Effect of Opsonization of Zymosan on the Chemiluminescent Response of Haemocytes in the Pacific oyster Crassostrea gigas

  • The effect of opsonization of zymosan with serum on the repiratory burst of haemocytes isolated from the Pacific oyster Crassostrea gigas was investigated. The reactive oxygen species (ROS) produced by the respiratory burst of haemocytes in response to the opsonized or unopsonized zymosan were measured using chemiluminescence (CL). The CL values were increased according to the increment of haemocyte number. The degree of CL amplification by increase of haemocytes from $0.1{\times}10^6$ to $0.5{\times}10^6$ cells/ml was 3-5 times, but comparatively low amplification was elicited by increase of haemocytes from $0.5{\times}10^6$ to $1{\times}10^6$ cells/ml. Chemiluminescences produced by the haemocytes in response to the zymosan opsonized with oyster serum were considerably higher than the CL produced by the haemocytes in response to the unopsonized zymosan.

Effects of Rudbeckia laciniata Extract on Phagocytosis of Serum-Opsonized Zymosan Particles in Macrophages (대식세포의 혈청으로 식균된 자이모잔의 탐식능에 대한 삼잎국화 추출물의 효과)

  • Phagocytosis is a primary and an essential step of host defense, and is triggered by the interaction of particles with specific receptor of macrophages. In this study, we investigated the effect of extracts of Rudbeckia laciniata (RLE) on the phagocytic activity of macrophage, by monitoring the phagocytosis-associated signal transduction. RLE markedly increased phagocytosis of serum-opsonized zymosan particles (SOZ), while phagocytosis of IgG-opsonized zymosan particles (IOZ) or none-opsonized zymosan particles (NOZ) remained unaffected. However, RLE did not affect the binding of opsonized zymosan particles (OZ) with the cell surface of macrophage. This suggests that RLE may regulate SOZ-induced intracellular signaling during phagocytosis of macrophage. To confirm this hypothesis, we investigated whether RLE was involved in the RhoA-mediated signal transduction during phagocytosis of SOZ. Inhibitors of the RhoA-mediated signaling pathway, such as Y-27632 (for ROCK), ML-7 (for MLCK), and Tat-C3 (for RhoA), totally blocked phagocytosis of SOZ enhanced by RLE, as well as phagocytosis of SOZ. Additionally, RhoA activity was markedly increased when cells were treated with RLE, suggesting that RLE could increase the phagocytic activity of macrophage via RhoA-ROCK/MLCK signal pathway. Thus, RLE may be used to develop functional foods for immunity.

Immunomodulating Effects on Macrophage of Rhamnan Sulfate Extracted from Monostroma nitidum (홀파래에서 추출한 Rhamnan sulfate의 Macrophage에 의한 면역조절효과에 관한 연구)

  • The rhamnan sulfate extracted from green algae seaweed, Monostroma nitidum was characterized as activity in vitro culture assay with macrophages from mice. Rhamnan sulfate indicated that F-4-3 fraction enhanced glucose consumption, as well as the production of nitrogen dioxide and tumor necrosis factor(TNF). F-4-3 fraction was also augmented IL-1 secretion from those macrophages. Effects of the pretreatment of peritoneal macrophages with rhamnan sulfate F-4-3 fraction and several polysaccharides as relative standard on the production of H2O2 induced with unopsonized zymosan A were examined. Pretreatment with polysaccharides inhibited the zymosan A mediated H2O2 production by macrophages. The phorbol myristate acetate (PMA) mediated H2O2 production was not affected by the pretreament. These result suggested that pretreatment of rhamnan sulfate interfered with the interaction of macrophages zymosan A. Rhamnan sulfate inhibited zymosan A mediated production of H**O** by macrophages and F-4-3 Fraction was also activator of macrophages.

Platelet-Activating Factor Potentiates the Activity of Respiratory Burst and Interleukin-1 in Rat Alveolar Macrophages

  • The objective of the present study was to test the effect of platelet-activating factor (PAF) on rat alveolar macrophages. PAF alone did not stimulate superoxide secretion from alveolar macrophages. However, PAF $(10^{-5}\;M)$ significantly enhanced phagocytic activator zymosan-induced superoxide secretion from alveolar macrophages. This enhancement of PAF plus zymosan was 30% above the sum of the separate effects of PAF and zymosan. Similarly, PAF $1.3{\times}(10^{-5}\;M)$ was not a direct stimulant of alveolar macrophages, as it had no stimulatory effect on chemiluminescence generation, but potentiated zymosan-induced activation of chemiluminescence, i.e., 162% above the separate effects of each stimulant. PAF $10^{-16}{\pm}10^{-6}\;M$ also failed to stimulate IL-1 production from alveolar macrophages. In contrast, when both PAF $10^{-10}\;M$ and lipopolysaccharide(LPS) $(1 {\mu}g/ml)$ were added together at the initiation of the culture, IL-1 production was significantly increased indicating the potentiative effects of PAF on IL-1 production by alveolar macrophages. Collectively, these data suggest that PAF alone does not activate the release of bioactive products from alveolar macrophages. However, PAF appears to act as a priming mediator that potentiates stimuli-induced macrophage activity. These novel actions of PAF prove its role as a potent mediator of inflammatory and immune responses in the lung.

  • The change of membrane fluidity in rockfish (Sebastes schlegeli) phagocytes during respiratory burst was investigated. Fluorescence polarization (FP) was used as a measure of membrane fluidity, and 1-(4-trimethylaminophenyl)-6-phenyl-1 .3 ,5-hexatriene (TMA.-DPH) was used us a fluorescent probe. The significantly higher FP values in phagocytes stimulated With zymosan or phurbol myristate acetate (PMA) than unstimulated control phagocytes suggests that membrane fluidity of phagocytcs is decreased during the respiratory burst. The faster decrease of FP value in PMA stimulated phagocytes than in zymosan sumulated phagocytes may be due to bypass of the receptor-mediated stages of functional modulation. which is needed in zymosan stimulated phagocytes.

  • Polyphenol QGR (quercetin 3-O-beta-(2"-galloyl)-rhamnopyranoside) was isolated from Persicaria lapathifolia (Polygonaceae). In this study, we investigated inhibitory effects of QGR and its building blocks (quercitrin, quercetin and gallic acid) on superoxide anion and NO productions in unopsonized zymosan-stimulated murine macrophages Raw264.7. QGR and quercetin showed potent inhibitory effects on unopsonized zymosan-induced superoxide anion production with IC50 values of 3.2 uM and 4.8 uM, respectively. (omitted)

  • This experiment is carried out for test whether the addition temperament drugs of Danchisoyo-san have an anti-inflammatory effect and have suppression effect on immunocyte in the state of inflammation which induced by carageenan and zymosan. The freezing lyophilization powder which were extracted from Danchisoyo-san divided low dose group(200mg/kg-DSL) and high dose group(500mg/kg-DSH) and after melting in water, it was orally administered to the mouse. Compared with inflammation induced group which were induced by triggering-inflammation reagent carageenan and zymosan and normal contrast group, we measured the edema decrement effect, macrophage and spleen cell activation. 1. Danchisoyo-san has suppress inflammatory reaction induced by carrageenan. 2. Danchisoyo-san has suppress increasing activation of abdominal cavity macrophage cell in the carrageenan induced inflammation. 3. Danchisoyo-san has suppress increasing activation of spleen cell in the carrageenan induced inflammation. 4. Danchisoyo-san has suppress increasing activation of abdominal cavity macrophage in the zymosan induced inflammation. 5. Danchisoyo-san has suppress increasing activation of spleen cell in the zymosan induced inflammation. Based on the above result, Danchisoyo-san was improved its suppression effect to the inflammatory reaction through the suppression of spleen cell and macrophage activation. So we concluded that Danchisoyo-san is prospected as a anti-inflammatory agent to cure inflammation.

Alteration of Biochemical Responses in Activated Human Neutrophils by ATP and Adenosine (활성화된 사람 중성 백혈구에서 ATP와 Adenosine 처리에 따른 생화학적 반응의 변경)

  • In both resting and opsonized zymosan activated neutrophils, ATP stimulated superoxide generation, whereas adenosine inhibited it slightly. The superoxide generation in activated neutrophils to ATP was greater than that of resting neutrophils. In $Ca^{++}$ free medium, inhibitory effect of adenosine on superoxide generation was detectable, whereas ATP did not have any effect. The stimulatory effect of ATP on superoxide generation was inhibited by adenosine in a dose dependent manner. Neither ATP nor adenosine had any effect on NADPH oxidase acitivity. Effects of ATP or adenosine on superoxide generation were more prominent than that by other triphosphate nucleotides or nucleosides. ATP and ADP further stimulated $Ca^{++}$ uptake and increased cytosolic free $Ca^{++}$ level in neutrophils activated by opsonized zymosan, but adenosine inhibited a $Ca^{++}$ mobilization. Verapamil effectively and tetrodotoxin slightly inhibited an increase of cytosolic free $Ca^{++}$ level induced by ATP. Inhibitory effect of either verapamil or tetrodotoxin on superoxide generation in the ATP plus opsonized zymosan-activated neutrophils was greater than in the cells activated by opsonized zymosan alone. Tetraethylammonium chloride had no apparent effect on superoxide generation. CCCP, 2,4-dinitrophenol, diphenylhydantoin and procaine all inhibited superoxide generation in neutrophils activated by opsonized zymosan. Among these, CCCP only inhibited a stimulatory effect of ATP. ATP further stimulated a loss of sulfhydryl groups in activated neutrophils, whereas adenosine had no effect on it. These results suggest that functional responses of neutrophils may be regulated at least partly by purines. ATP and adenosine may further after functional responses of activated neutrophils through their effect on $Ca^{++}$ uptake, membrane phosphorylation and oxidation of soluble sulfhydryl groups.

  • In the age of puberty or 20-30th young people who are sensitive to outward appearance, acne is serious problem at beauty and has social and psychological influence on that people. So this experiment is carried out for test whether the addition temperament drugs of Yungyopaedock-san(YP) have an anti-inflammatory effect and have suppression effect on immunocyte in the state of inflammation which induced by acne. The results was as follows. 1. YP has suppress inflammatory reaction induced by carageenan. 2. YP has suppress increasing activation of abdominal cavity macrophage in the carageen and zymosan induced inflammation. 3. YP has suppress increasing activation of spleen cell in the carageenan and zymosan induced inflammation. Based on the above result, YP was improved its suppression effect to the inflammatory reaction through the suppression of spleen cell and macrophage activation. So we concluded that YP is prospected as a anti-inflammatory agent to cure inflammation induced by ance.

  • Objective : Erysipelas is an acute inflammation caused by pyogenic bacteria. This mainly involves the upper part of dermis. It begins as erythematous patches with tenderness, followed by fever, headache, chills and fatigue etc. It may results in edema, obstruction of lymphatics and sepsis. So this experiment is carried out for test whether the Bojeasodok-um subtracted Scrophulariae Radix, Lasiosphaera seu Calvatia, Isatidis Radix added indigo Naturalis, Lithospermi Radix have an anti-inflammatory effect and have suppression effect on immunocyte in the state of inflammation which induced by Erysipelas. Method : Experimental animals made use of 4-5 week-age(weight 20-25g) ICR(male) mouse. In the breeding farm, the lighting time was controlled from 7:00 am till 7:00 pm, the temperature was controlled So we concluded that BS is prospected as an anti-inflammatory agent to cure inflammation induced bywithin 18-23$23^{\circ}C$ and water and food were not limited.The freezing lyophilization powder which were extracted from Bojesodok-Um divided low dose group(200mg/kg-BSL) and high dose group(500mg/kg-BSH) and after melting in water, it was orally administered to the mouse.
