Material Discrimination Using X-Ray and Neutron

  • Background: A nondestructive test is commonly used to inspect the surface defects and internal structure of an object without any physical damage. X-rays generated from an electron accelerator or a tube are one of the methods used for nondestructive testing. The high penetration of X-rays through materials with low atomic numbers makes it difficult to discriminate between these materials using X-ray imaging. The interaction characteristics of neutrons with materials can supplement the limitations of X-ray imaging in material discrimination. Materials and Methods: The radiation image acquisition process for air-cargo security inspection equipment using X-rays and neutrons was simulated using a GEometry ANd Tracking (Geant4) simulation toolkit. Radiation images of phantoms composed of 13 materials were obtained, and the R-value, representing the attenuation ratio of neutrons and gamma rays in a material, was calculated from these images. Results and Discussion: The R-values were calculated from the simulated X-ray and neutron images for each phantom and compared with those obtained in the experiments. The R-values obtained from the experiments were higher than those obtained from the simulations. The difference can be due to the following two causes. The first reason is that there are various facilities or equipment in the experimental environment that scatter neutrons, unlike the simulation. The other is the difference in the neutron signal processing. In the simulation, the neutron signal is the sum of the number of neutrons entering the detector. However, in the experiment, the neutron signal was obtained by superimposing the intensities of the neutron signals. Neutron detectors also detect gamma rays, and the neutron signal cannot be clearly distinguished in the process of separating the two types of radiation. Despite these differences, the two results showed similar trends and the viability of using simulation-based radiation images, particularly in the field of security screening. With further research, the simulation-based radiation images can replace ones from experiments and be used in the related fields. Conclusion: The Korea Atomic Energy Research Institute has developed air-cargo security inspection equipment using neutrons and X-rays. Using this equipment, radiation images and R-values for various materials were obtained. The equipment was reconstructed, and the R-values were obtained for 13 materials using the Geant4 simulation toolkit. The R-values calculated by experiment and simulation show similar trends. Therefore, we confirmed the feasibility of using the simulation-based radiation image.

Microbial Structure and Community of RBC Biofilm Removing Nitrate and Phosphorus from Domestic Wastewater

  • Using a rotating biological contactor modified with a sequencing bath reactor system (SBRBC) designed and operated to remove phosphate and nitrogen [58], the microbial community structure of the biofilm from the SBRBC system was characterized based on the extracellular polymeric substance (EPS) constituents, electron microscopy, and molecular techniques. Protein and carbohydrate were identified as the major EPS constituents at three different biofilm thicknesses, where the amount of EPS and bacterial cell number were highest in the initial thickness of 0-100${\mu}m$. However, the percent of carbohydrate in the total amount of EPS decreased by about 11.23%, whereas the percent of protein increased by about 11.15% as the biofilm grew. Thus, an abundant quantity of EPS and cell mass, as well as a specific quality of EPS were apparently needed to attach to the substratum in the first step of the biofilm growth. A FISH analysis revealed that the dominant phylogenetic group was $\beta$- and $\gamma$-Proteobacteria, where a significant subclass of Proteobacteria for removing phosphate and/or nitrate was found within a biofilm thickness of 0-250${\mu}m$. In addition, 16S rDNA clone libraries revealed that Klebsiella sp. and Citrobacter sp. were most dominant within the initial biofilm thickness of 0-250${\mu}m$, whereas sulfur-oxidizing bacteria, such as Beggiatoa sp. and Thiothrix sp., were detected in a biofilm thickness over 250${\mu}m$. The results of the bacterial community structure analysis using molecular techniques agreed with the results of the morphological structure based on scanning electron microscopy. Therefore, the overall results indicated that coliform bacteria participated in the nitrate and phosphorus removal when using the SBRBC system. Moreover, the structure of the biofilm was also found to be related to the EPS constituents, as well as the nitrogen and phosphate removal efficiency. Consequently, since this is the first identification of the bacterial community and structure of the biofilm from an RBC simultaneously removing nitrogen and phosphate from domestic wastewater, and it is hoped that the present results may provide a foundation for understanding nitrate and phosphate removal by an RBC system.

방사선 조사가 백서 악하선 줄무늬관세포에 미치는 영향에 관한 투과전자현미경적 연구 (AN ELECTRON MICROSCOPIC STUDY OF THE IRRADIATION EFFECTS ON THE STRIATED DUCT CELLS OF THE SUBMANDIBULAR GLAND IN RATS)

  • The purpose of this study was to investigate the effects of irradiation on the striated duct cells of the rat submandibular gland ductal tissues which control the characteristics of saliva. For this study, the experimental group was composed of 36 irradiated Sprague Dawley strain rats divided into 8 subgroups 1 hour, 2 hours, 3 hours, 6 hours, 12 hours, 24 hours, 48 hours, 72 hours after irradiation. 4 non-irradiated rats were used as the control group. The experimental animals were singly irradiated with a dose of 18Gy gamma ray to their head and neck region by the Co-6- teletherapy unit and sacrificed after each experimental duration. The specimens were examined with a light microscope with an H-E stain and with a trans- mission electron microscope. The results of this study were as follows. In the light micrograph, a severe atrophic change occurred in the striated duct cells at 2hours after irradiation and gradual recovery occurred from 6 hours after irradiation. 2. The nuclear chromosomes of the striated duct cells were changed granular at 2 hours after irradiation. Recovery was observed at 6 hours after irradiation. Nuclear bodies were also observed from 3 hours after irradiation. 3. The mitochondria of the striated duct cells had indistinct cristae at 2 hours after irradiation, and were degenerated or swollen at 3 hours after irradiation. They recovered, however, from 6 hours, with an increasing number at 48 hours and a regular arrangement was observed at 72 hours after irradiation. 4. The microvilli showed atrophic changes at 2 hours after irradiation and were almost lost at 3 hours after irradiation. They were observed again from 48 hours after irradiation. 5. The rough endoplasmic reticulum and golgi body were not apparent at 1 hour after irradiation and were dilated with degeneration 2 hours after, but intact rough endoplasmic reticulum were observed from 3 hours after irradiation and developed well at 24 hours after irradiation. By the result of this study, showing a mild change in the functional morphology of the salivary striated duct cells immediately following irradiation, it is considered that the many complications which occur after radiation therapy, will disappear in time with the histological and the functional recovery of the glandular tissues.

인산농도가 소의 법랑질에서 부식깊이와 브라켓 전단결합강도에 미치는 영향 (EFFECTS OF PHOSPHORIC ACID CONCENTRATION ON DEPTH OF ETCH AND SHEAR BOND STRENGTH OF ORTHODONTIC BRACKETS TO BOVINE ENAMEL)

  • 이 연구는 부식농도가 전단결합강도와 법랑질 표면의 부식깊이에 미치는 영향을 관찰하기 위하여 표면을 편평하게 연마한 소의 법랑질을 $0%,\;5%,\;10%,\;20%,\;30%,\;40%,\;50%,\;60%,\;70%,\;80%$$85\%$ 인산용액으로 60초간 부식시키고, 브라켓 접착의 전단결합강도와 법랑질 표면의 부식깊이와 조도를 측정하고, 주사전자현미경으로 부식면을 검경하였으며, 이상에서 얻은 자료를 통계처리하고 평가하여 다음과 같은 결론을 얻었다. 법랑질 표면의 부식깊이는 인산액의 농도가 $40\%$ 내지 $50\%$에 도달할 때까지 증가하다가 그 이후의 농도증가와 함께 점진적으로 감소하였다. 법랑질 표면의 부식깊이와 부식면의 조도 사이에 상관성이 없었으며, 부식면의 조도와 전단결합강도 사이에 중등도의 상관성($\gamma$=0.63)을 보였다. 인산액의 농도가 $5\%$에서 $40\%$에 도달할때까지 균질의 편평한 부식면을 나타내는 소견을 보였으나 $50\%$$60\%$에서 불균질의 과도한 그리고 편평하지 못한 부식면소견을 보였으며, $10\%$에서 $60\%$까지의 인산액에 의한 법랑질 부식 후 레진 접착제의 전단결합강도 사이에는 유의차가 얼었으며, $5\%$$70\%$의 인산액에 의한 것은 전자보다 작았으나 구강내에서 교정력에 견딜수 있는 전단결합강도를 나타낼 것으로 추정된다.

열발광, 전자스핀공명 및 DNA Comet 분석에 의한 대두의 방사선 조사 여부 검지 특성 (Detection Characteristics of TL, ESR and DNA Comet for Irradiated Soybeans)

  • 방사선 조사된 대두의 검지방법을 연구하기 위하여 국산 및 중국산 대두에 대하여 $0.5{\sim}4.0\;kGy$의 감마선을 조사하고 열발광(TL), 전자스핀공명(ESR) 및 DNA comet 특성을 비교 검토하였다. TL 측정에서 비조사구는 $280^{\circ}C$ 부근에서 매우 낮은 glow curve를 나타내었고, 감마선 조사구는 $200^{\circ}C$ 부근에서 조사선량에 의존적인 glow curve를 나타내었다. 재조사(re-irradiation) 방법에 의한 TL ratio$(TL_1/TL_2)$의 비교는 TL 측정 결과의 정확도를 높여 주었다. 원산지별 TL 특성 비교에서 국산을 중국산에 비해 높은 TL intensity를 나타내었다(p<0.01). 대두 껍질을 사용한 ESR 측정에서는 조사시료에서 cellulose radical 유래의 특이한 signal(g = 2.02374, 1.98715)을 보여주었고 중국산 시료의 ESR signal이 더 높게 나타났다. 비조사 대두의 DNA comet은 tail이 없거나 아주 짧은 tail을 가진 전형적인 intact cell을 나타내었다. 그러나 0.5 kGy 이상의 감마선 조사 시료에서는 조사선량에 의존적으로 comet의 tail length 증가와 더불어 comet의 크기 및 농도의 변화가 일어났다. 이상의 결과에서 TL, ESR 및 DNA comet의 분석은 대두의 방사선 조사 여부 확인을 가능하게 하였으며, 원산지별 검지특성의 차이는 거의 없었다.

건포도와 건바나나의 감마선 조사와 저장기간에 따른 열발광 및 전자스핀공명 특성과 관능적 품질 (Characteristics of Thermoluminescence and Electron Spin Resonance and Organoleptic Quality of Irradiated Raisin and Dried Banana During Storage)

  • 방사선 조사(1~4 kGy)와 저장 기간(4$^{\circ}C$, 6개월)에 따른 건조과일의 TL 및 ESR검지 특성과 관능적 품질을 평가하였다. 건포도에서 분리된 mineral의 TL측정에서 비조사구는 200~30$0^{\circ}C$ 부근에서 매우 낮은 intensity의 glow curve(TLI)를 나타내었으나, 1 kGy 이상의 조사구에서는 특유의 peak가 18$0^{\circ}C$ 부근에서 나타나면서 선량 의존적으로 intensity가 증가되었다($R^2$=0.9684). 이로써 방사선 조사구와 비조사구의 판별이 6개월 동안 가능하였다. 또한 재조사(1 kG)에 의해 측정된 TL$_2$signal을 활용하여 TL ratio(TL$_1$/TL$_2$)를 계산하였을 때 검지결과의 신뢰도를 높일 수 있었다. 건 바나나의 ESR 측정에서 1 kGy 이상의 조사 시료는 crystalline sugar radical 유래의 전형적인 multicomponent line을 보여 주어 조사 여부의 판별이 가능하였고 signal의 강도는 선량 의존적($R^2$= 0.8977)으로 6개월 동안 비교적 안정하였다. 방사선 조사된 건조과일은 6개월까지 상품성이 잘 유지되었으므로 TL 및 ESR 분석은 건포도와 건 바나나의 검지 관리에 적합한 방법임이 확인되었다.

야생 팽나무버섯의 생리활성 분석 (Analysis of physiological activities of wild Flammulina velutipes)

  • 충북지역에서 수집한 야생 팽나무버섯의 식품소재로의 가능성을 조사하기 위해 동결 건조한 후 일반성분 및 전자공여능, 총폴리페놀 함량, ACE 저해도와 ${\alpha}$-glucosidase 저해도등 생리활성을 분석하였다. 대조군으로 갈뫼팽이버섯과 4종의 수집 야생 팽나무버섯의 일반성분은 건물중으로 분석하였으며, 조단백질은 17.1~21.3 g/100g, 조지방은 4.1~4.9g/100g, 탄수화물은 65.9~70.5g/100g, 조섬유는 7.9~18.1g/100g, 조회분은 5.7~8.4g/100g 범위를 나타내었다. 전자공여능은 63.35~69.31%로 시료간 큰 차이가 없는 것으로 나타났으며, 총폴리페놀 함량은 9.83에서 11.14 mg/g로 대조구와 유사하였다. ACE 저해도는 대조구인 갈뫼 팽이버섯과 수집종인 CBMFV-02가 각각72.91%, 71.44%로 높은 값을 나타내었으며, ${\alpha}$-glucosidase 저해도는 CBMFV-01이 46.55%로 가장 높은 값을 나타내었다.

법랑질 표면의 탈회 및 재광화 후 EPMA (electron probe micro-analysis)를 이용한 칼슘, 인, 불소 변화의 정량적 분석 (A QUANTITATIVE STUDY OF THE CHANCE OF CALCIUM, PHOSPHATE, FLUORIDE USING EPMA AFTER IN VITRO DEMINERALIZATION AND REMINERALIZATION OF HUMAN TOOTH ENAMEL)

  • The aim of this in vitro study was to analyze the composition of human tooth enamel in terms of three components, Ca, P, and F after demineralization and remineralization in acid buffer solution. A total of 8 human premolars without any defects and cracks were selected and buccal and lingual sides of the teeth were cleaned with an ultrasonic device and pumice without fluoride 5$\times$5mm windows were opened, and other areas were completely covered with 3-coats of nail varnish to prevent from being in contact with demineralized and remineralized solutions. After demineralization process, each tooth was sectioned into two slices, highly polished one of them with$\gamma$-alumina, and then analyzed the composition of the demineralized tooth with EPMA(electron probe micro-analyzer). The other slices were put into the remineralized solution for 10 days, polished, and analyzed in the same manner. These data were statistically analyzed with one sample t-test(p<0.05). The results were as follows. 1. Normal tooth enamel consists of 49.76% Ca, 39.80% P, and 0.28% F. 2. After demineralization, percentage of Ca and P ratio were decreased by about 5.57 and 5.07% respectively. Percentage of F ratio was also decreased by about 0.01%, which was not statistically significant. 3. After remineralization, percentage of Ca, P increased about by 4.47 and 4.35% respectively Percentage of F decreased by about 0.01%, which was not statistically significant. In conclusion, remineralized solution used in our study has the potential to induce the uptake the Ca and P into the pore sites of the demineralized enamel. But, in the oral cavity. there were rapid temperature change, organic matrix that inhibits the movement of the ions, and limitation of continuous contact with this remineralized solution. Therefore, further in vivo study is necessary.

복합적층판(復合積層板)의 기계적(機械的) 체결부(締結部)에 관한 파손연구(破損硏究) (Damage Study on the Mechanical Fastening in Laminated Composites)

  • 기계적 체결방법은 분해 및 조립이 용이하고 본드접착에 의한 결합보다 신뢰도가 높은 장점이 있으나 원공으로 인한 높은 응력집중을 초래하고 국부적으로 집중된 응력 재분포의 메카니즘이 등방성 재료와는 달리 매우 복잡하고 실적 데이타 및 실험자료가 거의 없어, 복합재료구조물의 결합방법으로 많은 연구 검토를 필요로 하고 있다. 본 연구에서 사용한 시편은 $[0^{\circ}/45^{\circ}90^{\circ}/-45^{\circ}]_s$ 적층판으로 W(시편의 폭)/d(핀의 직경) 및 L(edge의 거리)/d를 변화시키면서 실험을 수행하여 기하학적 형상 및 적층강도 변화에 따른 파손강도 및 파손양상을 측정하여 그 결과를 이론해석 값과 비교하였다. 실험결과에 따라 최적기법으로 만든 새로운 변수 $\alpha,\;\beta,\;\gamma$를 재료상수로 취급하여 W/d및 L/d와 체결강도와의 관계식을 이용하면, 동일한 재료로 제작한 적층판의 체결강도를 8%이내의 오차로 예측 가능하다. 초음파 탐상법(Ultrasonic C-scanning)으로는 손상영역 탐색 및 손상영역 확대과정을 파악하고, X-ray로는 몇 단계 하중상태에서 손상부를 촬영하였고, SEM(Scanning electron microscopes)을 이용 원공주위 파손부의 균열의 진전상태를 미시적으로 관찰하였다. 파손강도 및 파손양상에 대한 실험결과와 FEM이론해석 모델에 예측한 값과의 차이는 $L/D=2{\sim}3$인 경우를 제외하고는 대부분 일치하였다.

PSL, ESR 및 TL 측정에 의한 복합조미식품의 방사선 조사여부 검지 (The Detection of Irradiated Composite Seasoning Foods by Analyzing Photostimulated Luminescence (PSL), Electron Spin Resonance (ESR) and Thermoluminescence (TL))

  • 미생물적 품질개선이 요구되는 복합조미식품 2종(beef broth 분말, pork bone extract 분말)을 대상으로 $0\~10kGy$의 감마선을 조사하고 광자극 발광(PSL), 전자스핀공명(ESR) 및 열발광(TL) 측정에 의한 조사여부검지 가능성을 분석하였다. 조사선량에 따른 PSL photon count를 측정해 본 결과, 비조사 beef broth와 pork bone 분말은 각각 503.0과 429.5의 값을 보여 비조사 시료에서 측정되는 700이하의 값이 확인되었다. 그러나 $1\~10kGy$ 조사시료에서는 모두 5,000 이상의 photon count가 측정되어 이들 복합조미식품은 PSL 분석에 의해 방사선 조사여부를 신속하게 screening할 수 있었다. 또한 조사시료의 ESR 분석에서는 beef broth와 Pork bone 분말에서 signal의 g-value가 2.0065와 2.0067로 각각 측정되었고, 조사선량에 따라 ESR signal intensity가 증가하는 경향으로 beef broth 분말 $R^2=0.9226$, pork bone extract 분말 $R^2=0.9280$의 상관을 보였으나 방사선 유래의 signal은 확인되지 않았다. TL분석 결과 beef broth분말의 경우에서만 $170^{\circ}C$ 부근에서 방사선 조사 유래의 glow curve가 나타났으며, 재조사에 의한 TL ratio $(TL_1/TL_2)$는 비조사군 0.05이하, 1kGy 이상 조사군 1.0 이상으로 방사선 조사 여부의 검지가 가능하였다.
