  • Genetic transformation of streptomyces gaespitosus by plasmid plJ 702 was camied out. Optimal conditions for the protoplast preparation of streptomyces casepitosus, its regeneration, and its transformation by plJ 702 were evaluated. Addition of 2% glycine to the culture broth was optimal for protoplast yield. Formation and regeneration of protoplasts were most efficient when the mycelium were harvested at between late log and stationary growth phase. The regeneration frequency of the protoplasts was 15% when the protoplats were regenerated on R2YE agar media containing 0.5M sucrose. Under the best condition for protoplats (M.W. 4,000) treatment for 2 minutes.

Studies on Protoplast Isolation and Regeneration of Lyophyllum ulmarium (느티만가닥 버섯의 원형질체 분리와 재생에 관한 연구)

  • This experiment was undertaken to investigate proper conditions for protoplast isolation and regeneration from the mycelia of Lyophyllum ulmnrium. Protoplast isolation and regeneration are influenced by a variety of factors such as enzyme, osmotic stabilizer, reaction time and age of mycelia. A combination of Novozyme 234 (10mg/ml) and cellulase Onozuka R-10 (10 mg/ml) with 0.6 M $MgSO_4$ was most effective for isolation of the protoplasts. The optimum reaction time of the mycelia with the lytic enzymes was 3.5~4 hours at $28^{\circ}C$ in shaking condition at 120 strokes per min. High yield of the protoplasts were obtained from its 4~5 days old mycelia on complete agar media. Its protoplasts were regenerated to normal hyphae. Regeneration media with 0.6 M sucrose were proper for regeneration of the protoplasts. Their regeneration frequency on complete agar media was 2.3~2.7%.

Utilization of Cyclodextrin in Biotransformation by Digitalis lanata Cell Cultures (Digitalis lanata 세포배양에 의한 생물학적 변환에서의 cyclodextrin의 이용)

  • Addition of cyclodextrin in the biotransformation of digitoxin into digoxin by Digitalis lanata cell suspension cultures enhanced the conversion yield. Presence of cyclodextrin also supported good stability of the intermediate product, digoxin, for long time. Among several kinds of cyclodextrins, ${\beta}$-cyclodextrin provided the best results. It was found that the optimum form of cyclodextrin utilization was the external addition of iclusion complexes between digitoxin and ${\beta}$-cyclodextrin at 1: 2 molar ratio from the beginning of biotransformation. With the optimized conditions, addition of ${\beta}$-cyclodextrin enhanced the production of digoxin up to 1.55 fold. In this case, not only digitoxin consumption was increased, but also the production of by-product was reduced.

  • The Chitin Thiocarbonate-Fe(II)-H2O2 redox initiator system was investigated for the graft copolymerization of acrylonitrile(AN) and acrylic acid(AA) monomers onto chitin powder. The reactions with vinyl monomers onto chitin were carried out under various the graft copolymerization conditions to elucidate the polymerization behavior in terms of graft yield. Reactions of chitin-acrylonitrile graft copolymer with hydroxyl amine hydrochloride and those with sodium hydroxide were conducted in order to obtain chitin-(amidoxime-co-acrylonitrile) and chitin-(acrylate-co-acrylamide) graft copolymers, respectively. The reaction efficiency was observed to depend on the alkali concentration, time, temperature, and the reactant concentrations. The prepared chitin derivatives were evaluated to find potential applications for use in wastewater treatments for adsorption and desorption of heavy metal ions as well as acidic and basic dyes.

  • Aluminum hydrate gels were prepared from the mixtures of bauxite and ammonium sulfate by wet acid process. Optimum conditions for obtaining the maximum yield( 99%) of aluminum hydrates from the same amount of bauxite were confirmed as follows ; 1. Mixing ratio ; addition of 25mole% of ammonium sulfate to 1mole of bauxite. 2. Calcination ; heated at 350℃ for 1hr. 3. Extraction ; leached at 95℃ in 1% H2SO4 for 90min. 4. pH of precipitating solution; slight below 7.0. Amorphous aluminum hydrates were precipitated at the pH lower than 8.5, but the precipitates crystallized to bayerite at the pH was 10. Mean diameter of α-Al2O3 powders which were obtained by calcining the aluminum hydrates was below 0.2㎛, and EDS analysis revealed than SiO2 was it's primary impurity.

  • For most engineering materials are influenced by the dominant mechanism resisting crack extention under large scale yielding conditions. Continuum mechanics analysis shows that fracture toughness, in addition to depending on young's modulus, flow stress strain hardening exponent, and yield strain, should be nearly proportoinal to the effective fracture ductility obtained for the stress state characteristic for region ahead of the crack; plane stress or plane strain. It's known that, in most ductile materials, crack propagation of the material strongly governed by the $J_{IC}$ value, which is still difficult to determine for it's complicate and treble-some determinative process. This paper, on the assumption that, initiation of crack tip strain field reaches on the relationships between the critical value of J-integral ($J_{IC}$) and the local fracture strain(${\varepsilon}_c$) in uniaxial tensile test in the region of maximun reduction areas was described.

  • This paper is to be researched into ozone generation character of Bi-Ti-Si type high dielectric yield ceramic catalyst discharge tube. And conditions of basic experiment are the outside diameter of discharge tube : 52 mm, the length of discharge tube : 350 mm, the frequence : 900 Hz, the temperature of cooling water : 25 $^{\circ}C$, quantity of flow : 5, 10, 20 l/min, pressure : 1.2, 1.4, 1.6 atm, and distance of discharge gap : 0.4, 0.6, 0.8 mm. Ozone generation characteristics were measured to consumption power. At quantity of flow : 20 l/min, discharge gap : 0.6 mm, pressure : 1.6, and consumption power : 150 W, Maximum ozone generation efficiency of 175 g/kWh was obtained. Maximum ozone generation efficiency was measured below the flow quantity of 20 l/min at below pressure of 1.6 atm. However, Maximum ozone generation efficiency was measured over the flow quantity of 20 l/min at over pressure of 1.6 atm.

  • For developing digital printing textiles, special pretreatment processes are necessary. These processes include developing formulation of coating agent and coating processes. The pretreatment were investigated with the variation concentration of anti-migration agent, fixation chemical etc.. The printing qualities according to pretreatment conditions were studied with color yield, printed capital letter sharpness and washing fastness. It was observed that the concentration of anti-migration agent, fixation agent(alkali) was closely related to printing qualities. For developing industrial technology of cellulosic digital printing textiles, optimum viscosity of pretreatment coating formulation is very important factor.

  • The environmental change is affecting the farm products like tomato, and pepper, etc. This affects to lead smart farming yield. What is more, this inconstant conditions cause the farms to be infected by variety diseases. Therefore ICT technology is needed to detect and prevent the crops from being effected by diseases. This article suggests the procedure to help producer for identifying farms disease based on the detected image. This detects the kind of diseases with comparing the trained image data before and after disease emergence. First step monitors an image of farms and resize it. Its features are extracted on parameters such as color, and morphology, etc. The next steps are used for classification to classify the image as infected or non-infected. on the bassis of detection algorithm.

Performance Analysis of R-1270(Propylene) Refrigeration System Using Internal Heat Exchanger (내부 열교환기를 이용한 R-1270용 냉동시스템의 성능 분석)

  • This paper considers the influence of internal heat exchangers on the efficiency of a refrigerating system using R-1270. These internal heat exchangers(liquid-gas or suction-line heat exchangers) can, in some cases, yield improved system performance while in other cases they degrade system performance. A steady state mathematical model is used to analysis the performance characteristics of refrigeration system with internal heat exchanger. The influence of operating conditions, such as the mass flowrate of R-1270, inner diameter tube and length of internal heat exchanger, to optimal dimensions of the heat exchanger is also analyzed. The main results were summarized as follows : the mass flowrate of R-1270, inner diameter tube and length of internal heat exchanger, and effectiveness have the influences on the cooling capacity, compressor work and RCI(Relative Capacity Index) of this system. With a thorough grasp of it is possible to design the R-1270 compression refrigeration cycle using internal heat exchanger.
