The expression of Corynebacterium ammoniagenes purF was analyzed by utilizing a plasmid carrying a cat gene fused to the purF promoter region. Adenine and guanine repressed the expression of the purF gene by 20~30% but hypoxanthine did not exert such repressive effect. The expression purF was maximal at the late log phase and remained constant throughout the stationary phase. Promoter $P_{180}$ which was developed in C. glutamicum was also functional in C. ammoniagenes, achieving maximal activity at the late log phase. The promoter outperformed Escherichia coli $P_{tac}$ promoter by 40~50% level. DNA-affinity purification identified a protein which could bind to the promoter region of the purF gene. The protein showed high similarity to the CRP-family transcriptional regulator encoded by NCgl0120 in C. glutamicum. The size of the screened protein agreed with the expected protein size from the ORF NCgl0120. The corresponding gene in C. ammoniagenes encoded a 42 kDa polypeptide composed of 400 amino acids with expected pI of 4.9. The encoded protein showed 14.1% and 15.8% identity with E. coli and Bacillus subtilis PurR, respectively, suggesting that the isolated protein might be a novel type of regulatory protein involved in the regulation of purine metabolism.

The rising concentration of atmospheric carbon dioxide is causing ocean acidification and global warming. The seahorse is an important species in marine ecosystems and fishery markets, however, their populations have recently decreased due to ocean acidification. As a result, we examined changes in the physiological responses of the spotted seahorse Hippocampus kuda when it was exposed to acidic sea water (pH 6.0, 6.5, and 7.0) and normal seawater (pH 8.0 as the control) over a period of 15 days. As the pH decreased, the seahorses' body weight and length also decreased. Components in body of ash, the crude lipids and crude proteins also differed significantly with changes in pH, due to stress caused by the seahorses' exposure to the acidic water conditions. The superoxide dismutase levels were significantly lower in the pH 6.0 and 6.5 groups than they were in the pH 7.0 and pH 8.0 groups. However, the catalase and glutathione levels were significantly higher in the acidic sea water groups. We suggest that decreasing the pH level of rearing water induces a stress response in H. kuda, damaging their ability to maintain their homeostasis and energy metabolism. Antioxidant enzymes are generally sensitive to acidic stress; in this study, the antioxidant activity was significantly affected by the pH level of the rearing water. These results indicate that physiological stress, induced by exposure to acidification, induces an antioxidant reaction, which can reduce general components in the body and the growth of H. kuda.

The objective of this study was to determine the effect of antioxidant enzyme activity and radical scavenging activities of cucumber grown greenhouse. The analytic method of antioxidant activities were measured by estimating DPPH free radical scavenging, nitrite scavenging ability activity and ABTS radical scavenging ability. The DPPH free radical scavenging activity of Jangjukcheongjang was the highest in most of the growth stage. The Nitrite scavenging ability at pH 1.2 was more than 40% in all cultivars and growth stages. The ABTS radical scavenging ability of Jangjukcheongjang and Nulpureuncheongjang showed relatively little higher than Janghyeongnakhap and Sinjoeunbaekdadagi. The SOD activity showed higher activity than 95% in all cultivars and growth stages. The activity of CAT was highest in the Nulpureuncheongjang of 2 (12~16cm) growth stage, and the APX activity of Nulpureuncheongjang and Janghyeongnakhap showed a relatively high activity. The POX activity showed distinctly different trends depending on the growth stage, and that is, the activity at harvest stage was significantly reduced. These results suggest that the cucumber had the potent biological activities, and that in the future, the availability of cucumber will be increase in the field of high-value cosmetic and food products.
The aim of this study was to investigate the effect of dental therapeutic agent on conduction velocity and threshold current of intradental A- and C-fibers in the cat. Inferior alveolar nerve of cat anesthetized with sodium pentobarbital was exposed and dissected until response of functional single pulp nerve until could be evoked by monopolar electrical stimulation of the crown of the lower left canine teeth. 10ms rectangular pulse was used to determine the threshold current and 1ms rectangular pulse was used to determine conduction velocity. After application of calcium chloride (1, 2, 6M), calcium hydroxide mixed with saline, potassium chloride (0.2, 0.8, 1.6M), eugenol, zinc oxide eugenol to the cavity on the labial surface, conduction velocity and threshold current of single pulp nerve unit were compared with the control. In 10 cats, 24 $A{\delta}$- and 11 C- pulp nerve units were recorded. The mean conduction velocities of $A{\delta}$- and C-fibers were 7.5m/sec (SD=5.8) and 1.2m/sec (SD=0.4), respectively. The mean threshold current was $12.3{\mu}A$ (SD=5.3) for $A{\delta}$-fibers and $24.9{\mu}A$ (SD=8.1) for C-fibers. 1, 2, 6M calcium chloride caused decrease of conduction velocity and remarkable increase of threshold current in $A{\delta}$- and C-fibers. The effect of calcium hydroxide mixed with saline was similar but smaller than calcium chloride solution. 0.2M potassium chloride had insignificant effect. In 0.8M potassium chloride, the threshold current was increased although conduction velocity was not affected. In 1.6M potassium chloride, the threshold current was increased and the conduction velocity was slowed down. Spontaneous activity was recorded frequently for first 5 min but gradually reduced both in $A{\delta}$- and C-fibers. Eugenol had irreversible effect on pulp nerve in that initially there were not certain changes in the conduction velocity and threshold current of $A{\delta}$- and C-fibers, but the responses to electrical stimulation were abruptly disappeared after sustained application and were not recovered. Contrary to eugenol, zinc oxide eugenol did not caused significant increase of the threhold current and caused time dependent decrease of the conduction velocity, and did not show any irreversible change.
This study examined the neurotoxicity of aluminum sulfate (AS), an environmental pollutant, and the protective effect of Phrymaleptostachya var. asiatica HARA (PLVAH) extract on the neurotoxicity induced by AS in the cultured C6 glioma cells. For this study, the cell viability and antioxidative effects, such as electron donating (ED) activity, lipid peroxidation (LP) activity, and superoxide anion-radical (SAR) scavenging activity, were analyzed. AS decreased the cell viability significantly in a dose-dependent manner and the $XTT_{50}$ value was measured at $120.0{\mu}M$ of AS. The neurotoxicity of AS was determined to be mid-toxic by Borenfreund and Puerner's toxic criteria. In addition, the catalase (CAT), antioxidant enzyme remarkably increased the cell viability injured by AS-induced neurotoxicity in these cultures. Regarding the protective effect of the PLVAH extract on AS-induced neurotoxicity, PLVAH extract significantly increased the ED ability, and the inhibitory ability of the LP and SAR scavenging ability. These findings suggest that oxidative stress is involved in the cytotoxicity of AS, and the PLVAH extract effectively protected against AS-induced neurotoxicity by its antioxidative effects. Natural resources, such as the PLVAH extract may be a putative therapeutic agent for the treatment of the toxicity induced by heavy metallic compounds, such as AS correlated with the oxidative stress.
The present study aims to examine the effect of turbid water on fishes in streams which branch into a turbid water area (Yeongyang-gun) and a non-turbid water area (Cheongsong-gun), and finally flow into the Imha reservoir. In a comparison of water quality, the chemical status of the water showed higher pH, DO and SS in the turbid water area than in the non-turbid water area. Also, high density of clay minerals such as vermiculite (V) and illite (I), which is from clay mineral leakage during rainfall, was detected in turbid water, resulting in an increase of turbidity. Fishes inhabiting the turbid water showed irregular spaces in gill lamella, cell separation, edema, and clubbing in epithelial tissues. Also, the gill surface showed roughness and plenty of muddy debris substances inside the gills. The Bowman's space was expanded because of contraction of the glomerulus in the Bowman's space of the kidney tissues. Antioxidant enzymes such as SOD, CAT, GPX, and GST showed higher activities in the specific tissues, muscles and kidney, of fishes living in turbid water than in the non-turbid area. We suggested that; first, the antioxidant activities were increased due to removal of harmful radicals generated in fish bodies in the turbid water area, second, long-time exposure of these histological changes in the tissues might have induced secondary lesion accompanying the inaccurate physiological constancy of fishes.
This study examined the antioxidative effects of Cornis fructus (CF) extract on cultured C6 glioma cells. For this purpose, cytotoxicity analysis of chromium trioxide ($CrO_3$) was performed and the protective effects of the CF extract on $CrO_3$-induced cytotoxicity was examined after the C6 glioma cells were cultured for 48 hours. The antioxidative effects, such as electron donating activity (EDA) and lactate dehydrogenase (LDH) activity were also analyzed. In this study, $CrO_3$ decreased the cell viability in a dose dependent manner. The $XTT_{50}$ value was determined to be $33{\mu}M$ after the cells were treated for 48 hours at a concentrations of $20{\sim}40{\mu}M$$CrO_3$. The catalase (CAT) antioxidant increased significantly the cell viability that had been decreased by $CrO_3$-induced cytotoxicity. Regarding the protective effect of the CF extract, the cell viability of the CF extract was increased significantly compared to that of $CrO_3$ only. In addition, the CF extract showed antioxidative effects, such as EDA and an inhibitory effect on the LDH activity. These findings suggest that the cytotoxicity of $CrO_3$ is correlated with oxidative stress, and the CF extract effectively prevented $CrO_3$-induced cytotoxicity through the antioxidative effects. In conclusion, natural products, such as the CF extract may be a useful therapeutic agent for the prevention or treatment of toxicity induced by heavy metals via oxidative stress.
The cultural salons in Beiping were very unique phenomenon in the 1930's Chines culturalhistory. These salons were created in the Beiping's urban environment with generosity toscholarship and art. The typical examples of these were the 'Lin Huiyin's salon' by Lin Huiyinand 'Poetry reading salon' by Zhu Gunagqian. 'Lin Huiyin's salon' became the representative salonin Beiping before the Sino-Japanese War broke out in 1937. In 'Lin Huiyin's salon' they heldmeeting in an impromptu, free and somewhat distracted atmosphere without any specific theme. Onthe other hand, Zhu Guangqian's 'Poetry reading salon', which included scholars, critics, poets andtheoreticians centered on Peking University and Tsinghua University, tendered to be academicbecause its main purpose was to test the "successful readability of new poetry". These two culturalsalons have received a positive evaluation in terms of promoting the 'Beiping school' andactivating the literary world, but the members who attended these salons were satirized as bluffersand hypocrites in the novels "The living room of our wife"(1933) and "The cat"(1946) which werewritten by Bingxin and Qian Zhougshu, respectively. In this article, I examined the positiveinfluences on cultural topography at the time and showed how they were reflected to the intelligentintellectuals who did not participate in the salons. Based on these contemplations, I also had achance to look at the 1930's salons correctly through ambivalent viewpoint.

BACKGROUND/OBJECTIVES: Owing to health concerns related to the consumption of traditional snacks high in sugars and fats, much effort has been made to develop functional snacks with low calorie content. In this study, a new recipe for Korean rice cookie, dasik, was developed and its antioxidative, lipid-lowering, and anti-inflammatory effects and related mechanisms were elucidated. The effects were compared with those of traditional rice cake dasik (RCD), the lipid-lowering effect of which is greater than that of traditional western-style cookies. MATERIALS/METHODS: Ginseng-added brown rice dasik (GBRD) was prepared with brown rice flour, fructooligosaccharide, red ginseng extract, and propolis. Mice were grouped (n = 7 per group) into those fed a normal AIN-76 diet, a high-fat diet (HFD), and HFD supplemented with RCD or GBRD. Dasik in the HFD accounted for 7% of the total calories. The lipid, reactive oxygen species, and peroxynitrite levels, and degree of lipid peroxidation in the plasma or liver were determined. The expression levels of proteins involved in lipid metabolism and inflammation, and those of antioxidant enzymes were determined by western blot analysis. RESULTS: The plasma and hepatic total cholesterol concentrations in the GBRD group were significantly decreased via downregulation of sterol regulatory element-binding protein-2 and 3-hydroxy-3-methylglutaryl-CoA reductase (P < 0.05). The hepatic peroxynitrite level was significantly lower, whereas glutathione was higher, in the GBRD group than in the RCD group. Among the antioxidant enzymes, catalase (CAT) and glutathione peroxidase (GPx) were significantly upregulated in the GBRD group (P < 0.05). In addition, nuclear factor-kappaB (NF-${\kappa}B$) expression in the GBRD group was significantly lower than that in the RCD group. CONCLUSIONS: GBRD decreases the plasma and hepatic cholesterol levels by downregulating cholesterol synthesis. This new dasik recipe also improves the antioxidative and anti-inflammatory status in HFD-fed mice via CAT and GPx upregulation and NF-${\kappa}B$ downregulation. These effects were significantly higher than those of RCD.

Objectives : CRATAEGII FRUCTUS(山査) is known as the substance which delays aging by the antioxidant effects. The purpose of this study is to investigate the effects of CRATAEGII FRUCTUS(山査) on antioxidant enzyme activities such as Thiobarbituric acid reactive substance(TBARS), Superoxide dismutase(SOD), Catalase(CAT), Glutathione peroxidase (GSH-px) in rat erythrocytes and blood plasma. Methods : Sprague-Dawley rats were divided into 3 groups, Normal group (supplied enough water and feeds only, Normal Group), D-galatose administered group(injected D-galatose 50mg/kg, 1time/day for 6 weeks, Control Group) and CRATAEGII FRUCTUS(山査) administered group (D-galactose 50mg/kg and CRATAEGII FRUCTUS(山査) extracts 85.0mg/200g 1time/day for 6 weeks, SS Group). Rats were sacrificed and TBARS, SOD, CAT, GSH-px, neutral fat and cholesterol were measured in rat erythrocytes and blood plasma. Results : There was no significant difference in the level of TBARS in the blood plasma between each experimental group. Red blood cell SOD activities were significantly different in each group. They were significantly decreased in the Control group compared to those of Normal and there was an increasing phenomenon in the SS group compared to the Control group. There was a significant difference in the activities of the red blood cell - glutathione peroxides in each group. There was a significant increase in the SS group compared to the Control group. Red blood cell catalase activities was no significant difference in each group. Plasma total lipid concentration was significantly different in each group. It was significantly increased in the Control group compared with the Normal group and it was decreasing in the SS group compared to the control group. Plasma triglyceride was not significantly different in each group. Plasma total cholesterol and Plasma HDL -cholesterol concentrations in the blood plasma were not significantly different in each group. Conclusions : According to the above results, it is considered that CRATAEGII FRUCTUS(山査) is effective in inhibiting lipid peroxides and increasing the activities of antioxidative(anti aging) enzyme in D-galactose induced aging rat.
