This experiment was performed to study the structure of esophageal mucousa and the histochemical properties of glycosaminoglycans of esophageal mucous cells in four teleostean species, i.e., Agramus agramus, Inimicus japonicus, Epinephelus chlorostigma and Helicolenus dactylopterus. To observe the structure of eosphageal mucosa, hematoxylin-eosin(H-E) staining was used. The glycosaminoglycans was stained with PAS(periodic acid schiff), alcain blue(AB) pH 2.5, AB pH 1.0, aldehyde fuchsin(AF) pH 1.7, AF pH 1.0, AB pH 2.5-PAS, AB pH 1.0-PAS and AF pH 1.7-alcian blue pH 2.5. As for the amount and histochemical composition of glycosaminglycans in Agramus agramus, most of mucous secreting columnar cell and mucous cells contain large and moderate amount of neutral glycosaminoglycans. A few of mucous cells having small amount of neutral glycosamino-glycans and minimal amount of sulfated glycosamino-glycans. In Inimicus japonicus, the esophageal mucous cells were composed of most of medium sized and large mucous cells with moderate amount of neutral glycosaminoglycanonly only, a few of medium sized and large mucous cells and most of small mucous cells with considerable amount of neutral glycosaminoglycans and minimal to small amount of nonsulfated glycosaminoglycans, and a feq of small mucous cells with small amount of neutral glycosaminoglycans and minimal amount of sulfated glycosaminoglycans. In Epinephelus chlorostigma, most of medium sized and large mucous cells were mixed small amount of neutral glycosaminoglycans with sulfated glycosaminoglycans, a few of which were contained with moderate or considerable amount of neutral glycosaminoglycans with sulfated glycosaminoglycans, while most of small mucous cells containing considerable amount of neutral glycosaminoglycans and small to moderate or considerable to minimal amount of nonsulfated glycosaminoglycans(sialomucin) a few of which containing a mixture of considerable amount of neutral glycosaminoglycans and considerable amount of nonsulfated glycosaminoglycans(sialomucin)only. In Helicolenus dactylopterus, most of medium sized and large mucous cells, mixing with moderate to considerable amount of neutral glycosaminoglycans and small to moderate amount of sulfated glycosaminoglycans, while most of small mucous cells with considerable amount of nonsulfated glycosaminoglycans (sialomucin), a few of which having a mixture of considerable amount of neutral glycosaminoglycans and considerable or small amount of nonsulfated glycosaminoglycans(sialomucin).

Lipopolysaccharide (LPS) and cell wall protein-A (CWP-A) were extracted from the cell wall of Vibrio vulnificus, Escherichia coli and Salmonella typhimurium. LPSs and CWP-As were injected into rat and the changes of the following blood components were examined. The change of the number of white blood cell (WBC), red blood cell (RCB), platelet (PLT), blood urea nitrogen (BUN) and blood glucose in rat blood and interferon (IFN) activity change by LPS and CWP-A were measured. WBC, RETI, PTT, and BUN were increased and RBC and blood glucose were increased slightly, but PLT was decreased.

A human-specific retroposon SINE-R.C2 has been derived from a human endogenous retrovirus HER V-K 10. It is absent in the genome of nonhuman primates and present within the third intron of the human C2 gene that is located in the class III region of the major histocompatibility complex. In the present study, we determined the regional location of the human C2 gene. The analysis of the Genebridge 4 radiation hybrid mapping panel using PCR amplification located the C2 gene between D6S1422 (10.1 cR) and CHLC.GATA4A03 (21.3) with a lod score of>3.0. This allowed us to localize C2 gene on the human chromosome 6 band p21.31.

Genetic Variation of Hemoglobin and Ganglioside Monooxygenase in Korean Sapsarees (Canis familiaris)Genetic variation of hemoglobin and erythrocyte ganglisoside monooxygenase were analyzed in Korean nature dog Sapsarees by horizontal starch gel electrophoresis and thin layer chromatography. Hemoglobin has three phenotypes A, B and AB, which are controlled by two codominant alleles, {TEX}$Hb^{a}${/TEX} and {TEX}$Hb^[b}${/TEX}, at one autosomal locus. Gene frequencies of {TEX}$Hb^{a}${/TEX} and {TEX}$Hb^{b}${/TEX} were 0.537 and 0.4625. Ganglioside monooxygenase has two phenotypes, dominant and recessive. They are controlled by a dominant allele {TEX}$Gmo^{a}${/TEX} and a recessive allele {TEX}$Gmo^{g}${/TEX}. Frequencies of {TEX}$Gmo^{a}${/TEX} and {TEX}$Gmo^{g}${/TEX} were 0.5477 and 0.4523.

Adenovirus which is an important infectious viral agent in respiratory and alimentary tract was investigated in Pusan, 1999. Fifteen cases of adenovirus were detected from stools and throat swabs of suspected patients. Two cases of enteric adenovirus were detected from a 5 years old boy and a 6-month-old boy. Thirteen cases of respiratory adenoviruses were detected from children aged under 10 years old and one adult. From respiratory specimens, 1 case of adenovirus type 2, 1 case of type 5, and 11 cases of type 3 were found. Enterotype 41 was detected from fecal preparations. Adenoviruses appeared mostly during winter months, January, February and December. Adenovirus showed a slowly progressive cytopathic effect on HEp-2 cells, Vero cells and BGM cells at 37
$^{\circ}C$ , in a 5-7%$CO_{2}$ incubation. An electron microscopic observation exhibited non-enveloped icosahedron with a diameter of 70nm. No significant differences on cytopathic effect and morphological features have been found from specimens of either alimentary tract or respiratory secretions. -
Cytokine deprivation-induced apoptosis can abrogate by the appropriate survival factors. Because the mechanism of Interleukin (IL)-2 deprived apoptotic cell death remains unclear, we here show the apoptosis in CTLL2 cells correlates with an increase of the activity of caspase3-like protease(s). Inhibition of caspase3-like protease(s) with caspase protease inhibitors (Z-VAD, Z-EVD, and Z-LPD) blocks typical apoptotic morphological abnormalities in CTLL2 cells. Interestingly, Bcl-{TEX}$X_{L}${/TEX} protein was decreased by IL-2 deprivation in the cells. These results suggest that caspase3-like protease(s), not caspase1, plays an important role in apoptosis execution of CTLL2 cell death.

To understand the effects of the fraction from Chrysanthemum coronarium L. (CC), we prepared five different types of samples, denoted here as CCMM, CCMH, CCMEA, CCMB and CCMA. We studied the effects of these samples on the phase transition of liposomal membranes by high-sensitivity differential scanning calorimetry (nano-DSC). We used dipalmitoylphosphatidylcholine (DPPC) bilayers which make most stable liposomes among the other phosphatidylcholines. When the samples were added to the bilayers, the phase transition temperatures of DPPC liposomes incorporated with CCMH and CCMEA were decreased by 1.5 and 2
^{\circ}C$ , while the other three fractions showed less tendencies. The CCMH and CCMEA fractions markedly affected the thermotropic properties of DPPC liposomes, broadened and shifted the thermograms of DSC. It also significantly reduced the size of cooperative unit of the transition. In all cases, there was no change in enthalpy of transition within the concentration range of the CC fractions studied. We concluded that the incorporation of the CCMH and CCMEA into DPPC liposomes was preferentially located in the hydrophobic core of DPPC bilayers compared to the other three fractions CCMM, CCMB and CCMA. These results suggest that certain substances in CCMH and CCMEA fractions might have biologically significant effects on the fluidity of biological membrane. -
Maekmoondong-Tang is prescription recorded on Zang‘s
${ulcorner}$ Synopsis of the Golden Chamber${\lrcorner}$ for the first time. It can be used in abnormal rising of qi, asthma, swelling throat ect. So it is observed influence of Maekmoondong-Tang on respiratory aspect of allergic asthma patient, we have significant result and report it. We used egg-white and ovalbmuin (OVA) to induce asthma in experimental animals for observe what maekmoondong-Tang have an effect on asthma. And it is observed respiratory aspect and obtained next conclusions. 1. Number of total respiration in group that is prescribed Maekmoondong-Tang slightly increased than in control group. 2. Number of abnormal respiration in group that is prescribed Maekmoondong-Tang decreased than in control group significantly. -
Trichostatin A (TSA) is a Streptomyces product, which inhibits the enzyme activity of histone deacetylase. It is also known as an inducer of apoptosis in several human cancer cell lines. In this study, we investigated the mechanism of apoptosis induced by TSA in MDA-MB-231 human breast carcinoma cells. The cytotoxicity of TSA on MDA-MB-231 cells was assessed by MTT assay. The cell viability was decreased dose-dependently and the IC\ulcorner value was about 100 ng/ml after 48 h treatment with TSA. Morphological change and DNA ladder formation, the biochemical hallmarks of apoptotic cell death, were observed after treatment of TSA in a concentration-dependent manner, which was accompanied with cleavage of poly(ADP-ribose) polymerase and
$\beta$ -catenin, and activation of caspase-3. TSA treatment up-regulated the expression of a cyclin-dependent kinase inhibitor p21 (Wafl/Cip1) protein, a key regulatory protein of the cell cycle. However, there is no detectable change of both Bcl-2 and Bax expressions. These results demonstrated that TSA might inhibit cell growth through apoptosis in human breast carcinoma MDA-MB-231 cells. -

Long terminal repeat (LTR) of human endogenous retrovirus K family (HERV-K) has been found to be coexpressed with sequences of closely located genes. We examined the transcribed HERV-K LTR elements in human liver and kidney tissues. Using the cDNA synthesized from mRNA of human liver and kidney, we performed PCR amplification and identified six HERV-K LTR elements. Those LTR elements showed a high degree of sequence similarity (93.3∼96.6%) with human-specific LTR. A phylogenetic tree obtained by the neighbor-joining method revealed that HERV-K LTR elements (Liv-1, 2, 3 and Kid-1, 2, 3) were belonged to group I. Our data suggests that HERV-K LTR elements are active on human liver and kidney tissues and may represent a source of genetic variation connected to human disease. -
The RAD4 gene of Saccharomyces cerevisiae is essential for the incision step of UV-induced excision repair. A yeast RAD4 gene has been previously isolated by functional complementation. In order to identify the RAD4 homologous gene from fungus Coprinus cinereus, we have constructed cosmid libraries from electrophoretically separated chromosomes of the C. cinereus. The 13 C. cinereus chromosomes were resolved by pulse-field gel electrophoresis, hybridized with S. cerevisiae RAD4 DNA, and then isolated homologous C. cinereus chromosome. Here, we report the cloning and characterization of fungus C. cinereus homolog of yeast RAD4 gene. Southern blot analysis confirmed that C. cinereus contains the sequence homologous DNA to RAD4 gene and this gene exists as a single copy in C. cinereus genome. When total RNA isolated from C. cinereus cells was hybridized with the 3.4 kb BglII DNA fragment of the S. cerevisiae RAD4 gene, a 2.5 kb of transcript was detected. The isolated gene encodes a protein of 810 amino acids.

We examined the effects of Kamgil-Tang on the process of lipopolysaccharide (LPS)-induced unclear factor (NF)-
${\kappa}$ Bp65 and inhibitory (I)-${\kappa}$ B${\alpha}$ alteration in RAW 264.7 cell and acute lung injury in rats. Immunoblot analysis showed that LPS-induced degradation of I-${\kappa}$ B${\alpha}$ in RAW 264.7 was inhibited by pretreatment of Kamgil-Tang. The total cells of bronchoalveolar lavage fluid by LPS challenge markedly decreased in the Kamgil-Tang pretreatment rats. Kamgil-Tang pretreatment caused also a decline in neutrophils infiltration into interstitium of the lung. In the alveolar macrophages and neutrophils, decreased NF-${\kappa}$ Bp65 and inducible nitric oxide synthase and increased I-${\kappa}$ B${\alpha}$ immunoreaction were detected in Kamgil-Tang pretreated rats compared with LPS alone treated ones. It may be concluded that Kamgil-Tang attenuates the development of LPS-induced inflammation by reduction of NF-${\kappa}$ Bp65 activation and neutrophil-mediated acute lung injury. Kamgil-Tang would be useful as a therapeutic agent for endotoxin-induced lung disease.
