Bowfin-과 Shark-neuropeptide $\gamma$의 구조 및 수축효과 (Structure and Contractile Activity of the Bowfin- and Shark-neuropeptide $\gamma$)

  • Tachykinin peptide의 구조와 생리활성간의 상관관계를 조사하기 위해 고상법으로 합성한 어류 유래의 neuropeptide $\gamma$(Mammalian-, Bowfin- 그리고 Shark-neuropeptide $\gamma$)를 사용하였다. 이들의 이차 구조를 알아보기 위해 circular dichroism (CD)을 이용하였다. CD 연구 결과에 따르면, mammalian-neuropeptide $\gamma$, bowfin-neuropeptide $\gamma$와 shark-neuropeptide $\gamma$는 완충액과 인공지질막 조건하에서 random구조를 나타내었다. 또한 이들 neuropeptide $\gamma$의 장관에 대한 수축 활성을 조사하기 위해 guinea-pig의 회장, rat의 십이지장과 carp intestine을 사용하였다. Carp intestine에 서 bowfin-neuropeptide $\gamma$>shark- neuropeptide $\gamma$>mammalian-neuropeptide $\gamma$순으로 활성 이 나타났다. 그러나, guinea-pig 회장과 rat 십이지장에 대해서 mammalian-NP$\gamma$의 활성이 어류 유래의 neuropeptide $\gamma$들보다 높게 나타났다. 이러한 결과들은 neuropeptide $\gamma$가 종-특이적인 활성을 나타냄을 제시한다.

난치성 결핵 환자의 단핵구에서 IFN-$\gamma$ 활성화 효과 및 IFN-$\gamma$ 수용체의 숫적 변화에 대한 연구 (The Priming Effect of IFN-$\gamma$ and Numbers of IFN-$\gamma$ Receptors in Patients with Chronic Refractory Tuberculosis)

  • 연구배경: 결핵에 대한 면역 반응은 T-림프구와 단핵 식세포를 주축으로 하는 세포성 면역 반응이 주률 이루는데 여기에 IFN-$\gamma$와 TNF-$\alpha$등의 cytokine 이 관여하고 있다. IFN-$\gamma$는 주로 T-림프구에서 분비되어 활성화된 단핵구에서의 TNF-$\alpha$생성을 증가시키는 기전을 통해 결핵균 증식을 억제하고 치유에 이르는 과정을 매개하는 것으로 알려져 있다. 효과적인 화학요법에도 불구하고 난치성 결핵으로 이행하는 기전에 IFN-$\gamma$가 관여하는지 규명하고자 본 연구를 시행하였다. 대상 및 방법: 정상 대조군과 최근 결핵으로 진단받아 치료를 시작한 환자, 난치성 결핵 환자에서 혈액을 채취하여 단핵구를 분리하고 아무런 자극을 하지 않은 경우, 12시간 LPS($l{\mu}g/ml$)만으로 자극을 한 경우, IFN-$\gamma$($2{\mu}g/ml$)로 2시간 전처치한 후 12 시간 LPS 자극한 경우로 니누어 TNF-$\alpha$의 농도를 측정, IFN-$\gamma$ 활성화 효과에 변화가 있는지를 살펴보았고 IFN-$\gamma$ 수용체 항제를 이용, IFN-$\gamma$ 활성화 효과의 변화가 IFN-$\gamma$ 수용체의 숫적 변화와 관련이 있는지를 확인하였다. 결과: IFN-$\gamma$ 활성화 효과를 알아보기 위해 LPS 만으로 지극한 경우와 IFN-$\gamma$ 전처치 후 LPS로 자극한 경우에 생성된 TNF-$\alpha$의 비를 구하여 보았을 때 정상 대조군 $13.5{\pm}7.6$, 최근 결핵환자로 진단받은 군 $10.8{\pm}6.4$, 난치성 결핵 환자군 $6.7{\pm}3.9$의 결과를 보였다. 난치성 결핵환자군의 경우 정상대조군에 비해 IFN-$\gamma$의 활성화 효과가 통계적으로 유의하게 감소되어 있음을 알 수 있었다 (p=0.002). IFN-$\gamma$ 수용체수의 감소는 대상 환자 중 난치성 결핵 환자 1명에서 관찰되었다. 결론: 난치성 결핵으로의 이행에 있어 단핵구에서의 IFN-$\gamma$ 활성화 효과의 감소가 중요한 역할을 할 것으로 생각되며 IFN-$\gamma$ 활성화 효과의 감소는 일부 환자의 경우, IFN-$\gamma$ 수용체의 감소와 관련되어 생기는 것으로 판단된다.

Repression of $PPAR{\gamma}$ Activity on Adipogenesis by $17{\beta}$-estradiol in Differentiated 3T3-L1 Cell

  • In our previous report, we showed that $PPAR{\gamma}$ does not influence adipogenesis in females with functioning ovaries, indicating that $PPAR{\gamma}$ activity on adipogenesis is associated with sex-related factors. Among the sex-related factors, estrogen has been recognized as a major factor in inhibiting adiposgenesis in females. Thus, we hypothensized that $17{\beta}$-estradiol (E) inhibits 3T3-L1 cell adipogenesis by preventing $PPAR{\gamma}$ activity. E decreased triglyceirde accumulation in differentiated 3T3-L1 cells compared with control group. E also decreased the expression of $PPAR{\gamma}$ mRNA as well as $PPAR{\gamma}$ dependent adipocyte-specific genes, such as adipocyte fatty acid binding protein and tumor necrosis factor $\alpha$. In addition, E not only decreased luciferase reporter activity by $PPAR{\gamma}$, but also transfection of estrogen receptor $\alpha$ ($ER{\alpha}$) or $ER{\beta}$ led to decreases in $PPAR{\gamma}$ reporter gene activation. Moreover, E-activated ERs significantly decreased the luciferase reporter gene activation induced by $PPAR{\gamma}$ transfection, suggesting that estrogen-activated ERs inhibit $PPAR{\gamma}$-dependent transactivation. Accordingly, our results demonstrate that E inhibits the action of $PPAR{\gamma}$ on adipogenesis through E activated ER, providing evidence that lack of estrogen may potentiate $PPAR{\gamma}$ action on adipogenesis.

Gamma 및 Generalized Gamma 분포 모형에 의한 적정 설계홍수량의 유도 (I) -Gamma 분포 모형을 중심으로- (Derivation of Optimal Design Flood by Gamma and Generalized Gamma Distribution Models(I) - On the Gamma Distribution Models -)

  • This study was conducted to derive optimal design floods by Gamma distribution models of the annual maximum series at eight watersheds along Geum , Yeong San and Seom Jin river Systems, Design floods obtained by different methods for evaluation of parameters and for plotting positions in the Gamma distribution models were compared by the relative mean errors and graphical fit along with 95% confidence interval plotted on Gamma probability paper. The results were analyzed and summarized as follows. 1.Adequacy for the analysis of flood flow data used in this study was confirmed by the tests of Independence, Homogeneity and detection of Outliers. 2.Basic statistics and parameters were calculated by Gamma distribution models using Methods of Moments and Maximum Likelihood. 3.It was found that design floods derived by the method of maximum likelihood and Hazen plotting position formular of two parameter Gamma distribution are much closer to those of the observed data in comparison with those obtained by other methods for parameters and for plotting positions from the viewpoint of relative mean errors. 4.Reliability of derived design floods by both maximum likelihood and method of moments with two parameter Gamma distribution was acknowledged within 95% confidence interval.

ON GRAPHS ASSOCIATED WITH MODULES OVER COMMUTATIVE RINGS

  • Let M be an R-module, where R is a commutative ring with identity 1 and let G(V,E) be a graph. In this paper, we study the graphs associated with modules over commutative rings. We associate three simple graphs $ann_f({\Gamma}(M_R))$, $ann_s({\Gamma}(M_R))$ and $ann_t({\Gamma}(M_R))$ to M called full annihilating, semi-annihilating and star-annihilating graph. When M is finite over R, we investigate metric dimensions in $ann_f({\Gamma}(M_R))$, $ann_s({\Gamma}(M_R))$ and $ann_t({\Gamma}(M_R))$. We show that M over R is finite if and only if the metric dimension of the graph $ann_f({\Gamma}(M_R))$ is finite. We further show that the graphs $ann_f({\Gamma}(M_R))$, $ann_s({\Gamma}(M_R))$ and $ann_t({\Gamma}(M_R))$ are empty if and only if M is a prime-multiplication-like R-module. We investigate the case when M is a free R-module, where R is an integral domain and show that the graphs $ann_f({\Gamma}(M_R))$, $ann_s({\Gamma}(M_R))$ and $ann_t({\Gamma}(M_R))$ are empty if and only if $$M{\sim_=}R$$. Finally, we characterize all the non-simple weakly virtually divisible modules M for which Ann(M) is a prime ideal and Soc(M) = 0.

Neuropeptide $\gamma$의 구조 및 생리활성 (Conformation and Biological Activity of the Neuropeptide $\gamma$)

  • 생리활성을 지닌 신경펩타이드의 구조와 활성간의 관계를 규명하기 위하여 고상법으로 합성한 세 종류의 neuropeptide $\gamma$(mammalian-, trout- 그리고 goldfish-neu-ropeptide $\gamma$)를 사용하여 연구하였다. Circular dichroism spectra에 의하면 mammalian-, trout-와 goldfish-neurope-ptide $\gamma$는 완충액 조건하에서 모두 random한 구조를 나타내었다. 중성 및 산성 지질 존재 하에서, mammalian과 trout-neuropeptide $\gamma$는 여전히 random한 구조를 취했다. 그러나, goldfish-neuropeptide $\gamma$는 중성 및 산성지질하에서 부분적으로 $\alpha-helix$ 구조를 나타내었다. 장관 수축활성 에 있어서는 carp 장관, guinea-pig 회장 그리고 rat 십이지장에 대하여 비교하였다. Carp에 대해서는 goldfish-neuropeptide $\gamma\;\simeq$ trout-neuropeptide $\gamma\;>$ mammalian- neuropeptide $\gamma$ 순으로 활성이 나타났다. 그러나, guinea-pig 회장과 rat 십이지장에 대해서 mammalian-neuropeptide $\gamma$는 어류 유래성 neuropeptide g들 보다 높은 수축활성을 나타내었다. 이러한 결과들은 neuropeptide $\gamma$들이 종-특이적인 활성을 나타낸다는 것을 제시한다.

  • The ${\gamma}/{\gamma}^{\prime}$ two-phases, commonly known as a eutectic structure, are observed in the ${\gamma}$ interdendritic region of a Ni-base superalloy. However, the growth behavior of the ${\gamma}/{\gamma}^{\prime}$ two-phases, whether it is of eutectic or peritectic nature, has not been decidedly established. Directional solidifications were, thus, performed with the planar interface at a low growth rate of $0.5{\mu}m/s$ in order to promote macro segregation. Directional solidification started with the ${\gamma}$ planar interface and the ${\gamma}^{\prime}$ phase nucleated on the ${\gamma}$ planar interface at the solidification fraction of 0.75. The ${\gamma}/{\gamma}^{\prime}$ two-phases showed the ${\gamma}^{\prime}$ rod structure as major phase and the ${\gamma}$ minor phase between ${\gamma}^{\prime}$ rods, and the volume fraction of the ${\gamma}$ phase changed continuously with an increasing solidification fraction. The two-phase ${\gamma}/{\gamma}^{\prime}$ is seen as the coupled peritectic.

AMPK γ 유전자의 표피세포극성 유지기능 규명 (AMPK γ is Required for Maintaining Epithelial Cell Structure and Polarity)

  • AMPK는 catalytic ${\alpha}$ subunit과 regulatory ${\beta}$${\gamma}$ subunit으로 구성된 인산화 효소로, 그 동안 생체 내 중요 대사 조절자로써 연구되어 왔으나, 최근 유전학 연구를 통해 지금까지 밝혀지지 아니한 새로운 생체기능을 가짐이 밝혀졌다. 본 연구에서 초파리 유전학 기법을 활용하여 AMPK ${\gamma}$ subunit 유전자가 결손된 모델 초파리를 제작 하여 연구한 결과, AMPK ${\gamma}$ 유전자 결손 시 초파리 embryo의 표피형성이 심각하게 저해됨을 발견하였고, 조직학적 실험을 통해 표피세포의 극성이 AMPK ${\gamma}$ 유전자 결손 초파리에서 손상되어 있음을 확인하였다. 또한 세포극성을 조절하는 중요 분자인 MRLC의 인산화 또한 AMPK ${\gamma}$ 유전자 결손 시 저해되었으며, AMPK ${\gamma}$ 유전자 재도입 시 MRLC인산화와 표피세포의 극성이 모두 회복됨이 확인되어, 초파리 표피세포의 극성유지에 AMPK ${\gamma}$ 유전자가 필수적 임을 확인하였다.

BOUNDED MOVEMENT OF GROUP ACTIONS

  • Suppose that G is a group of permutations of a set ${\Omega}$. For a finite subset ${\gamma}$of${\Omega}$, the movement of ${\gamma}$ under the action of G is defined as move(${\gamma}$):=$max\limits_{g{\epsilon}G}|{\Gamma}^{g}{\backslash}{\Gamma}|$, and ${\gamma}$ will be said to have restricted movement if move(${\gamma}$)<|${\gamma}$|. Moreover if, for an infinite subset ${\gamma}$of${\Omega}$, the sets|{\Gamma}^{g}{\backslash}{\Gamma}| are finite and bounded as g runs over all elements of G, then we may define move(${\gamma}$)in the same way as for finite subsets. If move(${\gamma}$)${\leq}$m for all ${\gamma}$${\subseteq}$${\Omega}$, then G is said to have bounded movement and the movement of G move(G) is defined as the maximum of move(${\gamma}$) over all subsets ${\gamma}$ of ${\Omega}$. Having bounded movement is a very strong restriction on a group, but it is natural to ask just which permutation groups have bounded movement m. If move(G)=m then clearly we may assume that G has no fixed points is${\Omega}$, and with this assumption it was shown in [4, Theorem 1]that the number t of G=orbits is at most 2m-1, each G-orbit has length at most 3m, and moreover|${\Omega}$|${\leq}$3m+t-1${\leq}$5m-2. Moreover it has recently been shown by P. S. Kim, J. R. Cho and C. E. Praeger in [1] that essentially the only examples with as many as 2m-1 orbits are elementary abelian 2-groups, and by A. Gardiner, A. Mann and C. E. Praeger in [2,3]that essentially the only transitive examples in a set of maximal size, namely 3m, are groups of exponent 3. (The only exceptions to these general statements occur for small values of m and are known explicitly.) Motivated by these results, we would decide what role if any is played by primes other that 2 and 3 for describing the structure of groups of bounded movement.

Influence of Interferon-${\gamma}$ Deficiency in Immune Tolerance Induced by Male Islet Transplantation

  • Background: Traditionally, interferon-${\gamma}$ (IFN-${\gamma}$) was regarded as a pro-inflammatory cytokine, however, recent reports suggested role of IFN-${\gamma}$ in immune tolerance. In our previous report, we could induce tolerance to male antigen (HY) just by male islet transplantation in wild type C57BL/6 mice without any immunological intervention. We tried to investigate the influence of IFN-${\gamma}$ deficiency on tolerance induction by male islet transplantation. Methods: To examine the immunogenicity of male tissue in the absence of IFN-${\gamma}$, we transplanted male IFN-${\gamma}$ knock-out (KO) skin to female IFN-${\gamma}$ KO mice. Next, we analyzed male IFN-${\gamma}$ KO islet to streptozotocin-induced diabetic female IFN-${\gamma}$ KO mice. And, we checked the functionality of grafted islet by graft removal and insulin staining. Results: As our previous results in wild type C57BL/6 mice, female IFN-${\gamma}$ KO mice rejected male IFN-${\gamma}$ KO skin within 29 days, and did not reject male IFN-${\gamma}$ KO islet. The maintenance of normal blood glucose level was dependent on the presence of grafted male islet. And the male islet recipient did not reject 2nd challenge of male islet graft also. Conclusion: Deficiency of IFN-${\gamma}$ does not have influence on the result of male skin graft and male islet transplantation. Conclusively, male islet transplantation induced T cell tolerance is not dependent on the presence of IFN-${\gamma}$.
