Reliable experimental data as a key factor for design of mechanical structures

  • The experimentally determined mechanical behavior of the material under the prescribed service conditions is the basis of advanced engineering optimum design. To allow experimental data on the behavior of the material considered, uniaxial stress tests were made. The aforementioned tests have enabled the determination of mechanical properties of material at different temperatures, then, the material's resistance to creep at various temperatures and stress levels, and finally, insight into the uniaxial high cyclic fatigue of the material under different applied stresses for prescribed stress ratio. Based on fatigue tests, using modified staircase method, fatigue limit was determined. All these data contributes the reliability of the use of material in mechanical structures. Data representing mechanical properties are shown in the form of engineering stress-strain diagrams; creep behavior is displayed in the form of creep curves while fatigue of the material is presented in the form of S-N (maximum applied stress versus number of the cycles to failure) curve. Material under consideration was 18CrNi8 (1.5920) steel. Ultimate tensile strength and yield strength at room temperature and at temperature of $600^{\circ}C$: [${\sigma}_{m,20/600}=(613/156)MPa$; ${\sigma}_{0.2,20/600}=(458/141)MPa$], as well as endurance (fatigue) limit at room temperature and stress ratio of R = -1 : (${\sigma}_{f,20,R=-1}=285.1MPa$).

Influence of column yielding on degree of consolidation of soft foundations improved by deep mixed columns

  • Laboratory and field data showed that deep mixed (DM) columns accelerated the rate of consolidation of the soft foundations. Most analyses of consolidation of DM column-improved foundations so far have been based on the elastic theory. In reality, the DM columns may yield due to the stress concentration from the soft soil and its limited strength. The influence of column yielding on the degree of consolidation of the soft foundation improved by DM columns has not been well investigated. A three-dimensional mechanically and hydraulically-coupled numerical method was adopted in this study to investigate the degree of consolidation of the DM column foundation considering column yielding. A unit cell model was used, in which the soil was modeled as a linearly elastic material. For a comparison purpose, the DM column was modeled as an elastic or elastic-plastic material. This study examined the aspects of stress transfer, settlement, and degree of consolidation of the foundations without or with the consideration of the yielding of the DM column. A parametric study was conducted to investigate the influence of the column yielding on the stress concentration ratio, settlement, and average degree of consolidation of the DM column foundation. The stress concentration ratio increased and then decreased to reach a constant value with the increase of the column modulus and time. A simplified method was proposed to calculate the maximum stress concentration ratios under undrained and drained conditions considering the column yielding. The simplified method based on a composite foundation concept could conservatively estimate the consolidation settlement. An increase of the column modulus, area replacement ratio, and/or column permeability increased the rate of consolidation.

Investigation of Ge2Sb2Te5 Etching Damage by Halogen Plasmas (할로겐 플라즈마에 의한 Ge2Sb2Te5 식각 데미지 연구)

  • Effect of Ge2Sb2Te5 (GST) chalcogen composition on plasma induced damage was investigated by using Ar ions and F radicals. Experiments were carried out with three different modes; the physical etching, the chemical etching, and the ion-enhanced chemical etching mode. For the physical etching by Ar ions, the sputtering yield was obtained according to ion bombarding energy and there was no change in GST composition ratio. In the plasma mode, the lowest etch rate was measured at the same applied power and there was also no plasma induced damage. In the ion-enhanced chemical etching conditions irradiated with high energy ions and F halogen radicals, the GST composition ratio was changed according to the density of F radicals, resulting in higher roughness of the etched surface. The change of GST composition ratio in halogen plasma is caused by the volatility difference of GST-halogen compounds with high energy ions over than the activation energy of surface reactions.

Study on the Effects of Supplemented Factors on the Production of Vitamin $B_{12}$ by Propionibacterium shermanii (Propionibacterium shermanii에 의한 비타민 $B_{12}$의 생성에 영향을 미치는 배지첨가물들에 대한 연구)

  • Following the study on the fermentation conditions influencing the production of vitamin $B_{12}$ by Propionibacterium shermanii(Korean J Biotechnol. Bioeng. 7,126-131, 1992), the effects of some factors supplemented in the medium on the production of vitamin $B_{12}$ were studied. Maximum production of vitamin $B_{12}$ was observed when $Co^{+2}$ was supplemented at the concentration of 2-4 ppm in the fermentation medium. Increase of the supplemented $Co^{+2}$ to 12 ppm did not inhibit the growth of the organism, but it accelerated the lysis of the organism. In the literature, peptone was reported to activate the biosynthesis of vitamin $B_{12}$. Examination of the effect of peptone on the growth and the production of vitamin $B_{12}$ showed that at early stage more vitamin$B_{12}$ was observed in the supplemented medium, but no difference was observed in the later stage of fermentation. Examination of the time for addition and the amount of 5,6-dimethylbenzimidazole, a precursor known to influence the production of vitamin $B_{12}$, showed that a maximum yield of vitamin $B_{12}$ was observed when 15 mg/L was added to the fermentation medium after 2 days' incubation. The effect was comparable with the increase of the production of vitamin $B_{12}$ when the fermentation condition was changed to aerobic condition after 2 days' culture under anaerobic condition.

Biosynthesis of trifolin, a bioactive flavonoid by biotransformation (생물전환으로 생리활성물질인 trifolin의 생합성)

  • Kaempferol 3-O-galactoside (Trifolin), a member of the flavonol group, has been reported to have anticancer effects against promyelocytic leukemia, histocytic lymphoma, skin melanoma and lung cancer. Trifolin has been extracted and used from several plants, but the extraction process is complicated and the final yield is low. Biotransformation is an alternative tool to produce high value-added chemicals from inexpensive compounds. To synthesis trifolin from naringenin, three genes (PeFLS and OsUGE-PhUGT) were introduced into Escherichia coli, respectively. In order to synthesis trifolin from naringenin, a co-culture fermentation system was established by optimizing the cell concentration, biotransformation temperature and medium, isopropyl-β-D-thiogalactoside (IPTG) concentration, substrate supply concentration, and recombinant protein induction time. The established optimal conditions for trifolin production were a 3:1 ratio of BL-UGTE to BL-FLS, induction of recombinant protein at 25 ℃ for 4 h after addition of 2.0 mM IPTG, biotransformation at 30 ℃, and supply of 300 μM naringenin. Through the optimized co-culture fermentation system, trifolin was biosynthesized up to 67.3 mg/L.

Immobilization of GH78 α-L-Rhamnosidase from Thermotoga petrophilea with High-Temperature-Resistant Magnetic Particles Fe3O4-SiO2-NH2-Cellu-ZIF8 and Its Application in the Production of Prunin Form Naringin

  • To efficiently recycle GH78 thermostable rhamnosidase (TpeRha) and easily separate it from the reaction mixture and furtherly improve the enzyme properties, the magnetic particle Fe3O4-SiO2-NH2-Cellu-ZIF8 (FSNcZ8) was prepared by modifying Fe3O4-NH2 with tetraethyl silicate (TEOS), microcrystalline cellulose and zinc nitrate hexahydrate. FSNcZ8 displayed better magnetic stability and higher-temperature stability than unmodified Fe3O4-NH2 (FN), and it was used to adsorb and immobilize TpeRha from Thermotoga petrophilea 13995. As for properties, FSNcZ8-TpeRha showed optimal reaction temperature and pH of 90℃ and 5.0, while its highest activity approached 714 U/g. In addition, FSNcZ8-TpeRha had better higher-temperature stability than FN. After incubation at 80℃ for 3 h, the residual enzyme activities of FSNcZ8-TpeRha, FN-TpeRha and free enzyme were 93.5%, 63.32%, and 62.77%, respectively. The organic solvent tolerance and the monosaccharides tolerance of FSNcZ8-TpeRha, compared with free TpeRha, were greatly improved. Using naringin (1 mmol/l) as the substrate, the optimal conversion conditions were as follows: FSNcZ8-TpeRha concentration was 6 U/ml; induction temperature was 80℃; the pH was 5.5; induction time was 30 min, and the yield of products was the same as free enzyme. After repeating the reaction 10 times, the conversion of naringin remained above 80%, showing great improvement of the catalytic efficiency and repeated utilization of the immobilized α-L-rhamnosidase.

Upcycling the Spent Mushroom Substrate of the Grey Oyster Mushroom Pleurotus pulmonarius as a Source of Lignocellulolytic Enzymes for Palm Oil Mill Effluent Hydrolysis

  • Mushroom cultivation along with the palm oil industry in Malaysia have contributed to large volumes of accumulated lignocellulosic residues that cause serious environmental pollution when these agroresidues are burned. In this study, we illustrated the utilization of lignocellulolytic enzymes from the spent mushroom substrate of Pleurotus pulmonarius for the hydrolysis of palm oil mill effluent (POME). The hydrolysate was used for the production of biohydrogen gas and enzyme assays were carried out to determine the productivities/activities of lignin peroxidase, laccase, xylanase, endoglucanase and β-glucosidase in spent mushroom substrate. Further, the enzyme cocktails were concentrated for the hydrolysis of POME. Central composite design of response surface methodology was performed to examine the effects of enzyme loading, incubation time and pH on the reducing sugar yield. Productivities of the enzymes for xylanase, laccase, endoglucanase, lignin peroxidase and β-glucosidase were 2.3, 4.1, 14.6, 214.1, and 915.4 U g-1, respectively. A maximum of 3.75 g/lof reducing sugar was obtained under optimized conditions of 15 h incubation time with 10% enzyme loading (v/v) at a pH of 4.8, which was consistent with the predicted reducing sugar concentration (3.76 g/l). The biohydrogen cumulative volume (302.78 ml H2.L-1 POME) and 83.52% biohydrogen gas were recorded using batch fermentation which indicated that the enzymes of spent mushroom substrate can be utilized for hydrolysis of POME.

Capsule Endoscopy in Refractory Diarrhea-Predominant Irritable Bowel Syndrome and Functional Abdominal Pain

  • Background/Aims: Capsule endoscopy is a diagnostic method for evaluating the small bowel lumen and can detect undiagnosed lesions. The aim of this study was to evaluate the diagnostic yield and clinical impact of capsule endoscopy in patients with refractory diarrhea-predominant irritable bowel syndrome and functional abdominal pain. Methods: This study involved a retrospective analysis of prospectively collected data, maintained in a database. Patients with refractory diarrhea-predominant irritable bowel syndrome and functional abdominal pain within the period of March 2012 to March 2014 were included. Capsule endoscopy was used to detect small bowel pathologies in both groups. Results: Sixty-five patients (53.8% female) fulfilled the inclusion criteria and had a mean (${\pm}$standard deviation) age of $50.9{\pm}15.9$ years. Clinically significant lesions were detected via capsule endoscopy in 32.5% of the patients in the abdominal pain group and 54.5% of the patients in the diarrhea group. Overall, 48% of patients had small bowel pathologies detected during the capsule endoscopy study. Inflammatory lesions and villous atrophy were the most frequent lesions identified in 16.9% and 15.3% of patients in the abdominal pain and the diarrhea groups, respectively. Conclusions: Routine use of capsule endoscopy in patients with irritable bowel syndrome should not be recommended. However, in patients with refractory conditions, capsule endoscopy may identify abnormalities.

A Preliminary Research of the Bifacial PV System Under Installation Conditions (설치환경 및 조건에 따른 양면수광형 태양광발전시스템의 기초 특성 연구)

  • Nowadays the bifacial PV system market and its applications are increasing rapidly. The performance of the bifacial PV system take advantage of its rear surface irradiance. Also, the ground albedo, PV module tilt and azimuth, PV module installation height, shading effect and module temperature are factors of bifacial PV system performance. This paper investigates how the performance of bifacial PV system is influenced by above factors. First, we analyzed the energy yield depending on PV module installation by simulation. Secondly, we compare energy performance evaluation of monofacial and bifacial module on different weather condition by experiment. Thirdly, we tested the albedo effect and checked operating characteristics using Dupont Tyvek material for the bifacial PV module. Fourthly, we check the shading effect of bifacial PV module on bypass diode operating. Finally, we applied the bifacial PV module in the nearby subway station for the noise reduction barrier using a qualified simulation program. In summary, we confirm that the energy performance superiority of the bifacial PV module has a lot of application use including road. Also, we have confirmed the bifacial module and inverter design should be considered by rear surface irradiance.

Effect of Water-impregnation on Steam Explosion of Pinus densiflora

  • This study addresses the biorefinery feedstock from Pinus densiflora. This raw material is a major tree species in the Republic of Korea; it is renewable, has cost-effective, and is readily available. In this study, steam explosion of P. densiflora was performed in a reactor at $225^{\circ}C$ and with 1 to 13 min reaction times with or without previous water impregnation. The combined severity factor (Ro), which is an expression relating the reaction temperature and reaction time used in the steam explosion treatment, ranged from 3.68 to 4.79. The influence of both impregnation and steam explosion conditions were investigated by examining color variations, chemical composition, and mass balance on the pretreated solids. The results showed that steam-exploded P. densiflora that was not impregnated with water exhibited significantly darker color (chroma 28.8-41.4) than water-impregnated and steam-exploded P. densiflora (chroma 18.8-37.3). The increased ${\alpha}$-cellulose and lignin contents were detected as the severity factor increased. Furthermore, the ${\alpha}$-cellulose and lignin contents in the non-impregnated/steam-exploded P. densiflora were higher than those in the water-impregnated/steam-exploded P. densiflora. However, the decreased holocellulose content was detected as the severity factor increased. In mass balance, the holocellulose yield from water-impregnated/steam-exploded P. densiflora was higher than that from the non-impregnated P. densiflora.
