A fusion protein, consisting of a human epidermal growth factor as the recognition domain and human angiogenin as the toxin domain, can be used as a targeted therapeutic against breast cancer cells among others. The fusion protein was expressed as an inclusion body in recombinant E. coli, yet when the conventional solution-phase refolding process was used the refolding yield was very low due to severe aggregation, probably because of the opposite surface charge resulting from the vastly different pl values of each domain. Accordingly the solid-phase refolding process, which exploits the ionic interactions between a solid matrix and the protein, was tried, however the ionic binding yield was also very low regardless of the resins and pH conditions used. Therefore, to provide a higher affinity toward the solid matrix, six Iysine residues were tagged to the N-terminus of the hEGF domain. When cation exchange resins, such as heparin- or CM-Sepharose, were used as the matrix, the adsorption capacity increased 2.5~3-fold and the subsequent refolding yield increased nearly 15-fold compared to the conventional process. A similat result was also obtained when an Ni-NTA metal affinity resin was used.
Objective: Aim of the current comparative study was to evaluate production outputs, reproduction efficiency and functional traits in dual-purpose Fleckvieh and Braunvieh cows, reared under temperate European conditions. Methods: A data-set from 414 Fleckvieh and 42 Braunvieh cows and 799 lactations was analysed. ID tag number, milk yield per milking session, number of steps/interval and milk conductivity were recorded and collected daily using AfiMilk 3.076 A-DU software (Afimilk Ltd., Kibbutz, Israel). Production and milk quality data were taken from the results of the official performance recordings and the reproductive outputs of cows were recorded by the research stations veterinarians. Comparisons between the two genotypes were carried out using the one way analysis of variance protocol, with categorical factor being considered the breed of cows. All the statistical inferences were carried out using Statistica software (StatSoft Inc., Tulsa, OK, USA). Results: Fleckvieh cows significantly outperformed ($p{\leq}0.05$) the Braunvieh herd, with average milk yields of $5,252.1{\pm}35.79kg$ and $4,897.6{\pm}128.94kg$, respectively. Age at first calving was significantly ($p{\leq}0.01$) influenced by the breed, with Fleckvieh heifers being more precocious ($32.8{\pm}0.29mo$) compared to those of Braunvieh breed ($35.7{\pm}0.84mo$). Reproduction efficiency as defined by the number of inseminations per gestation, calving interval, dystocia, days dry and days open, was not influenced by genotype (p>0.05). Incidences of sub-clinical mastitis, clinical mastitis, lameness and abortions were not influenced by the breed factor (p>0.05). Stay-ability of cows was significantly ($p{\leq}0.001$) influenced by genotype, with Braunvieh cows having an average age at culling of $117.88{\pm}11.78$ months compared to $90.88{\pm}2.89$ months in Fleckvieh. Conclusion: Overall, results have shown that genotype significantly influenced milk yield, age at first calving and longevity.
Thirteen strains or crosses of Lentinus edodes obtained from home and abroad were compared for their mycelial characteristics on artificial media. Eleven of them were selected and inoculated on logs in order to study for their productivity, and the morphological characteristics of the mushrooms were investigated. 1) Seven strains, i. e., 11001, 12001, 13005, 13012, 13015, 12004 and 20101 were grown well on the sawdust medium. 2) The best mycelial growth and establishment of infection on logs were obtained from strains 20101 and 11001. 3) The highest yield of the mushrooms on logs during the six year periods were obtained from strains 11001, 12001 and 20101. 4) The highest yield of the mushrooms usually was obtained on the fourth year after inoculation. 5) The fruit body productivity in terms of dry weight recovery was the highest in strains 13005 and 20101. 6) The most productive period of the mushrooms during the years depended on strains. Most prevalent types of the strains were summer and autumn growing types, and more mushrooms were produced during these periods. 7) Morphological characteristics of the mushroom were more or less dependent on environmental conditions. 8) Thicker fruit bodies and relatively uniform shapes of the mushroom were produced by strain 20101.

We have carried out the experiment to investigate optimal drainage time of barley seeding synchronized with rice harvesting in paddy field. Number of emerged barley seedling on moist paddy field was less than that of common paddy field. It is very hard to plow using combine owing to long time to work on wet condition compared to drying condition of paddy field. According to the drainage time, growth and yield of barley were not significantly different. However, late drainage time decreased the spikes per $m^2$. Rice yield and 1,000 husked rice grain weight were not significant depending on drainage times at 15 days, 20 days and 25 days before barley seeding. The range of 1,000 husked rice grain weight was 21.7~22.2 g on different drainage conditions. In results, on considering of soil hardness and time of labor, the optimal contents of soil moisture was about 33%. This study reveals that drainage time at 20 days before barley seeding is proper to increase seedlings and yield.
The use of thin plate increases due to the need for light weight in large ship. Thin plate is easily distorted and has residual stress by welding heat. Therefore, the thin plate should be carefully joined to minimize the welding deformation which costs time and money for repair. For one effort to reduce welding deformation, it is very useful to predict welding deformation before welding execution. There are two methods to analyze welding deformation. One is simple linear analysis. The other is nonlinear analysis. The simple linear analysis is elastic analysis using the equivalent load method or inherent strain method from welding experiments. The nonlinear analysis is thermo-elastic analysis which gives consideration to the nonlinearity of material dependent on temperature and time, welding current, voltage, speed, sequence and constraint. In this study, the welding deformation is analyzed by using thermo-elastic method for PCTC(Pure Car and Truck Carrier) which carries cars and trucks. PCTC uses thin plates of 6mm thickness which is susceptible to welding heat. The analysis dimension is 19,200mm(length) * 13,825mm(width) * 376mm(height). MARC and MENTAT are used as pre and post processor and solver. The boundary conditions are based on the real situation in shipyard. The simulations contain convection and gravity. The material of the thin block is mild steel with $235N/mm^2$ yield strength. Its nonlinearity of conductivity, specific heat, Young's modulus and yield strength is applied in simulations. Welding is done in two pass. First pass lasts 2,100 second, then it rests for 900 second, then second pass lasts 2,100 second and then it rests for 20,000 second. The displacement at 0 sec is caused by its own weight. It is maximum 19mm at the free side. The welding line expands, shrinks during welding and finally experiences shrinkage. It results in angular distortion of thin block. Final maximum displacement, 17mm occurs around welding line. The maximum residual stress happens at the welding line, where the stress is above the yield strength. Also, the maximum equivalent plastic strain occurs at the welding line. The plastic strain of first pass is more than that of second pass. The flatness of plate in longitudinal direction is calculated in parallel with the direction of girder and compared with deformation standard of ${\pm}15mm$. Calculated value is within the standard range. The flatness of plate in transverse direction is calculated in perpendicular to the direction of girder and compared with deformation standard of ${\pm}6mm$. It satisfies the standard. Buckle of plate is calculated between each longitudinal and compared with the deformation standard. All buckle value is within the standard range of ${\pm}6mm$.

This study was focused on the comparison of the variations in the yield and chemical composition of Chamaecyparis obtusa leaf oil obtained under different pressure conditions of the supercritical carbon dioxide extraction (SCE), and by hydro-distillation. SCE was carried out varying the pressure in the range of 100~400 bar at $40^{\circ}C$. The chemical composition of C. obtusa leaf oils was determined by gas chromatography-mass spectroscopy (GC-MS) analysis. The maximum yield of 4.4% (relative to the initial mass of oven dry mass) was obtained in the extraction under 300 bar pressure, which was higher than that of the hydro-distillation method (1.9%). The contents of sesquiterpenes in the extracts obtained by the SCE were higher than those of the essential oils of C. obtusa by the hydro-distillation. The sesquiterpenes in the SCE extracts made up approximately 39%~46% of the total, followed by monoterpenes, diterpene, and lignan. The contents of each constituent in the supercritical carbon dioxide extracts were varied on the extraction pressure. Therefore, these results showed that the extraction condition of SCE had significant effect on the yield of C. obtusa oils and its chemical composition.

We examined the effects of light colors (wavelength) and light quantities on the yield ratio and quality of cultivated cowpea sprouts. All light colors resulted in a lower cowpea sprout yield ratio compared to the untreated condition (darkness), but were similar in hard seed ratio. All light colors promoted the growth of the epicotyl and root when compared to the untreated condition, but limited the growth of the hypocotyl. White light (458 nm) significantly improved grade by increasing the lightness of the cotyledon and the hypocotyl and the yellowness of the hypocotyl. The Fe content of cowpea sprouts was higher in those grown under red light (632 nm), and the total amino acid content was higher for those grown under yellow light (560 nm), white light (458 nm), and blue light (460 nm) compared to plants grown in the untreated condition (darkness). The yield ratio of cowpea sprouts was lower in the yellow light condition (560 nm) at lower light quantity, but no differences were observed at other light colors and quantities. The lightness and yellowness of cowpea sprouts was higher in the yellow light (560 nm) and red light (632 nm) at lower light quantity, redness was lower. No significant differences were observed in the content of normal and inorganic components according to the light quantities of each light color, except that Fe content was higher in sprouts grown under red light (632 nm) as light quantity increased. Total amino acid content was slightly higher in sprouts grown under white light (458 nm) and blue light (460 nm) as light quantity increased.
Enzymatic synthesis of fructose-based sugar fatty acid esters, such as methyl fructoside oleic acid mono and diester, was investigated using methyl fructoside as a sugar starting material. For the production of methyl fructoside fatty acid monoester by solvent process, 2-methyl 2-propanol was found to be a god reaction medium resulting a higher yield and productivity due to its high sugar solubility. The yield and productivity of methyl fructoside oleic acid monoester were 70% and 12.6g/L-hr, respectively, when molar ratio of methyl fructoside, initial concentration of methyl fructoside, enzyme(Novozym 435) content, and reaction temperature were 3:1, 200g/L, 1%(w/v), and $60^{\circ}C$, respectively. Methyl fructoside oleic acid diester was prepared by lipase-catalyzed diacylation of methyl fructoside and oleic acid in the solvent-free process. Maximum yield of 98% and productivity of 140g/L-hr were achieved when molar ratio(methyl fructoside and oleic acid) of 1:2 enzyme content of 10%(w/v) and reaction temperature of $70^{\circ}C$ were applied for the operating conditions under a reduced pressure of 20∼200 mmHg.
This experiment was conducted to investigate the optimum nitrogen fertilization conditions for the production of high-quality Rhodiola rosea L. in forcing culture. Up until 48 kg N/10 a of both urea and ammonium sulfate, dry matter yield of root of Rhodiola rosea L. tended to increase with increase in application rates, however, it decreased thereafter in higher application rates. In the case of urea, the content of salidroside in the root of the Rhodiola rosea L. appeared to decrease rapidly from the application rates of 64 kg N/10 a and over. Meanwhile, the content of salidroside in the root tended to decrease gradually with the application rates exceeding 64 kg N/10 a of ammonium sulfate. The optimum fertilization rates of urea and ammonium sulfate was 45-8-20-10-10 (N-P-K-Ca-Mg) kg/10 a according to the curvilinear regression equation. However, considering the nitrogen accumulation in soil, nitrogen translocation into the plant, and dry matter yield and content of salidroside in the root of Rhodiola rosea L., the optimum fertilization rates of urea and ammonium sulfate would be 40-8-20-10-10 kg/10 a and 35-8-20-10-10 kg/10 a, respectively.
Effects of several different petroleum fractions (LGO, HGO, VGO, Diesel oil, SP(E), HGO-wax, L/M-wax), stepwise addition of calculated amounts of HGO at defined intervals, recycling of spent media on cell growth of Candida tropicalis KIST 351 were studied using $2.5{\ell}$ fermenter by batch process. In addition, continuous cultivation of the yeast was also performed in the light of biomass production using $28{\ell}$ fermenter with LGO. 1) Cell concentration, yield on the basis of gas oil and n-paraffin with the petroleum fractions were in the range of $11{\sim}15g/{\ell}$, $10{\sim}12%$ and $77{\sim}82%$, respectively. 2) By stepwise addition of the gas oil, cell concentration and yield on the oil were increased up to 18.9 g/land 13%, respectively. 3) Spent medium slowed emulsifying ability of hydrocarbon and stimulating effect on the cell growth. Without additional supplementation of $Mg^{++}$ up to 20% of spent medium could be reused, while by adding of the $Mg^{++}$, 50% of medium could be recycled. 4) Optimum condition of continuous cultivation for biomass production was attained at the dilution rate of $D=0.1{\sim}0.125\;hr^{-1}$. Maximum yield coefficient on consumed n-paraffin was 0.94 at $D=0.1\;hr^{-1}$, however, 24% of supplied n-paraffin in the media was not utilized at this dilution rate.
