대마 유전자원의 지방산 조성과 감마리놀렌산 함량변이 (Variation on Fatty Acid Profile Including ${\gamma}-Linolenic$ acid among Hemp (Cannabis sativa L.) Accessions)

  • 종실의 유분함량이 많아 유료작물로 전용이 가능한 대표적인 섬유작물인 대마 종실의 지방산조성의 분석을 통하여 감마리놀렌산 함유여부 및 함량과와 품종간 차이를 조사하였다. 45 수집종의 대마 종실에는 감마리놀렌산의 경우 평균 2.1% 함유되어 있었으며 이중 IH4 등 7 품종이 $3.1%{\sim}3.8%$로 높으면서 경장이 $31{\sim}122\;cm$로 작아 종실용 품종 육성재료로 유망시 되었다. 도입된 대마의 지방산 조성 변이에서 감마리놀렌산 비율 범위 및 변이계수는 각각 $0.8%{\sim}3.8%$, 42.9%로 품종간 변이가 가장 컸으나 리놀레산은 각각 $51.4%{\sim}61.8%$, 4.2%로 가장 적었다 개화일수와 경장간에는 고도의 정의 상관이었고, 개화일수와 알파리놀렌산 비율 및 경장과 감마리놀렌산 비율간에는 부의 상관이었으며 환각성분인 THC와 다른 형질들 간에는 유의적인 상관이 없었다. 팔미트산과 감마리놀렌산 비율간에는 부의상관, 올렌산과 알파리놀렌산 그리고 리놀레산과 알파리놀렌산은 고도의 부의 상관을 나타내었다. 감마리놀렌산 함량이 높은 도입종들은 대체로 수집지가 중앙러시아 등 유럽이었고 개화일수가 $29{\sim}44$일로 짧았다.

  • 우리나라에서 생산되고 있는 60여종의 약용식물의 지방산 조성을 조사하여 식물체의 부위 와 종류에 따른 지방산 조성의 특성을 관찰한 결과 아래와 같은 결과얻었다. 1. 달맞이꽃종자, 컴프리잎, 지치뿌리에서 각각 총지방산함량의 9.1, 3,5, 7,2%의 GLA가 검출되었으며 달맞이 꽃종자가 컴프리잎이나 지치뿌리보다 GLA의 절대량이 많았다. 2. 들깨종자, 참깨종자, 호박종자등의 지방산이 양(量)과 불포화도면에서 좋은 자원임이 확인되었다. 3. 식물체의 잎, 줄기, 뿌리에서의 지방산조성은 종자의 경우에 비하여 18 : 3의 분포 비율이 현저히 높게 나타났으며 18 : 0의 비율은 비교적 같게 나타났다.

Fermentation Process Development of Recombinant Hansenula polymorpha for Gamma-Linolenic Acid Production

  • Development of the strain and the fermentation process of Hansenula polymorpha was implemented for the production of ${\gamma}$-linolenic acid ($GLA,\;C18:3{\Delta}^{6,9,12}$), an n-6 polyunsaturated fatty acid (PUFA) that has been reported to possess a number of health benefits. The mutated ${\Delta}^6$-desaturase (S213A) gene of Mucor rouxii was expressed in H. polymorpha under the control of the methanol oxidase (MOX) promoter. Without the utilization of methanol, a high-cell-density culture of the yeast recombinant carrying the ${\Delta}^6$-desaturase gene was then achieved by fed-batch fermentation under glycerol-limited conditions. As a result, high levels of the ${\Delta}^6$-desaturated products, octadecadienoic acid ($C18:2{\Delta}^{6,9}$), GLA, and stearidonic acid ($C18:4{\Delta}^{6,9,12,15}$), were accumulated under the derepression conditions. The GLA production was also optimized by adjusting the specific growth rate. The results show that the specific growth rate affected both the lipid content and the fatty acid composition of the GLA-producing recombinant. Among the various specific growth rates tested, the highest GLA concentration of 697 mg/l was obtained in the culture with a specific growth rate of 0.08 /h. Interestingly, the fatty acid profile of the yeast recombinant bearing the Mucor ${\Delta}^6$-desaturase gene was similar to that of blackcurrant oil, with both containing similar proportions of n-3 and n-6 essential fatty acids.

감마-리놀렌산 생산 곰팡이의 탐색 (Screening of ${\gamma}-Linolenic$ Acid-Producing Fungi)

  • 감마-리놀렌산 생산을 목적으로 균주 보관소에서 구입한 Mucorales목 15균주와 실험실에서 분리한 사상균 10균주의 균체성장, 지질생성량, 감마-리놀렌산 함량 및 펠렛 크기등을 비교 조사하였다. 조사한 균주중 토양에서 분리한 FB-354균주가 진탕배양시 균체성장이 양호하고, 지질함량이 건조균체중 29.9%, 감마-리놀렌산의 함량이 총 지방산중 16.8%로 높고 직경 1mm미만의 적은 펠렛을 형성하는 등 감마-리놀렌산의 생산균주로서의 양호한 특성을 나타내었다. 이 균주는 형태학적 특성을 조사한 결과 Mucor sp.로 동정되었다. 이 균주가 생산하는 지질은 81.1%가 중성지질, 7.2%가 당지질 그리고 11.8%가 인지질로 밝혀졌으며, 감마-리놀렌산은 당지질에는 거의 없고 중성지질에 전체량의 대부분이 존재하였으며 인지질에는 인지질 획분 지방산의 21.4%를 차지하여 상대적으로 다량 존재한 것으로 나타났다.

Gas Chromatograph-Mass Spectrometer를 이용한 γ-Linolenic Acid(GLA) 생성 균주탐색 및 확인 (Screening of GLA (γ-Linolenic Acid) from Fungi by Gas Chromatography and Mass Spectroscopy)

  • 가스크로마토그라프와 질량분석기 (GC-MS)를 이용하여 감마리놀렌산(GLA) 생산 균주를 선발하였다. KACC에 보존 중인 Phytophthora 속, Pythium 속 및 Mucor 속 및 Rhizopus 속 등 4속에 속하는 44개 균주를 GC-FID를 통하여 GLA를 분석하였다. 4속 39종의 곰팡이 균주에서 GLA를 확인하였으며, M. hiemalis f. sp. hiemalis KACC 40264 균주에서는 GLA의 조성비가 전체 지방산의 24.8%로서 가장 높은 비율을 나타냈다. 속간에 전체적인 GLA 함량은 Mucor 속(14.2%)과 Rhizopus 속(14.3%)의 Zygomycota가 상대적으로 높은 함량을, 그리고 Phytophthora 속(3.3%)과 Pythium 속(3.0%)의 Oomycota는 낮은 함량을 나타냄으로서 뚜렷한 양분현상을 나타냈다. 지방산총량에서도 Mucor 속(15.4 mg/100 ml)과 Rhizopus 속(7.1 mg/100 ml)가 Phytophthora 속(2.6 mg/100 ml)와 Pythium 속(4.5 mg/100 ml)보다 높게 나타나서 전체적으로 지방산을 생산하는 유용한 균류자원으로 평가되었다. Mucor 속 내에서도 계통별로 큰 차이를 나타냈는데 18.2%의 GLA를 생성함으로 지방산총량이 50.8 mg/100 ml까지 이른 Mucor blumbeus KACC 40935가 실제적으로 가장 활력이 좋은 균주로 평가되었다.

Effects of Dietary Lysine and Gamma-linolenic Acid Levels on Growth Performance and Meat Quality in Finishing Pigs

  • This study investigated the effect of dietary lysine and gamma-linolenic acid(GLA) levels on growth performance, carcass traits, and meat quality in finishing pigs. Pigs were provided with feed containing two different levels of lysine(0.45% and 0.75%) with three different levels of gamma-linolenic acid(0.0, 0.3, and 0.6%). Average daily gain(ADG) was significantly lower (p<0.01) in pigs provided with the lower level of lysine. In contrast, feed/gain(p<0.01), diet cost/gain(p<0.05), and intramuscular fat(p<0.01) were all significantly higher in pigs fed the lower level of lysine. Similarly, meat color scores(CIE L*, a*, and b*) and cooking loss were significantly higher(p<0.01) in pigs fed the lower level of lysine, whereas shear force(kg/2.5 inch2)was not affected by dietary lysine. The addition of GLA had no significant effect on any of the parameters measured. The results indicate that providing pigs with 0.45% lysine in their diet may help to increase intramuscular fat content, allowing the industry to produce pork products that meet consumer needs in Korea.

Metabolic Flux Distribution for $\gamma$-Linolenic Acid Synthetic Pathways in Spirulina platensis

  • Spirulina produces $\gamma$-linolenic acid (GLA), an important pharmaceutical substance, in a relatively low level compared with fungi and plants, prompting more research to improve its GLA yield. In this study, metabolic flux analysis was applied to determine the cellular metabolic flux distributions in the GLA synthetic pathways of two Spiru/ina strains, wild type BP and a high­GLA producing mutant Z19/2. Simplified pathways involving the GLA synthesis of S. platensis formulated comprise of photosynthesis, gluconeogenesis, the pentose phosphate pathway, the anaplerotic pathway, the tricarboxylic cycle, the GLA synthesis pathway, and the biomass syn­thesis pathway. A stoichiometric model reflecting these pathways contains 17 intermediates and 22 reactions. Three fluxes - the bicarbonate (C-source) uptake rate, the specific growth rate, and the GLA synthesis rate - were measured and the remaining fluxes were calculated using lin­ear optimization. The calculation showed that the flux through the reaction converting acetyl­CoA into malonyl-CoA in the mutant strain was nearly three times higher than that in the wild­type strain. This finding implies that this reaction is rate controlling. This suggestion was sup­ported by experiments, in which the stimulating factors for this reaction $(NADPH\;and\;MgCl_{2})$ were added into the culture medium, resulting in an increased GLA-synthesis rate in the wild type strain.

Sexually Matured Female Mice showed Higher γ-Linolenic Acid (GLA) in the Muscle Tissue

  • The sexual maturation occurred by the changes of steroid hormones was known to sex-dependently and/or age-dependently regulate the lipid metabolism in various animal species. Our current study demonstrates that lipid and its functional fatty acids can be changed depending on the status of sexual maturation. Of the functional fatty acids, ${\gamma}$-linolenic acid (GLA; 18:3n-6) is an important factor for maintaining human health. The purpose of our study was to investigate the level of GLA in mice with different stages of sexual maturation. To this end, the longissimus muscle (LM) of immature (3-week-old) and mature (7-week-old) female mice was analysed for the fatty acid composition by gas chromatography. Furthermore, both gene and protein level of ${\Delta}6$ desaturase (FADS2) which is involved in GLA metabolism by real time PCR and Western blotting, respectively. Mature females showed greater (P<0.05) serum $17{\beta}$-estradiol (E2) level and LM GLA contents than immature group. The mRNA and protein levels of FADS2, which converts precursor linoleic acid into GLA, were higher (P<0.05) in mature female mice than in immature mice. In conclusion, these results show that sexual maturation of female mice induces GLA and FADS2 contents in LM.

유통중인 건강기능식품의 감마-리놀렌산 및 오메가-3 지방산 함량 모니터링 (Monitoring of compositions of gamma-linolenic and omega-3 fatty acids in some functional foods consumed in market)

  • The compositions of health functional food products (HFFP; 18 products) containing gamma linolenic acid (GLA; $C_{18:3}$, n-6) and omega-3 fatty acids (EPA and DHA) were investigated. The contents of index components (especially, GLA and omega-3) in HFFPs were monitored by GC-FID analysis. Among the GLA products (sample No. 1~8), the content of GLA in most samples (except sample No.6) ranged from 8.04 to 9.98 g/100 g. These results were suitable for the reference standard (more 7.0 g/100 g) of HFF. In the omega-3 products (sample No. 10 and 15) derived from harp seal oil (HSO), the total contents of EPA and DHA were 14.21-15.98 g/100 g, respectively. These values were suitable for the reference standard (more 12.0 g/100 g) of HFF. Besides, among the omega-3 products (sample No.9, 11~14, 16~18) derived from fish oil, the total content of EPA and DHA ranged from 24.11 to 31.20 g/100 g. These results were suitable for the reference standard (more 18.0 g/100 g) of HFF. In the result of TLC analysis, the HFFPs of 18 were mainly composed of triacylglycerols (TAGs). The content of trans fatty acid in 18 HFFPs was detected in less than 0.30 g/100 g. For the detection of trans fatty acid, $^1H$-NMR (600 MHz) can be used because chemical shift of trans fatty acid was observed at 5.3 ppm in this study.

γ-LA-Supplementation to IVC for IVP Bovine Embryos

  • The present study aimed to examine the effects of ${\gamma}$-linolenic acid (GLA) supplementation to in vitro culture (IVC) medium on in vitro developmental competence, freezability and morphology of in vitro matured and fertilized bovine embryos. In vitro produced (IVP) bovine zygotes were cultured in IVC medium supplemented with 0 (negative control), 15, 31, 62, 125, 250, 500 or 1,000 ppm GLA, 250 ppm linoleic acid albumin (LAA) and without any supplement as a control. Day 6 blastocysts derived from culture control were cultured in IVC medium containing either 62, 250 GLA or 250 LAA for 24 h, and at Day 7 were subjected to freezing or morphological examination by electron microscope. GLA 15 showed a tendency to have a higher cleavage rate at Day 2 (70.3%) than other groups. The hatching rate at Day 9 in LAA (38.2%) was significantly higher than the control and all treatment groups (p<0.05), while the blastocyst rate in LAA (32.4%) did not differ from those of 15 (30.5%), 31 (27.1%), and 62 GLA (33.1%) or the control (35.1%). GLA in concentrations of 125, 250, 500, and 1,000 ppm had significantly detrimental effect on the blastocyst rate compared to 15, 31 and 62 ppm GLA, LAA, and control groups (p<0.05). In contrast, the highest post-thaw survival rate (100%) was observed in the control group (p<0.01). Large lipid droplets were observed in the cytoplasm of trophoblastic cells, even in the control, but were abundant in GLA groups. Taking the results of the study into consideration, the addition of GLA to the culture medium for IVP bovine embryos at the dose of 15 ppm increased the developmental competence of zygotes and enhanced the cleavage rate up to Day 2. However, blastulation rate and post-thaw survival were not increased when GLA was added to the culture media.
