Numerical Study of Turbulent Flow Around a Rotating Cylinder with Surface Roughness (표면에 계단이 부착된 회전하는 실린더 주위 난류유동의 전산해석)

  • Erosion-corrosion in a pipe system often occurs at fittings, valves, and weld beads where flow separation and reattachment yield high turbulence intensity. Thus identifying their correlations would be the first step towards resolving the erosion-corrosion problems associated with industrial applications. Bremhorst of the Univ. of Queensland, Australia, proposed that a rotating cylinder with surface roughness (two backward-facing steps periodically mounted on a circular cylinder) be an economical and tractable tool which can generate extreme flow conditions for erosion-corrosion study. In this work, DNS has been carried out for turbulent flows around the same rotating cylinder as his experimental apparatus. Our result shows that a region of intense turbulence intensity and high wall-shear stress fluctuation is formed along the cylinder surface in the recirculating region behind the step, where high mass-transfer capacity is also experimentally observed. Since corrosion is mass-transfer controlled, our finding sheds light on the direction of future corrosion research.

  • Stress distribution and interfacial debonding process at the interface between a rubber mold and a powder compact were analyzed during unloading under cold isostatic pressing. The Cap model proposed by Lee and Kim was used for densification behavior of powder based on the parameters involved in the yield function of general Cap model and volumetric strain evolution. Cohesive elements incorporating a bilinear cohesive zone model were also used to simulate interfacial debonding process. The Cap model and the cohesive zone model were implemented into a finite element program (ABAQUS). Densification behavior of powder was investigated under various interface conditions between a rubber mold and a powder compact during loading. The residual tensile stress at the interface was investigated for rubber molds with various elastic moduli under perfect bonding condition. The variations of the elastic energy density of a rubber mold and the maximum principal stress of a powder compact were calculated for several interfacial strengths at the interface during unloading.

  • A settlement prediction method based on shear wave velocity measurements and soil nonlinearity was recently developed and verified by means of centrifuge tests. However, the method was only applicable to heavily overconsolidated soil deposits under enlarged yield surfaces. In this study, the settlement evaluation method was refined to consider the stress history of the sublayer, based on an overconsolidation ratio evaluation technique, and thereby incorporate irrecoverable plastic deformation in the settlement calculation. A relationship between the small-strain shear modulus and overconsolidation ratio, which can be determined from laboratory tests, was adopted to describe the stress history of the subsurface. Based on the overconsolidation ratio determined, the value of an empirical coefficient that reflects the effect of plastic deformation over the elastic region is determined by comparing the overconsolidation ratio with the stress increment transmitted by the surface design load. The refined method that incorporate this empirical coefficient was successfully validated by means of centrifuge tests, even under normally consolidated loading conditions.

  • The production of maltose utilizing swollen extrusion starch seems to have many technical advantages, such as, high reaction rate and high yield, production of high purity concentrated maltose, and low energy consumption, over the conventional method utilizing liquefied starch. The characteristics of maltose formation in heterogeneous enzyme reaction system comtaining swollen extrusion starch was investigated using fungal $\alpha $-amylase. The influence of extrusion conditions on structure of extruded starch, such as, degree of gelatinization, water absorption index, and water solubility index was analyzed. The relationship between the structural features and maltose forming reaction was investigated, and the result was analyzed in terms of surface reaction of insoluble extruded swollen starch. The characteristics of maltose formation from swollen sxtrusion starch was compared using endo-type fungal $\alpha $-amylase and exo-type $\beta $anylase, and the structural trasformation of extruded starch was also observed to clarify the reaction mechanism.

  • Percentage of Bakanae infection of rice seedlings grown in nursery box was higher than rice seedlings grown in protected semi-irrigated nursery regardless of seed treatment. Rice seedlings grown in nursery box had higher Bakanae symptom ratio than that of the rice seedlings grown in protected semi-irrigated nursery in the paddy field conditions after transplanting. Grain yield of rice transplanted from nursery box was lower than that of the rice in protected semi-irrigated nursery.

  • In order to increase the production of glutathione by maximizing the expression of recombinant gsh plasmids, two genes responsible for the biosynthesis of glutathione were cloned. A gshI gene was cloned onto pBR322 plasmid as 3.6Kb PstI DNA fragment from E. coli K-12 chromosomal DNA. Also gshII gene was cloned onto pUC13 plasmid as 2.2Kb PstI-BamHI DNA fragment. In order to improve the glutathione producing activity more efficiently, various recombinant plasmids containing tandem repeated gshI genes or both genes in various copy number onto the same vector were constructed. E. coli cells harboring pGH501 plasmid (pUC8-gshI$\cdot$I$\cdot$II) showed the highest glutathione synthesizing activity. The conditions for glutathione production with an ATP-generating system such as acetate kinase reaction of E. coli cells or glycolytic pathway of yeast cells were examined using the E. coli cells harboring the pGH501 plasmid. When the acetate kinase reaction of E. coli cells was used as an ATP generating system, 20mM of L-csteine was converted into glutathione with a yield of $100\%$.

Improvement of Antibiotic-Producing Streptomyces (항생물질 생산 방선균의 역가 개량에 관하여)

  • An improved procedure for the rapid purification of glucose-6-phosphate dehydrogenase from extracts of Saccharomyces cerevisiae was developed by using affinity chromatography. Among six affinty media tested, NADP$^{+}$-agarose and Affi-gel Blue were more effective than others (i.e., Affi-gel Red, AMP-agarose, ATP-agarose, and NAD$^{+}$-agarose). Conditions to desorb the enzyme bound to the affinity media were examined to increase the purity as well as yield. The best result was obtained when the column was developed with a linear gradient of KCl (0-1.0M). In case of Affi-gel Blue, introduction of NAD$^{+}$ (15mM) washing step prior to the salt gradient was most effective to remove NAD$^{+}$-binding proteins. For a large scale preparation of G-6-P dehydrogenase higher recovery was obtained by Affi-gel Blue than NADP$^{+}$-agarose, however, the purity of the enzyme was decreased by 10 times if the former was used as the affinity medium. The capacity of Affi-gel Blue for G-6-P dehydrogenase was found to be 5 times higher than that of NADP$^{+}$-agarose. Furthermore Affi-gel Blue could be reused repeatedly and its preparation is relatively easier and less expensive than NADP$^{+}$-agarose.X> +/-agarose.

Selection of Protoplasts-Derived Cell Lines for High Yields Indole Alkaloids from Suspension Cultures of Vinca (Catharanthus roseus) (일일초(Catharanthus roseus) 현탁배양으로부터 원형질체유래 Indole Alkaloid 고생산성 세포주 선발)

  • To produce economically important indole alkaloids by cell cultures, we selected protoplastsderived clones (protoclones) of vinca (Catharanthus roseus) for high yields of catharanthine and ajmalicine. Protoplasts were enzymatically isolated from suspension-cultured cells. The highest plating efficiency (1%) was obtained when protoplasts were plated at a density of 1$\times$105 protoplasts/ml in a culture medium solidified with 0.4% Seaplaque agarose. The growth rates of 40 protoclones subcultured on a solid medium varied over a wide range. Protoclone VPC-6, which had the highest growth rate, was observed to produce relatively high yields of catharanthine and ajmalicine when cultured in a liquid medium. Although the original cell line did not produce catharanthine at a detectable level by HPLC, protoclone VPC-10 produced it at a level of 5.9$\mu\textrm{g}$/g fresh weight of cells for 10 days of culture. Under the same conditions, protoclone VPC-15 produced ajmalicine at a level of 133.6$\mu\textrm{g}$/g, of which productivity was improved about ,3 times than that of the original cell line. The results indicate that differences in the growth rate and indole alkaloid yield among the protoclones reflect the somaclonal variation in suspnsion-cultured cells.

참당귀 뿌리 배양에 있어서 root segmentation과 식물생장조절제가 뿌리 생장과 decursinol angelate 생산에 미치는 영향

  • Decursinol angelate, a new anticancer agent, was produced by root cultures of Angelica gigas Nakai. In addition, difference of specific yields between primary and secondary root was investigated. It was found that specific yield of secondary root was much higher than that of primary root at various conditions, so that it was thought that the formation and growth of secondary root were feasible. From this point of view, effects of root segmentation and plant growth regulators (NAA, IBA) on root morphology and decursinol angelate production were examined. Root segmentation increased secondary root mass and product formation. On the other hand, addition of NAA or IBA at various concentrations promoted secondary root formation and production of decursinol angelate significantly. Five-fold increase of production was obtained at 4 mg/L of IBA compared to control without NAA and IBA.

In-line Automatic Defect Repair System for TFT-LCD Production

  • An automated circuit repair system was developed for enhancing the yield of nondefective liquid crystal panels, focusing on the resist patterns on the circuit material layer of thin-film transistor (TFT) substrates prior to etching. The developed system has an advantage over the parallel conventional system: In the former, the repair conditions depend on the type of resist whereas in the latter, the repair parameters must be fine-tuned for each circuit material. The developed system consists of a resist pattern defect inspection system and a pattern repair system for short and open defects. The repair system performs fine corrections of abnormal areas of the resist pattern. The open-repair system is equipped with a syringe to dispense resist. To maintain a stable resist diameter, a thermal insulator was installed in the syringe system. As a result, the diameter of the dispensed resist became much more stable than when no thermal insulator was used. The resist diameter was kept within the target of $400{\pm}100{\mu}m$. Furthermore, a prototype system was constructed, and using a dummy pattern, it was confirmed that the system worked automatically and correctly.
