The Effects of the low power intensity He-Ne IR laser on the change of the Serum Immunoglobulin components in burn rat (저 에너지 레이저 강도가 화상흰쥐의 혈청 면역글로불린의 성분 변화에 미치는 효과)

  • This studies were to investigate the effects of low power intensity Helium-Neon Infra-Red(He-Ne IR) laser on the changes of the serum immunoglobulin(1g) components in burn rats, The fourty eight Spraque-Dawley adult male rats were assigned to the 8 groups: the experimental groups(4), the burn control groups(2) and the control groups(2) There was made three degree burn by the 250mW IR on the back of each rats, from 3 days after being burned, the experimental laser groups were irradiated low power He-Ne IR laser for 3 minutes and 4.5 minutes every day during the 7days and 21 days. The results were as follows: The concentrations of immunoglobulin G(Ig G) in serum of burn rats on the treated with during the 3minutes laser for 7 days were higher than those of burn and 4.5 minutes laser groups. There were significantly decreased the change of the level of immunoglobulin M(Ig M) in serum of burn rats on the treated with 3 minutes laser for 7 days to the control group, but were significantly increased on the 3minutes and 4.5 minutes laser groups for 21 days to the those of burn and control groups, and the levels of the Ig M on the 3minutes laser group for 21 days were higher than those of 7 days. The concentrations of immunoglobulin E(Ig E) in serun of the burn rats on the experimental and burn groups were significantly lower than those in control group. As above results, The changes of the level of immonoglobulin components in serum have meaningful role in the healing process on the treated with the low pewer intensity of HE-Ne IR laser.

Effect of Salvianolic Acid B on Cardiac Muscle Damage Following Exhaustive Exercise in Rats (고강도 운동부하에 의한 흰쥐의 심장근육 손상에 대한 Salvianolic Acid B의 효능에 대한 연구)

  • Objectives: This study examined the effects of salvianolic acid B (SAB) on exhaustive exercise-induced cardiac muscle damage to rats. Methods: The study was carried out with 12-week-old young adult male Sprague-Dawley rats. Thirty-six rats were divided into 3 groups; normal (n=12), exhaustive exercise group (ExS, n=12) and exhaustive exercise with SAB (ExS+SAB, n=12). Five days before exhaustive exercise, SAB were medicated for 5 days in ExS+SAB group. Rats in ExS and ExS+SAB group were forced to swim for 150 minutes and then they were sacrificed, while rats in normal group were sacrificed at rest. After that, blood was collected and cardiac muscle tissue damage indices were analyzed. Results: Serum aspartate transaminase activity and lactate dehydrogenase activity were significantly lower in ExS+SAB group than in ExS group. Serum creatine phosphokinase activity of ExS+SAB was significantly lower than ExS group. However, the content of serum creatinine had no difference between ExS and ExS+SAB group. In the H&E stained left ventricle myocardium, ExS group showed signs of myocardial damage such as sporadic fragmentation of myocardial fibers, interstitial edema, cytoplasmic eosinophilia and neutrophils infiltration. However, ExS+SAB group alleviated the severity of the signs of myocardial damage. In the myocardial dihydroethidium staining, optical density was remarkably decreased in ExS+SAB group compared to ExS group. Furthermore, the up-regulation of Bax/Bcl-2 ratio was observed in ExS+SAB group compared with ExS group. Conclusions: The above results suggest that SAB may protect cardiac muscle damage via antioxidant activity and prevention of apoptosis.

Differentiation and Distrbution of the Choline Acetyltransferase-immunoreactive Nerve Cells in the Magnocellular Preoptic Nucleus of the Rat Forebrains during the Postnatal Development

  • This study was performed to investigate the differentiation and distribution of choline acetyltransferase (ChAT}-immunoreactive cells in the magnacellular preoptic nucleus (MCPO) of the postnatal and adult rat forebrains, utilizing techniques of immunocytochemistry. According to the cell shape and the ratio of long axis versus short axis of cell soma, the ChATimmunoreactive nerve cells in the MCPO were classified into six types: 1) round, 2) oval, 3) elongated, 4) fusiform, 5) triangular, and 6) polygonal types. Frequency distributions of the oval and round nerve cells on the postnatal day (PND) 0 were observed to be high. But in the adult, frequency distributions of the same cells were shown to decrease. Compared to those of the postnatal rats, frequency distributions of elongated, fusiform, triangular, and polygonal nerve cells in the adult were increased. The total mean volumes of ChAT-immunoreactive cell somata in the MCPO of PND 0 rat were the lowest, while those in the PND 17 rat were shown to be the highest and decreased in the adult. The soma volumes of the immunoreactive cells at the PND 17 were evenly distributed, but those in the other developmental stages (e.g. PND 7 and adult) appeared to exhibit unimodal distributions. On the electron micrography, the free ribosomes, polysomes, and rough endoplasmic reticula (RER) of the nerve cells in the MCPO of PND 21 rat forebrains were immunoreactive to ChAT in the tissues untreated with triton X-100. According to the observations in the present study, it is considered that the ChAT-immunoreactive nerve cells in the MCPO of the rat forebrains are differentiated throughout the following processes during the postnatal development: 1) increase in cell soma volumes, 2) development of neurites, 3) increase in the frequency of differentiated cell types, and 4) decrease in cell soma volumes. The ribosomes, polysomes, and RER are considered to be closely related to the intracellular localization and biosynthesis of the ChAT but not Golgi complex.

  • This studies to investigate the effects of ultrasound and swimming on the changes of the serum creatine kinase, latate dehydrogenase, aspatate aminotransferase, alanine aminotransferase, alkaline phosphatase, in peripheral nerve and muscle injury rats. The forty Sprague-Dawley adult male rats were assigned to the 4 groups: the experimental groups(3), and the peripheral nerve and muscle injury control group(1). There was made artificial injured by ischial nerve and muscle of each rats the each experimental ultrasound group and swimming group were treated from 3 days after being injuried for the 5 minutes and 10 minutes every day during the 10 days and 16 days respectively. The results were as follows: 1. The activities of creatine kinase, lactate dehydrogenase were significantly decreased on of the ultrasound and swimming groups for 10 days. All experimental group were significantly decreased for 16 days. there were significantly decreased the swimming group and ultrasound group were to the 10 days group. 2. The activities of asparatate aminotransferase, alalnine aminotransferase on the ultrasound group were significantly increased to the injured control group, there were significantly decreased the ultrasound group for 16 days group were to the 10 days group and there were significantly increased the swimming group for 16 days group were to the 10 days group. 3. The activities of alkaline phosphatase changes were no difference all experimental groups, there were significantly increased for 16 days group to the 10 days group. From these results it may be concluded that the effects of the ultrasound and swimming, the changes of the serum activities creatine kinase , lactate dehydrogenase of (; the objective indicates) to the muscle regeneration process of the nerve and muscle injured rats.

Effect of 6-Hydroxydopamine (6-OHDA) on the Expression of Testicular Steroidogenic Genes in Adult Rats

  • A neurotoxin, 6-hydroxydopamine (6-OHDA) has been widely used to create animal model for Parkinson's disease (PD). The present study was undertaken to examine whether depletion of brain dopamine (DA) stores with 6-OHDA can make alteration in the activities of the testicular steroidogenesis in adult rats. Young adult male rats (3 months old) were received a single dose of 6-OHDA (200 ${\mu}g$ in 10 ${\mu}{\ell}$/animal) by intracerebroventricular (icv) injection, and sacrificed after two weeks. The mRNA levels of steroidogenesis-related enzymes were measured by qRT-PCRs. Serum testosterone levels were measured by radioimmunoassay. Single icv infusion of 6-OHDA significantly decreased the mRNA levels of CYP11A1 (control:6-OHDA group=$1:0.68{\pm}0.14$ AU, p<0.05), CYP17 (control:6-OHDA group=$1:0.72{\pm}0.13$ AU, p<0.05). There were no changes in the mRNA levels of $3{\beta}$-HSD (control:6-OHDA group=$1:0.84{\pm}0.08$ AU) and $17{\beta}$-HSD (control: 6-OHDA group=$1:0.63{\pm}0.20$ AU), though the levels tended to be decreased in the 6-OHDA treated group. Administration of 6-OHDA decreased significantly the mRNA level of StAR when compared to the level of saline-injected control animals (control:6-OHDA group=$1:0.72{\pm}0.08$ AU, p<0.05). Treatment with single dose of 6-OHDA remarkably lowered serum testosterone levels compared to the levels of control group (control:6-OHDA group=$0.72{\pm}0.24:0.13{\pm}0.03ng/m{\ell}$, p<0.05). Taken together with our previous study, the present study demonstrated that the activities of hypothalamus-pituitary-testis hormonal axis could be negatively affected by blockade of brain DA biosynthesis, and suggested the reduced reproductive potential might be resulted in the animals. More precise information on the testicular steroidogenic activities in PD patients and PD-like animals should be required prior to the generalization of the sex steroid hormone therapy to meet the highest standards for safety and efficacy.

EFFECT OF PHENOBARBITAL AND / OR SKF 525-A ON THE METABOLISM AND ACUTE TOXICITY OF PARATHION IN ADULT FEMALE PATS (자성 흰쥐의 파라치온 급성독성 및 대사에 미치는 페노바르비탈 및 SKF-525-A의 영향)

  • Effects of altering hepatic mixed-function oxidase (MFO) enzyme activities on the metabolism and acute toxicity of parathio were investigated in adult female rats. In vitro hepatic metabolism of parathion to paraoxon was increased by phenobarbital pretreatment (50 mg/kg/day, ip, for 4 consecutive days) and SKF 525-A (50 mg/kg, ip, 1 hr prior to sacrifice) decreased paraoxon formation indicating that phenobarbital induces that form(s) of cytochrome P-450 catalyzing conversion of parathion to paraoxon. Degradation of paraoxon to p-nitrophenol was increased by phenobarbital pretreatment, but not affected by SKF 525-A suggesting that MFO activities play only a minor role in the detoxification of the active metabolite of this insecticide. The phenobarbital-induced increase in paraoxon formation was partially antagonized by SKF 525-A. Significant activity for both parathion activation and paraoxon degradation was also observed in the lung preparation, however, this extrahepatic parathion and paraoxon metabolizing activity was not induced by phenobarbital or inhibited by SKF 525-A pretreatment. Phenobarbital pretreatment increased paraoxon level in livers of rats when measured 3 hr following parathion injection (2 mg/kg, ip). SKF 525-A did not alter parathion or paraoxon levels in brain, blood and liver. Phenobarbital pretreatment decreased the toxicity of parathion (4mg/kg, ip) or paraoxon (1.5 mg/kg, ip) as determined by decreases in lethality and inhibition of brain and lung acetylcholinesterases. An additional SKF 525-A treatment failed to decrease the protective effects of phenobarbital against parathion or paraoxon toxicity. These results suggest that some unknown factors other than hepatic MFO induction are involved in the protective action of phenobarbital against parathion and paraoxon toxicity.

  • The average life expectancy has continuously increased with the development of medical industry. As women get older, they experience the deficit of estrogen caused by decreased function of ovarian. Specifically, they suffer from temporary fever because of unstable vasomotion and, in the long term experience, degeneration of urogenital organs, tooth loss, arteriosclerosis, demantia, snores and so on. Because of these symptoms the estrogen medical supply is getting required. Along with this, many studies have conducted to find out substitutional medicals without any side effects. This research was carried out to find out such substitutable materials. For the purpose of study, we examined the change of estrogen using the rats fed with the pupa powder immediately before the eclosion, eclosed adult, the pupa extract immediately before the eclosion, and the silkworm powder prepared at $3^{rd}$ day of $5^{th}$ instar. The result showed no significant chance in weight, feeding and water intake quantity among variables. However, the estrogen hormone secretion effects was very high at 73 pg/ml in the rats fed with the freeze-dried powder of the Yeonnokjam male pupa prepared immediately before the eclosion, compared with the control at 59 pg/ml. The next was Yangwonjam male adult at 71 pg/ml.

Serial Expression of Hypoxia Inducible Factor-$1{\alpha}$ and Neuronal Apoptosis in Hippocampus of Rats with Chronic Ischemic Brain

  • Objective : The purpose of this study is to investigate serial changes of hypoxia-inducible factor $1{\alpha}$ (HIF-$1{\alpha}$), as a key regulator of hypoxic ischemia, and apoptosis of hippocampus induced by bilateral carotid arteries occlusion (BCAO) in rats. Methods : Adult male Wistar rats were subjected to the permanent BCAO. The time points studied were 1, 2, 4, 8, and 12 weeks after occlusions, with n=6 animals subjected to BCAO, and n=2 to sham operation at each time point, and brains were fixed by intracardiac perfusion fixation with 4% neutral-buffered praraformaldehyde for brain section preparation. Immunohistochemistry (IHC), western blot and terminal uridine deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay were performed to evaluate HIF-$1{\alpha}$ expression and apoptosis. Results : In IHC and western blot, HIF-$1{\alpha}$ levels were found to reach the peak at the 2nd week in the hippocampus, while apoptotic neurons, in TUNEL assay, were maximal at the 4th week in the hippocampus, especially in the cornu ammonis 1 (CA1) region. HIF-$1{\alpha}$ levels and apoptosis were found to fluctuate during the time course. Conclusion : This study showed that BCAO induces acute ischemic responses for about 4 weeks then chronic ischemia in the hippocampus. These in vivo data are the first to show the temporal sequence of apoptosis and HIF-$1{\alpha}$ expression.

Effects of Soy Isoflavone Intake on Nitrite Content and Antioxidant Enzyme Activities in Male Rats Fed High-Fat Diet (고지방 섭취 흰쥐에서 대두 이소플라본 섭취가 혈액 내 Nitrite 함량과 항산화 효소 활성에 미치는 영향)

  • This study was performed to investigate the effect of soy isoflavone on plasma nitrite concentration and the antioxidant enzyme activities of erythrocyte and the liver using adult male rats fed high fat diet. Seven-week old male Sprague Dawley rats were divided into three groups and fed high fat diet (15% beef tallow, 1 % cholesterol; control: IF0) or high fat diets containing isoflavone 80 ppm (IF80) or 320 ppm (IF320) for 10 weeks. Plasma nitrite concentration as a vasodilator, and antioxidant enzyme activities in erythrocytes and the liver were measured. Plasma nitrite concentration was increased by 45% and 35%, respectively, in IF80 and IF320 than in IF0 group. Erythrocyte catalase, glutathione peroxidase (GPx) and glutathione reductase (GR) activities increased by 31 %, 30% and 40% in IF320 compared to IF0 group. Especially, erythrocyte GR activity increased by 61 % in IF80 group. However, catalase activity in the liver was decreased in IF80 group. GPx and GR activities in the liver were not differ among groups. The results suggest that soy isoflavone have the protective effect against risk factors related with cardiovascular disease by improving vasodilator factor, nitrite, and antioxidant enzyme activities in blood.
