Effects of GamiChungYi-tang on the Caerulein-induced Acute Pancreatitis in Rats (Caerulein으로 유발된 흰쥐의 급성 췌장염에 대한 가미청이탕(加味淸胰湯)의 효과)

  • In this study, we aimed to investigate the effect of GamiChungYi-tang(GCY-t) on caerulein-induced acute pancreatitis (AP). It is performed by detecting oxidative stress markers and observing histopathological examination. Thirty adult male rats(Sprague-Dawley) were divided into six groups as follows: normal (NOR,n=5), caerulein-induced (CON,n=5), caerulein+Cefotaxime Sodium(CT,n=5), caerulein+GCY-t (130 mg/kg, CHA,n=5), caerulein+GCY-t (260 mg/kg, CHB,n=5) and caerulein+GCY-t (520 mg/kg, CHC,n=5) groups. Pancreatic tissues of rats from all groups were removed for apoptosis and light, and electron microscopic examination. Blood of rats from all groups were collected for oxidative stress markers inspection and pathological examination. Pancreatic oxidative stress markers were evaluated by the measurements of leukocyte, serum amylase and platelet activating factor (PAF), Interleukin-6 (IL-6) levels were determined spectrophotometrically. CON group has a significant increase (p<0.05) in amylase compared with NOR, but CT and CHA, CHB, CHC groups reduced the levels of these enzyme. The levels of Platelet activating factor (PAF) were increased in CON compared with NOR, but decreased in CT and CHA, CHB, CHC groups compared with CON. Interleukin-6 (IL-6) levels were increased significantly in CON compared with NOR, but reduced in CT and CHA, CHB, CHC groups. In the observations of Optical microscopy and electron microscopy, The experimental groups showed the significant decreases in pancreatic tissue inflammation, edema, vacuolization, necrosis compared to the control group. After all, GCY-t is potentially capable of limiting pancreatic damage produced during AP by restoring the fine structure of acinar cells and tissue.

Effect of High Voltage Pulsed Current Stimulation on Surgical Wound of Rat (고전압맥동전류자극이 흰쥐 창상 치유에 미치는 영향)

  • The purpose of this study was to determine the effects of high voltage pulsed current (HVPC) stimulation on wound healing. Thirty-four Sprague-Dawley adult female rats were assigned to experimental and control groups. Each rats were anesthetized with pentotal sodium, and a 10 mm full-thickness incision was made on the back. From 24 hours after surgery, the rats of experimental groups were stimulated with HVPC, $140{\mu}s$, 120 pps, 30-40 V for 30 minutes, daily. The rats were sacrified 2 days, 4 days and 6 days after stimulation, respectively. The skin was removed, and processed for light microscopic examination. The length of incisional wounds were measured by microcaliper, and nucleolar organizer regions were counted under light microscope. For the histological observations the specimens were stained with Hematoxylin and Eosin, Masson Trichrome, Gomori Reticulum and Ag-NOR. There was a significant decrease in the length of incisional wound in experimental group compared with control group at 6 days HVPC stimuation (p<0.05). In experimental group, wound were significant difference (p<0.01) between the duration of post surgery. The mean numbers of nucleolar organizer regions per nucleus were significantly increased in the experimental group at 6 days HVPC stimulation (p<0.05), and were significantly difference (p<0.01) between the duration of post surgery in experimental group. Histological examination of the wound site suggested a more rapid of epithelialization and collagen formation between experimental groups compared with control groups. The result may indicated that the HVPC with $140{\mu}s$, 120 pps, 30-40 V for 30 minutes promoted surgical wound healing in the rat.

Effects of 915 MHz Radiofrequency Identification Electromagnetic Field Exposure on Neuronal Precursor Cells in the Dentate Gyrus of Adult Rat Brains

  • To explore the effects of radiofrequency electromagnetic field on the fate of neuronal cells, we investigated whether exposure to 915 MHz radiofrequency identification (RFID) caused morphological changes in neuronal cells in rat hippocampal dentate gyrus (DG). A reverberation chamber was used as a whole-body RFID exposure system. Rats were assigned to two groups: sham- and RFID-exposed groups. Rats in the RFID-exposed group were exposed to RFID at 4 W/kg specific absorption rate (SAR) for 8 hours daily, 5 days per week, for 2 weeks. Morphological evaluation of DG was performed using immunohistochemistry with doublecortin (DCX) as a neuronal precursor cell marker and neuronal nuclei (NeuN) as a mature neuronal cell marker. No significant morphological changes in DCX+ or NeuN+ cells in the DG of RFID-exposed rats were observed. These results suggest that RFID exposure induces no significant change in DCX+ neuronal precursor or NeuN+ neuronal cells in DG of rats.

Effect of Poncirus Trifoliata on Colonic Motility in Spinal Cord Injured Rats (척수손상 흰쥐에서 대장 운동에 대한 지실의 효과)

  • Objectives : The purpose of this study was to investigate the effect of Poncirus Trifoliata(PT) on improvement of fecal impaction in spinal cord injured(SCI) rats. Methods : Fifteen adult Sprague-Dawley female rats were used weighing 200~250 g. A complete spinal cord transection was performed surgically at the T10 cord level. Experimental groups were assigned into 3 groups: Control(n=5), SCI+vehicle(n=5) and SCI+PT(n=5). PT was administered 100mg/kg in 0.5ml every 24 hours from 1st operation day to 7th day. We measured the body weight and food intake as well as the number and the weight of fecal pellet every morning. After 1 week of operation, whole colon was divided into proximal and distal segments under anesthesia. Each segment of colon was mounted with longitudinal direction in a organ bath. We measured spontaneous contraction and compared the area under the curve in each segments. Enhanced responses were observed by acetylcholine($10^{-6}M$), 40 mM KCl solution, L-NAME($10^{-4}M$). Results : The fecal number and weights were significantly higher in the group of SCI+PT than SCI+vehicle group(p<0.05). In organ bath study, area under the curves of the spontaneous contraction in SCI+vehicle and SCI+PT groups were significantly increased compared to control group. Contractility of distal colon in response to acetylcholine or KCl in SCI+vehicle group was significantly decreased compared to other groups(p<0.05). Conclusions : These results suggest that PT might be useful to promote bowel emptying in spinal cord injured rats.

Effect of Dietary Fat and Genistein on Lipid Metabolism and Antioxidant Activity in Hyperlipidemic Male Rats induced High Fat Diet (고지방식이로 유도된 고지혈증 모델 흰쥐에서 지방과 제니스테인 섭취가 지질대사 및 항산화능에 미치는 영향)

  • This study was conducted to investigate whether dietary factors, normal fat and genistein leads to beneficial improvement of lipid metabolism and oxidative stress in adult hyperlipidemic male rats. Seven wk-old male SD rats were fed high fat diet (15% fat, 1% cholesterol) for 4 wks for induction of hyperlipidemic model rat. Weight-matched rats were then assigned to four groups according to dietary fat level (7% or 15% fat) and genistein contents (0 or 320 mg/kg diet). Food intake was significantly decreased by both high fat intake and genistein supplementation compared with normal fat intake and genistein no supplementaion. But weight gain was significantly decreased by genistein supplementation in normal fat intake compared with the other groups. Total lipid, total cholesterol and triglyceride in serum and liver were significantly decreased by normal fat intake compared with high fat intake. But total cholesterol in liver was significantly increased by genistein supplementation in both high fat and normal fat intake. TBARS in serum and liver was less produced by normal fat intake compared with high fat intake but TBARS in liver was significantly increased by genistein supplementation compared with genistein no supplementation in normal fat intake. Glutathione reductase activity in erythrocytes was significantly reduced by genistein supplementation in normal fat intake compared with the other groups. Glutathione peroxidase and glutathione reductase activities in liver were significantly inhibited by normal fat intake compared with high fat intake. Catalase activity in liver was significantly increased by genistein supplementation compared with genistein no supplementation in high fat intake. Nitrite was significantly decreased by normal fat intake compared with high fat intake. These results suggest that normal fat intake has the treatment effect against risk factors related with cardiovascular disease by reducing lipid profiles, lipid peroxidation. And genistein shows action as a antioxidant replacing antioxidant enzymes but also may act as prooxidant causing the production of TBARS.

The Differentiation of the Cholinergic Nerve Cells at the Meynert Basal Nucleus of the Basal Forebrains in the Growth Period of Rat (흰쥐 전뇌 기저부 Meynert기저핵에서 출생 후 발육기간에 따른 콜린성 신경세포의 분화)

  • This study was performed to investigate the distribution and differentiation on the immunoreacted cells of the ChAT (choline acetyltransferase) at the Meynert basal nucleus of the forebrains in the growth periods of rat, using the immunohistochemistric method. According to the cell shape and the ratio of long axis vs short axis of cell soma, the ChAT antibody reacted nerve cells in the Meynert basal nucleus of the rats were classified into six types. In the adult rats, the FD (frequency distributions) of round, oval and elongated cells were maximum. The FD of these types were shown to be progressively decreased during the postnatal development. In addition, the FD of elongated nerve cells in were observed in the adult rats respectively. This was thought to be the same phenomenon as those in the round and oval cells . The total mean volume of ChAT antibody reacted cell somata was lowest in the PND (postnatal days) 7 rats and was highest in the PND 21 rats. But, those were decreased to the adult. These results suggest that ChAT antibody reacted nerve cells grow up to PND 21 and then, differentiate into the various types by neurites outgrowing. On the electron micrography, the adult rat forebrain cells were obtained to be well developed ribosomes, polysomes , rough endoplasmic reticula and mitochondria. The immunreactivities were observed in ribosomes, polysomes, rough endoplasmic reticula and outer membrane of mitochondria. Golgi complexes were poorly developed and not showed jmmunreactivity. The ribosomes , polysomes and endoplasmic reticula are considered to be closely related to the inter cellular localization and biosynthesis of the ChAT but not Golgi complex. According to the results in the present study, it is considered that the ChAT-immunoreacted nerve cells in the Meynert basal nucleus of the rat forebrains are differentiated throughout the postnatal development with following processes of changes; 1) the cholinergic nerve cells develop postnatally 2) cell soma volumes gradually increase during the early postnatal days 3) and then, cells differentiate into the various types by projecting the neurites to the appropriate area after PND 21.

Effects of Flos Sophora japonica Extract on the Changes of Cerebral Hemodynamics

  • The purpose of this study was to measure the changes of regional cerebral blood flow(rCBF) and blood pressure(BP) in rats, following the intravenous injection of Flos Sophora japonica L.(FSJ) water extract. The measurement was continually monitored by Laser-Doppler Flowmeter and pressure transducer in anaesthetized adult Sprague-Dawley rats for 2 hours to 2 hours and a half through the data acquisition system. FSJ increased the changes of rCBF in rat significantly. The rCBF of FSJ did not change by pretreated propranolol, atropine, L-NNA($N^G$-nitro-L-argininine) and indomethacin. But the rCBF of Flos Sophora japonica L. was increased by pretreated methylene blue. FSJ decreased the changes of BP, significantly. The BP of FSJ did not change by pretreated propranolol, atropine, L-NNA and indomethacin. But the BP FSJ was decreased by pretreated methylene blue. There results indicated that FSJ can increase the rCBF and decrease the BP, that is related to guanylyl cyclase activity.

The Effect of Extract from Several Herbs Grown Naturally in Namwon Province on Wound Treatment (남원자생 허브의 혼합추출물이 창상치유에 미치는 영향)

  • Purpose: This study was aim to examine the effect of extract from several herbs on wound treatment which are grown naturally in Namwon province. Methods: Adult male Sprague-Dawley rats were assigned to 6 of 2 group: control group (C, n=6), experimental group (E, n=6). Rats in E group had 3 sessions (everyday) for 4 days or 8 days of the extract treatment which was composed of German chemomile, Teatree and Lavender (CTL). Results: CTL extract on wound-induced mice showed significantly the increase of wound treatment rate (WTR), epidermal gap (EP) and collagen density (CD), compared with control. Conclusion: CTL extract may be a putative resources for cure or treatment of wound.

p53 Protein Expression Area as a Molecular Penumbra of Focal Cerebral Infarction in Rats

  • Objective : The authors investigate the spatial characteristics of apoptotic genes expressed around the focal cerebral infarction, and attempted to explain the penumbra with them. Methods : A delayed focal cerebral infarction was created in twelve adult Sprague-Dawley rats. We performed the immunohistochemical staining for the apoptosis, bcl-2 and p53 proteins and measured the local cerebral blood flow [CBF] at the infarction core area and peri-infarct area pre- and intra-operatively. The peri-infarct area was divided into six sectors by distance from the infarction border. Results : The size [$mm^2$] of apoptosis, bcl-2, and p53 areas were $3.1{\pm}1.2$, $4.7{\pm}2.1$, and $6.8{\pm}2.4$, respectively. Apoptosis, bcl-2 or p53 positive cells were concentrated at the peri-infarct area adjacent to the infarction core. Their numbers reduced peripherally, which was inversely proportional to the local CBF. The p53 area seems to overlap with and larger than the ischemic penumbra. Conclusion : The p53 positive area provides a substitutive method defining the penumbra under the molecular base of knowledge.

The Change of Spinal Neuron Excitability by Cranial Electrostimulation(CES) in Rats (백서에서 두개전기자극에 의한 척수신경원 흥분성의 변화)

  • The purpose of this study was to determine the effect of spinal motor neuron excitability by cranial electrostimulation(CES). The fifteen Sparague-Dawley adult male rats were assigned to the three groups; GroupI(control), GroupII(low rate CES), GroupIII(high rate CES). Spinal motor neuron excitability was measured to use a computerized H reflex. The results of this study was as follows; M latency, M amplitude and H latency were no significant difference in all groups on repeated measured ANOVA(p>.05) but low rate CES and high rate CES groups were lower than ether group in comparative measurement of H amplitude and Hmax/Mmax ratio(p<.05). These results lead to the conclusion that spinal neuron excitability was influenced by CES. These results suggest that CES had the capability to lower spinal motor neuron excitability used synaptic blockade in spinal segment.
