The Isolation and Fusion of Pea and Barley Mesophyll Protoplasts (완두와 보리의 엽내세포 원형질체 분이 및 융합)

  • The optimal conditions for the protoplast isolation from the leaves of pea (Pisum sativum L. cv. Sparkle) and barley (Hordeum vulgare L. cv. Baecdong) were determined in order to achieve a somatic hybridization between two species. It was revealed that the use of 0.5M sorbitol as an osmoticum was appropriate for pea. The yield of intact protoplasts was the highest (40%) when pea leaves were incubated in the enzyme solution for 4 hours. In case of barley, the optimal concentrations of cellulase, pectinase and mannitol as the enzyme solution were 2%, 1% and 0.35M, respectively. And the yield of barley protoplasts was the highest(87%) when leaves were incubated in this enzyme solution for 3.5 hours. A fusion of protoplasts from pea and barley was induced by PEG treatment enriched with calcium salts within 60 minutes.

  • Polyethylene maleate (PEM) and epoxy resin cinnamate (BEC) were prepared to investigate their photosensitivity. Various samples coated on glass plate were exposed to light under various conditions and steeped in the same solvent as used for coating, and then the yield of residual film (W/$W_0$) was calculated. The yield (W/$W_0$), which was closely related to the sensitivity of the film, was affected by the degree of polymerization of the film, light source, sensitizers land their concentration. In polymer homologs, the sensitivity depended upon degree of polymerization. Most effective sensitizers for PEM and BEC among those used here were benzanthrone and 2,6-dichloro-4-nitroaniline.

  • In this paper, the result of study in artificial lighting uniformity ratio yield method subject to lecture room are reported. It is desirable that the application of equation ① U=Ei/Ex×100 cannot be used to the mean value in place to be effective of light and darkness. And it is desirable that the application of equation ② U=Ei/Ea×100 or equation ③ U=(Ea-Ei)/Ea×100 be used to the mean Value equality illumination conditions. Also, requiring preparation of judgement standard, uniformity ratio equation agree with actual circumstances in this country. It must be considered intensity of illumination and specificity of space in established standard.

Viscozyme L aided flavonoid extraction and identification of quercetin from Saururus chinensis (Lour.) Baill

  • In order to enhance the extraction efficiency of flavonoid from Saururus chinensis, carbohydrate-hydrolyzing enzyme Viscozyme L aided extraction techniques have been studied. Then flavonoid composition, as well as quercetin, were also identified using UV/Vis, HPLC/MS, and 1H-NMR. The results showed that favorable extraction conditions were Viscozyme L concentration of 0.25 mg/g, pH 4.2, reaction at 45 ℃ for 12 h. Under the favorable extraction condition, total flavonoid yield (37.9 mg/g) and quercetin yield (0.86 mg/g) increased by about 2.0 and 9.6 times, respectively, compared to control group. Interestingly, as a significant flavonoid of S. chinensis, flavonoid glycones rutin was hydrolyzed to aglycones quercetin by Viscozyme L. These findings provide scientific and theoretical support for the development quercetin-rich products, which was quickly absorbed by the human body than rutin.

A Study of Greenstuff Cultivation Using Enzyme Treated Nightsoil Liquid Manure (효소처리 분뇨액비를 이용한 작물재배의 실험적 연구)

  • The cultivtion efficiencies of enzyme treated nightsoil liquid manure were investigated. As a result of applying the liquid manure prepared by the nightsoil digestion due to aerobic or anoxic conditions with enzyme treatement to cultivate greenstuffs as compared with the controls (conventional cultivation) due to application of none enzyme treated fertilizer, the yield was markedly increased. In the cases of kidney bean, seedleaves were increased over 2~4 times and stem growth and diameter was increased 40% as compared the controls. Finally, cultivation yield of liquid manure prepared by the nightsoil aerobic or anoxic digestion with enzyme treatment were more increased as compared with the controls.

  • Superloading yield surface concept is newly introduced together with subloading yield surface conception in order to describe full gradation continuously of the mechanical behavior of soils from typical sand through intermediate soil to typical clay (All Soils). Finite deformation theory has been applied to the soil skeleton-pore water coupled continuum mechanics, which enables us to discuss things in a perpetual stream from stable state to unstable state like from deformation to failure and vice versa like from liquefaction to post liquefaction consolidation of sand (All States). Incremental form of the equation of motion has been employed in the continuum mechanics in order to incorporate a rate type constitutive equation, which is "All Round" enough to predict ground behavior under both static and dynamic conditions. The present paper is the shortened version of the lecture note delivered in 2008 Theoretical and Applied Mechanics Conference, Science Council Japan, but with newly developed application examples.

Reactive Ion Etching of a-Si for high yield and low process cost

  • In this paper, amorphous semiconductor and insulator thin film are etched using reactive ion etcher. At that time, we experiment in various RIE conditions (chamber pressure, gas flow rate, rf power, temperature) that have effects on quality of thin film. The using gases are $CF_4,\;CF_4+O_2,\;CCl_2F_2,\;CHF_3$ gases. The etching of a-Si:H thin film use $CF_4,\;CF_4+O_2$ gases and the etching of $a-SiO_2,\;a-SiN_x$ thin film use $CCl_2F_2,\;CHF_3$ gases. The $CCl_2F_2$ gas is particularly excellent because the selectivity of between a-Si:H thin film and $a-SiN_x$ thin film is 6:1. We made precise condition on dry etching with uniformity of 5%. If this dry etching condition is used, that process can acquire high yield and can cut down process cost.

Design, Synthesis and Catalytic Property of L-Proline Derivatives as Organocatalysts for Direct Aldol Reaction

  • A series of chiral prolinamide compounds with pyridine-2, 6-dicarboxylic acid moieties derived from L-proline have been designed and synthesized, their catalytic properties for direct asymmetric aldol reactions were also studied in this article. These catalysts gave the aldol product in high yield (87%) and high enantioselectivity, up to 85%, of the anti-structure at room temperature but gave disappointing results at a lower temperature or when additive was added. Conditions, including solvents, temperature and additives were screened for the reactions. Moreover, the influence of presence of water on yield and stereoselectivity was also discussed.

  • The behaviour of elasto-plastic planar arches subjected to dynamic loads in presented. The governing equations are formulated through the dynamic equations and compatibility conditions. The latter is established by applying the generalized conjugate segment analogy. Bending moments at the nodes and axial forces in the members are considered as primary variables in the elastic regime. They are supplemented by the rotations at the nodes and dislocations in the elements when plastic hinges occur. Newmark-${\beta}$ method is adopted in the time marching process. The interaction diagram of each element is treated as the yield surface for the element and the associated flow rule is enforced as plastic flow occurs. The method provides good prediction of dynamic response of elasto-plastic arches while requiring small core storage and short computer time.

Comparative studies of gasification potential of agro-waste with wood and their characterization

  • In this work, an experimental study of the gasification on wood was carried out in downdraft type fixed bed Gasifier attached with 10 kW duel fuel diesel engine. The main objective of the study was to use wood as the biomass fuel for downdraft Gasifier and to evaluate the operating parameter of gasifier unit to predict its performance in terms of gas yield and cold gas efficiency. The influence of different biomass on fuel consumption rate, gas yield and cold gas efficiency was studied. Composition of producer gas was also detected for measuring the lower heating value of producer gas to select the feed stock so that optimum performance in the existing gasifier unit can be achieved. Under the experimental conditions, Lower heating value, of producer gas, cold gas efficiency and gas yields, using wood as a feed stock, are $4.85MJ/m^3$, 46.57% and $0.519m^3/kg$.
