홍삼추출물이 마우스 복강 대식세포 Hydrogen Peroxide 생산에 미치는 영향 (Effects of Red Ginseng Extracts on Hydrogen Peroxide Production of Murine Prtitoneal Macrophages)

  • 홍삼의 추출물인 50% ethanol extract, crude saponin, 그리고 lipid soluble fraction이 마우스 대식세포의 oxidative burst를 유발할 수 있는지 여부를 알아보고자 in vitro와 in vivo에 각각의 추출물을 처치하고 hydrogen peroxide 생산을 DCFH-DA를 이용한 형광분광광도법으로 측정하였다. 형광분광법에 의한 hydrogen peroxide의 측정을 최적화하기 위한 DCFH-DA의 농도는 3.2$mu extrm{m}$이었고, oxidative burst를 유도하기 위한 zymosan A, PNA의 최적 농도는 각각 100$\mu\textrm{g}$, 250'기호'를 사용하였다. In vitro의 경우, 홍삼의 3가지 추출물은 모두 oxidative burst를 유발하지 못하였지만, zymosan A로 유발한 경우에는 50% ethanol extract에서 가장 높은 hydrogen peroxide를 생산하였다. In vivo 실험에서는, lipid soluble extract에서만 유의하게 증가한(P<0.01) oxidative burst를 유발하였고, ginsenoside(saponin)가 어느 정도 포함되어 있는 50% ethanol extract와 crude saponin은 대조군에 배하여 유의하게 낮은(P<0.05) hydrogen peroxide를 생산하였다. 이는 ginsenoside가 마우스의 nitric oxide 생산을 억제한다는 다른 연구자들의 보고와 일치하는 결과이다. Oxidative burst를 유발한 lipid soluble extract에는 phenol계 화합물, polyactylence계 화합물, 미량성분 등이 함유되어 있으므로 차후 연구를 통하여 과연 어느 성분이 hydrogen peroxide를 증가시키는지 규명하는 것이 필요하다.

관절염 모델에서 가미청열사습탕(加味淸熱瀉濕湯)의 진통 및 소염 효과에 관한 연구 (Anti-nociceptive and Anti-inflammatory Effects of Gami-cheongyulsaseub-tang in Arthritic Model)

  • Objectives This study was carried out to find the effects of Gami-cheongyulsaseub-tang (hereinafter referred to GCST) on the inhibition of zymosan-induced pain in rats and collagen II-induced arthritis (CIA) in DBA/1J mouse. Methods As an acute inflammatory pain model, peripheral inflammation was induced by intraplantar injection of zymosan into the right hind paw in rats and then the hyperalgesia and pain regulating factors in spinal cord were analyzed. As a chronic inflammation model, the mixture of collagen II and complete Freund's adjuvant was treated into mice to establish rheumatoid arthritis and then body weight, thickness of hind paw, pathological change of spleen, immunological rheumatoid factor (IgG1, IgG2a, IgG2b, IgM and anti-collagen II), pro-inflammatory cytokines, and bone injury were analyzed. Results In the acute inflammatory pain model, GCST significantly inhibited the thermal and mechanical hyperalgesia and the pain regulating factors, including Fos, CD11b, PKA and PKC, in the spinal cord with a dose-dependent manner. In the chronic rheumatoid arthritis model, GCST administration decreased arthritic index and paw edema as compared with CIA control group. In particular, GCST reduced significantly the serum levels of total IgG2a, IgG2b, IgM, and specific anti-collagen II, but not total IgG1. GCST also resulted in the attenuation of bone injury and spleen enlargement/adhesion in CIA mice. Moreover, the secretion of pro-inflammatory cytokines TNF-${\alpha}$ and IL-$1{\beta}$ in CIA mice was significantly reduced by GCST in a dose-dependent manner. Conclusions Comparison of the results in this study showed that GCST had anti-nociceptive and immunomodulatory effects. These data imply that GCST can be used as an effective drug for not only rheumatoid arthritic pain but also other auto-immune diseases.

어류혈청의 보체에 의한 살균작용 (Bactericidal action by complement of fish serum)

  • 어류의 방어기구를 밝히기 위하여, 잉어, 틸라피아 및 넙치의 혈청 중에 존재하는 보체의 살균작용에 대하여 실험하였다. 실험어의 모든 정상혈청으로 E. coli는 살균되었고 가열하여 보체를 불활성화시킨 혈청에서는 살균되지 않았다. zymosan으로 처리한 실험어의 정상혈청에서는 살균작용이 나타나지 않았으므로 정상혈청 중의 살균작용에도 보체의 대체경로가 중요한 것으로 생각되었다. EGTA로 처리한 틸라피아와 넙치의 정상혈청에서는 살균작용이 지속되고 있었으나, 잉어의 살균작용은 소실되는 점으로 봐서 잉어의 대체경로에는 $Ca^{2+}$이 요구되는 것으로 나타났다. 실험어의 정상혈청이 병원성이 있는 E. tarda에 대해서는 살균작용을 나타내지 않았고 병원성이 없는 E. coli에 대해서는 살균작용을 나타냈다. E. coli와 E. tarda는 면역혈청 중에서 모두 살균되었고 그 살균속도는 정상혈청에 비하여 빠른 것으로 나타났다.

Mouse Air Pouch Model에서 침전극 저주파치료로 유도된 소염 작용에 관한 연구 (Study for Related Mechanism of Anti-Inflammatory Effect Induced by Neddle electrode electrical stimulation in Mouse Air Pouch Model.)

  • In oriental medicine, manual-acupuncture and electroacupuncture (EA) have been widely utilized to cure several inflammatory diseases such as arthritis. We designed this experiment to find neurochemical mechanism related to electroacupuncture induced anti-inflammatory effect on mouse air pouch model. EA with both low frequency (1 Hz) and high frequency (120 Hz) was treated after induction of inflammation in air pouch using injection of zymosan. To verify the role of opioid system in electroacupuncture-induced anti-inflammatory effect, naloxone (10 mg/kg) was pretreated. In addition, idazoxan (5 mg/kg) was pre-treated to evaluate the possible effect of endogenous adrenergic system in autonomic system on EA induced anti-inflammatory effect. As results of this study, naloxone pretreatment did not change the anti-inflammatory effect evoked by high frequency EA, while low frequency EA(1 Hz) induced anti-inflammatory effect was dramatically suppressed by naloxone pretreatment. These data indicated that endogenous opioid system might be extensively involve in anti-inflammatory effect evoked by not high frequency, but low frequency EA. However, idazoxan pretreatment did not produce any modulatory effect on both low and high frequency EA induced anti-inflammatory effect, suggesting that EA induced anti-inflammatory effect was not mediated by endogenous adrenergic system. In conclusion, these data strongly suggested that EA induced anti-inflammatory effect is mediated by endogenous opioid system, not endogenous adrenergic system.

칼슘 길항제가 활성화된 호중구에서의 $O_{\overline{2}}$의 생성, NADPH oxidase활성도 및 탐식작용에 미치는 영향 (Effects of Calcium Antagonists on Superoxide Generation, NADPH Oxidase Activity and Phagocytic Activity in Activated Neutrophils)

  • 면역보체가 결합되어 있는 zymosan 또는 열로 응집된 IgG에 의하여 활성화된 호중구에서의 NADPH oxidase 의존적인 $O_{\overline{2}}$의 생성과 탐식작용은 칼슘의 흡수과정과 일치하였다. 활성화된 백혈구의 반응은 세포외 칼슘 농도에 따라 항진되었으며, 이는 칼슘의 킬레이트제인 EGTA나 EDTA에 의하여 유의하게 억제되었다. 활성화된 백혈구로부터 $O_{\overline{2}}$의 생성은 dantrolene과 chlorpromazine에 의하여 억제되었다. 칼슘 길항제인 bepredil, diltiazem, verapamil, nifedipine, nimodipine은 효과적으로 활성화된 백혈구의 칼슘흡수, $O_{\overline{2}}$ 생성 그리고 탐식 작용을 억제하였고 NADPH oxidase 활성도 또한 억제하였다. 그러므로, 칼슘 길항제는 칼슘 유입을 억제하거나 칼슘의 세포내 재분포 및 NADPH oxidase 반응계에 작용하여 활성화된 백혈구에서의 $O_{\overline{2}}$의 생성과 백혈구의 탐식작용을 억제할 것으로 시사되었다.

Ginsenoside Rg3 promotes inflammation resolution through M2 macrophage polarization

  • Background: Ginsenosides have been reported to have many health benefits, including anti-inflammatory effects, and the resolution of inflammation is now considered to be an active process driven by M2-type macrophages. In order to determine whether ginsenosides modulate macrophage phenotypes to reduce inflammation, 11 ginsenosides were studied with respect to macrophage polarization and the resolution of inflammation. Methods: Mouse peritoneal macrophages were polarized into M1 or M2 phenotypes. Reverse transcription-polymerase chain reaction, Western blotting, and measurement of nitric oxide (NO) and prostaglandin $E_2$ levels were performed in vitro and in a zymosan-induced peritonitis C57BL/6 mouse model. Results: Ginsenoside $Rg_3$ was identified as a proresolving ginseng compound based on the induction of M2 macrophage polarization. Ginsenoside $Rg_3$ not only induced the expression of arginase-1 (a representative M2 marker gene), but also suppressed M1 marker genes, such as inducible NO synthase, and NO levels. The proresolving activity of ginsenoside $Rg_3$ was also observed in vivo in a zymosan-induced peritonitis model. Ginsenoside $Rg_3$ accelerated the resolution process when administered at peak inflammatory response into the peritoneal cavity. Conclusion: These results suggest that ginsenoside $Rg_3$ induces the M2 polarization of macrophages and accelerates the resolution of inflammation. This finding opens a new avenue in ginseng pharmacology.

Effects of In Vitro Exposure to Silica on Bioactive Mediator Release by Alveolar Macrophages

  • Alveolar macrophages play a pivotal role in the pathogenesis of silicosis since the macrophages may release a wide variety of toxic and inflammatory mediators as well as mitogenic growth factors. In the present study, the effects of in vitro exposure to silica on release of various mediator such as reactive oxygen species, platelet activating factor(PAF), and interleukin-1 (IL-1) by alveolar macrophages were examined. First, hydrogen peroxide release from alveolar macrophages was monitored by measuring the change in fluorescence of scopoletin in the absence or presence of graded concentration of silica. Significantly enhanced release of hydrogen peroxide was observed at 0.5 mg/ml and above. A maximal enhancement of 10 fold above control was observed at 5 mg/ml silica. Similarly, in vitro exposure to silica also significantly stimulated the generation of chemiluminescence from alveolar macrophages at 0.5 mg/ml and above with n maximal enhancement of 8 fold at 5 mg/ml silica. Second, PAF release from alveolar macrophages after 30 min incubation at $37^{\circ}C$ in absence or presence of zymosan and silica was determined by measuring $^{3}H-serotonin$ release ability of the conditioned macrophage supernates from platelets. 5 mg/ml zymosan as a positive control fur the PAF assay increased PAF release by 19 % of total serotonin release. Furthermore, silica also resulted in significant enhancement of the PAF release compared with that in unstimulated (control) cells, i.e., $17.7{\pm}5.8%$ and $24.0{\pm}4.9%$ of total serotonin release at 5 mg/ml and 10 mg/ml silica, respectively, which represents the release of nanomole levels of PAF. Lastly, IL-1 production by alveolar macrophages was analysed following their stimulation with lipopolysaccharide (LPS) and silica by their capacity to stimulate thymocyte proliferation. $10\;{\mu}g/ml$ LPS resulted in an 11 fold increase in IL-1 production. In comparison, $50\;{\mu}g/ml$ silica resulted in a 4 fold increase in IL-1 release. These data indicate that in vitro exposure of alveolar macrophages to silica activates the release of various bioactive mediators such as reactive oxygen species, PAF and IL-1 which thus contribute to amplification of inflammatory reactions and regulation of fibrotic responses by the lung after inhalation of silica.

Novel Antihypertension Mechanism of 𝛽-Glucan by Corin and ANP-Mediated Natriuresis in Mice

  • Many of the 𝛽-glucans are known to have antihypertensive activities, but, except for angiotensin-converting enzyme II inhibition, the underlying mechanisms remain unclear. Corin is an atrial natriuretic peptide (ANP)-converting enzyme. Activated corin cleaves pro-ANP to ANP, which regulates water-sodium balance and lowers blood pressure. Here, we reported a novel antihypertensive mechanism of 𝛽-glucans, involved with corin and ANP in mice. We showed that multiple oral administrations of 𝛽-glucan induced the expression of corin and ANP, and also increased natriuresis in mice. Microarray analysis showed that corin gene expression was only upregulated in mice liver by multiple, not single, oral administrations of the 𝛽-glucan fraction of Phellinus baumii (BGF). Corin was induced in liver and kidney tissues by the 𝛽-glucans from zymosan and barley, as well as by BGF. In addition to P. baumii, 𝛽-glucans from two other mushrooms, Phellinus linteus and Ganoderma lucidum, also induced corin mRNA expression in mouse liver. ELISA immunoassays showed that ANP production was increased in liver tissue by all the 𝛽-glucans tested, but not in the heart and kidney. Urinary sodium excretion was significantly increased by treatment with 𝛽-glucans in the order of BGF, zymosan, and barley, both in 1% normal and 10% high-sodium diets. In conclusion, we found that the oral administration of 𝛽-glucans could induce corin expression, ANP production, and sodium excretion in mice. Our findings will be helpful for investigations of 𝛽-glucans in corin and ANP-related fields, including blood pressure, salt-water balance, and circulation.

Korean Mistletoe (Viscum album Coloratum) Extract Induces Eel (Anguilla japonica) Non-specific Immunity

  • Background: The immunomodulatory effects of Korean mistletoe (Viscum album Coloratum) on the innate immune responses of eel (Anguilla japonica) were studied. Methods: Mistletoe, Freund’s complete adjuvant (FCA), or phosphate-buffered saline (PBS) as a control was injected into eel peritoneal cavities. Results: Nitroblue tetrazolium (NBT)-positive cells in the head kidney of eel were significantly augmented by the second day post-injection of mistletoe. Reactive oxygen intermediates (ROI) were more produced in mistletoe-injected fish kidney leucocytes than in FCA-injected ones. The level of lysozyme activity in the serum of fish 2 days after injection with mistletoe was also significantly higher than that in the serum of the control fish. The optimal concentration of mistletoe in inducing the highest serum lysozyme activity was revealed to 500${\mu}$g/200 g of fish. In phagocytic activity assay, mistletoe-sensitized eel kidney phagocytes captured more zymosan than did the control fish. Conclusion: Korean mistletoe appeared to be a good activator of the non-specific immune responses of eel.
