Survey of the Presence of Aflatoxins in Compound Feeds and Feed Ingredients (국내생산 사료의 Aflatoxin 오염도 조사)

  • Contamination of aflatoxins(AFs) was monitored in 447 compound feeds and 138 feed ingredients samples distributed in South KOREA in 2006 and 2007. The degree of $AFB_1\;and\;AFB_2$ contamination in compound feed was 20% and 3%, respectively. The levels of detection were ranged from 0.48 to 10.46 ppb for $AFB_1$ and from 0.25 to 0,42 ppb for $AFB_2$. Thirty eight percent of compound feeds were contaminated with $AFB_1$ at concentration between 0.43 and 5.52 ppb and $AFB_2$ was detected in 2% of compound feeds at levels ranging 0.26-0.40 ppb. The highest degree of $AFB_1$ contamination was observed in compound feeds for beef cattle (75%) followed by for dairy cattle (72%) and in bran among feed ingredients (30%). Bran exhibited the highest level of $AFB_1$contamination (3.1 ppb). Vegetable proteins and compound feeds for dog showed relative lower degree of contamination at 2.9 and 1.9 ppb, respectively. $AFG_1\;and\;AFG_2$ were not detected in any compound feeds and feed ingredients samples.

DNA Adduct Formation and Expression of Ras Gene in the Liver of Rats Treated with Aflatoxins at Various Levels (랫드의 간에서 다양한 농도의 아플라톡신 투여에 의한 DNA Adduct의 형성과 Ras의 발현양상)

  • Aflatoxins are produced by Aspergillus flavus, parasiticus that grows in improperly stored cereals. Aflatoxin $B_1\;(AFB_1)$ is a potent hepatocarcinogen in a variety of experimental animals including human beings. In spite of a high attention to the hepatocarcinogenecity of aflatoxins, the relative toxicity of other types $(AFB_2,\;AFG_1\;and\;AFG_2)$ of the toxins is not fully clarified. Sprague-Dawley male rats were orally administered with $AFB_1,\;AFB_2,\;AFG_1\;and\;AFG_2$ at the dose of 250, 1250, and $2500\;{\mu}g/kg$ body weight. Animals were then killed at 12, 24 or 48 hrs following aflatoxin adminstration. Subsequently the relative weight of liver was measured and histopathological examination on the liver was performed. Level of 8-OxodG and expression of ras gene in the liver was determined. The relative liver weights at high doses of $AFB_1\;and\;AFG_1$ was significantly low. The treatment of $AFB_1$ at the high dose of $2500\;{\mu}g/kg$ showed vacuolar degeneration and centrilobular hepatic necrosis with inflammatory cells. The pathological changes by $AFB_2\;AFG_1,\;and\;AFG_2$ were not clearly found. The formation of 8-OxodG by $AFB_1$ increased in a dose-dependent manner up to 24 hrs after a single treatment of $AFB_1$ thereafter decreased to the level of the control. The treatments of $AFB_2\;AFG_1,\;and\;AFG_2$ showed an inconsistent pattern in the formation of 8-OxodG in the liver of rats with increasing time. The expression of ras oncogene in the liver by $AFB_1$ at the dose of $1250\;{\mu}g/kg$ was increased twice compared to the control. The treatments of $AFB_2\;AFG_1,\;and\;AFG_2$ at all doses decreased the expression of ras in the liver. These results in the present study indicate that $AFB_1$ among aflatoxins with low comparable levels is the most toxic as determined by early biomarkers such as 8-OxodG formation and ras expression. However, the levels of 8-OxodG and ras as biomarkers were not useful to predict the relative hepatocarcinogenicity of aflatoxins to $AFB_1$ in the present model. Further studies are required to look for other biomarkers to predict carcinogenic potency of aflatoxins.

Anticancer and Immuno-activity of Methanol Extract from Onion Kochujang (양파고추장 메탄올추출물의 항암 및 면역활성)

  • Antitumor activities of methanol extract from onion Kochujang (MEOK) were investigated by using aflatoxin $B_{1}-mediated$ Salmonella typhimurium mutagenicity and the model of cytotoxicity on the cancer cell lines. Their immune activities were also investigated using mouse spleen cells and macrophage cell lines, respectively. MEOK showed the enhanced antimutagenicity in a dose-dependent manner. relative to the control group. MEOK decreased over $20\%$ of the proliferation of the A549(lung cancer cell) and MCF(breast cancer cell) cell lines at $1,000\;{\mu}g/mL$ concentration compared with the control cells. The proliferation of mouse spleen cells and the NO production in marcrophage cell lines treated MEOK were increased in a dose-dependent manner, respectively. The activities were higher than that of the control(no added onion) Kochujang.

Investigation of Unintentionally Hazardous Substance in Teas (식수대용차 재료의 비의도적 유해물질 오염도 조사연구)

  • This study was conducted to survey levels of heavy metals, benzopyrene and mycotoxins in teas purchased from online markets, supermarkets, and traditional markets in Gwangju, Korea. We examined the arsenic, cadmium and lead content in heavy metals. We also tested for benzo(a)pyrene, which is classified as a carcinogen. In addition, we tested for the mycotoxins aflatoxin (AFB1, AFB2, AFG1, AFG2), ochratoxin A (OTA), fumonisin (FUB1, FUB2), and zearalenone (ZON). According to our results, heavy metals were detected within the allowable range (below 5.0 mg/kg for lead) and all samples met with the standards for tea as presented in the Korean Food Code. The Benzo(a)pyrene showed a detection rate of 12.8% in 16 of 125 samples and mycotoxins showed 23.2% in 29 of 125 samples. However, safe levels regarding benzo(a)pyrene and mycotoxin in tea were not ascertainable since the standards are not set in the Korean Food Code.

Survey of Yogurt Powder Storage in Ambient Export Countries A Safety Evaluation Standard Compliance and Comparative Analysis

  • Yogurt powder is fermented milk processed in the form of dry yogurt, and has advantages such as stability, storability, convenience, and portability. China and Vietnam are important export target countries because of the increased demand for dairy products. Therefore, we surveyed dairy product standardization in order to establish an export strategy. Lactic acid bacteria counts are unregulated in Korea and Vietnam. In China, lactic acid bacteria counts are regulated at $1{\times}10^6$ colonyforming units (CFU)/mL and detected at $6.24{\pm}0.33\;Log\;CFU/mL$. All three countries have regulated standards for total bacterial counts. In China, total bacterial counts of milk powder are regulated to n=5, c=2, m=50,000, M=200,000 and detected at $6.02{\pm}0.12\;Log\;CFU/mL$, exceeding the acceptable level. Lactic acid bacterial counts appeared to exceed total bacterial counts. Coliform group counts, Staphylococcus aureus, Listeria monocytogenes, and Salmonella species were not detected. Acidity is not regulated in Korea and Vietnam. In China, acidity was regulated to over $70^{\circ}T$ and detected $352.38{\pm}10.24^{\circ}T$. pH is unregulated in all three countries. pH was compared to that of general fermented milk, which is 4.2, and that of the sample was $4.28{\pm}0.01$. Aflatoxin levels are not regulated in Korea and China. In Vietnam, aflatoxin level is regulated at 0.05 ppb. Therefore, all ingredients of the yogurt powder met the safety standards. This data obtained in this study can be used as the basic data in assessing the export quality of yogurt powder.

Ochratoxin A Contamination of Red Chili Peppers from Chile, Bolivia and Peru, Countries with a High Incidence of Gallbladder Cancer

  • Our previous study detected aflatoxins in red chili peppers from Chile, Bolivia, and Peru, each of which have a high incidence of gallbladder cancer (GBC). Since the aflatoxin B1 concentration was not so high in these peppers, it is important to clarify the presence of other mycotoxins. Here we attempted to determine any associations between the concentrations of aflatoxins and ochratoxin A (OTA) in red chili peppers, and the corresponding GBC incidences. We collected red chili peppers from three areas in Peru: Trujillo (a high GBC incidence area), Cusco (an intermediate GBC incidence area), and Lima (a low GBC incidence rate), and from Chile and Bolivia. Aflatoxins and OTA were extracted with organic solvents. The concentrations of aflatoxins B1, B2, G1, and G2, and OTA were measured by high-performance liquid chromatography. The values obtained were compared with the incidence of GBC in each area or country. All of the red chili peppers from the three areas showed contamination with aflatoxins below the Commission of the European Communities (EC) recommended limits ($5{\mu}g/kg$), but the OTA contamination of two samples was above the EC recommended limit ($15{\mu}g/kg$). The mean concentrations of OTA in the peppers from Chile (mean $355{\mu}g/kg$, range < $5-1,059{\mu}g/kg$) and Bolivia (mean $207{\mu}g/kg$, range $0.8-628{\mu}g/kg$), which has a high incidence of GBC, were higher than that in Peru ($14{\mu}g/kg$, range < $5-47{\mu}g/kg$), which has an intermediate GBC incidence. The OTA contamination in the red chili peppers from Chile, Bolivia, and Peru was stronger than that of aflatoxins. Our data suggest that OTA in red chili peppers may be associated with the development of GBC.

Effects of Ammoniation or Supplementation of BHT, CTC, and Probiotics to Moldy Corn on the Performance of Broiler Chickens (곰팡이 쓴 옥수수를 사용할때 Ammonia 처리와 BHT, CTC, Probiotics의 첨가가 육계에 미치는 영향)

  • In order to study the effect of feeding moldy corn in the feed and to compare the efficacy of several antidotical treatments, a feeding trial was conducted using broiler chickens. Ground yet low corn was adjusted to contain 20% moisture and then incubated at 35$^{\circ}C$ for 20 days. The number of fungi increased from 11${\times}$10$^4$/g to 42 10$\^$7/g on incubation but aflatoxins (B$_1$, B$_2$, G$_1$, G$_2$) were not detected by TLC test. Moldy corn was included in the experimental diets at the level of 7% with various treatments. Thirty six hatched male broiler chickens were divided into 6 groups and each group was placed for 4 weeks on one of the following treatments: Fresh corn, Moldy corn, Ammoniated (1.5%) moldy corn, Moldy corn+BHT (0.1%), Moldy corn CTC (100ppm) and Moldy corn+Probiotics. Weight gains of the birds fed Ammoniated moldy corn diet, Moldy corn+Probiotics dist and Moldy corn+CTC diet were greater by 6.1%. 7.6% and 3.9% respectively than those fed Moldy corn diet. The feed efficiencies of Moldy corn+Probiotics diet and Moldy corn+CTC diet were better than those of Moldy corn diet. Dry matter contents of the livers of the birds fed Ammoniated moldy corn diet, Moldy corn+BHT diet and Moldy corn diet were lower than those of other treatments. Proteion contents of the livers of the birds fed Moldy corn diet, Moldy corn +CTC diet and Moldy corn+BHT diet were lower than those of other treatments. Moldy corn tended to increase fat content of the livers.

Analysis of Total Aflatoxins in Spices and Dried Fruits (향신료 및 건조과실류 중 총 아플라톡신의 분석)

  • We used fluorescence detector to analyse total aflatoxins (G1, G2, B1, B2) with TFA (Trifluoroacetic acid) derivation method and PHRED (Photochemical reactor enhanced detection) method. PHRED method was superior in reproduction and convenience, but TFA derivation method was superior in selectivity and sensitivity. The recovery rate of aflatoxin B1, B2, G1 were more than 80%, and G2 was more than 70%. The detection limit of B1, B2, G1 and G2 were respectively 0.05, 0.05, 0.2 and $0.1\;{\mu}g/kg$. Confirmed method was used to analyse total aflatoxins in total 316 items as 9 kinds 137 dried fruits and 10 kinds 179 spices. By the result, Aflatoxins were detected in 27 dried fruits (19.7%) and in 87 spices (48.6%).

Detection of Aflatoxin $M_1$ in Human and Porcine Urine and Its Risk Assessment (인축의 뇨 중 aflatoxin $M_1$의 오염분석 및 위해성 평가)

  • To conduct a risk assessment of $AFB_1$ intake, $AFM_1$, which is a metabolite of $AFB_1$ in the human and porcine urine, was determined by competitive direct ELISA (cdELISA). The detection limit of cdELISA using anti-$AFM_1$ antibody and $AFB_1$-HRP conjugate was 10 pg/mL. The recoveries of $AFM_1$ were 117-167% after the addition of $AFM_1$ in the human urine in a range of 3-100 pg/mL. 165 samples (95.5%) of those obtained from 172 persons evidenced measurable levels of urinary $AFM_1$. The detected $AFM_1$ ranges were 0-11.6 pg/mL and the average level of $AFM_1$ contamination was 2.74${\pm}$ 1.89 pg/mL. The estimated amount of $AFM_1$ excretion in the human urine was 3.97 ng/day and the estimated $AFB_1$ intake amount was 79.4 ng/day. The probable daily intake (PDI) of $AFB_1$ by the subjects was estimated to be 1.28 ng/kg bw/day, which was higher than the tolerable daily intake (TDI, 0.15 ng/kg bw/day). In the case of porcine urine, the $AFM_1$ ranged between 0.97-26.7 pg/mL and the average contaminated $AFM_1$ was 10.62${\pm}$4.39 pg/mL. The estimated amount of $AFM_1$ excretion in the porcine urine was 27.6 ng/day, and the estimated $AFB_1$ intake amount was 551 ng/day.

Administration of Mycotoxins in Food in Korea (식품 중 곰팡이독소 안전기준 관리)

  • Total aflatoxin ($B_1+B_2+G_1+G_2$) maximum levels of 15 ${\mu}g/kg$ ($B_1=10\;{\mu}g/kg$) were set for grain, beans, peanut, nuts & their processed food (grinding, cutting etc.), processed cereal product & processed bean product, confectionaries (peanut or nut-containing food), soybean paste, red pepper paste, dried red pepper, processed com products for popcorn and steamed rice. The maximum levels for aflatoxin $M_1$ are 0.5 ${\mu}g/kg$ for raw milk and milks before manufacturing processing. The patulin maximum level is 50 ${\mu}g/kg$ in apple juice and apple juice concentrate (including concentrate to use as raw material and converted by concentration multiple). The ochratoxin A is managed at the maximum levels of 5 ${\mu}g/kg$ in wheat, barley, rye, coffee beans and roasted coffee, 10 ${\mu}g/kg$ in instant coffee and raisin, 2 ${\mu}g/kg$ in Grape juice, concentrated grape juice as reconstituted and wine. The fumonisins ($B_1+B_2$) maximum levels are 4000 ${\mu}g/kg$ in com, 2000 ${\mu}g/kg$ in com processed food (grinding, cutting etc.) and com powder, 1000 ${\mu}g/kg$ in processed com products. Standards for mycotoxins in food have been established and the mycotoxin risk in food is managed reasonably and scientifically, based on risk assessment and exposure analysis.
