Analysis of Growth Characteristics and Yield Components According to Rice Varieties Between on Irrigated and Partially Irrigated Rice Paddy Field (수리불안전답에서의 벼 품종별 생육 및 수량구성요소 특성 변이 분석)

  • Drought caused by global climate change is one of serious problems for rice cultivation. However, it was little reported the impact of drought on rice cultivation in Korea. In here, to assess impact of drought on rice varieties in Korean climate condition, growth characteristics and yield components of rice were compared on irrigated and partially irrigated rice paddy field. First, we have chosen 11 rice varieties including 'Saeilmi' and 'Shindongjin' which are widely cultivated in Korea. For partially irrigated rice paddy treatment, we have withheld irrigation from 25 days after transplanting and water supply was totally dependent on rainfall for rice cultivation. When we examined early plant height and tiller number of these varieties on partially irrigated rice paddy were reduced 1.6% to 18.4% and 10.4% to 33.1%, respectively, and these reduction rate were highly correlated with yield loss in our experimental conditions. Among rice yield components, panicle number was decreased 10.5% to 30.1% according to rice varieties and reduced panicle number was highly correlated with yield loss. Grain number per panicle, grain filling rate and 1,000 seeds weight did not have correlation with yield loss of rice varieties. These result means that growth stage, especially the tillering stage, is seriously affected by drought on rice cultivation in Korea. And we suggest that 'Saeilmi', 'Ilmi' and 'Ilpum' are good for rice cultivation on drought prone rice field in Korea.

Improvement of Radiosynthesis Yield of [11C]acetate ([11C]아세트산의 방사화학적 수율 증가를 위한 연구)

  • Purpose $[^{11}C]$acetate has been proved useful in detecting the myocardial oxygen metabolism and various malignancies including prostate cancer, hepatocellular carcinoma, renal cell carcinoma and brain tumors. The purpose of study was to improve the radiosynthesis yield of $[^{11}C]$acetate on a automated radiosynthesis module. Materials and Methods $[^{11}C]$acetate was prepared by carboxylation of grignard reagent, methylmagnesium chloride, with $[^{11}C]$$CO_2$ gas, followed by hydrolysis with 1 mM acetic acid and purification using solid phase extraction cartridges. The effect of the reaction temperature ($0^{\circ}C$, $10^{\circ}C$, $-55^{\circ}C$) and cyclotron beam time (10 min, 15 min, 20 min, 25 min) on the radiosynthesis yield were investigated in the $[^{11}C]$acetate labeling reaction. Results The maximum radiosynthesis yield was obtained at $-10^{\circ}C$ of reaction temperature. The radioactivities of $[^{11}C]$acetate acquired at $-10^{\circ}C$ reaction temperature was 2.4 times higher than those of $[^{11}C]$acetate acquired at $-55^{\circ}C$. Radiosynthesis yield of $[^{11}C]$acetate increased with increasing cyclotron beam time. Conclusion This study shows that radiosynthesis yield of $[^{11}C]$acetate highly dependent on reaction temperature. The best radiosynthesis yield was obtained in reaction of grignard reagent with $[^{11}C]$$CO_2$ at $-10^{\circ}C$. This radiolabeling conditions will be ideal for routine clinical application.

Optimization of Submerged (Ginseng Root Culture Conditions for the Production of Saponin (사포닌 생산을 위한 인삼 root 액체배양조건의 최적화)

  • This study was carried out to determine the optimal liquid medium composition of ginseng (Panax ginsengC.A. Meyer) root induced by growth regulators in order to improve the yield of saponin production. Submerged culture conditions were optimized using the fractional factorial design with 4 factors and 3 levels by a RSM computer program The effects of various pH values of medium, sucrose, nitrogen and phosphate concentration on the saponin content of the ginseng root were investigated. The optimum phosphate concentration determined by a partial differentiation of the model equation, pH of medium, sucrose and nitrogen concentration were phosphate 93 mg/L, pH 5.5, sucrose 5% and nitrogen 50 mg/L, respectively. Under these conditions, the predicted saponin content of ginseng root was estimated at 0.308%.

The use of JIP test to evaluate drought-tolerance of transgenic rice overexpressing OsNAC10

  • In this study, the JIP test was exploited to assess drought-tolerance of transgenic rice overexpressing OsNAC10. Two types of promoters, RCc3 (root-specific) and GOS2 (constitutive), were used to drive the transcription factor OsNAC10, a gene involved in diverse functions including stress responses. Three-month-old plants were exposed to drought for 1 week and their fluorescence kinetics was evaluated. Our results showed that drought-treated non-transgenic plants (NT) have higher fluorescence intensity at the J phase (2 ms) compared to transgenic plants, indicating a decline in electron transport beyond the reduced plastoquinone ($Q_A^-$). As manifested by negative L bands, transgenic plants also showed higher energetic connectivity and stability over NT plants under drought conditions. Also, the pool size of the end electron acceptor at the photosystem I was reduced more in NT than in transgenic plants under drought conditions. Furthermore, the transgenic plants had higher $PI_{total}$, a combined parameter that reflects all the driving forces considered in JIP test, than NT plants under drought conditions. In particular, the $PI_{total}$ of the RCc3:OsNAC10 plants was higher than that of NT plants, which was in good agreement with their differences in grain yield. Thus, the JIP test proved to be practical for evaluating drought-tolerance of transgenic plants.

  • Since the middle of the 70s, the countries with the marine resources declared the exclusive water zone of two hundred miles. As a result, our country lacked many foreign fishing grounds. This awakened our perception of fishing grounds. The fishing ground is an important factor in the economical effectiveness of fisheries. The fishing ground must have the following basic conditions. 1. It must be the place the fishery resources inhabit and wander. 2. Their catching or aquaculturing must be possible by the technical methods. 3. It must have economic worth in the management. The fishing ground that achieves the above basic conditions is affected by the following factors. This is the factor of the fishing ground location. A. Natural factor 1). Suitable climate 2).Abundant resources 3). Good quality of the sea-bottom 4). Not to be pollutted area B, Economic factor 1). Be adjacent to fishing port and fisheries market 2). Good working conditions 3). Be abundant of economical resources C. Social factor 1). Be possible to work legally 2). Be permitted in the political relations 3). Be acquainted with the customs of foreign countries The fishing ground location is mainly affected by the natural factor of the above factors. The abundance of resources depends on the natural resources. The resources in the fishing ground have the cycle of developing period, maturing period, and decaying period according to the fishing efforts. The point that we sustainably yield the maximum of fishery resources is the changing point of maturing period and decaying period. We call this point MSY, so the resources of fishing ground should be managed by the MSY. Therefore, when we select the fishing ground location. We should judge the natural factor, the economic factor and the social factor synthetically according to the situation and environment, so we can achieve the efficient management.

Dissolution Conditions of Solid Radioactive Wastes Generated from NPP for the Analysis of Radionuclides Using a Closed-vessel Microwave Acid Digestion System (원전 발생 고체 방사성폐기물 내 핵종 분석을 위한 극초단파 산분해 장치를 이용한 용액화 조건)

  • The optimal conditions are obtained for the decomposition of solid radioactive wastes, including ion exchange resin, zeolite, active charcoal, and sludge from nuclear power plant. In the process of decomposing the radioactive wastes were used the microwave acid digestion method with mixed acid. The solution after acid digestion by the following method was colorless and transparent. Each solution was analyzed with ICP-AES and AAS and the recovery yield for 5 different elements added the simulated radioactive wastes were over 94%. As an effective pre-treatment, the proposed microwave acid digestion conditions concerning the chemical trait of each radioactive waste are expected to be generally applied to above-mentioned radioactive wastes from nuclear power plant hereafter.

  • Polyethylene terephthalate (PET) has been widely applied in polymers and packaging industries to produce synthetic fibers, films, drink bottles or food containers. Therefore, it has become one of the major plastic wastes. In this article, glycolysis known as one of the main methods in PET chemical recycling was investigated using a glycol to break down the polymer into a monomer. Glycolysis of PET and ethylene glycol was performed in a micro-tubing reactor under various conditions. The effect of glycolysis conditions on the product distribution was investigated at experimental conditions of the EG/PET ratio of 1~4, the reaction time of 15~90 min and the reaction temperature of $250{\sim}325^{\circ}C$ with Mn and Cu catalysts. The highest yield of bis (2-hydroxyethyl) terephthalate monomer (BHET) was obtained as 89.46 wt% under the condition of the reaction temperature of $300^{\circ}C$ and the time of 30 min using 10 wt% $Cu/{\gamma}-Al_2O_3$ catalyst, with the PET and ethylene glycol ratio of 1 : 2.

The Incidence of Muscle Abnormalities in Broiler Breast Meat - A Review

  • The dramatic improvements in the growth rate and breast muscle size and yield in broilers through the intensive genetic selection, and the improvement in nutrition and management over the past 50 years have introduced serious abnormalities that influenced the quality of breast meat. The abnormalities include pale-soft-exudative (PSE) conditions, deep pectoral muscle (DPM) myopathy, spaghetti meat (SM), white striping (WS), and woody breast (WB) that have serious negative implications to the broiler meat industry. The incidences of PSE and DPM have been known for several decades, and their prevalence, etiology and economic impact have been well discussed. However, other abnormalities such as SM, WS and WB conditions have been reported just for few years although these conditions have been known for some time. The newly emerging quality issues in broilers are mainly associated with the Pectoralis major muscles, and the incidences have been increased dramatically in some regions of the world in recent years. As high as 90% of the broilers are affected by the abnormalities, which are expected to cause from $200 million to $1 billion economic losses to the U.S. poultry industry per year. So, this review mainly discusses the histopathological characteristics and biochemical changes in the breast muscles with the emphasis on the newly emerging abnormalities (SM, WS, and WB) although other abnormalities are also discussed. The impacts of the anomalies on the nutritional, functional, mechanical and sensory quality of the meat and their implications to the poultry industry are discussed.

Study of the Productivity of Aflatoxin through the Interaction of Bacillus subtilis & Aspergillus(I) (Bacillus subtilis와 Apergillus flavus의 상호작용에 의한 aflatoxin 생성능에 관한 연구 1)

  • In order to investigate the production of aflatoxin in various conditions such as pH, moisture and temperature, 27 smaples were inoculated with Aspergillus flavus, and in addition 3 smaples were inoculated with the mixture of Aspergillus flavus and Bacillus subtilis and cultured under the conditions such as $20^{\circ}C$ and 30% moisture contents. The following results were obtained : 1) Aflatoxin production was the highest at pH 5.0 and relatively high at pH 7.0. Its production was decreased significantly when pH reached 9.0. 2) The yield of aflatoxin was shown comparatively high level at 30% moisture contens. The higher moisture contents was, the lower aflatoxin production was. 3) The highest level of aflatoxin production was at $20^{\circ}C$, and comparatively high level was at $30^{\circ}C$. However, its production was fairly low at $40^{\circ}C$. 4) The highest crude aflatoxin production was 5,093ppm $(B_1,\;1.912\;ppm;\;B_2,\;0.521\;ppm;\;G_1,\;2.119\;ppm;\;G_2,\;0.541\;ppm)$ at 30% moisture, pH 5.0 and $20^{\circ}C$ and the lowest one 2.197 ppm $(B_1,\;0.793\;ppm;\;B_2,\;0.185\;ppm;\;G_1,\;0.102\;ppm;\;G_2,\;0.381\;ppm)$ at 63% moisture, pH 9.0 and $40^{\circ}C$ 5) When Aspergillus flavus and Bacillus subilis were cultured together under the conditions such as $20^{\circ}C$ and 30% moisture, aflatoxin production was decreased by 27% comparing with the culture of Aspergillus flavus alone.

Influence of roasting conditions on the flavor quality of sesame seed oil (참깨 볶음조건이 참기름의 향미에 미치는 영향)

  • Sesame seeds were roasted for 30, 60, 90, and 120 min at different temperatures (100, 200, and $300^{\circ}C$) and extracted to investigate an adequate condition for producing the high quality sesame oil. Sesame seeds roasted at $200^{\circ}C$ for 90 min gave the high yield of oil. The oil contained the low content of brownish-black precipitates and exhibited an excellent organoleptic quality when judged by descriptive sensory analysis. Thirty one volatile flavor compounds, which are the largest number of volatiles among the oil samples prepared, were identified from the oil sample. The oil contained relatively high concentrations of furfurals (sweet candy-like flavor) and pyrazines (roasted-like flavor), that are considered as good contributors to sesame seed oil flavor, and low concentations of aldehydes $(C5{\sim}C10)$ and ketones, which are considerd as bad contributors (oxidized fat-like and painty-like flavors). These results suggest that the roasting condition of $200^{\circ}C$ for 90 min was the best for the oil production in terms of the overall aroma and taste quality under the test conditions (Received July 13, 1993; accepted November 4, 1993).
