육두구의 생리활성에 관한 연구 (Functional Properties of Nutmeg)

  • This study investigated the antioxidative and antimicrobial activities of nutmeg (water, ethanol extract and essential oil). The total phenol contents of water, ethanol extract and essential oil were 3.4%, 16.9%, and 3.2%, respectively. Hydrogen donating abilities of water, ethanol extract and essential oil at 1,000 ppm were 4.9%, 41.8% and 6.8%, respectively. The antioxidative activities in linoleic acid substrates were in the order of BHT > ethanol > extract > ${\alpha}$-tocopherol > essential oil > water extract. The antioxidative activities in linoleic acid emulsion substrates were in the order of BHT > water extract > essential oil > ethanol extract > ${\alpha}$-tocopherol. In antimicrobial activity, ethanol extract showed growth inhibition effect against Micrococcus luteus, Bacillus cereus and Salmonella enteritidis, and the essential oil showed growth inhibition effect against Micrococcus luteus. However, no antimicrobial activity of water extract was observed. The nitrite-scavenging abilities of water, ethanol extract and essential oil at 1,000 ppm were 4.5%, 28.8% and 98.8%, respectively, and the ACE inhibitory activities were 0.2%, 11.0% and 10.0%, respectively.

Antioxidative Activity of Acylated Anthocyanin Isolated from Fruit and Vegetables

  • The antioxidative activity of seven different acylated anthocyanin pigments isolated from grape, sweet potato, eggplant and red cabbage was evaluated bvy using linoleic acid autoxidation and rat liver microsomal systems. The acylated anthocyanins were isolated and purified by Amberlite SAD-7, ODS and Sephadex LH-20 column omatography, and preparative HPLC. Most of acylated anthocyanisns exhibited antioxidative activity as strong as $\alpha$-tocoperol, and especially peonidin3-O--(6-O-trans-caffeyl)-2-O-(6-O-trans-feuloylglucopyranosyl)-$\beta$-D-glucopyranosyl-5-O-$\beta$-D-glucopyranoside from purple sweet potato showed the strongest activity, comparable to BHA (not significant, p<0.05) in the linoleic acid system. Meanwhile, two acylated anthocyanins from the pericarps of grape and eggplant inhibited considerably the MDA formation from rat liver microsomal lipid peroxidation induced by FeSO$_4$/$H_2O$$_2$. In particular, malvidin 3-O-(6-O-p-coumaroyl)-$\beta$-D-glucopyranosde from grape pericarps showed the strongest antioxidant activity, comparable to $\alpha$-tocopherol (not significant, p<0.05). These results suggest that the acylated anthocyanins from fruits and vegetables can be used as potential dietary ntioxidants and natural colorants.

목단피 추출물의 항균 및 항산화 작용 (Antimicrobial and Antioxidative Activities from Moutan Cortex Extract)

  • 목단피의 메탄을 추출물과 용매 분획물을 이용하여 항균작용, 프리라디칼 소거작용 및 항산화 작용을 검색하고, HPLC와 GC를 이용하여 페놀화합물을 분석하였다. 항균작용은 에칠아세테이트 분획에서 가장 높게 나타났으며, 그람 음성균보다는 그람 양성균에 대하여 더욱 효과적이었다. 에칠아세테이트 분획의 MIC를 측정한 결과, 그람 양성균에 대하여는 156-1250$\mu\textrm{g}$/ml, 그람 음성균에는 2500-5000$\mu\textrm{g}$/ml 이었다. DPPH 라디칼에 대한 프리라디칼 소거활성과 linoleic acid를 이용한 항산화 시험에서도 에칠아세테이트 분획이 가장 높은 활성을 나타내었다. 에칠아세테이트 분획의 항산화 활성은 $\alpha$-tocopherol과 비슷하였다. 에칠아세테이트 분획으로부터 U와 HPLC를 이용하여 3종의 페놀화합물을 확인하였으며, 그 함량은 p-hydroxybenzoic acid 1.35%, methyl gallate 14.61%, gallic acid 4.01% 이었다.

In vitro Antioxidant Effect of Aster scaber Thunb. Extract

  • Methanol extract and solvent fractions of Aster scaber Thunb. were tested to elucidate antioxidative characteristics in vitro. Methanol extract showed strong inhibition on linoleic acid autoxidation and conjugated diene production. Hexane and diethyl ether fractions showed stronger chelating effect on $10^{-4}M\;FeCl_3$ than the other fractions. Butanol fraction had the strongest capacity on $10^{-3}M\;CuSO_4$ similar to ${\alpha}$-tocopherol. Synergistic effect of butanol fraction with histidine, methionine, and lysine on linoleic acid autoxidation had been observed. Contents of total phenol, vitamin E, vitamin C, and carotenoid were evaluated. From the results, Aster scaber Thunb. could be expected to play a role as an antioxidant in vivo.

가시오가피 잎·뿌리·추출물의 이화학적 특성 및 항산화 효과 (Antioxidative Activity and Chemical Characteristics of Leaves, Roots, Stems and Fruits Extracts from Acanthopanax senticosus)

  • 가시오가피 잎, 뿌리, 줄기 및 열매의 수용성, 에탄올 및 메탄올 추출물의 생리활성 물질 분석과 항산화 활성(DPPH free radical scavenging 활성, Cu/Fe-환원력, 간 조직 microsome 생체막 및 linoleic acid 과산화지질, Tyrosinase 저해활성)을 측정 하였다. 가시오가피 잎의 열수 추출물에서 추출 수율 16.04%, 폴리페놀 화합물 함량2.67% 및 플라보노이드 함량 1.43%로 가장 높았다. 가시오가피 잎, 뿌리, 줄기 및 열매의 주요 미네랄은 K, Ca 및 Mg 이었다. DPPH free radical scavenging 활성, Cu/Fe-환원력, 간 조직 microsome 생체막 및 linoleic acid의 과산화 지질 측정에 의한 항산화 활성은 가시오가피 잎, 뿌리, 줄기 및 열매 모두에서 나타났으며, 시료 처리 농도 의존적으로 활성이 증가되는 것으로 나타났다. 이상의 실험 결과에서 가시오가피 잎, 뿌리, 줄기 및 열매 모두에서 높은 항산화 활성이 있었으며, 이는 폴리페놀 화합물과 플라보노이드와 같은 생리활성 성분을 많이 함유하고 있는 것과 깊은 관련성이 있는 것으로 향후 건강기능식품이나 기능성화장품의 천연 항산화제 소재 개발 관련 연구의 기초자료로 활용될 수 있을 것으로 사료되어 진다.

측백나무 잎.열매 추출물의 이화학적 특성 및 항산화 효과 (Antioxidative Activity and Chemical Characteristics of Leaf and Fruit Extracts from Thuja orientalis)

  • 측백나무 잎 및 열매 물, 에탄올 및 메탄올 추출물의 생리활성 물질(총 폴리페놀 화합물, 플라보노이드, 미네랄, 지방산 조성) 분석과 항산화 활성(DPPH free radical scavenging 활성, Cu/Fe-환원력, 간 조직 microsome 생체막 및 linoleic acid 과산화지질)을 측정하였다. 측백나무 잎의 메탄올 추출물에서 추출 수율(12.90%), 폴리페놀 화합물(16.02%) 및 플라보노이드(0.25%) 함량이 가장 높았다. 측백나무 잎 및 열매의 주요 미네랄은 Ca, K, 및 Mg이었다. 측백나무 잎의 주요 지방산은 palmitic acid 및 lauric acid였으며, 열매는 palmitic acid 및 decanoic acid가 높은 함량을 보였다. DPPH free radical scavenging 활성, Cu/Fe-환원력, 간 조직 microsome 생체막 및 linoleic acid의 과산화 지질 측정에 의한 항산화 활성은 측백나무 열매보다는 잎 추출물에서 높았으며, 시료 처리 농도 의존적으로 활성이 증가되는 것으로 나타났다. 이상의 실험 결과에서 측백나무 잎의 메탄올 추출물에서 높은 항산화 활성이 있었으며, 이는 폴리페놀 화합물과 플라보노이드와 같은 항산화 활성을 나타내는 생리활성 성분을 많이 함유하고 있는 것으로 나타나 향후 건강기능식품이나 화장품의 천연 항산화제 소재개발에 유용하게 사용될 것으로 사료된다.

식이 지방산이 혈소판 인지질의 지방산 조성, 혈장 Thromboxane B2의 농도 및 혈소판 응집에 미치는 영향 (Effect of Dietary Fatty Acids on Fatty Acid Composition of Platelet Phospholipids, Thromboxane B2 Formation, and Platelet Aggregation in Men)

  • The degree of platelet aggregation, thromboxane B2(TXB2)formation and fatty acid composition of platelet phospholipids(PL) were investigated in 24 healthy male subjects who for five weeks consumed either corn oil(CO) rich in linoleic acid(LA), perilla oil (PO) rich in $\alpha$-linoleic acid($\alpha$-LAN), or canola oil(CNO) rich in oleic acid(OA) as a major fat source. Total fat intake was 30% of total calories and prescribed oil intake of each dietary group was 50% of the total fat intake. In the CO group, significantly decreased contents of polyunsaturated fatty acids(PUFA), n-6 PUFA, n-3 PUFA and eicosapentanoic acid(EPA) were observed, and significantly increased contents of OA and saturated fatty acids(SFA) were observed in platelet PL after 3 weeks and 5 weeks of dietary treatment. In the PO group, contents of OA and docosahexanoic acid(DHA) were increased, and the ratio of n-6/n-3 was decreased significantly in platelet PL after dietary treatment. The CNO group showed significatnlty decreased contents of PUFA, P/S ratio, n-6 PUFA, LA,(EPA+DHA)/arachidonic acid(AA), and significantly increased SFA contents after 3 weeks of the oil-based diet. The dietary-induced effects on fatty acid composition of platelet PL were observed mostly after 3 weeks of the oil-based diet. The dietary-induced effects on fatty acid composition of platelet PL were observed mostly after 3 weeks. Plasma TXB2 levels were increased after 3 and 5 weeks of dietary treatment. However, only the CO and CNO groups showed significantly increased plasma TXB2 levles after 3 and 5 weeks of dietary treatment. However, only the CO and CNO groups showed significantly increased plasma TXB2 levels after 5 weeks of experimental diets, when compared with initial values. Degree of platelet aggregation increased only in the CO group after dietary treatment. As a result, at week 5 the degree of platelet aggregation of the CO group was significantly higher than those of the PO and CNO groups. Among the three oil-based diets, the PO-based diet seems to have beneficial effects on atherosclerosis by influencing plasma TXB2 levels and the degree of platelet aggregation, while the CO-based diet showed the most adverse effects. Our results imply that plasma TXB2 levels might be affected by dietary fatty acid composition.

솔잎 추출물의 항산화 효과 (Antioxidative Effects of Pine (Pinus denstifora) Needle Extracts.)

  • 식물성 성분 중에서 생리활성 인자를 탐색할 목적으로 솔잎의 수용성 추출물에 의한 항산화 활성을 DPPH법, thiocyanate법, TBA법 및 microsome 생 체 막 지 질 과산화물 생성정도를 TBARS법으로 측정 하고, 총 폴리페놀 함량을 측정하였다. 솔잎 수용성 추출물의 총 폴리페놀 화합물 함량은 1.61%이었으며, DPPH 측정법에서는 짙은 자색의 탈색되는 정도로 나타내는 전자 공여능이 솔잎 수용성 추출물의 0.1% 농도에서 BHT와 비슷한 수준으로 항산화 활성이 높게 나타났다. 간 microsome을 이용한 생체막 지질 과산화 억제정도는 솔잎 수용성 추출물의 0.1% (67.7%)>0.05% (63.1%)>0.01%(28.2%) 순으로 농도에 비례하여 증가하였다. Linoleic acid 산화 실험계를 이용한 thiocyanate법에서는 0.01%, 0.05%, 0.1%의 농도에서 모두 반응 7일째까지 대조구에 비해 매우 강한 항산화 활성을 보였으며, TBA법에서는 반응 6일째까지 는 0.1%>0.05%>0.01%의 순으로 농도 증가에 따라 항산화 활성도 높게 나타났다. 이상의 결과에서 솔잎 수용성 추출물 중에는 in vitro 항산화 실험에서 항산화 활성을 나타내는 생리활성 성분이 존재하는 것으로 볼 수 있으므로 천연 항산화제로서의 가능성을 시사하였다.

  • 유지의 자동산화과정중의 ${\alpha}-$, ${\gamma}-Tocopherol$${\gamma}-tocotrienol$에 대한 주요 산화생성물로 quinone이 확인되었으며, 미량의 dimer 혹은 trimer로 추정되는 성분이 검출되었다. 생성된 quinone은 micro column을 사용하여 분취한 후 np-와 rp-HPLC로 분석하였고 MS로 동정하였다.

Biosynthesis of Conjugated Linoleic Acid and Its Incorporation into Ruminant's Products

  • Bio-hydrogenation of $C_{18}$-unsaturated fatty acids released from the hydrolysis of dietary lipids in the rumen, in general, occurs rapidly but the range of hydrogenation is quite large, depending on the degree of unsaturation of fatty acids, the configuration of unsaturated fatty acids, microbial type and the experimental condition. Conjugated linoleic acid (CLA) is incompletely hydrogenated products by rumen microorganisms in ruminant animals. It has been shown to have numerous potential benefits for human health and the richest dietary sources of CLA are bovine milk and milk products. The cis-9, trans-11 is the predominant CLA isomer in bovine products and other isomers can be formed with double bonds in positions 8/10, 10/12, or 11/13. The term CLA refers to this whole group of 18 carbon conjugated fatty acids. Alpha-linolenic acid goes through a similar bio-hydrogenation process producing trans-11 $C_{18:1}$ and $C_{18:0}$, but may not appear to produce CLA as an intermediate. Although the CLA has been mostly derived from the dietary $C_{18:2}$ alternative pathway may be existed due to the extreme microbial diversity in the reticulo-rumen. Regardless of the origin of CLA, manipulation of the bio-hydrogenation process remains the key to increasing CLA in milk and beef by dietary means, by increasing rumen production of CLA. Although the effect of oil supplementation on changes in fatty acid composition in milk seems to be clear its effect on beef is still controversial. Thus further studies are required to enrich the CLA in beef under various dietary and feeding conditions.
