This experiment of which aim contribute to plan irrigation system so as to increase forage crop yields, was conducted to estimate evapotranspiration amount of forage crops and to find out system of consumed water in a pasture-ground. The results obtained by this study are as follows: 1. The general weather conditions which, were closely related to the evapotrannpiration of forage crops were nearly same as those of the average year with the exception that temperature of May and June were slightly low. 2. According to the investigation of potential evapotranspirations (P.E) or forage crops and its changes during growing periods, changes of tenday P.E. were high significant according to the harvesting period. P.E of Alfalfa of which yield was the largest was the biggest. Althrough the correlations between P.E. and meteorological factors were irregular oming to three-time harvesting, correlation between ten-day evapotraspiration amount and copper plated pan evaporation or solar radiation was high positive significant. 4. Predicting formulas of P.E. were led by weather factors, and also relatione between P.E. and weather factors were showed as figure. from the these formulas, P.E. may be calculated by weather factors. 5. Predicting formulas of P.E. were led by mean temperature and copper plated pan evaporation, and by mean temperature and solar radiation. As computed values and measured values showed in figure, these formulas were high signiflent. 6. In the total consumed soil water duration of 10 days which, was non-rain period from 12th to 21th of August, Alfalfa was the largest 48.1mm, second, Orchard grass 40.1mm and Fescue 37.6mm, and Ladino clover was the smallest 37.1mm, also, order of each forage crop yield amound. was same to the abov. Order of soil moisture extraction rate of soil layer of all the for forage crops dulation of ten-day was soil layer 1 which was largest, soil layer 2, 3, and 4 Reviewing the the first five-day and the second five day, in the first five-day, order of that of all the forage crops was same to the above, but in the second five-day, that of soil layer 2 or 3 was more than the of soil layer 1.

Most of agricultural structures are consisted of complex components and exposed to various boundary conditions. There have been no ways to express those structures exactly for model experiment. As an alternative, 3D printer can produce any type of solid model. However, there are limited informations related to structural experiments using 3D printer. The object of this study gives the basic informations to structural engineers who try to use 3D printer for model experiment. When PLA was used as a supplier for 3D printer, the outcomes showed less heat deformation to compare with ABS. To test the material properties, two kinds of experiments (three-point flexibility test and compression test) were executed using universal testing machine. In three-point flexibility test, plastic hinge and its deformation were developed as observed in material such as steel. The behavior was in a linear elastic state, and elastic bending modulus and yield force were evaluated. In the compression test using unbraced columns with hinge-hinge boundary condition, the constant yield forces were observed regardless of different lengths in all columns with same section size, whereas the compressive elastic modulus was increased as the length of column was increased. The suggested results can be used for model experiments of various agricultural structures consisted of single material.
Native crystalline chitin was hydrolyzed in an agitated bead reaction system using crude chitinase excreted from Aspergillus fumigatus JC-19. The reaction was enhanced significantly, and the concentration and yield of reducing sugar after 48 hours were measured to be 35.42 g/I (w/v) and 0.64, respectively, around 1.86 times higher than those of the conventional system that was carried out without glass beads. The effect of reaction conditions, such as the amounts of chitin, chitinase and glass beads, and the size of glass bead, were examined. Ball milled chitin was also hydrolyzed in the agitated bead reaction system, the conversion yield and reaction rate of ball milled chitin for 24 hours increased up to 0.87 and 48.02 g/I, respectively. Chitinase showed relatively high stability in the agitated bead reaction system, particularly in the presence of enzyme stabilizer, $Ca^{++}$, which played a critical role in preventing the deactivation of chitinase by the physical impact of glass beads. The variations of the structural features of chitin during the reaction were followed by SEM and X-ray diffraction, and the enhanced hydrolysis reaction was caused by both the fragmentation of chitin particles and the destruction of the crystalline structure owing to the synergic effects of the attrition of glass beads and the hydrolytic action of chitinase.

Six nonpathogenic fungal strains isolated from alkaline soils of Buenos Aires Province, Argentina (Acremonium murorum, Aspergillus sidowii, Cladosporium cladosporoides, Neurospora tetrasperma, Purpureocillium lilacinum (formerly Paecilomyces lilacinus), and Westerdikella dispersa) were tested for their ability to produce keratinolytic enzymes. Strains were grown on feather meal agar as well as in solid-state and submerged cultures, using a basal mineral medium and "hair waste" as sole sources of carbon and nitrogen. All the tested fungi grew on feather meal agar, but only three of them were capable of hydrolyzing keratin, producing clear zones. Among these strains, P. lilacinum produced the highest proteolytic and keratinolytic activities, both in solid-state and submerged fermentations. The medium composition and culture conditions for the keratinases production by P. lilacinum were optimized. Addition of glucose (5 g/l) and yeast extract (2.23 g/l) to the basal hair medium increased keratinases production. The optimum temperature and initial pH for the enzyme production were $28^{\circ}C$ and 6.0, respectively. A beneficial effect was observed when the original concentration of four metal ions, present in the basal mineral medium, was reduced up to 1:10. The maximum yield of the enzyme was 15.96 $U_c/ml$ in the optimal hair medium; this value was about 6.5-fold higher than the yield in the basal hair medium. These results suggest that keratinases from P. lilacinum can be useful for biotechnological purposes such as biodegradation (or bioconversion) of hair waste, leading to a reduction of the environmental pollution caused by leather technology with the concomitant production of proteolytic enzymes and protein hydrolyzates.
Objectives SJ004 is a natural herbal medicine that contains Acyranthes japonica Nakai and Eucommia ulmoides Oliver traditionally used for joint and spinal diseases. This study aimed to establish an efficient method of extracting SJ004 to standardize using the yield, high-performance liquid chromatography (HPLC), and antioxidant assay. Methods SJ004 was extracted with distilled water, 70% and 100% of ethyl alcohol (EtOH). The method validation of 20-hydroxyecdysone and pinoresinol diglucoside was determined by HPLC-photo diode array and the content of SJ004 was calculated. The antioxidant activity of each extract was compared and measured using total flavonoids, total phenolic compounds, 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid), and ferric reducing antioxidant power according to the standard protocol. Results The yield was highest in pure water extract and lowest in 100% EtOH. But, the content of marker compounds indicating 20-hydroxyecdysone and pinoresinol diglucoside was highest in 100% EtOH extract. In the physiological activity measurement using antioxidant activity, 100% ethanol extract was highest. The limit of detection indicating 20-hydroxyecdysone and pinoresinol diglucoside were analyzed 0.33 ㎍/mL, 0.1616 ㎍/mL, and the limit of quantification were analyzed 1.01 ㎍/mL and 0.49 ㎍/mL respectively. Conclusions The experimental results showed that the extraction conditions have a significant effect on content of marker compounds and antioxidant activity. As a result of method validation, SJ004 was standardized by 20-hydroxyecdysone and pinoresinol diglucoside.
One of the characteristics of polycrystalline graphene that determines its material properties is grain size. Mechanical properties such as Young's modulus, yield strain and tensile strength depend on the grain size and show a reverse Hall-Petch effect at small grain size limit for some properties under certain conditions. While there is agreement on the grain size effect for Young's modulus and yield strain, certain MD simulations have led to disagreement for tensile strength. Song et al. showed a decreasing behavior for tensile strength, that is, a pseudo Hall-Petch effect for the small grain size domain up to 5 nm. On the other hand, Sha et al. showed an increasing behavior, a reverse Hall-Petch effect, for grain size domain up to 10 nm. Mortazavi et al. also showed results similar to those of Sha et al. We suspect that the main difference of these two inconsistent results is due to the different modeling. The modeling of polycrystalline graphene with regular size and (hexagonal) shape shows the pseudo Hall-Petch effect, while the modeling with random size and shape shows the reverse Hall-Petch effect. Therefore, this study is conducted to confirm that different modeling is the main reason for the different behavior of tensile strength of the polycrystalline structures. We conducted MD simulations with models derived from the Voronoi tessellation for two types of grain size distributions. One type is grains of relatively similar sizes; the other is grains of random sizes. We found that the pseudo Hall-Petch effect and the reverse Hall-Petch effect of tensile strength were consistently shown for the two different models. We suspect that this result comes from the different crack paths, which are related to the grain patterns in the models.

Optimum processing conditions for vinegar fermentation using fig, pear and persimmon were determined. Alcohol contents in the fermentatio broth of crushed fruits of fig, pear and persimmon were 7.5%, 5.1% and 6.8%, respectively. Alcohol contents increased up to 14.3~15.1% by adding 24% of sugar to the fruit juices. The total acidity of 7.04%, 3.30% and 3.66% were obtained for fig, pear and persimmon, respectively, through acetic acid fermentation of fruit juices containing 8% ethanol. Acetic acid yield increased by shaking during fermentation for pear and persimmon broth. Acetic acid yield increased 1.80~1.92 times by adding 0.5% of yeast extract to the fermentation broth of pear and persimmon. After fermentation, each fruit vinegar was clarified up to 93.1~97.4 of light transmittance by using 0.6% of kaki shibu for 4 days at 1$0^{\circ}C$. After aging for 60 days at 1$0^{\circ}C$, the acidity of fruit vinegar decreased slightly. Tannin content of persimmon vineger was remarkably higher than the other, while light absorbance of pear vinegar was higher than the other vinegars. Acetic acid was identified as the main volatile organic acid in the fruit vinegars, while propionic, isobutyric and isovaleric acids were identified as the minors. The content of non-volatile organic acids in the pear vinegar was higher than that in the persimmon vinegar. Sensory evaluation results indicate that the fig vinegar was preferred to the pear vinegar in the aspects of color, flavor and overall acceptability, but the fig vinegar had a strong background taste. Sensory scores of the persimmon vinegar increased significantly by pasteurization, but those of the fig and pear vinegars did not by pasteurization.
Rice straw is the main grain straw and is produced in large quantities every year in Korea. Pretreatment of lignocellulosic biomass using soaking process was carried out mild conditions at atmospheric pressure and temperature of $60^{\circ}C$. We found enzymatic hydrolysis condition of pretreated biomass. In case of a rice straw, compared with previous lignocellulosic biomass, we found that hydrolysis time was a shorter than others. Hydrolysis of SAA-treated rice straw has shown conversion rate was higher at $50^{\circ}C$. Hydrolysis was ended between 40~48 hour. Glucose conversion rate was higher when enzyme loading is 65 FPU/ml and 32 CbU/ml. When substrate concentration was 5%(w/v), it was that conversion rate was 83.8% after hydrolysis for 72 hr. In simultaneous saccharification and fermentation(SSF) experiment about SAA-treated rice straw, ethanol productive yield was highest from $40^{\circ}C$. The yield of that time was 33.05% from 48 hour.
Aureobasidium pullulans produced high concentration of polyols extracellularly in the media of sucrose, glucose and mannose as sole carbon source. Mannitol was the main polyol produced during the late exponential and stationary phases of growth together with small quantities of glycerol. Sucrose and glucose were rather rapidly metabolized to mannitol among carbon sources examined where the initial glucose concentration showed no difference in the amount of mannitol. In contrast 20%(w/v) of sucrose was the most appropriate concentration tested. However, the yield of mannitol based on substrate used($Y_{p/s}$) was independent on the initial concentration, and the mean value of mannitol yield in 10% glucose and sucrose media was 0.144 and 0.188, respectively. Mannitol production was reduced in response to an elevated water stress imposed by salts within the range from 0.25 to IM of NaCl or KCl as stress solutes. However, glycerol contents and its ratio to mannitol were increased at the conditions of high salinity. Based on the results, extracellular mannitol produced by A. pullulans probably resulted partly from osmoregulation(in case of glycerol) and mainly from, as known to occur in most of fungi, enzymatic reduction of the corresponding hexoses through phosphate pathway.
The Simultaneous Saccharification and Fermentation(SSF) of ethanol production from potato starch studied with respect to growth pH, temperature, substrate concentration. The glucoamylase and Saccharomyceses cerevisiae have a capacity to carry out a single stage SSF process for ethanol production. The characteristics, termed as starch hydrolysis, accumulation of glucose, ethanol production and biomass formation, were affected with variation in pH, temperature and starch concentration. The maximum ethanol concentration of 12.9g/l was obtained using a starch concentration 30g/l, which represent an ethanol yield of 86%. The optimum conditions for the maximum ethanol yield were found to be a temperature of 38, pH of 4.0 and fermentation time of 18hr. Thus by using the control composite design, it is possible to determine the accurate values of the fermentation parameters where maximum production of ethanol occurs.
