Microbial Studies on the Korean Traditional Soju -Part 1. Characteristics of Fungal Amylases Produced by the Isolate from the Native Youngkwang Koji- (우리나라 재래 소주에 관한 미생물학적 연구 -제1보. 영광 토종국에서 분리한 곰팡이의 Amylase 활성-)

  • The most active strain for the amylase activity from the native Youngkwang koji, was isolated and identified as Aspergjllus awamori. The optimal conditions for the production of ${\alpha}-amylase$ and glucoamylase were investigated and the properties of these enzymes were also examined. Maximum yields of ${\alpha}-amylase$ and glucoamylase were obtained at $30^{\circ}C$, pH 5.0 for days. The production of these two enzymes were increased with the addition of maltose, urea, $K_2HPO_4$, $MgSO_4{\cdot}7H_2O$, and $CaCl_2{\cdot}2H_2O$. The activities of these enzymes were maximized at $50^{\circ}C$, $pH\;5{\sim}6$. The heat stabilites of ${\alpha}-amylase$ and glucoamylase were maintained at $50^{\circ}C$ for 20min and 40min, respectively. Also, the addtion of $MgSO_4{\cdot}7H_2O$, $K_2HPO_4$, and $CaCl_2{\cdot}2H_2O$ salt increased the heat stabilities of these enzymes.

Saccharification of Uncooked Starch (무증자 전분의 당화에 관한 연구)

  • For the eventual alcohol production from uncooked starch, an efficient saccharification process was examined by using the combined action of steeping, pectin depolymerase, ${\alpha}-amylase$ and glucoamylase. The total sugar content of rice, sweet potato and tapioka used were 4.53, 4.26, and 3.92 mmole/g sample. $70\;{\pm}\;10%$ of the total sugar was hydrolyzed when cooked starch was saccharified under the condition which is currently used in industry. The smaller starch particle was used, the more saccharification was obtained. Efficient saccharification was obtained by treatment with 5% $H_2SO_4$ (sample: working volume = 1:2) at $60^{\circ}C$ for 12 hr. Optimization was carried out for the saccharification of uncooked starch by the combined action of pectin depolymerase, ${\alpha}-amylase$, and glucoamylase. The conditions are: pectin depolymerase; pH 4.5, $45^{\circ}C$, 2 hr, ${\alpha}-amylase$; pH 6.0, $60^{\circ}C$, 1 hr, and glucoamylase; pH 3.5, $60^{\circ}C$, 1 hr. Simultaneous treatment of the combined action of macerating, liquifying and saccharifying enzymes yielded better result than stepwise treatment of 3 enzymes. Degrees of saccharification of uncooked tapioka, rice and sweet potato were 82, 90.5, and 84.5%, respectively on the basis of total sugar, under the optimized conditions.

  • The porous silica membrane was prepared from Si(${OC}_2H_5)_4-H_2O$ system by sol-gel method. To investigate the characteristics of gels and porous silica membrane, we examined gels and porous silica membrane using TG-DTA, X-ray diffractometer, IR spectrophotometer, BET, SEM and TEM. The optimum mole ratio of Si(OC$_2$H$_{5}$)$_4$ : $H_2O$ $C_2$H$_{5}$OH for porous silica membrane was 1 : 4.5 : 4. The porous silica membrane was obtained by heat treatment of the gel above 700 $^{\circ}C$. The specific surface area of sintered gel was 3.8 $m^2$/g to 902.3 $m^2$/g at 100 $^{\circ}C$ to 1100 $^{\circ}C$ The pore size of sintered gel was in the range 20 $\AA$~ 50$\AA$. The particle size of sintered gel was 15 nm to 30 nm at 30$0^{\circ}C$ to 700$^{\circ}C$. The performance of the porous silica membrane was investigated for the separation of $H_2$/$N_2$ gas mixture. Gas separation through porous silica membrane depends upon Knudsen flow and surface flow. The veal separation factor($\alpha$) of $H_2$/$N_2$ was 5.17 at 155.15 cmHg and $25^{\circ}C$. The real separation factor($\alpha$), head separation factor($\beta$), and tail separation factor( $\bar{B}$) increased as the pressure of permeation cell Increased.sed.

Effect of pH values and inoculation amounts for α-glucosidase inhibitory activity in mulberry leaf fermentation

  • Mulberry leaves containing 1-deoxynojirimycin (DNJ) have been recognized as a potentially important source for prevent or treat hyperglycemia. However, DNJ content of natural mulberry leaf are as low as 0.1%. Thus, the most effective method for increasing ${\alpha}$-glucosidase inhibitory activity with the DNJ high-production is needed. In this study, we investigated the influence of ${\alpha}$-glucosidase inhibitory activity according to different pH values (6-9) and inoculation amounts (0.1-0.5%) when Bacillus subtilis cultured on mulberry leaf powder media. We confirmed that ${\alpha}$-glucosidase inhibitory activity was difference according to culture conditions of different pH values, inoculation amounts, and fermentation times. The results of mulberry leaf fermentation according to pH values and inoculation amounts were shown that the optimal conditions for ${\alpha}$-glucosidase inhibitory activity were defined as pH 7 and 9, inoculation amount 0.4%, and incubation until 2 to 4 days. These results can be provided a basic data for the optimal culture conditions increasing ${\alpha}$-glucosidase inhibitory activity from mulberry leaf fermentation.

Characterization of ${\alpha}$-Galactosidase and ${\beta}$-Glucosidase by Weissella cibaria (Weissella cibaria가 생산하는${\alpha}$-Galactosidase 및 ${\beta}$-Glucosidase의 특성)

  • A strain producing ${\alpha}$-galactosidase and ${\beta}$-glucosidase was isolated from Kimchi. The isolated strain was identified as Weissella cibaria by 16S rDNA analysis and designated as Weissella cibaria K-M1-4. The enzyme activity of ${\alpha}$-galactosidase and ${\beta}$-glucosidase reached the maximum in the soy medium at $37^{\circ}C$ for 24 hr. The enzymes were purified by ethanol fractionation, DEAE sepharose fast flow, and sephacryl S-100HR column chromatography. ${\alpha}$-Galactosidase specific activity was shown by 576 Units/mg protein and the yield was 3.5% of the total activity of crude extracts. ${\beta}$-glucosidase specific activity was shown by 480 Units/mg protein and the yield was 2.9% of the total activity of crude extracts. The optimum temperature for ${\alpha}$-galactosidase was $60^{\circ}C$ and 43% of its original activity remained when it was treated at $80^{\circ}C$ for 30 min. For ${\alpha}$-galactosidase shows the optimum pH of 8.0 and is fairly stable between pH5.0 and pH9.0. The enzyme activity was increased in the presence of $Fe^{2+}$ and $Cu^{2+}$. The value of Km and Vmax for the enzyme were 0.98 mM and $1.81{\mu}$mole/min, respectively. The ${\beta}$-glucosidase has the optimum temperature of $50^{\circ}C$ and 46% of its original activity remained when it was treated at $80^{\circ}C$ for 30min. Its optimum pH of 7.0 and is fairly stable between pH5.0 and pH9.0. The enzyme activity was increased in the presence of $Fe^{2+},\;Co^{2+}$ and $Cu^{2+}$. The value of Km and Vmax for the enzyme were 1.24 mM and $6.81{\mu}$mole/min, respectively.

Changes in Ovarian and Placental 20α-hydroxysteroid Dehydrogenase Activity during the Pregnancy in the Rat

  • The enzyme $20{\alpha}$-hydroxysteroid dehydrogenase ($20{\alpha}$-HSD) catabolizes progesterone to $20{\alpha}$-dihydroprogesterone ($20{\alpha}$-OHP), and is appeared in rat corpora luteal and placenta. A polled samples of 10-15 placental and ovarian tissues collected from each individual rat were subjected to measurement of $20{\alpha}$-HSD activity. A $20{\alpha}$-HSD activity in the cytosol fraction was based on the generation of NADPH. In this study, it is designed to study cytosolic $20{\alpha}$-HSD activity in rat ovarian and placenta during pregnancy, and its relationship to embryonic mortality. It was found that from days 5 to 18 of pregnancy the $20{\alpha}$-HSD activities steady by decreased but at parturition time rapidly increased in ovary. On the other hand, placental cytosolic $20{\alpha}$-HSD activities were high detected from days 8 to 10 of pregnancy, not detectable from days 11 to 20 of pregnancy, but again very high at the time of parturition. Analysis of DEAE column chromatography revealed that two different types of $20{\alpha}$-HSD (HSD-1 and HSD-2) were found with similar activity in the placental cytosol on day 10 of pregnancy. The number of fetuses on day 10 of pregnancy was 15.4 and decreased significantly to 12.9 on day 12. The results suggested that expression of $20{\alpha}$-HSD in the placental tissues seems to be related the number of fetal survived in the specific time (days 11 and 12) which spontaneous fetal loss occurs.

Production of 2-O--$\alpha$-D-Glucopyranosyl L-Ascorbic Acid by Cyclodextrin Glucanotransferase from Bacillus sp. JK-43 (Bacillus sp. JK-43의 Cyclodextrin Glucanotransferase에 의한 2-O-$\alpha$-D-Glucopyranosyl L-Ascorbic Acid 생산에 관한 연구)

  • The 2-O-$\alpha$-D-glucopyranosyl L-ascorbic acid (AA-2G) which was enzymatically glucosylated with the cyclodextrin glucanotransferase (CGTase) [EC 2.4.1.19] from Bacillus sp. JK-43 has been reported previously. The presnet experiments examined the optimal conditons for the productio of AA-2G from AA and soluble starch, and characterized the properties of the CGTase from Bacillus sp. JK-43. The reaction mixture for the maximal production of AA-2G was followings; 12% total substrate concentration, 1,400 usits/mL of CGTase and a mixing ratio of 2 : 3(g or AA : g of soluble starch). Under this condition, 1.76mM of AA-2G, which corresponded to 2.53% yield based on AA, was produced after incubation for 24hrs at 45$^{\circ}C$ (pH 5.5). The optimum pH and temperature for the CGTase activity were 6.0 and 45$^{\circ}C$, respectively. The enzyme was stable at pH 5.5 to 9.5, and at temperature up to 5$0^{\circ}C$. The thermostability of the enzyme could be enhanced up to 6$0^{\circ}C$ by the addition of 30mM CaCl2.

Factors Affecting on Pregnancy Rate of Recipients Following Transfer of Hanwoo Embryos Produced In Vivo (한우 체내 수정란 이식 후 수태율에 미치는 요인)

  • This study was performed to improve the pregnancy rates of recipients following transfer of bovine embryos produced in vivo. Superovulation response didn't showe significant differences between each season (4.18 in spring; 4.36 in summer; 5.50 in fall; 4.38 in winter). Pregnancy rate was significantly different (p<0.05) between fresh (43.4%) and frozen embryos (17.2%). In administration of hCG to recipients, the pregnancy rate of fresh embryos (45.7%) was slightly higher than that of control (35.3%), but the pregnancy rates of frozen embryos in control group (25.0%) was higher than that of hCG group (16.0%). When synchrony of recipient and embryo was -2, -1, 0 and 1, the pregnancy rates were 20.0, 45.0, 30.3 and 26.3%, respectively. The pregnancy rates of recipients synchronized by naturally or $PGF_{2{\alpha}}$, CIDR/$PGF_{2{\alpha}}$ and E/P/CIDR/$PGF_{2{\alpha}}$/E treatments were 35.3, 48.0, 29.0 and 40.0%, respectively. Gestation lengths and birth weights of female and male calf were 288 and 290.5 days, 28.3 and 30.0 kg, respectively. The results were showed that the superovulation response was not affected by seasons, and also pregnancy rate didn't increase by administration of hCG, synchrony of embryo and recipients, synchrony methods. Further study and concern should be focused on improving the embryo freezing and pregnancy rate for commercial embryo transfer.

QUENCHING OF TYPE II PHOTOSENSITIZER IN THEIR TRIPLET STATES BY $\alpha$-TOCOPHEROL VIA AN ELECTRON TRANSFER REACTION

  • Occurrence of an electron (or H atom equivalent to one electron plus H+) transfer from $\alpha$- tocopherol $\alpha$(TOH) to a number of photosensitizers in theri triplet states were investigated by monitoring the ESR signal of $\alpha$-chromaoxyl radical ($\alpha$(TO.) in ethanolic solutions of $\alpha$TOH and the sensitizers under continuous illumination. Every sensitizer molecule examined, such as protocholorophyllide (Pchl), hematoporphyrin and rose bengal which are generally regarded as efficient type II photosensitizers and thus have long-lived triplet states, was found to actively participate in an electro transfer reaction with $\alpha$TOH even under air-saturation conditions, generating $\alpha$TOH complex as an intermediate in a fashion of Michaelis-Menten type of reaction. For the reaction of $\alpha$TOH with triplet Pchl, the rate law was derived by applying the steady approximation for the binary complex, triplet Pchl-$\alpha$TOH , which turned out to be well consistent with the kinetic data.

In this paper, we consider the operator $T_{{\alpha},{\beta}}=V^{2{\alpha}}{\mathcal{L}}^{-{\beta}}_2$ and its conjugate $T^*_{{\alpha},{\beta}}$, where $0<{\alpha}{\leq}{\beta}{\leq}1$. We establish the $(L^p,\;L^q)$-boundedness of operator $T_{{\alpha},{\beta}}$ and $T^*_{{\alpha},{\beta}}$, respectively, we also show that $T_{{\alpha},{\beta}}$ is bounded from Hardy type space $H^1_{L_2}({\mathbb{R}}^n)$ into $L^{p_2}({\mathbb{R}}^n)$ and $T^*_{{\alpha},{\beta}}$ is bounded from $L^{p_1}({\mathbb{R}}^n)$ into BMO type space $BMO_{{\mathcal{L}}1}({\mathbb{R}}^n)$, where $p_1={\frac{n}{4({\beta}-{\alpha})}}$, $p_2={\frac{n}{n-4({\beta}-{\alpha})}}$.
