤 저하활성이 있는 것으로 밝혀진 것은 이번이 처음으로 금후 이들 물질과 이들 물질을 함유하고 있는 식물 활용에 필요한 자료를 제공하였다고 사료된다.
