Preparation and Analysis of Yeast Cell Wall Mannoproteins, Immune Enhancing Materials, from Cell Wall Mutant Saccharomyces cerevisiae

  • Yeast cell wall matrix particles are composed entirely of mannoprotein and ${\beta}-glucan$. The mannoproteins of yeast cell wall can systemically enhance the immune system. We previously purified and analyzed alkali-soluble ${\beta}-glucans$ [${\beta}$-(1,3)- and ${\beta}$-(1,6)-glucans] [10]. In the present study, a wild-type strain was first mutagenized with ultraviolet light, and the cell wall mutants were then selected by treatment with 1.0 mg/ml laminarinase (endo-${\beta}$-(1,3)-D-glucanase). Mannoproteins of Saccharomyces cerevisiae were released by laminarinase, purified by concanavalin-A affinity and ion-exchange chromatography. The results indicated that the mutants yielded 3-fold more mannoprotein than the wild-type. The mannoprotein mass of mutant K48L3 was 2.25 mg/100 mg of yeast cell dry mass. Carbohydrate analysis revealed that they contained mannose, glucose, and N-acetylglucosamine. Saccharomyces cerevisiae cell wall components, mannoproteins, are known to interact with macrophages through receptors, thereby inducing release of tumor necrosis factor alpha ($TNF-{\alpha}$) and nitric oxide. Mannoprotein tractions in the present study had a higher macrophage activity of secretion of $TNF-{\alpha}$ and nitric oxide and direct phagocytosis than positive control ($1{\mu}g$ of lipopolysaccharide). In particular, F1 and F3 fractions in mannoproteins of K48L3 enhanced and upregulated the activity of nitric oxide secretion and macrophage phagocytosis by approximately two- and four-fold, respectively.

식용달팽이 β-Galactosidase의 정제와 생화학적 특성 (Purification and Characterization of the β-Galactosidase from Edible Snail)

  • 달팽이 내장을 마쇄, 추출, 염석 및 투석 후 얻은 조효소액을 두 번의 이온교환 크로마토그래피 및 두 번의 겔여과 크로마토그래피를 거쳐 최종적으로 얻은 $\beta$-galactosidase의 비활성도는 18.8 units/mg protein이었고, 정제도는 31.3%배, 수율은 20.8%이었다. $\beta$-Gallactosidase의 분자량을 측정하기 위해 겔 여과와 전기영동을 실시한 결과, native molecular weight가 약 144,000으로, SDS-PAGE 결과, 약 72,000으로 나타나 이 효소는 동일한 polypeptide로 구성된 dime로 추측되었으며, 등전점은 pI 4.1이었다. $\beta$-Galactosidase의 최적 pH와 온도는 각각 pH 3.0과 6$0^{\circ}C$로 측정되었으며, pH 2.0~8.0에서 80%이상의 효소 활성을 나타내었고, 온도 30~5$0^{\circ}C$에서 안정되었다. 모든 금속이온과 fructose, glucose, galactose, maltose 및 xylose는 $\beta$-galactosidase의 저해제로 작용하였다.

쌀귀리 가루의 입도별 이화학적 특성 및 항산화 활성 (Effect of particle size of naked oat flours on physicochemical and antioxidant property)

  • 본 연구에서는 영양학적 가치가 높은 쌀귀리의 식품산업 이용률을 높이고자 쌀귀리를 입도별로 제조하여 이들의 이화학적 특성 및 항산화 활성을 비교하였다. 쌀귀리의 조단백질, 조지방 및 조회분 함량이 각각 15.7%, 10.0% 및 1.8%이었으며, 입도가 작아질수록 이들의 함량은 감소하였다. 주요 유리아미노산은 비필수아미노산 중에서는 cystine(58.3 mg%), ${\beta}$-aminoisobutyric acid(53.8 mg%) 등, 필수아미노산 중에서는 phenylalanine(8.6 mg%)과 histidine (12.2 mg%)이었으며, 입도가 작아질수록 함량은 감소하였다. 총 전분, 아밀로스, 손상전분, 총 식이섬유, ${\beta}$-glucan 및 총 유리당 함량은 각각 56.4%, 21.4%, 11.7%, 11.0%, 4.7% 및 2,555.3 mg%이었으며, 입도가 작아질수록 총 전분, 아밀로스 및 손상전분의 함량은 감소한 반면에 총 식이섬유와 ${\beta}$-glucan의 함량은 증가하였다. 색도는 L 값이 75.3, a 값이 0.3 및 b 값이 13.1이었으며, 입도가 작아질수록 L 값은 증가하였으나 a와 b 값은 감소하였다. 호화특성은 최고점도가 263.8 RVU, 최저점도가 155.6 RVU 및 최종점도가 399.3 RVU이었으며, 입도가 작아질수록 최고점도와 최종점도는 증가하였다. TPC, TFC 및 항산화 활성(DPPH radical assay, ABTS radical cation assay 및 reducing power)은 각각 $237.8{\mu}g/g$, $90.9{\mu}g/g$, 57.3%, 53.4% 및 0.23이었으며, 입도가 작아질수록 항산화 성분 및 항산화 활성은 감소하였다. 한편 TPC와 TFC에 관한 각 항산화 활성의 결정계수(correlation determination coefficient, $R^2$) 값이 각각 0.87 이상과 0.81 이상으로 나타나 항산화 성분과 항산화 활성은 상관성을 보였다.

금은화가 LPS로 유발된 급성 폐 손상에 미치는 영향 (Effects of Lonicerae Flos Extracts on LPS-induced Acute Lung Injury)

  • Objective : The object of this study was to observe the effects of Lonicerae Flos (LF) aqueous extracts on lipopolysaccharide (LPS)-induced rat acute lung injury. Method : Five different dosages of LF extracts were orally administered once a day for 28 days before LPS treatments, and then all rats were sacrificed after 5 hour-treatment of LPS. Eight groups of 16 rats each were used in the present study. The following parameters caused by LPS treatment were observed ; body weights, lung weights, pulmonary transcapillary albumin transit, arterial gas parameters (pH, $PaO_2$ and $PaCO_2$) bronchoalveolar lavage fluid (BALF) protein lactate dehydrogenase (LDH), and proinflammatory cytokines tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin-$1{\beta}$ (IL-$1{\beta}$) contents, total cell numbers, neutrophil and alveolar macrophage ratios, lung malondialdehyde (MDA), myeloperoxidase (MPO), proinflammatory cytokines TNF-${\alpha}$ and IL-$1{\beta}$ contents. In addition, the histopathologic changes were observed in the lung in terms of luminal surface of alveolus, thickness of alveolar septum, number of polymorphonuclear neutrophils. Result : As results of LPS-injection, dramatical increases in lung weights, pulmonary transcapillary albumin transit increases, increases in $PaCO_2$, decreases in pH of arterial blood and $PaO_2$, increases of BALF protein, LDH, TNF-${\alpha}$ and IL-$1{\beta}$ contents, total cells, neutrophil and alveolar macrophage ratios, TNF-${\alpha}$ and IL-$1{\beta}$ contents increases were detected with decreases in LSA and increases of alveolar septum and PMNs numbers, respectively as compared with intact control. These are means that acute lung injuries (resembling acute respiratory distress syndrome) are induced by treatment of LPS mediated by inflammatory responses, oxidative stress and related lipid peroxidation in the present study. However, these LPS-induced acute lung injuries were inhibited by 28 days continuous pretreatment of 250 and 500mg/kg of LF extracts. Because of lower three dosages of LF treated groups, 31.25 and 62.5 and 125mg/kg did not showed any favorable effects as compared with LPS control, the effective dosages of LF in LPS-induced acute lung injuries in the present study, is considered as about 125mg/kg. The effects of 250mg/kg of LF extracts showed almost similar effects with ${\alpha}$-lipoic acid 60mg/kg in preventing LPS-induced acute lung injuries. Conclusion : It seems that LF play a role in protecting the acute respiratory distress syndrome caused by LPS.

트리폴리인산염을 이용한 산성광산배수 내 칼슘 및 철이온 제거 (Removal of Ca2+ and Fe3+ in Acid Mine Drainage by Tripolyphosphates)

  • 이 연구는 산성광산배수(AMD, Acid Mine Drainage)를 대상으로 트리폴리인산나트륨의 적용을 평가한다. 경북에 위치한 문경 석탄탄광으로부터 발생한 AMD와 트리폴리인산나트륨의 회분식 반응실험에서 얻어진 결과에 근거하면, AMD를 처리하기 위한 트리폴리인산나트륨의 최적주입량은 $4.7{\times}10^{-3}mole$이었다. $Ca^{2+}$의 경우 농도가 $16.4mg/{\ell}$에서 처리 후 $5.6mg/{\ell}$로 감소하여 제거율은 65.9%이고, $Fe^{3+}$의 경우 농도가 $3.7mg/{\ell}$에서 처리 후 $0.02mg/{\ell}$로 감소하여 제거율은 99.5%이다. 그러나 $SO{_4}^{2-}$의 경우 농도가 $526.8mg/{\ell}$에서 $566.5mg/{\ell}$ 범위로 증가나 감소경향이 나타나지 않았다. 트리폴리인산나트륨을 사용한 결과, AMD 내 $Na^+$의 농도는 $549.8mg/{\ell}{\sim}599.3mg/{\ell}$이고 정인산염은 $6.82mg/{\ell}{\sim}7.60mg/{\ell}$였다. 트리폴리인산나트륨과 AMD의 반응에서 발생한 침전물을 SEM, XRF, XRD로 분석한 결과 침전물의 형태는 인회석${\gg}{\beta}$-인산삼칼슘>산화철$(Fe(OH)_3)$인 것으로 판단된다. 결과적으로 트리폴리인산염의 사용은 AMD에서 $Fe^{3+}$, $Ca^{2+}$ 제거와 pH 완충에 있어서 우수한 것으로 나타났다.

Amino Acid, Amino Acid Metabolite, and GABA Content of Three Domestic Tomato Varieties

  • To determine the nutritional value of domestic tomatoes, the levels of amino acids, amino acid metabolites, and the bioactive compound ${\gamma}-aminobutyric-acid$ (GABA) were analyzed in three domestic tomato varieties (Rafito, Momotaro, and Medison). Eighteen free amino acids were found, and total free amino acid content was 3,810.21~4,594.56 mg/100 g (dry weight). L-glutamic acid (L-Glu) was the most abundant amino acid, ranging from 1,866.60 mg/100 g for Momotaro to 2,417.45 mg/100 g for Medison. The next most abundant amino acids were L-glutamine (L-Gln) and L-aspartic acid (L-Asp). The three tomato varieties had a good balance of all the essential amino acids except tryptophan. Total essential amino acid content was 274.26~472.71 mg/100 g (dry weight). The following amino acid metabolites were found: L-carnitine (L-Car), hydroxylysine (Hyl), o-phosphoethanolamine (o-Pea), phosphoserine (p-Ser), ${\beta}-alanine$ (${\beta}-Ala$), N-methyl-histidine (Me-His), ethanolamine (EtNH2),and L-citrulline(L-Cit). Large quantities of GABA were found in all three varieties: 666.95-868.48 mg/100g (dry weight). These results support the use of these tomato varieties as nutritious food materials.

The Effects of Pulsatilla Koreana NAKAI Pharmacopuncture Therapy on the Chunchu (ST25) for Dextran Sulfate Sodium-Induced Colitis in Mice

  • Background: This research was performed to investigate the effects of Pulsatilla Koreana NAKAI pharmacopuncture (PPA) therapy on intestinal disease in rats with dextran sulfate sodium (DSS)-induced colitis. Methods: The subjects were divided into five groups : A control group, saline group, pharmacopuncture group PPA1 ($0.2mg/1kg/40{\mu}{\ell}$), pharmacopuncture group PPA2 ($0.5mg/1kg/40{\mu}{\ell}$), and pharmacopuncture group PPA 3($1mg/1kg/40{\mu}{\ell}$). The experimental model of colitis was induced by infection of dextran sulfate sodium (DSS) for eighteen days. After colitis was induced, PPA therapy was practiced on the Chunchu (ST25) once every two days for a total six times. Thereafter Disease Activity Index (DAI), colon length, damage to the colonic mucosa, body weight, IL-6, IL-10, $IL-1{\beta}$, $IFN-{\gamma}$, $TNF-{\alpha}$, $TGF-{\beta}1$, IL-23 and IL-17 were measured. Results: The results were as follows. 1. DAI was significantly decreased in the PPA groups. 2. Colon length was significantly increased in the PPA groups. 3. Damage of colonic mucosa was observed less in the PPA groups. 4. Body weight was significantly increased in the saline group and the PPA groups. 5. The PPA2 group showed a significant decrease in the intensity of IL-6, $IL-1{\beta}$, $IFN-{\gamma}$ and $TNF-{\alpha}$ levels and the mean of IL-23. 6. The PPA3 group showed a significant increase in the intensity of IL-10 and $TGF-{\beta}1$ levels. 7. No significant differences were shown in the mean of IL-17. Conclusion: These results suggest that PPA therapy on Chunchu (ST25) can be used as an effective treatment for inflammatory bowel disease.

Assessment of Phytochemicals, Quality Attributes, and Antioxidant Activities in Commercial Tomato Cultivars

  • To assess South Korean commercial tomato cultivars, regular and cherry tomato cultivars were grown in the greenhouse and evaluated for color attributes, titratable acidity, pH, total soluble solids, carotenoids (lycopene and ${\beta}$-carotene), total phenols, flavonoids, vitamin C, and antioxidant activity. Significant differences (p < 0.05 using Duncan's multiple range test (DMRT)) were observed in the levels of most phytochemicals, quality parameters, and antioxidant activity among the twenty South Korean tomato cultivars tested. Lycopene and ${\beta}$-carotene contents varied significantly (p < 0.05 using DMRT), from $0.95mg{\cdot}100g^{-1}$ to $5.12mg{\cdot}100g^{-1}$ and $0.65mg{\cdot}100g^{-1}$ to $3.56mg{\cdot}100g^{-1}$ of fresh weight, respectively. ${\beta}$-carotene contents exhibited the highest genetic variation (59.2%), followed by naringenin (52.8%) and other phytochemicals. Most of the cherry tomato cultivars had statistically higher levels (p < 0.05 using DMRT) of carotenoids, phenols, flavonoids, vitamin C, and antioxidant activity compared to the regular tomato varieties, suggesting their higher nutritional value. Lycopene content was highest in the cultivars YoYo, Jicored, Titi-Chal, TY-Endorphin, and Rubyking. Cultivars Rubyking, TY-Endorphin, and Titi-Chal also showed relatively higher antioxidant activities in three assays: 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and ferric ion reducing antioxidant power (FRAP) assays. All the antioxidants, except luteolin, were positively correlated with antioxidant activities; the highest correlation was observed between total phenol and antioxidant activities, followed by the correlation between rutin and vitamin C. Cultivars identified to have superior nutritional status would be useful in tomato breeding programs to further improve quality and health benefits of tomatoes for the fresh and processed markets.

Xanthophyllomyces dendrorhous 변이군주에 의한 Carotenoids 생산 발효의 특성 연구 (Fermentation Kinetics for Production of Carotenoids by ${\beta}$-ionone Resistant Mutant of Xanthophyllomyces dendrorhous)

  • 본 연구에서는 적효모 X. dendrorhous KCTC 7704로부터 여러 ${\beta}$-ionone 내성 변이균주를 선별하였다. 야생균 KCTC 7704는 ${\beta}$-ionone 0.021 mM 농도에서 생육이 현저히 저하되었지만, NTG처리 후 ${\beta}$-ionone 0.1 mM 농도에서 선별된 변이균주는 ${\beta}$-ionone 0.15 mM에서도 70% 이상의 상대 생육율을 나타내는 매우 강한 ${\beta}$-ionone 내성을 갖고 있었다. 여러 ${\beta}$-ionone 농도에서 선별한 변이균주들을 $20^{\circ}C$에서 4일간 회분식 발효로 배양하여 그 특성을 조사하였다. 선별된 가장 우수한 변이균주는 야생균주보다 카로티노이드 생성능이 2.3배 향상(즉 $1.2{\mu}g$ of total carotenoids per mg of dry cells)되었으며 유기산과 같은 대사산물은 거의 생성하지 않았다. 여러 탄소원 들에 대한 비교 발효특성 연구 결과 과당이나 자당을 사용했을 때봐 비교하여 포도당 배지에서 최종 균체농도 및 총 카로티노이드 생성량이 많았다. 포도당이 제한되는 연속발효(dilution rate $0.04h^{-1}$) 실험을 통하여 pH의 영향을 조사한 결과 균체농도 및 총 카로티노이드 생성은 pH 4.0 조건하에서 최적인 것을 알 수 있었다.

Particle Bombardment에 의해 전처리 된 참나리(Lilium lancifolium Thunb.) 캘러스의 Agrobacterium tumefaciens을 통한 형질전환 (Agrobacterium Mediated Transformation from Callus Pretreated with Particle Bombardment in Lilium lancifolium Thunb.)

  • 형질전환 효율을 높이기 위하여, NPTII와 GUS유전자를 포함하고 있는 pIG121Hm을 미세 입자에 코팅하여 particle bombardment를 수행한 후 pIG121Hm가 도입된 Agrobacterium tumefaciens EHA101와 3일간 공동 배양하였다. 그 후 캘러스를 1mg/L 2,4-D, 0.1mg/L BAP, 100mg/L kanamycin, 그리고 200mg/L carbenicillin이 포함된 MS배지로 옮겨 4주간 성장시킨 후 kanamycin저항성 캘러스를 선발하여 GUS 발현을 관찰하였고, PCR 분석을 통하여 700 bp의 NPT II 유전자가 도입된 것을 확인하였다. Particle bombardment 후 Agrobacterium과 공동배양 한 처리구가 Agrobacterium과 공동배양만 한 처리구보다 GUS발현 분석에 의한 형질전환 효율이 3배정도 더 높았다 형질전환 된 재분화 식물체를 얻기위해 다시 선발된 kanamycin저항성 캘러스는 1mg/L NAA와 1mg/L BAP가 포함된 재분화 배지에 옮겨져 4주 후 형질전환 된 소인경을 형성하였다.
