2-Methoxy-1,4-naphthoquinone (MNQ) regulates cancer key genes of MAPK, PI3K, and NF-κB pathways in Raji cells

  • 2-Methoxy-1,4-naphthoquinone (MNQ) has been shown to cause cytotoxic towards various cancer cell lines. This study is designed to investigate the regulatory effect of MNQ on the key cancer genes in mitogen-activated protein kinase, phosphoinositide 3-kinase, and nuclear factor κB signaling pathways. The expression levels of the genes were compared at different time point using polymerase chain reaction arrays and Ingenuity Pathway Analysis was performed to identify gene networks that are most significant to key cancer genes. A total of 43 differentially expressed genes were identified with 21 up-regulated and 22 down-regulated genes. Up-regulated genes were involved in apoptosis, cell cycle and act as tumor suppressor while down-regulated genes were involved in anti-apoptosis, angiogenesis, cell cycle and act as transcription factor as well as proto-oncogenes. MNQ exhibited multiple regulatory effects on the cancer key genes that targeting at cell proliferation, cell differentiation, cell transformation, apoptosis, reduce inflammatory responses, inhibits angiogenesis and metastasis.

Effects of Phosphate and Two-Stage Sous-Vide Cooking on Textural Properties of the Beef Semitendinosus

  • Comparing the effects of sodium tripolyphosphate (STPP) concentrations of 0.2% and 0.4% on beef semitendinosus is the objective of the current investigation. The samples were cooked at varied temperatures (45+60℃ and 45+70℃) and times (1.5+1.5 h and 3+3 h) using staged cooking. The colour properties, cooking loss, water retention, shear force, water-holding capacity, sarcoplasmic, and myofibrillar solubility, and total collagen were investigated. The cooking time and temperature affected the water-holding capacity, cooking loss, CIE L*, CIE a*, CIE b*, myofibrillar, and sarcoplasmic solubility, with lower temperature and short time having the lower detrimental effect. However, the significant effect can be intensified after the addition of STPP with higher water-holding capacity and tender meat obtained with 0.4% phosphate concentration at any cooking conditions. The STPP lowered the collagen content and increased the protein solubility of myofibrillar and sarcoplasmic, which this degradation is used as a good indicator of tenderness.

Preparation and Evaluation of Paclitaxel Solid Dispersion by Supercritical Antisolvent Process (초임계유체를 이용한 파클리탁셀고체분산체의 제조 및 평가)

  • Paclitaxel is a taxane diterpene amide, which was first extracted from the stem bark of the western yew, Taxus brevifolia. This natural product has proven to be useful in the treatment of a variety of human neoplastic disorders, including ovarian cancer, breast and lung cancer. Paclitaxel is a highly hydrophobic drug that is poorly soluble in water. It is mainly given by intravenous administration. Therefore, The pharmaceutical formulation of paclitaxel ($Taxol^{(R)}$; Bristol-Myers Squibb) contains 50% $Cremophor^{(R)}$ EL and 50% dehydrated ethanol. However the ethanol/Cremophor EL vehicle required to solubilize paclitaxel in $Taxol^{(R)}$ has a pharmacological and pharmaceutical problems. To overcome these problems, new formulations for paclitaxel that do not require solubilization by $Cremophor^{(R)}$ EL are currently being developed. Therefore this study utilized a supercritical fluid antisolvent (SAS) process for cremophor-free formulation. To select hydrophilic polymers that require solubilization for paclitaxel, we evaluated polymers and the ratio of paclitaxel/polymers. HP-${\beta}$-CD was used as a hydrophilic polymer in the preparation of the paclitaxel solid dispersion. Although solubility of paclitaxel by polymers was increased, physical stability of solution after paclitaxel/polymer powder soluble in saline was unstable. To overcome this problem, we investigated the use of surfactants. At 1/20/40 of paclitaxel/hydrophilic polymer/ surfactant weight ratio, about 10 mg/mL of paclitaxel can be solubilized in this system. Compared with the solubility of paclitaxel in water ($1\;{\mu}g/mL$), the paclitaxel solid dispersion prepared by SAS process increased the solubility of paclitaxel by near 10,000 folds. The physicochemical properties was also evaluated. The particle size distribution, melting point and amophorization and shape of the powder particles were fully characterized by particle size distribution analyzer, DSC, SEM and XRD. In summary, through the SAS process, uniform nano-scale paclitaxel solid dispersion powders were obtained with excellent results compared with $Taxol^{(R)}$ for the physicochemical properties, solubility and pharmacokinetic behavior.

Risk of Treatment Related Death and Febrile Neutropaenia with Taxane-Based Adjuvant Chemotherapy for Breast Cancer in a Middle Income Country Outside a Clinical Trial Setting

  • Background: The risk of treatment-related death (TRD) and febrile neutropaenia (FN) with adjuvant taxane-based chemotherapy for early breast cancer is unknown in Malaysia despite its widespread usage in recent years. This study aims to determine these rates in patients treated in University Malaya Medical Centre (UMMC). Patients and Methods: Patients who were treated with adjuvant taxane-based chemotherapy for early breast cancer stages I, II or III from 2007-2011 in UMMC were identified from our UMMC Breast Cancer Registry. The TRD and FN rates were then determined retrospectively from medical records. TRD was defined as death occurring during or within 30 days of completing chemotherapy as a consequence of the chemotherapy treatment. FN was defined as an oral temperature > $38.5^{\circ}C$ or two consecutive readings of > $38.0^{\circ}C$ for 2 hours and an absolute neutrophil count < $0.5{\times}10^9/L$, or expected to fall below $0.5{\times}10^9/L$. Results: A total of 622 patients received adjuvant chemotherapy during this period. Of these patients 209 (33.6%) received taxane-based chemotherapy. 4 taxane-based regimens were used namely the FEC-D, TC, TAC and AC-PCX regimens. The commonest regimen employed was the FEC-D regimen accounting for 79.9% of the patients. The FN rate was 10% and there was no TRD. Conclusion: Adjuvant taxane-based chemotherapy in UMMC for early breast cancer has a FN rate of 10%. Primary prophylactic G-CSF should be considered for patients with any additional risk factor for FN.

Designing and Developing the Agricultural Information Management System of North Korea

  • In North Korea, there has been the considerable loss of human lives every yew due to the deficiency of foods. Thus, in order to reduce such damages, a research project should be launched to provide various information for cooperation with North Korean government, and to develop proper agricultural management system. Furthermore, based on the water resources information map generated by KOWACO (Korea Water Resources Corporation) and the environmental information system developed by MOE (Ministry of Environment), an agricultural information infrastructure of North Korea and a management system need to be effectively performed. Therefore, this research is mainly to develop the Agricultural Information Management System of North Korea (NKAIMS), which can collect, manage and analyze agricultural information and water resources utilization status of North Korea, and further support to make relevant decisions and establish the agricultural land-use plans. This research has three phases. The major outcome of the first phase is collecting the agricultural and water resources utilization data such as soils, rivers, streams, collective farms, etc., designing and building database, and developing integrated management system considering the users' requirements. The main work of the second phase is improving and reinforcing database such as adding the information of dams, land-over data, bridges, tunnels, satellite images, etc., inspecting and renewing such as importing detail attribute information of reservoirs, and improving system for more conveniently using. The third phase will be to supplement more useful functions such as statistic analysis, continually inspecting and improving database, and developing web-based system. The product of this research supports collecting and analyzing relevant data to facilitate easier agricultural activities and support effective decision making for food production in the preparation of unification. Moreover, through designing database considering sharing information and system expendability, it can support systematic data usability of agricultural information and save cost for data management.

  • A total of 45 scale insects belonging to 29 genera of five families were recognized on 15 kind of flowering plants on the basis of specimens collected in 2003 and specimens deposited in the Insect Collection of NIAST in Korea. Three species were recognized on Korean forsythia, four on chrysanthemum, 15 on common camellia, seven on kobus magnolia, three on rose of sharon, one on Indian lilac, six on rose, seven on rhododendrons, two on bamboo palm, three on benjamin tree, 12 on evergreen euonymus, five on Japanese yew, two on corn plant, six on orchids and two on cacti. Pulvinaria floccifera (Westwood) and Pseudaonidia paeoniae (Cockerell) on common camellia and Pulvinaria citricolar Kuwana on rose of sharon are reported for the first time from the Korean Peninsula.

This duplex plasma surface tri-atments of chromizing and plasma nitriding have induced a lormation of a duplex-lrcated surfacr hyer of approximat~ls 70-80 $\mu\textrm{m}$thickncss with a iargcly improved microiiardnrss up to approxiniateW 1500Hv(50gf). The main cause for the lage improvment in the surface hardncss is altribilted to [.he fact that CrN and $Fe_xN$ phases are created successfully by ccliromizins and plasma nilriding treatment. High tenipera1,urc wear resislance of the duplex-treated mild steel and HI3 steels at $600^{\circ}C$ was examined. Comparing the duplex-treated specimens with the specimens treated only by chromizing, the rcsults shovmi that, thc wear volume of the duplex-treated mild skcl and 1113 stcel aSt.er a wear test, at $600^{\circ}C$ were reduced hy a Iactor of 8 and 3, respectively. Characteristics of the CrS phase by duplrx treatment were compared with $CrN_x$,/TEX> film by ion plating and the wear behaviors of CrN film lormed by two different nroccsses arc nea.riy identical.

Purification of Paclitaxel and Its Derivatives by Supercritical Fluid Chromatography(SFC) (초임계 유체 크로마토그래피(SFC) 방법에 의한 Paclitaxel 및 그 유도체의 분리 정제)

  • Studies were carried out to elucidate the effects of pressure, temperature and mobile phase composition on supercritical $CO_2$ chromatographic separations of paclitaxel, baccatin III, 10-deacetyl baccatin III, 7-epi-10-deacetyltaxol, cephalomanine, and 10-deacetyltaxol. High resolutions of paclitaxel, 10-deacetylbaccatin III, 10-deacetyltaxol were observed with optimized pressure, temperature, and mobile phase composition. The highest resolution between paclitaxel and 10-deacetylbaccain III was observed at 275 kg/$\textrm{cm}^2$, $40^{\circ}C$ with the mobile phase composition of gradient mixture of 3.9-3.6 mL/min $CO_2$, 0.1-0.4 mL/min methanol for 20 min. Resolutions of baccatin III, capalomannine, and 7-epi-10-deacetyltaxol were found to be low in this study. On-line coupled SFE/SFC process was applied to isolate paclitaxel from yew tree powder. As a consequence, paclitaxel with a purity of 95% was obtained with a recovery yield of 38%.

Taxol Production in Taxus spp. Cell Culture : Semisynthesis of Taxol from Baccatin III in Cell Cultures of Taxus baccta Pendula (주목 세포배양에 의한 Taxol 생산 : 유럽 주목의 세포배양에서 추출한 Baccatin III로부터 Taxol의 반합성)

  • Anticancer agent taxol was synthesized from baccatin III which was extracted from cell cultures of European yew, Taxus baccata Pendula. Callus and suspension cultures of T. baccata Pendula showed the formation baccatin III. The content of baccatin III in cell cultures reached 0.015% (w/w) on dry weight basis. The semisynthetic approaches were made with baccatin III and taxol side chain. To prepare taxol side chain, (-)-N-((S)-2-hydroxy-1-phenylethyl) benzamide was synthesized first from (S)-(+)-phenylglycine. Then (-)-N-((IS,2S)-2-hydroxy-1-phenyl-3-butenyl) benzamide was synthesized with vinyl magnesium bromide. The synthesis of (2R, 3S)-(-)-2-(1-ethoxyethoxy)-3-phenyl-3-(phenylmethanamido) propanoic acid with RuCl3, catalyst was the final step to prepare taxol side chain. The semisynthetic reactions yielded 0.0002% taxol, 0.0005% 7-epi-10-deacetyltaxol, and unidentified taxanes on dry weight basis. It is suggested that the semisynthesis of taxol from baccatin III could be an alternative source of taxol and related taxanes.

Identification and Purification of New Brassinosteroid-Conjugates in Arabidopsis thaliana (애기장대 내 새로운 브라시노스테로이드 결합체의 분리 및 동정)

  • Metabolism of $^3H$-castasterone in the presence of $^{32}P$-ATP was examined by an enzyme solution prepared from A. thaliana after a reversed phased HPLC, after which a polar metabolite labeled by both $^3H$ and $^{32}P$ was obtained, suggesting that $^3H$-CS is phosphorylated by $^{32}P$-ATP. To elucidate the structure of the phosphorylated CS, the same enzyme assay was carried out with non-isotopes labeled CS and ATP. In GC-MS analysis the metabolite gave a molecular ion at m/z 664 as a bismethanboronate, suggesting the metabolite is a CS phosphate. Treatment of wheat germ acid phosphatase that hydrolyzed phosphoester bond gave the same mass spectrum and GC retention time in GC-MS analyses, confirming that the metabolite is phosphorylated CS. This is the first example of phosphorylated conjugates of CS in plants.
